Functional insights into the biosynthesis of antifungal mandimycin.

BACKGROUND: Escalating fungicide resistance intensifies crop losses, demanding sustainable solutions. We report Streptomyces netropsis AK308 from Xinjiang's Ailik Lake, which produces mandimycin, a polyene macrolide whose unique C-35 dideoxysaccharide is reported to enable phospholipid targeting, thereby potentially circumventing ergosterol-based resistance mechanisms. We evaluated its efficacy against 12 key pathogens and mapped its biosynthetic gene cluster to enable future development. RESULTS: The mandimycin mixture consisting of four congeners (A-D), showed broad-spectrum activity agains

Ion-activated mucoadhesive in situ gel incorporating terbinafine-loaded NLCs promotes ocular penetration and retention for enhanced fungal keratitis therapy.

Fungal keratitis (FK) is a sight-threatening infection with poor outcomes due to rapid tear clearance and insufficient corneal penetration. To overcome the limitations, we developed an ion-activated mucoadhesive in-situ gelling system (TH-NLC-ISG) integrating terbinafine-loaded nanostructured lipid carrier (TH-NLC) into a gellan gum/carboxymethyl chitosan matrix. The optimized formulation exhibited a particle size of 25.27 ± 0.75 nm. Upon contact with simulated tear fluid, the formulation underwent rapid sol-gel transition, achieving a 70-fold viscosity increase (34 °C, shear rate 1 s-1), crit

Keratomycosis: An insight into epidemiology, etiology, and antifungal susceptibility testing of causative agents at a tertiary care centre.

In India, mycotic keratitis accounts for 7%-63% of infectious keratitis. Ocular trauma being the primary predisposing factor for mycotic keratitis. The present prospective, observational study was conducted on the corneal scrapings from clinically suspected patients of keratomycosis. Samples were processed as per the standard mycological techniques. Antifungal susceptibility testing was performed as per CLSI guidelines M38A2 and M27A3 for mycelial fungi and yeast, respectively. Out of a total of 254 patients suspected to be suffering from mycotic keratitis based on clinical presentation, 68 (2

Current perspectives on ophthalmic mycoses.

Fungi may infect the cornea, orbit and other ocular structures. Species of Fusarium, Aspergillus, Candida, dematiaceous fungi, and Scedosporium predominate. Diagnosis is aided by recognition of typical clinical features and by direct microscopic detection of fungi in scrapes, biopsy specimens, and other samples. Culture confirms the diagnosis. Histopathological, immunohistochemical, or DNA-based tests may also be needed. Pathogenesis involves agent (invasiveness, toxigenicity) and host factors. Specific antifungal therapy is instituted as soon as the diagnosis is made. Amphotericin B by variou
