An influence of molecular weight, deacetylation degree of chitosan xerogels on their antimicrobial activity and cytotoxicity. Comparison of chitosan materials obtained using lactic acid and CO2 saturation.

This paper presents a comparison of the antimicrobial activity and cytotoxicity against L929 cells of chitosan xerogels prepared by dissolving the polymer in a solution of lactic acid (LA) or carbonic acid (CO2) and then freeze-drying. There was no simple relationship between the antimicrobial activity and cytotoxicity of the samples obtained using both techniques (LA and CO2). Chitosan materials obtained by the LA method in a 1:1 dilution were characterized by the highest cytotoxicity against L929 cells (∼20%). For the same diluted samples prepared using the CO2 saturation method, the viabili

Design and optimization of mucoadhesive chitosan microspheres for oesophageal delivery of fluconazole.

BACKGROUND: Oesophageal candidiasis remains challenging to treat owing to rapid transit of conventional oral formulations through the oesophagus, resulting in inadequate local drug delivery and reduced therapeutic efficacy. To address these limitations, the present study aimed to develop fluconazole-loaded chitosan microspheres (FCZ-CMs)-based orodispersible tablets (ODTs) that rapidly disintegrate in the oral cavity while releasing mucoadhesive microspheres capable of adhering to oesophageal mucosa, enabling sustained localized drug delivery. METHODS: FCZ-CMs were developed by the emulsion c

New Insights into the Mechanisms of Bitterness and Astringency in Chitosan Oligosaccharide Complexes.

Chitosan oligosaccharides (COS) possess valuable bioactive properties but exhibit undesirable bitter and astringent tastes that severely restrict their applications in functional foods. This study comprehensively elucidates the formation mechanisms of these taste defects by systematically investigating COS fractions with varying molecular weights. It was found that COS astringency intensifies with increasing molecular weight, while bitterness increases with decreasing molecular weight. The astringency mechanism primarily involves COS binding to salivary proteins via electrostatic interactions,

Advantages of the Combined Use of Cyclodextrins and Chitosan in Drug Delivery: A Review.

Cyclodextrins and chitosan are biomaterials largely used as pharmaceutical excipients due to their biocompatibility, biodegradability, and low/absent toxicity, associated with a number of favorable properties. In particular, cyclodextrins complexation is mainly utilized to improve the physicochemical and biological properties of drugs, including solubility, stability, and bioavailability, and to reduce their irritating effect. Nevertheless, some disadvantages related to the fast removal of the complex from blood circulation after in vivo administration, and possible competition effects for int

Exploring the potentials of chitosan as a promising carrier for methotrexate for beating cancer.

Cancer has long been considered to be one of the drivers of global mortality. The most prevalent malignancies are those of the breast, lungs, colorectal, and prostate. There is a different set of interventions that is used for treating cancer; chemotherapeutics, radiotherapeutics, and surgical interventions. However, these classic medications leave the body in crisis as a consequence of their tremendous harm. Consequently, researchers would not rely on those classic choices. Hopefully, chitosan (CS) and methotrexate (MTX) can offer a state-of-the-art delivery system in beating cancer. As CS ca

An Assessment of Fabrication, Properties, and Medical Applications of Chitosan-Nanometal Coatings.

Chitosan (chit) is a specific polymer, an oligosaccharide, that demonstrates several important properties alone or in combination with other compounds or elements. These important properties include being biocompatible with the human body and simultaneously effective in destroying bacteria. Moreover, it is a smart polymer, as it can change its properties when the pH value decreases below about 7. Therefore, chitosan is widely applied in dermo-cosmetics, but it is also intensively investigated for its use in food preservation and the targeted regeneration of teeth in dentistry. Due to these adv

A thermosensitive chitosan hydrogel: An attempt for the nasal delivery of dimethyl fumarate.

Dimethyl fumarate (DMF) is a drug that is orally administered for the treatment of relapsing-remitting multiple sclerosis. However, DMF causes gastrointestinal side effects and flushing in 43 % of patients, which significantly contributes to treatment discontinuation. To reduce side effects and increase patient compliance, the aim of this study was to develop a thermosensitive chitosan/glycerophosphate hydrogel for the nasal administration of DMF. A binary system of DMF with hydroxypropyl-β-cyclodextrin (HP-β-CD) was made and included in the hydrogel precursor solution. The precursor solution

The Role of Inhaled Chitosan-Based Nanoparticles in Lung Cancer Therapy.

Lung cancer is the leading cause of cancer-related mortality worldwide, largely due to the limited efficacy of anticancer drugs, which is primarily attributed to insufficient doses reaching the lungs. Additionally, patients undergoing treatment experience severe systemic adverse effects due to the distribution of anticancer drugs to non-targeted sites. In light of these challenges, there has been a growing interest in pulmonary administration of drugs for the treatment of lung cancer. This route allows drugs to be delivered directly to the lungs, resulting in high local concentrations that can
