Maternal periconceptional exposure to cigarette smoking and alcohol consumption and congenital diaphragmatic hernia.

BACKGROUND: Congenital diaphragmatic hernia (CDH) is a major birth defect that occurs when abdominal organs herniate through a diaphragmatic opening into the thoracic cavity and is associated with high mortality (>50%). The etiology of CDH is not well understood. METHODS: Using data from the National Birth Defects Prevention Study, we examined associations between CDH and maternal periconceptional exposure (1 month before through the third month of pregnancy) to cigarette smoking and alcohol. Interview reports of exposures were provided by mothers of CDH (n = 503) and unaffected control (n =

Maternal peanut exposure during pregnancy and lactation reduces peanut allergy risk in offspring.

BACKGROUND: Maternal allergy is believed to be a risk factor for peanut allergy (PNA) in children. However, there is no direct evidence of maternal transmission of PNA susceptibility, and it is unknown whether maternal peanut exposure affects the development of PNA in offspring. OBJECTIVE: To investigate the influence of maternal PNA on offspring reactions to the first peanut exposure, and whether maternal low-dose peanut exposure during pregnancy and lactation influences these reactions and peanut sensitization in a murine model. METHODS: Five-week-old offspring of PNA C3H/HeJ mothers (PNA-

Maternal exposure to the antidepressant fluoxetine impairs sexual motivation in adult male mice.

Depressive disorders have a worldwide high prevalence. Fluoxetine (FLX), a selective serotonin reuptake inhibitor (SSRI) antidepressant, has been widely prescribed for depression during pregnancy and/or lactation. Since serotonin is a neurotrophic factor, the use of FLX by mothers could disrupt brain development resulting in behavioral alterations in their progeny. The aim of the present study was to evaluate the effects of developmental FLX exposure on sexual behavior, as well as on endocrine parameters, of male mice. Swiss dams were treated daily, by gavage, with 7.5 mg/kg of FLX during preg

Effects of cocaine/polydrug exposure and maternal psychological distress on infant birth outcomes.

To assess teratogenic effects of cocaine exposure and maternal psychological distress on birth outcomes, we conducted a longitudinal prospective study of 415 infants (218 cocaine-exposed--CE, 197 nonexposed--NE). Drug exposure was determined through a combination of maternal self-report, urine, and meconium screens. Maternal psychological distress postpartum was evaluated through a standardized, normative, self-report assessment. An extensive set of confounding variables was controlled, including severity of exposure to alcohol, tobacco, marijuana and other drugs, maternal age, race, parity, n

Epigenome-wide association analysis of prenatal organophosphate ester flame retardant exposure in maternal blood implicates maternal immune and neuronal epigenetic alterations.

BACKGROUND: Organophosphate esters (OPEs) are widely used flame retardants and plasticizers prevalent in indoor air, dust, and water. Studies suggest prenatal OPE exposure is linked to adverse maternal and infant outcomes, but the biological mechanisms remain unclear. METHODS: We examined associations between nine urinary OPE metabolites measured in the third trimester and DNA methylation in maternal blood from 175 pregnant women in the MADRES cohort. 837,856 CpGs were profiled using the Illumina EPIC array. We assessed the effect of each OPE (in tertile or detection status) and the joint eff

Maternal, fetal, and neonatal toxicity and potency estimates of perfluorohexane sulfonate (PFHxS) from oral maternal exposure in the Sprague-Dawley rat.

Perfluorohexane sulfonate (PFHxS) is a legacy, long-chain per- and polyfluoroalkyl substance (PFAS) frequently detected in human serum and environmental media across the globe. Previously published developmental studies in rats indicated a general lack of effects in common endpoints like maternal and offspring body and liver weights at doses up to 50 mg/kg/d. To facilitate mixture-based studies of PFAS co-exposure, individual dose response parameters are necessary for model predictions. To address these data needs we exposed timed-pregnant Sprague-Dawley rat dams to PFHxS (3-125 mg/kg/d) via o

A job-exposure matrix for potential endocrine-disrupting chemicals developed for a study into the association between maternal occupational exposure and hypospadias.

A study to assess the association between the prevalence of hypospadias and maternal occupational exposure to potential endocrine-disrupting chemicals was carried out using data from the congenital anomaly register of the Office for National Statistics. The occupation of the mother is recorded in this register and to facilitate the assessment of maternal occupational exposure, a specific job-exposure matrix for potential endocrine-disrupting chemicals was developed. Seven categories of contaminants were evaluated (pesticides, polychlorinated organic compounds, phthalates, alkylphenolic compoun

Effects on the postnatal development of the mouse of preconception, postconception and/or suckling exposure to manganese via maternal inhalation exposure to MnO2 dust.

Female mice were exposed either to MnO2 dust (7 hours/day, 5 days/week) or filtered air (control group) for 16 weeks prior to conception. On day 1 of gestation, half of each of these groups was assigned randomly either to MnO2 dust or filtered air exposure until day 17 of gestation. To separate effects of prenatal maternal exposure to MnO2 from postnatal exposure of offspring to Mn via suckling, a complete fostering/-cross-fostering design was employed. Compared to control mothers, mothers exposed to MnO2 prior to conception produced significantly larger litters. Prenatal exposure resulted in

Geographical Pattern of Testicular Cancer Points to Maternal Exposures Behind Increasing Incidence.

Evidence suggests that testicular cancer (TC) originates from germ cell neoplasia in situ arising during fetal development, implying that prenatal environmental exposures may influence disease risk. We investigated whether maternal place of residence during pregnancy is associated with regional variation in TC incidence. We conducted a nationwide cohort study including 1,342,996 men born in Finland between 1960 and 1999. Participants were followed from birth until diagnosis of TC, death, emigration, or the end of follow-up (2021). A total of 2976 TC cases were identified. Information on munici

Effects of Maternal Tetramethyl Bisphenol F Exposure on Neurodevelopment and Behavior in Mouse Offspring.

Bisphenol A (BPA) has long been used in plastics, resins, and food packaging materials; however, extensive research has demonstrated its reproductive, developmental, and endocrine-disrupting effects. Consequently, BPA has been increasingly restricted and replaced with structural analogues. Among these, tetramethyl bisphenol F (TMBPF) has emerged as one of the most widely used substitutes, particularly in epoxy resins and food-can coatings. Although initially regarded as a safer alternative, accumulating evidence suggests that TMBPF may exert multiple toxicological effects, raising concerns abo

Maternal exposure to ambient air pollution and risk of congenital limb defects in offspring.

Limited evidence on exposure to maternal air pollutants before and after conception and the risk of congenital limb defects (CLDs) in offspring is inconsistent. We explored the dose‒response relationship between maternal air pollutant exposure before and after conception and the risk of CLDs and their subtypes and further examined whether individual sociodemographic characteristics modify these associations. We carried out a population-based cohort study in Wuhan, China, based on the government registration system. The CLDs and daily air pollutant concentration data were collected. Individual

Impact of Maternal Exposure to SARS-CoV-2 on Immunological Components of Breast Milk.

COVID-19, caused by SARS-CoV-2, has become a global public health threat. Although no replication-competent virus has been found in breast milk samples, breastfeeding practices during the pandemic were impacted. It is well known that breast milk is adapted to meet the needs of infants, providing the appropriate amounts of nutrients and various bioactive compounds that contribute to the maturation of the immune system and antioxidant protection, safeguarding infants against diseases. While its composition is variable, breast milk contains immune cells, antibodies, and cytokines, which have anti

Activation of the arylhydrocarbon receptor through maternal beta-naphthoflavone exposure in the neonatal kidney.

The kidneys of neonates are vulnerable to stressors due to their immature structure and function. Excess activation of the transcription factor arylhydrocarbon receptor (AhR) in the kidneys of neonates can cause severe hydronephrosis, as shown previously using 2,3,7,8-tetrachlorodibenzo-p-dioxin, an AhR agonist. In this study, we aimed to clarify the conditions under which AhR activation leads to hydronephrosis using beta-naphthoflavone (BNF), another potent agonist of AhR. Mouse dams were fed a BNF-containing diet, and the kidneys of their pups were examined. Maternal BNF exposure on postnata

Maternal Phthalate Exposure and Allergic Diseases in Children: A Meta-Analysis and Network Toxicology.

Several studies suggest a relationship between phthalates (PAEs) and allergic diseases in children. Therefore, we speculated that PAE exposure may be an important environmental factor causing allergic diseases. The present study employed meta-analysis and network toxicology to analyze the interactions and assess potential pathogenic pathways between prenatal and postnatal PAE exposure and childhood allergic diseases. This study found that prenatal PAEs exposure was positively associated with childhood wheezing and eczema (OR = 1.03, 1.05), and postnatal PAEs exposure was positively associated

Exposure to maternal nicotine in utero and/or via lactation alters craniofacial development in mice.

The Center for Disease Control's National Birth Defects Prevention Study data suggests that maternal nicotine use may increase the incidence of craniofacial birth defects and growth anomalies like craniosynostosis, cleft palate, and/or lip in offspring. Craniofacial growth proceeds by expansion at fibrous sutures and synchondroses. In the cranial base, synchondroses, which are cartilaginous joints, play a major role in craniofacial development including neurocranial expansion and facial outgrowth. Our previous data showed alterations in craniofacial structures with intrauterine exposure to nic

Exploring the Influence of Maternal Behavioural Activation and Inhibition on Predicting Fetal Alcohol Exposure.

Background: Prenatal alcohol exposure (PAE) can cause adverse physical and neurocognitive fetal development. Numerous underlying mechanisms of PAE-risk have been hypothesized; however, these have predominantly focused on health-, interpersonal-, and social-related factors. Evidence suggests that stable personality traits, such as behavioral activation (BA) and behavioral inhibition (BI) may contribute toward risk of alcohol misuse. We posit that BA and BI may therefore increase risk of PAE. The potential of BA and BI to underpin both pre- and during-pregnancy alcohol use was thus explored in a

The Effects of Maternal Endocrinopathies and Exposure to Endocrine Disruptors During Pregnancy on the Fetus and Newborn.

Maternal health has a profound impact on fetal development, influencing the risk of pediatric endocrine disorders both directly and indirectly through various biological and environmental mechanisms. Throughout pregnancy, several endocrine disorders can arise or be exacerbated due to the physiological changes that occur. An in-depth review of articles with evidence-based research discussing the significant effects of maternal endocrinopathies and endocrine disruptors on fetal development and infant health was conducted in this review paper. The most common endocrine disorder during pregnancy i
