Molecular and biochemical approaches of the trypanothione system in Leishmania spp.: A key player in parasite resistance to antimonial therapy.

The trypanothione system is a crucial antioxidant defense mechanism in Leishmania spp. The enzymes involved, including trypanothione reductase (TR), trypanothione synthetase (TS), tryparedoxin (TXN) and tryparedoxin peroxidase (TXNPx) are essential for maintaining the redox balance. This system plays a fundamental role in the biology of Leishmania spp., contributing to parasite resistance against metalloid-based treatments, such as trivalent antimony (Sb3+). The mechanisms underlying this resistance, particularly those linked to the functionality of the trypanothione system, have garnered incr

Two cases of deliberate elemental arsenic ingestion with no apparent toxicologic consequences.

INTRODUCTION: Arsenic is a metalloid that is highly toxic in its inorganic and organic forms. Ingestion of elemental arsenic is rare. CASE REPORTS: A 33-year-old woman ingested elemental arsenic 10 g purchased online. She experienced brief gastrointestinal upset but was otherwise asymptomatic. She underwent chelation with succimer until it became apparent that she had no toxicologic consequences. She remained asymptomatic at 19-day follow-up. Her spot urine inorganic arsenic concentrations were >5,000 µg/L, 109 µg/L, and 87 µg/L at 12 days, 36 days, and 19 days post-ingestion, respectively. A

Impact of sediment mobilization on trace elements release in Galician Rías (NW Iberian Peninsula): insights into aquaculture.

In the latest years, the concentration levels of certain metals and metalloids in the sediments of the Galician Rías have shown an increasing trend (e.g., As, Zn, Cu, Pb, Hg). These areas are also characterized by their richness in nutrients and their great aquaculture or mariculture activity, with the presence of more than 3500 mussel rafts in the Rías Baixas. The inner areas of the Galician Rías are subjected to activities that resuspend the sediment such as high levels of maritime traffic and dredging or cleaning operations. It is likely that a transfer of these elements to the water column

Nanoparticles-mediated entomotoxicology: lessons from biologica.

Nanotechnology has grown in importance in medicine, manufacturing, and consumer products. Nanoparticles (NPs) are also widely used in the field of insect pest management, where they show a variety of toxicological effects on insects. As a result, the primary goal of this review is to compile and evaluate available information on effects of NPs on insects, by use of a timely, bibliometric analysis. We also discussed the manufacturing capacity of NPs from insect tissues and the toxic effects of NPs on insects. To do so, we searched the Web of Science database for literature from 1995 to 2023 and

Priority and emerging micropollutants distribution from coastal to continental slope sediments: A case study of Capbreton Submarine Canyon (North Atlantic Ocean).

Incising continental margins, submarine canyons are key issue for understanding shelf/deep sea exchange of particulate pollutant, impact on marine ecosystem and global geochemical cycling. The occurrence and distribution of 100 priority and emerging micropollutants were investigated in sediments within the first 25 km of the Capbreton submarine area. The most predominant compounds were polycyclic aromatic hydrocarbons (PAHs), trace metals and metalloid (TMs) (e.g. mercury, lead and arsenic), synthetical musks (e.g. musk ketone, galaxolide), UV filters (e.g. octocrylene and 2-ethylhexyl 4-metho

Cadmium Pathways in Snails Follow a Complementary Strategy between Metallothionein Detoxification and Auxiliary Inactivation by Phytochelatins.

Metal detoxification is crucial for animals to cope with environmental exposure. In snails, a pivotal role in protection against cadmium (Cd) is attributed to metallothioneins (MTs). Some gastropod species express, in a lineage-specific manner, Cd-selective MTs devoted exclusively to the binding and detoxification of this single metal, whereas other species of snails possess non-selective MTs, but still show a high tolerance against Cd. An explanation for this may be that invertebrates and in particular snails may also synthetize phytochelatins (PCs), originally known to be produced by plants,

Arsenic Toxicity in Male Reproduction and Development.

Arsenic is a toxic metalloid that exists ubiquitously in the environment, and affects global health problems due to its carcinogenicity. In most populations, the main source of arsenic exposure is the drinking water. In drinking water, chronic exposure to arsenic is associated with increased risks of various cancers including those of skin, lung, bladder, and liver, as well as numerous other non-cancer diseases including gastrointestinal and cardiovascular diseases, diabetes, and neurologic and cognitive problems. Recent emerging evidences suggest that arsenic exposure affects the reproductive

Differences in phosphorus translocation contributes to differential arsenic tolerance between plants of Borreria verticillata (Rubiaceae) from mine and non-mine sites.

We have identified new arsenic-tolerant plant species Borreria verticillata (Rubiaceae) that has mine and non-mine populations at a highly contaminated site (CS) and an uncontaminated site (UCS), respectively, in Brazil. Plants of B. verticillata from both sites were cultivated at different As and P concentrations. At low P concentration, CS plants showed reduced As uptake, higher P translocation to shoots, higher constitutive levels of phenolic compounds in roots, and higher tolerance to this metalloid. At the lowest P and highest As concentration, CS plants showed higher biomass. In addition

Chronic exposure to arsenic in drinking water can lead to resistance to antimonial drugs in a mouse model of visceral leishmaniasis.

The Indian subcontinent is the only region where arsenic contamination of drinking water coexists with widespread resistance to antimonial drugs that are used to treat the parasitic disease visceral leishmaniasis. We have previously proposed that selection for parasite resistance within visceral leishmaniasis patients who have been exposed to trivalent arsenic results in cross-resistance to the related metalloid antimony, present in the pentavalent state as a complex in drugs such as sodium stibogluconate (Pentostam) and meglumine antimonate (Glucantime). To test this hypothesis, Leishmania do

Induction of anoikis by sodium arsenite in rat hepatoma FGC4 cells: comparison with cadmium chloride and implications for assessment of regulation of heat shock protein 70.

CONTEXT: Arsenic, a toxic metalloid with major health concerns, elicits upregulation of heat shock protein 70 (HSP70) in rat hepatoma FGC4 cells, together with evidence of detachment of viable cells from the growth substratum. OBJECTIVE: To determine if this cell detachment was linked to anoikis, and the impact of this on measurement of HSP70 expression. MATERIALS AND METHODS: FGC4 cells were exposed to sodium arsenite, and detached and attached cells were taken for assessment of cell viability, activation of procaspase-3, and expression of HSP70. RESULTS: Exposure to sodium arsenite led to

Influence of humic acid on the uptake of aqueous metals by the killifish Fundulus heteroclitus.

The role of humic acids, over a concentration range of 0 to 20 mg L(-1) , was investigated in the uptake of three metals (Cd, Cr, and Hg-as both inorganic Hg [Hg(II)] and methylmercury [MeHg]) and a metalloid (As) from the aqueous phase by the killifish (Fundulus heteroclitus). Cadmium uptake showed no relationship with humic acid concentration, whereas Cr, Hg(II), and MeHg uptake showed an inverse relationship, and As uptake increased with increasing humic acid concentration. Concentration factors were >1 for Cd, Hg(II), and MeHg at all humic acid concentrations, indicating killifish were mor

Isolation and characterization of Arabidopsis halleri and Thlaspi caerulescens phytochelatin synthases.

The synthesis of phytochelatins (PC) represents a major metal and metalloid detoxification mechanism in various species. PC most likely play a role in the distribution and accumulation of Cd and possibly other metals. However, to date, no studies have investigated the phytochelatin synthase (PCS) genes and their expression in the Cd-hyperaccumulating species. We used functional screens in two yeast species to identify genes expressed by two Cd hyperaccumulators (Arabidopsis halleri and Thlaspi caerulescens) and involved in cellular Cd tolerance. As a result of these screens, PCS genes were ide

Sodium antimony gluconate induces generation of reactive oxygen species and nitric oxide via phosphoinositide 3-kinase and mitogen-activated protein kinase activation in Leishmania donovani-infected macrophages.

Pentavalent antimony complexes, such as sodium stibogluconate and sodium antimony gluconate (SAG), are still the first choice for chemotherapy against various forms of leishmaniasis, including visceral leishmaniasis, or kala-azar. Although the requirement of a somewhat functional immune system for the antileishmanial action of antimony was reported previously, the cellular and molecular mechanism of action of SAG was not clear. Herein, we show that SAG induces extracellular signal-regulated kinase 1 (ERK-1) and ERK-2 phosphorylation through phosphoinositide 3-kinase (PI3K), protein kinase C, a

Hepatoprotective activity of liposomal flavonoid against arsenite-induced liver fibrosis.

Arsenic, the environmental metalloid toxicant, is known to induce oxidative damage to liver and produce hepatic fibrosis. The theme of our study was to optimize and evaluate the therapeutic efficacy of galactosylated liposomal flavonoidal antioxidant, quercetin (QC), in combating arsenic-induced hepatic fibrogenesis. The rats of the hepatic damage group were injected s.c. a single dose of sodium arsenite (NaAsO(2)) (100.06 microM/kg b. wt. in 0.5 ml of physiological saline). Hepatocytes and stellate cells were separated. Mitochondrial membranes were isolated from all those separated cells. Oxi

The cancer metallome continuum: a framework for understanding metal dysregulation from carcinogenesis to therapeutic resistance.

Metals and metalloids play essential roles in cancer biology, influencing redox balance, epigenetic regulation, immune responses, and cell death throughout tumour development, progression, and resistance to therapy. Despite this broad biological relevance, the systematic clinical integration of metallomics into oncology remains in its early stages. This review introduces the Cancer Metallome Continuum, a framework illustrating how metallomic reprogramming evolves from cancer initiation through progression to therapy-resistant states, and evaluates the analytical, systems-level, and translation

An integrative analysis of elementomics and oxidation-reduction potentials identifies redox-related metals associated with coronary artery disease and post-PCI outcomes in type 2 diabetic patients.

Exposure to metals has been associated with elevated oxidative stress, which may contribute to diabetic macrovascular complications. However, current methods for evaluating oxidative stress, which mainly rely on quantifying individual byproducts of oxidative damage, may not fully characterize the effects of metal exposure on global redox imbalance, especially in the setting of type 2 diabetes mellitus (T2DM). Here, by assessing global redox status using the novel marker oxidation-reduction potential (ORP) and performing elemental profiling of 35 plasma metals/metalloids in a large cohort of 31

The association among metals and changes in their concentrations in the treatment of lead poisoning in lead-exposed workers.

BACKGROUND: High-throughput metallomics studies on lead-exposed workers are rare, and the effects of lead exposure on other metals, as well as the loss of trace metals during such treatments, remain unclear. METHODS: We recruited 1091 lead-exposed and 75 lead-poisoned workers undergoing calcium disodium ethylenediaminetetraacetate (CaNa2EDTA) treatment. Urine samples were analyzed for 33 metals/metalloids using inductively coupled plasma mass spectrometry (ICP-MS). In lead-poisoned workers, first morning urine was collected daily over a 15-day two-course of lead elimination treatment, plasma

Advances in immunology of male reproductive toxicity induced by common environmental pollutants.

Humans are exposed to an ever-increasing number of environmental toxicants, some of which have gradually been identified as major risk factors for male reproductive health, even associated with male infertility. Male infertility is usually due to the reproductive system damage, which may be influenced by the exposure to contaminants such as heavy metals, plasticizers, along with genetics and lifestyle. Testicular immune microenvironment (TIM) is important in maintaining normal physiological functions of the testis, whether disturbed TIM after exposure to environmental toxicants could induce re
