Characterization of microsatellite loci in Phormia regina towards expanding molecular applications in forensic entomology.

Forensic entomology involves the use of insects and arthropods to assist a spectrum of medico-criminal investigations that range from identifying cases of abuse, corpse movements, and most commonly, post mortem interval estimates. Many of these applications focus on the use of blowflies given their predicable life history characteristics in their larval stages. Molecular tools have become increasingly important in the unambiguous identification of larval blowfly species, however, these same tools have the potential to broaden the array of molecular applications in forensic entomology to includ

Synanthropic flies as vectors of Cryptosporidium and Giardia among livestock and wildlife in a multispecies agricultural complex.

The capacity of synanthropic flies belonging to the families Calliphoridae, Sarcophagidae, and Muscidae to serve as mechanical vectors of Cryptosporidium and Giardia among livestock and wildlife in a multispecies agricultural complex in northwest Georgia (USA) was studied using fluorescent in situ hybridization (FISH) and immunofluorescent antibody (IFA) techniques. Flies from all three families were carrying viable Cryptosporidium oocysts and Giardia cysts internally and externally, and deposited them on trap surfaces. Furthermore, the contaminated flies and trap surfaces occurred in associat

Orbital myiasis secondary to neglected medial canthal basal cell carcinoma caused by Chrysomyabezziana.

A case of orbital myiasis caused by Chrysomya bezziana arising from a long-standing, untreated basal cell carcinoma of the medial canthus in a 52-year-old woman is reported. The patient presented with periocular swelling, foul-smelling discharge, bleeding, and visible larvae emerging from the ulcerated lesion. Computed tomography showed orbital soft-tissue involvement with extension into the ipsilateral nasal cavity and ethmoid sinus, and focal bony erosion. Management included prompt mechanical removal of larvae, local wound care, systemic antibiotics, and adjunctive oral and topical ivermect

Insect Antimicrobial Peptide Complexes Prevent Resistance Development in Bacteria.

In recent decades much attention has been paid to antimicrobial peptides (AMPs) as natural antibiotics, which are presumably protected from resistance development in bacteria. However, experimental evolution studies have revealed prompt resistance increase in bacteria to any individual AMP tested. Here we demonstrate that naturally occurring compounds containing insect AMP complexes have clear advantage over individual peptide and small molecule antibiotics in respect of drug resistance development. As a model we have used the compounds isolated from bacteria challenged maggots of Calliphorida

Observations on myiasis by the calliphorid, Bufolucilia silvarum, in the eastern American toad (Bufo americanus americanus) from southeastern Wisconsin.

Larvae of certain species of blowflies (Calliphoridae) cause myiasis in amphibians which may result in significant mortality, yet there are few reports from North America. In this study, we observed primary myiasis in a population of juvenile eastern American toads (Bufo americanus americanus) collected during May-July 1998 from southeastern Wisconsin (USA). Nine (6%) of 140 toads were infected by the green blow fly (Bufolucilia silvarum) with a mean intensity of 10.5 +/- 7.2 (range = 1-24). Weekly parasite prevalence and mean intensity remained low, ranging from 0-20% and 2 +/- 1.4 to 14 +/-

Influence of Clothing on Decomposition and Presence of Insects on Rabbit Carcasses.

The decomposition rate and insects attraction to carcasses are affected by a number of factors such as temperature, body mass, the presence of poisons, humidity, and insect activity. The present study was planned to investigate the influence of clothing on both the decomposition process and the presence of insects on rabbit carcasses. The study was performed in May 2016 and involved the use of 18 live mature rabbits (Oryctolagus cuniculus L.), which were euthanized by chloroform. Rabbit carcasses were divided into three replicates with six rabbits in each replicate. Rabbits in each replicate w

Toxicity and oviposition deterrence of essential oils of Clinopodium nubigenum and Lavandula angustifolia against the myiasis-inducing blowfly Lucilia sericata.

Cutaneous myiasis is a severe worldwide medical and veterinary issue. In this trial the essential oil (EO) of the Andean medicinal plant species Clinopodium nubigenum (Kunth) Kuntze was evaluated for its bioactivity against the myiasis-inducing blowfly Lucilia sericata (Meigen) (Diptera Calliphoridae) and compared with that of the well-known medicinal plant species Lavandula angustifolia Mill. The EOs were analysed and tested in laboratory for their oviposition deterrence and toxicity against L. sericata adults. The physiology of EO toxicity was evaluated by enzymatic inhibition tests. The ant

Phylogenetic inference of calyptrates, with the first mitogenomes for Gasterophilinae (Diptera: Oestridae) and Paramacronychiinae (Diptera: Sarcophagidae).

The complete mitogenome of the horse stomach bot fly Gasterophilus pecorum (Fabricius) and a near-complete mitogenome of Wohlfahrt's wound myiasis fly Wohlfahrtia magnifica (Schiner) were sequenced. The mitogenomes contain the typical 37 mitogenes found in metazoans, organized in the same order and orientation as in other cyclorrhaphan Diptera. Phylogenetic analyses of mitogenomes from 38 calyptrate taxa with and without two non-calyptrate outgroups were performed using Bayesian Inference and Maximum Likelihood. Three sub-analyses were performed on the concatenated data: (1) not partitioned; (

Pre-adult mortality in the blowfly Lucilia sericata.

Populations of the blowfly, Lucilia sericata (Meigen) (Diptera: Calliphoridae), have a considerable potential for rapid increase; the lifetime reproductive output of each adult female has been estimated to be between 130 and 172 eggs. Nevertheless, in the field, absolute population densities of this species are relatively low. To account for this difference, the levels of mortality affecting the eggs, feeding and wandering larvae and pupae of L. sericata were assessed in the field and laboratory. Percentage egg hatch was dependent on relative humidity with no egg eclosion at humidities below 5

Genomic analyses of a livestock pest, the New World screwworm, find potential targets for genetic control programs.

The New World Screwworm fly, Cochliomyia hominivorax, is a major pest of livestock in South America and Caribbean. However, few genomic resources have been available for this species. A genome of 534 Mb was assembled from long read PacBio DNA sequencing of DNA from a highly inbred strain. Analysis of molecular evolution identified 40 genes that are likely under positive selection. Developmental RNA-seq analysis identified specific genes associated with each stage. We identify and analyze the expression of genes that are likely important for host-seeking behavior (chemosensory), development of

Temperature-dependent lipid metabolism in the blow fly Lucilia sericata.

An understanding of how arthropods use energy is fundamental to explaining their diverse life histories and adaptation to specific environments. It is also of importance when attempting to predict the impacts of environmental change on patterns of development and phenology. Here, lipid use by the economically important agent of ovine myiasis, Lucilia sericata (Diptera: Calliphoridae), was quantified at a range of temperatures. During pupation, at temperatures above the minimum temperature required for development (9 °C), pupae depleted an average of 30% of their total lipid over the course of

Current status of the New World screwworm Cochliomyia hominivorax in Venezuela.

The geographical distribution and seasonality of the New World screwworm (NWS), Cochliomyia hominivorax Coquerel (Diptera: Calliphoridae), were monitored through the use of sentinel animals as part of a co-ordinated programme involving veterinarians and farmers, as well as undergraduate students and teachers from veterinary colleges in Venezuela. This surveillance activity made it possible to collect NWS egg masses or larvae from all 23 states in the country and to determine that the rainy season has a strong positive influence on the number of cases of myiasis caused by C. hominivorax in dogs

Traumatic myiasis of geese in Hungary.

Five geese flocks were surveyed to gather data on the prevalence and clinical manifestation of traumatic myiasis and the fly species involved. Myiasis was recorded in all the flocks and the total number of infested geese was 26 (ca. 0.1% of the total numbers). The first cases were observed at the end of May, the last ones in August. Most birds (16/26) were infested in August. Each affected goose had only one lesion, which was located more frequently on the wings (14/26) than on any other body. In seven geese, Wohlfahrtia magnifica (Diptera: Sarcophagidae) was the only myiasis-causing species.

The New World screwworm fly in Libya: a review of its introduction and eradication.

The New World screwworm fly, Cochliomyia hominivorax (Coquerel) (Diptera: Calliphoridae), became established in the Old World for the first time during 1988, in the Socialist People's Libyan Arab Jamahiriya. An obligate parasite of mammals in its larval stages, causing wound myiasis, it is one of the most serious insect pests of livestock in the Americas. To avert a major disaster for the livestock industry and wildlife of Africa and southern Europe, the Libyan Government and the Food and Agriculture Organization of the United Nations launched a massive screwworm eradication programme, based o

Necrophagous fly occurrence and primary colonizing species in forensic entomology cases from the Yeongnam Region, South Korea.

Forensic entomology is widely used in death investigations to estimate the minimum postmortem interval (mPMI). In South Korea, however, casework-based ecological data remain limited, particularly for indoor scenes. We established a collaborative framework with investigating officers in the Yeongnam region (Gyeongsangbuk-do, Daegu, and Busan) and compiled forensic entomology cases from 2019 to 2022 to summarize the occurrence patterns of necrophagous flies. Scene information and specimens were collected using a standardized field record form, and species were identified by morphological examina

Antibacterial and antifungal activity of excretions and secretions of Calliphora vicina.

Antibiotic-resistant bacteria pose a major threat to global health in the 21st century, requiring a quick, cheap and effective response from public health officials. This study evaluated the antimicrobial activity of native excretions/secretions (NES) produced by third instar (3 days old) larvae of Calliphora vicina using a protocol adapted from the Institute of Clinical and Laboratory Standards (CLSI). The microorganisms tested were: Escherichia coli, Klebsiella pneumoniae, Staphylococcus aureus and the fungus Candida albicans. After the incubation period, the suspensions were diluted and spr

Recovery of male DNA acquired from carrion-feeding insects in a simulated sexual assault scenario.

When bodies of sexual assault victims are discovered in advanced stages of decomposition, the recovery of direct DNA evidence is often compromised, posing a challenge in criminal investigations. This study aimed to determine insect colonization and succession patterns on pig carcasses inoculated with semen and to assess the feasibility of recovering Y-chromosomal short tandem repeat (Y-STR) DNA from fly larvae, fly pupae, and beetles. Three female pigs were used as models to simulate sexual homicide scenarios, and an indoor control experiment using chicken livers mixed with 3 mL of semen was a

Differential effects on ploidy levels and underreplication of Cochliomyia macellaria as a function of electron beam energy and gamma radiation sources.

Cochliomyia hominivorax (Diptera: Calliphoridae) is an obligate parasitic species and a source of major public health concern as an agent of myiasis in humans, livestock, and wildlife systems, causing millions of dollars in agricultural losses annually. A major approach for screwworm control and eradication is the sterile insect technique (SIT) using gamma radiation. Electron beam (eBeam) radiation is a safer and potentially less expensive alternative ionizing radiation source being explored for SIT. To test this method, late-stage pupae of Cochliomyia macellaria, a sister species of C. homini
