Pd-Catalyzed [2 + 2 + 2] Cyclization of Alkyne-cyclohexadienones and O-Akynyl Benzenesulfonamides for Construction of Fused Tricyclic Hydronaphthofurans.

A palladium-catalyzed [2 + 2 + 2] cyclization of 1,6-enynes with unsymmetrical alkynes has been successfully accomplished, resulting in the formation of a series of fused tricyclic hydronaphthofurans with high stereo- and regioselectivity in a single step. This reaction demonstrates 100% atomic economy and exhibits a broad substrate scope.

Alkyne Derivatives of SARS-CoV-2 Main Protease Inhibitors Including Nirmatrelvir Inhibit by Reacting Covalently with the Nucleophilic Cysteine.

Nirmatrelvir (PF-07321332) is a nitrile-bearing small-molecule inhibitor that, in combination with ritonavir, is used to treat infections by severe acute respiratory syndrome coronavirus-2 (SARS-CoV-2). Nirmatrelvir interrupts the viral life cycle by inhibiting the SARS-CoV-2 main protease (Mpro), which is essential for processing viral polyproteins into functional nonstructural proteins. We report studies which reveal that derivatives of nirmatrelvir and other Mpro inhibitors with a nonactivated terminal alkyne group positioned similarly to the electrophilic nitrile of nirmatrelvir can effici

Double Nitroxide Labeling by Copper-Catalyzed Azide-Alkyne Cycloadditions with Noncanonical Amino Acids for Electron Paramagnetic Resonance Spectroscopy.

Electron paramagnetic resonance spectroscopy in combination with site-directed spin labeling (SDSL) is an important tool to obtain long-range distance restraints for protein structural research. We here study a variety of azide- and alkyne-bearing noncanonical amino acids (ncAA) in terms of protein single- and double-incorporation efficiency via nonsense suppression, metabolic stability, yields of nitroxide labeling via copper-catalyzed [3 + 2] azide-alkyne cycloadditions (CuAAC), and spectroscopic properties in continuous-wave and double electron-electron resonance measurements. We identify p

Isoindoline-Based Nitroxides as Bioresistant Spin Labels for Protein Labeling through Cysteines and Alkyne-Bearing Noncanonical Amino Acids.

Electron paramagnetic resonance (EPR) spectroscopy in combination with site-directed spin labeling (SDSL) is a powerful tool in protein structural research. Nitroxides are highly suitable spin labeling reagents, but suffer from limited stability, particularly in the cellular environment. Herein we present the synthesis of a maleimide- and an azide-modified tetraethyl-shielded isoindoline-based nitroxide (M- and Az-TEIO) for labeling of cysteines or the noncanonical amino acid para-ethynyl-l-phenylalanine (pENF). We demonstrate the high stability of TEIO site-specifically attached to the protei

Functionalization of luminescent lanthanide-gallium metallacrowns using copper-catalyzed alkyne-azide cycloaddition and thiol-maleimide Michael addition.

The synthesis and characterization of {Ln[12-MCGaIIIN(eshi)-4]}2(iph)4 and {Ln[12-MCGaIIIN(shi)-4]}2(miph)4 metallacrowns (MCs), where shi3- is salicylhydroximate, eshi3- is 4-ethynylsalicylhydroximate, iph2- is isopthalate, and miph2- is 5-maleimidoisophthalate, is reported. The ethynyl functionality allows for coupling of MCs to azides using copper(I) catalyzed alkyne-azide cycloaddition (CuAAC), while the maleimido functionality allows for coupling of the MCs to thiol-bearing compounds. We demonstrate these coupling reactions using benzyl azide for the former and cysteamine for the latter,

Zinc-Catalyzed Alkyne Oxidation/C-H Functionalization: Highly Site-Selective Synthesis of Versatile Isoquinolones and β-Carbolines.

An efficient zinc(II)-catalyzed alkyne oxidation/C-H functionalization sequence was developed, thus leading to highly site-selective synthesis of a variety of isoquinolones and β-carbolines. Importantly, in contrast to the well-established gold-catalyzed intermolecular alkyne oxidation, over-oxidation can be completely suppressed in this system and the reaction most likely proceeds by a Friedel-Crafts-type pathway. Mechanistic studies and theoretical calculations are described.

A highly efficient access to spiroketals, mono-unsaturated spiroketals, and furans: Hg(II)-catalyzed cyclization of alkyne diols and triols.

Hg(II) salts are identified as highly efficient catalysts for the versatile construction of spiroketals from alkyne diols in aqueous conditions. Monounsaturated spiroketals and furans were accessed with equal ease when propargylic triols (or propargylic diols) were subjected to similar conditions. Even the semiprotected alkyne diols gave the corresponding spiroketals with the same ease in a cascade manner. The reactions are instant and high yielding at ambient temperatures. Regioselectivity issues are well addressed.

Simultaneous Construction of C-Se And C-S Bonds via the Visible-Light-Mediated Multicomponent Cascade Reaction of Diselenides, Alkynes, and SO2.

A visible-light mediated multicomponent cascade reaction of diselenides, alkynes, and sulfur dioxide was developed, in which multiple C-Se and C-S bonds were constructed, and unexpected β-sulfonylvinylselane compounds were generated with high selectivity for E configuration. β-Sulfonylvinylselane transformation into 1,4-oxathiine-4,4-dioxide and sulfonylethyne derivates was then investigated. A plausible mechanism involving a selenium radical-initiated cascade reaction and sulfur dioxide insertion was proposed.

Gold-catalyzed oxidation of terminal alkynes to glyoxals and their reactions with 2-phenylimidazo[1,2-a]pyridines: one-pot synthesis of 1,2-diones.

A novel one-pot protocol for the convenient and efficient synthesis of (2-phenylimidazo[1,2-a]pyridin-3-yl)alkane-1,2-diones (3) in good yields (32-88%) from 2-phenylimidazo[1,2-a]pyridines (1) and terminal alkynes (2) has been established with a wide range of substrate scope. A tandem reaction sequence containing gold-catalyzed double oxidations of terminal alkynes to generate glyoxals, nucleophilic addition of 2-phenylimidazo[1,2-a]pyridines to glyoxals to yield α-hydroxyl ketones, and oxygenation of the α-hydroxyl ketones to afford the final products 3 under air atmosphere is involved in th

Copper(I)-Catalyzed Formal [4 + 2] Cyclocondensation of ortho-Hydroxybenzyl Alcohol, Aromatic Terminal Alkynes, and Sulfonyl Azides: An Alternative Approach to 2-Sulfonyliminocoumarins.

In this paper, an alternative and efficient copper(I)-catalyzed synthesis of 2-sulfonyliminocoumarins is developed through a three-component reaction of ortho-hydroxybenzyl alcohol, alkynes, and p-toluenesulfonyl azide. The proposed route for access to the 2-iminocoumarin ring involves a [4 + 2] hetero-Diels-Alder reaction between ortho-quinone methide and ketenimine intermediates generated in situ.

Cobalt-catalyzed enantioselective C-H/N-H annulation of aryl sulfonamides with allenes or alkynes: facile access to C-N axially chiral sultams.

Herein we report a cobalt-catalyzed enantioselective C-H/N-H annulation of aryl sulfonamides with allenes and alkynes, using either chemical or electrochemical oxidation. By using O2 as the oxidant, the annulation with allenes proceeds efficiently with a low catalyst/ligand loading of 5 mol% and tolerates a wide range of allenes, including 2,3-butadienoate, allenylphosphonate, and phenylallene, resulting in C-N axially chiral sultams with high enantio-, regio-, and position selectivities. The annulation with alkynes also exhibits excellent enantiocontrol (up to >99% ee) with a variety of funct

Copper-Catalyzed Synthesis of 3-Silyl-1-silacyclopent-2-enes via Regio- and anti-Selective Addition of Silylboronates to Silicon-Containing Internal Alkynes.

A copper-catalyzed disilylative cyclization of silicon-containing internal alkynes with silylboronates has been developed for the synthesis of 3-silyl-1-silacyclopent-2-enes. The reaction proceeded regio- and anti-selectively under simple and mild conditions by employing a combination of nucleophilic silicon donors and electrophilic silicon acceptors. The reaction could also be extended to the synthesis of a 1-germacyclopent-2-ene and a silicon-centered spirocyclic compound by using appropriate alkyne substrates.

Enantioselective Ni/N-Heterocyclic Carbene-Catalyzed Redox-Economical Coupling of Aldehydes, Alkynes, and Enones for Rapid Construction of Acyclic All-Carbon Quaternary Stereocenters.

Acyclic quaternary carbon stereocenters exist widely in natural products and bioactive molecules, but their enantioselective construction remains a prominent challenge. In particular, multicomponent enantioselective couplings of simple precursors to acyclic all-carbon quaternary stereocenters are very rare. We describe herein an N-heterocyclic carbene (NHC)-Ni catalyzed redox-economical three-component reaction of aldehydes, alkynes, and enones that proceeds in a highly chemo-, regio-, and enantioselective manner. A wide variety of valuable acyclic α-quaternary chiral ketones were synthesized

Heterogeneous Carbon Nitrides Photocatalysis Multicomponent Hydrosulfonylation of Alkynes To Access β-Keto Sulfones with the Insertion of Sulfur Dioxide in Aerobic Aqueous Medium.

Although hydrosulfonylation of alkynes is an ideal process to generate β-keto sulfones, such an approach is rarely implemented. Here we reported a facile and efficient graphitic carbon nitride (p-g-C3N4) photocatalyzed hydrosulfonylation of alkynes with the insertion of sulfur dioxide in aerobic conditions. Controlled experiments and ESR results indicated both the superoxide radicals and valence band holes played an important role in the reaction. Further isotope experiments confirmed the oxygen atom of the products comes from H2O, while the O2 plays an important role in the reaction by quench

Mechanistic insights into the one-pot synthesis of propargylamines from terminal alkynes and amines in chlorinated solvents catalyzed by gold compounds and nanoparticles.

Propargylamines can be obtained from secondary amines and terminal alkynes in chlorinated solvents by a three- and two-component synthesis catalyzed by gold compounds and nanoparticles (Au-NP) under mild conditions. The use of dichloromethane allows for the activation of two C-Cl bonds and a clean transfer of the methylene fragment to the final product. The scope of the reaction as well as the influence of different gold(III) cycloaurated complexes and salts has been investigated. The involvement of gold nanoparticles generated in situ in the process is discussed and a plausible reaction mecha

Preparation of Customizable Ligand-Functionalized Chitin Biopolymer Adsorbents Utilizing the Azide-Alkyne "Click" Reaction for the Adsorption of Aqueous Cu(II).

Chitin is an abundant biopolymer, with 1-3 million tons potentially available annually from shellfish-processing waste, but remains underutilized because its insolubility in conventional solvents makes it difficult to process and functionalize. Herein, a three-step route to chitin adsorbents with interchangeable ligands is reported. Highly acetylated chitin was tosylated and converted to the C-6 azide, and amine- and hydroxyl-terminated ligands were attached to this common intermediate by the copper-catalyzed azide-alkyne cycloaddition (CuAAC) "click" reaction, yielding water-insoluble derivat

Inter- vs Intramolecular Aldehyde-Alkyne-Amine A3-Reaction in the Solution-Phase Synthesis of a Potent Macrocyclic Azapeptide CD36 Modulator.

Modern macrocyclic peptides feature unnatural elements that are used to improve the activity and pharmacokinetic properties. Chemistry beyond peptide synthesis must, however, be employed to introduce such elements. Notably, application of the copper-catalyzed aldehyde-alkyne-amine A3-reaction in solid-phase peptide synthesis (SPPS) enabled a diversity-oriented approach for macrocyclization and furnished the azapeptide cluster of differentiation-36 receptor (CD36) modulator 298 which exhibits potent anti-inflammatory and cardioprotective effects. Broadly Applicable Copper(I)-Catalyzed Alkyne Semihydrogenation and Hydrogenation of α,β-Unsaturated Amides Enabled by Bifunctional Iminopyridine Ligands.

A highly active bifunctional catalyst consisting of a copper(I)/N-heterocyclic carbene complex and a basic 2-iminopyridine subunit allows for copper hydride chemistry under low H2 pressure, achieving efficient catalysis reaching 1 bar (balloon pressure). The bifunctional catalyst tolerates a remarkable variety of functional groups in catalytic alkyne semihydrogenations. Furthermore, this catalyst design gives rise to a high reactivity that allows for the catalytic hydrogenation of α,β-unsaturated amides (a substrate class hitherto unreactive in copper hydride catalysis) at a low H2 pressure fo

Discovery and Identification of Scarce Terminal Alkyne-Containing Natural Products by Chemical Isotope Labeling-Assisted MALDI-TOF Mass Spectrometry.

Terminal alkyne-containing natural products (TANPs) are pharmacologically valuable but remain underexplored due to their low abundance, instability, and inefficient isolation methodologies. Herein, we employed a structure-guided screening and isolation strategy integrating chemical isotope labeling (CIL) and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry (MALDI-TOF MS) for TANPs discovery. Parallel plant extracts were labeled via copper(I)-catalyzed azide-alkyne cycloaddition (CuAAC) reaction using isotope labeling reagents ABI/d5-ABI, followed by MALDI-TOF MS ana
