Combining sterile insect releases with refuge areas to delay the evolution of resistance to Bt sugarcane: an agent-based modeling approach.

The strategic use of refuge areas is a well-known method for delaying the development of pest resistance to Bacillus thuringiensis (Bt) crop. A lesser-known method to control against resistance development is sterile insect releases. In this article, an agent-based simulation model is used to test the effectiveness of combining the use of Bt sugarcane, refuge areas, and sterile insect releases as an integrated strategy against Eldana saccharina Walker (Lepidoptera: Pyralidae) infestation and resistance development. Individual insects are modeled with their own genetic traits on a simulated sug

Enhancing mass production of Heterorhabditis bacteriophora: influence of different bacterial symbionts (Photorhabdus spp.) and inoculum age on dauer juvenile recovery.

The entomopathogenic nematode Heterorhabditis bacteriophora (Nematoda: Rhabditidae) is used in biological insect control. Their dauer juveniles (DJs) are free-living and developmentally arrested, invading host insects. They carry cells of their bacterial symbiont Photorhabdus spp. in the intestine. Once inside the insect´s hemolymph the DJs perceive a food signal, triggering them to exit the DJ stage and regurgitate the Photorhabdus cells into the insect's haemocoel, which kill the host and later provide essential nutrients for nematode reproduction. The exit from the DJ stage is called "recov

Active tactile exploration and tactually induced turning in tethered walking stick insects.

Many animals use their tactile sense for active exploration and tactually guided behaviors such as near-range orientation. In insects, tactile sensing is often intimately linked to locomotion, resulting in the orchestration of several concurrent active movements, including turning of the entire body, rotation of the head, and searching or sampling movements of the antennae. The present study aims at linking the sequence of tactile contact events to associated changes of all three kinds of these active movements (body, head and antennae). To do so, we chose the Indian stick insect Carausius mor

Insecticidal potential of Brevibacillus laterosporus against dipteran pest species in a wide ecological range.

In order to increase our understanding of the insecticidal potential of the entomopathogenic bacterium Brevibacillus laterosporus strain UNISS 18 against insect pests, investigations were conducted on a selection of dipteran species including fruit flies, house flies, blow flies, and mosquitoes, characterized by adaptations to very diverse habitats. According to lethal concentration (LC50) values, the common house mosquito Culex pipiens (LC50 = 0.10 × 106 spores/mL) and the yellow fever mosquito Aedes aegypti (LC50 = 0.18 × 106 spores/mL) were significantly more susceptible than the flies. Co-existence at a cost: The outcome of competition between Steinernema and Heterorhabditis within the same insect host.

Competition within a single insect between the two genera of entomopathogenic nematodes, Heterorhabditis and Steinernema, was explored using H. downesi, H. bacteriophora, S. carpocapsae, and S. feltiae. Insect larvae were infected with one genus (primary invader) and then exposed to the other genus (secondary invader) after 24, 48 or 72 h. We hypothesised that (1) Steinernema and Heterorhabditis will not both reproduce within the same insect (2) reproductive success of the secondary invader will depend on identity of the primary invader and length of occupation and (3) nematodes will avoid ins

Lipid metabolism in insect disease vectors.

More than a third of the world population is at constant risk of contracting some insect-transmitted disease, such as Dengue fever, Zika virus disease, malaria, Chagas' disease, African trypanosomiasis, and others. Independent of the life cycle of the pathogen causing the disease, the insect vector hematophagous habit is a common and crucial trait for the transmission of all these diseases. This lifestyle is unique, as hematophagous insects feed on blood, a diet that is rich in protein but relatively poor in lipids and carbohydrates, in huge amounts and low frequency. Another unique feature of

Targeting the malaria parasite inside the mosquito host.

Control of malaria uses a variety of tools including drugs, vaccines, and insecticidal strategies to reduce parasite transmission by mosquito vectors. A new tool that eliminates parasites in mosquitoes by delivering antimalarials to the insects rather than infected people is emerging as a simple, relatively inexpensive malaria control strategy. In this Chapter, we review antimalarials that show efficacy in mosquitoes, effective methods to deliver compounds into the insects, and strategies to optimize this new tool as a useful component of the arsenal of strategies to combat malaria.

Virtual dissection of Aedes aegypti mosquito using phase-contrast synchrotron microtomography.

In this paper, in-line phase-contrast synchrotron microtomography was used to study the morphology of adult Aedes aegypti. These specimens are vectors of several arboviruses, causing dengue, chikungunya, Zika and yellow fever. The morphological details of this insect species are still incomplete and insufficient. To address this gap, this study examined whole specimens of Aedes aegypti in the adult phase at high resolution. For this, the adult samples were scanned in the microtomography beamline (SYRMEP) at the Italian Synchrotron Light Laboratory (ELETTRA). The phase-contrast technique allowe

Current insecticide resistance patterns in mosquito vectors in Thailand.

Chemical pesticides are still commonly used in Thailand for control of agricultural pests and disease vectors. Organophosphates, carbamates and synthetic pyrethroids are commonly used for agricultural purposes, whereas synthetic pyrethroids have become more popular and predominate for public health use. The genetic selection of insecticide resistance (whether physiological, biochemical or behavioral) in pests and disease vectors has been extensively reported worldwide (Brown and Pal, 1971). The long-term intensive use of chemical pesticides to control insect pests and disease vectors is often

Toxicological effects of a difenoconazole fungicide on a non-target butterfly.

Agricultural intensification, driven by increasing food demand, is one of the main drivers of global biodiversity loss. Pesticides, including fungicides, may increase crop yield but pose substantial threats to the environment. However, our current knowledge on the effects of fungicides on biodiversity suffers from less research interest compared to other pesticides and a neglect of insect herbivores. We here investigate the effects of a commercially available, ready-to-use difenoconazole fungicide on a non-target insect, the butterfly Pieris napi, under controlled laboratory conditions. When b

Fine-tuning striated muscle performance: conserved sarcomere-level mechanisms across insect and vertebrate systems.

Striated muscles exhibit remarkable structural and functional specialization that enables precise control of force production, contractile kinetics, and energetic efficiency. Although vertebrate skeletal and cardiac muscles have been extensively studied, comparative analyses across animal phyla reveal that many molecular and biophysical principles governing muscle performance are deeply conserved. Insects, in particular, possess highly differentiated muscle types that provide powerful systems for dissecting the regulation of contraction, elasticity, and force generation at the level of the sar

Candidate gene screen in the red flour beetle Tribolium reveals six3 as ancient regulator of anterior median head and central complex development.

Several highly conserved genes play a role in anterior neural plate patterning of vertebrates and in head and brain patterning of insects. However, head involution in Drosophila has impeded a systematic identification of genes required for insect head formation. Therefore, we use the red flour beetle Tribolium castaneum in order to comprehensively test the function of orthologs of vertebrate neural plate patterning genes for a function in insect head development. RNAi analysis reveals that most of these genes are indeed required for insect head capsule patterning, and we also identified severa

Global distribution and sustainable management of Asian corn borer (ACB), Ostrinia furnacalis (Lepidoptera: Crambidae): recent advancement and future prospects.

The Asian corn borer (ACB), Ostrinia furnacalis (Guenée, 1854), is a serious pest of several crops, particularly a destructive pest of maize and other cereals throughout most of Asia, including China, the Philippines, Indonesia, Malaysia, Thailand, Sri Lanka, India, Bangladesh, Japan, Korea, Vietnam, Laos, Myanmar, Afghanistan, Pakistan and Cambodia. It has long been known as a pest in South-east Asia and has invaded other parts of Asia, Solomon Islands, parts of Africa and certain regions of Australia and Russia. Consequently, worldwide efforts have been increased to ensure new control strate

Directional selection to improve the sterile insect technique: Survival and sexual performance of desiccation resistant Anastrepha ludens strains.

The sterile insect technique (SIT) is an effective, environmentally friendly method for insect control whose success depends on the sexual performance and survival of sterile males. These two parameters are influenced by environmental conditions of target areas, and releasing insects with a higher tolerance to stressful environments can improve SIT efficiency. Directional selection can be used to produce insect strains with higher tolerance to extreme environmental conditions, such as low humidity, for extended periods. We evaluated, under field cage conditions, the sexual competitiveness, sex

Is the lobster cockroach Nauphoeta cinerea a valuable model for evaluating mercury induced oxidative stress?

Organic and inorganic forms of mercury are highly neurotoxic environmental contaminants. The exact mechanisms involved in mercury neurotoxicity are still unclear. Oxidative stress appears to play central role in this process. In this study, we aimed to validate an insect-based model for the investigation of oxidative stress during mercury poisoning of lobster cockroach Nauphoeta cinerea. The advantages of using insects in basic toxicological studies include the easier handling, rapid proliferation/growing and absence of ethical issues, comparing to rodent-based models. Insects received solutio

Aberrant splicing of a nicotinic acetylcholine receptor alpha 6 subunit is associated with spinosad tolerance in the thrips predator Orius laevigatus.

Susceptibility to insecticides is one of the limiting factors preventing wider adoption of natural enemies to control insect pest populations. Identification and selective breeding of insecticide tolerant strains of commercially used biological control agents (BCAs) is one of the approaches to overcome this constraint. Although a number of beneficial insects have been selected for increased tolerance to insecticides the molecular mechanisms underpinning these shifts in tolerance are not well characterised. Here we investigated the molecular mechanisms of enhanced tolerance of a lab selected st

Generating prophylactic immunity against arboviruses in vertebrates and invertebrates.

The World Health Organization recently declared a global initiative to control arboviral diseases. These are mainly caused by pathogenic flaviviruses (such as dengue, yellow fever and Zika viruses) and alphaviruses (such as chikungunya and Venezuelan equine encephalitis viruses). Vaccines represent key interventions for these viruses, with licensed human and/or veterinary vaccines being available for several members of both genera. However, a hurdle for the licensing of new vaccines is the epidemic nature of many arboviruses, which presents logistical challenges for phase III efficacy trials.

Tightly intertwined: Waterscapes prompt urgent reconsideration of aquatic insects and their role in agricultural landscapes.

In landscape ecology, the waterscape refers to permanent or temporary, running or stagnant surface waters within a terrestrial area. Across ecosystem boundaries, aquatic organisms and nutrients can reach terrestrial ecosystems, as formalised by the meta-ecosystem theory. Recent studies on aquatic insects emerging from temperate streams suggest that the extent of their biomass and fluxes across agricultural landscapes may have been neglected until now. Following a conceptual and empirical approach, we presently discuss how the temporal dynamics of floods coupled with the emergence and aerial fl

Construction of an improved baculovirus insecticide containing an insect-specific toxin gene.

Baculoviruses provide alternatives to chemicals for controlling insect pests and can be applied by spraying. Baculoviruses have a limited host range, but work relatively slowly. They are dissolved in the midgut of insect larvae to release infectious virions which enter gut epithelial cells and begin to replicate. Replication in other organs causes extensive tissue damage and eventually death. This process can take 4-5 days, but in the field may last for more than a week, allowing the larvae to feed for longer and thereby damaging the host plant. Baculovirus expression vectors expressing foreig
