Metabolomics analysis of Brassica rapa leaves and validation of its action as a natural remedy for pain, pyrexia and inflammation through modulating JAK/STAT3/MCP1 signaling pathway.

Brassica rapa L (Turnip), an edible plant, has been employed to remedy for various conditions like headaches, pyrexia, and inflammatory-related illness such as rheumatisms, and edema. Traditional usage offer anecdotal evidence, but comprehensive research is necessary to validate these claims scientifically. In order to provide a clear evidence of B. rapa's traditional application, this study aimed to evaluate the analgesic, anti-pyretic, and anti-inflammatory potentials of the methanolic extract of B. rapa leaves (BRMLE) along with comprehensive phytochemical characterization. The analgesic an

Nano-sulforaphane attenuates PhIP-induced early abnormal embryonic neuro-development.

BACKGROUND: 2-Amino-1-methyl-6-phenylimidazo[4,5-b]pyrimidine (PhIP), one of the most abundant heterocyclic aromatic amines (HAA) formed by cooking meat at high temperatures, may modify humans and rodents through the metabolic process prior to affecting nervous system development. In humans and rodents may be modified by metabolic processes and then affecting nervous system development. METHODS: In this paper, PhIP was used to prepare a chicken embryo model with abnormal embryonic nervous system defects. Sulforaphane (SFN) is a derivative of a glucosinolate, which is abundant in cruciferous v

Characterisation of a cyanide hydratase gene in the phytopathogenic fungus Leptosphaeria maculans.

A gene encoding a cyanide hydratase was cloned from an aggressive isolate of Leptosphaeria maculans, the fungus which causes blackleg disease of oilseed Brassica spp. This enzyme catalyses the breakdown of hydrogen cyanide to a less toxic compound, formamide. The predicted amino acid sequence of cyanide hydratase in L. maculans is 77% and 82% identical to cyanide hydratases from two other ascomycetes, Gloeocercospora sorghi and Fusarium lateritium, respectively. The gene is present as a single copy in the L. maculans genome, in both aggressive and non-aggressive isolates, although there is a r

Short-Term Effects of Broccoli-Derived Glucoraphanin on Recovery from Eccentric Muscle Damage: A Double-Blind Randomized Crossover Study.

Background/Objectives: Broccoli-derived glucoraphanin (a sulforaphane precursor that activates Nrf2 defenses) may aid repair; however, its short-term effects in humans remain unknown. This study aimed to evaluate whether short-term supplementation with broccoli-derived glucoraphanin improves recovery from exercise-induced muscle damage. We hypothesized that short-term supplementation with broccoli-derived glucoraphanin would attenuate exercise-induced muscle damage and accelerate recovery. Methods: In a randomized, double-blind, placebo-controlled crossover design, fifteen participants consume

Sulforaphane and related mustard oils in focus of cancer prevention and therapy.

The plant family Brassicaceae, formerly Cruciferae, contains mustard oil glycosides, from which mustard oils are enzymatically hydrolyzed. Mustard oils offer protection from pests, microorganisms and fungi. More than 120 different mustard oils with various biological functions are known. Since ancient times, these substances are used as natural antibiotics, antiviral drugs and antimycotics. The antioxidative effect of mustard oils contributes to protection from DNA damage. Epidemiological and experimental studies have shown preventive and therapeutic effects of crucifers or isolated substances

Drought-mitigation of Verticillium longisporum colonization in Brassica napus is associated with transcriptome and DNA methylation reprogramming.

Climate is becoming more variable, and the frequency and magnitude of drought and pathogen epidemics are changing. This creates a need to understand how limited water availability influences plant responses to biotic challenges. In Brassica napus L., drought decreases the disease progression and fungal colonization of the vascular pathogen Verticillium longisporum, yet the underlying regulatory mechanisms remain unclear. Here, we used an integrative approach combining transcriptome profiling, co-expression network analysis, and DNA methylation analysis to investigate host responses in hypocoty

Biochemical pathways linking adiposity, diet, and endometrial carcinogenesis.

Endometrial cancer (EC) arises from a convergence of metabolic, endocrine, and inflammatory disturbances largely driven by excess adiposity and diet-induced biochemical reprogramming. This review synthesizes molecular evidence linking adipose-derived estrogen excess, insulin/IGF-1 activation of PI3K/AKT/mTOR and RAS/MAPK pathways, adipokine imbalance, and NF-κB-dependent inflammation to EC initiation and progression. Mechanistic studies demonstrate that aromatase-mediated estradiol production in hypertrophic adipose tissue activates proliferative estrogen receptor-dependent transcriptional pro

Analysis of the Genes Involved in Thiocyanate Oxidation during Growth in Continuous Culture of the Haloalkaliphilic Sulfur-Oxidizing Bacterium Thioalkalivibrio thiocyanoxidans ARh 2T Using Transcriptomics.

Thiocyanate (N=C-S-) is a moderately toxic, inorganic sulfur compound. It occurs naturally as a by-product of the degradation of glucosinolate-containing plants and is produced industrially in a number of mining processes. Currently, two pathways for the primary degradation of thiocyanate in bacteria are recognized, the carbonyl sulfide pathway and the cyanate pathway, of which only the former has been fully characterized. Use of the cyanate pathway has been shown in only 10 strains of Thioalkalivibrio, a genus of obligately haloalkaliphilic sulfur-oxidizing Gammaproteobacteria found in soda l

Bioactivated Glucoraphanin Improves Cell Survival, Upregulating Phospho-AKT, and Modulates Genes Involved in DNA Repair in an In Vitro Alzheimer's Disease Model: A Network-Transcriptomic Analysis.

BACKGROUND/OBJECTIVES: Alzheimer's disease (AD) is one of the most common neurodegenerative diseases, for which a definitive cure is still missing. Recently, natural compounds have been investigated for their possible neuroprotective role, including the bioactivated product of glucoraphanin (GRA), the sulforaphane (SFN), which is highly rich in cruciferous vegetables. It is known that SFN alleviates neuronal dysfunction, apoptosis, and oxidative stress in the brain. In the light of this evidence, the aim of this study was to investigate the molecular effects of SFN pre-treatment in differentia

Role of Keap1-Nrf2 signaling in depression and dietary intake of glucoraphanin confers stress resilience in mice.

The transcription factor Keap1-Nrf2 system plays a key role in inflammation which is involved in depression. We found lower expression of Keap1 and Nrf2 proteins in the prefrontal cortex (PFC), CA3 and dentate gyrus (DG) of hippocampus in mice with depression-like phenotype compared to control mice. Serum levels of pro-inflammatory cytokines in Nrf2 knock-out (KO) mice were higher than those of wild-type mice, suggestive of enhanced inflammation in KO mice. Decreased brain-derived neurotrophic factor (BDNF) and its receptor tropomyosin-receptor-kinase B (TrkB) signaling in the PFC, CA3 and DG

Phytohormones: Heart of plants' signaling network under biotic, abiotic, and climate change stresses.

Industrialization has made the world increasingly unstable, subjecting plants to various constraints. As a consequence, plants are constantly experiencing biological, environmental, and climatic constraints, necessitating defense mechanisms to ensure their survival. Plants are vulnerable to various biotic factors, including insects, pathogens (bacterial, fungal, viral, and nematodes), weeds, and herbivores. They also face different abiotic and climate change challenges such as drought (regulated by genes like GH3, DREB, ZIFL1;3, etc), salinity, heavy metals, metalloids, ultraviolet radiations

Differential Costs of Two Distinct Resistance Mechanisms Induced by Different Herbivore Species in Arabidopsis.

Plants respond to herbivory with the induction of resistance, mediated by distinct phytohormonal signaling pathways and their interactions. Phloem feeders are known to induce plant resistance via the salicylic acid pathway, whereas biting-chewing herbivores induce plant resistance mainly via the jasmonate pathway. Here, we show that a specialist caterpillar (biting-chewing herbivore) and a specialist aphid (phloem feeder) differentially induce resistance against Pieris brassicae caterpillars in Arabidopsis (Arabidopsis thaliana) plants. Caterpillar feeding induces resistance through the jasmon

Evaluation of the standardized ileal digestibility of amino acids of rapeseed meals varying in protein solubility for Pekin ducks.

This study was conducted to determine whether protein solubility (PS) of rapeseed meals (RSM) can affect standardized ileal amino acid digestibility (SIDAA) in meat ducks. A total of 1,168, 14-days-old ducks were randomly allotted to 23 treatments (6 cages per diet, 8 ducks per cage) and 1 nitrogen-free diet treatment (8 cages, 8 ducks per cage) based on body weight. The 23 experimental diets consisted of a corn-soybean meal basal diet, and 22 diets containing 15% RSM: 85% basal diet. Titanium dioxide (0.5%) was included in all diets as an indigestible marker. On day 18, all ducks were euthani

Role of folate receptor genes in reproduction and related cancers.

Plants often respond to attack by insect herbivores and necrotrophic pathogens with induction of jasmonate-dependent resistance traits, but respond to attack by biotrophic pathogens with induction of salicylate-dependent resistance traits. To assess the degree to which the jasmonate- and salicylate-dependent pathways interact, we compared pathogenesis-related protein activity and bacterial performance in four mutant Arabidopsis thaliana lines relative to their wild-type backgrounds. We found that two salicylate-dependent pathway mutants (cep1, nim1-1) exhibited strong effects on the growth of

Fulvic acid-functionalized Fe3O4 nanoparticles enhance Chinese kale tolerance to saline-alkali stress via antioxidant and metabolic reprogramming.

BACKGROUND: In recent years, iron oxide nanoparticles (Fe3O4 NP) have emerged as a promising eco-friendly approach to mitigate various abiotic stresses. However, the role of surface-coated Fe3O4 NP with bio-derived organic compounds, particularly fulvic acid, in mitigating saline-alkali stress in Chinese kale plants has not yet been explored. In this study, we investigated for the first time the effects of green-synthesized Fe3O4 NP, both uncoated (UFe) and fulvic acid-coated (FFA), on mitigating saline-alkali stress in Chinese kale, with a specific focus on modulating physiological responses

Restoring circadian disrupted gut microbial metabolite rhythms with phytochemicals: a new avenue against metabolic disease.

The global epidemic of metabolic diseases-encompassing obesity, type 2 diabetes mellitus (T2DM), non-alcoholic fatty liver disease (NAFLD), and cardiovascular disease-represents a defining public health challenge of our era. The traditional model of simple caloric excess vs. expenditure has proven insufficient, giving way to a paradigm that acknowledges complex interactions between genetics, environment, and lifestyle, mediated by intricate physiological systems. Among these, the host circadian timing system and the gut microbiome have ascended as pivotal, deeply intertwined regulators of meta
