Metabolomics-Driven Elucidation of Cellular Nitrate Tolerance Reveals Ascorbic Acid Prevents Nitroglycerin-Induced Inactivation of Xanthine Oxidase.

Glyceryl trinitrate (GTN) has found widespread use for the treatment of angina pectoris, a pathological condition manifested by chest pain resulting from insufficient blood supply to the heart. Metabolic conversion of GTN, a nitric oxide (NO) pro-drug, into NO induces vasodilation and improves blood flow. Patients develop tolerance to GTN after several weeks of continuous use, limiting the potential for long-term therapy. The mechanistic cause of nitrate tolerance is relatively unknown. We developed a cell culture model of nitrate tolerance that utilizes stable isotopes to measure metabolism o

Synthesis and characterization of a novel organic nitrate NDHP: Role of xanthine oxidoreductase-mediated nitric oxide formation.

In this report, we describe the synthesis and characterization of 1,3-bis(hexyloxy)propan-2-yl nitrate (NDHP), a novel organic mono nitrate. Using purified xanthine oxidoreductase (XOR), chemiluminescence and electron paramagnetic resonance (EPR) spectroscopy, we found that XOR catalyzes nitric oxide (NO) generation from NDHP under anaerobic conditions, and that thiols are not involved or required in this process. Further mechanistic studies revealed that NDHP could be reduced to NO at both the FAD and the molybdenum sites of XOR, but that the FAD site required an unoccupied molybdenum site. C

Use of xanthine oxidase inhibitor febuxostat inhibits renal interstitial inflammation and fibrosis in unilateral ureteral obstructive nephropathy.

BACKGROUND: Renal interstitial fibrosis is the common pathway in progressive renal diseases, where oxidative stress promotes inflammation and macrophage infiltration. Febuxostat is a novel nonpurine xanthine oxidase (XO)-specific inhibitor for treating hyperuricemia. While some reports suggest a relationship between hyperuricemia and chronic kidney disease (CKD), the renoprotective mechanism of an XO inhibitor in CKD remains unknown. Recent reports have focused on XO as a source of oxidative stress. METHODS: Here, we investigate the potential of febuxostat to reduce fibrogenic and inflammator

Steady-state kinetics of the hypoxanthine-guanine-xanthine phosphoribosyltransferase from Tritrichomonas foetus: the role of threonine-47.

Tritrichomonas foetus, an anaerobic flagellated protozoan, causes urogenital trichomoniasis in cattle. Hypoxanthine-guanine-xanthine phosphoribosyl transferase (HGXPRTase), an essential enzyme in T. foetus required for salvaging exogenous purine bases, has been regarded as a promising target for anti-tritrichomonial chemotherapy. The steady-state kinetic analyses of synthesis and pyrophosphorolysis of IMP, GMP, and XMP and product inhibition studies have been used to elucidate the reaction mechanisms. Double-reciprocal plots of initial velocities versus the varying concentrations of one substr

Xanthine derivatives: comparison between suppression of tumour necrosis factor-alpha production and inhibition of cAMP phosphodiesterase activity.

Several in vitro and in vivo studies have demonstrated suppression of tumour necrosis factor-alpha (TNF-alpha) synthesis by pentoxifylline. In the present study we compared the effect of pentoxifylline with that of five other xanthine derivatives. We addressed two questions. First, what is the relative potency of those chemically related compounds in suppressing the lipopolysaccharide (LPS)-induced production of TNF-alpha in human mononuclear cells? Second, does suppression of TNF-alpha production by these xanthine derivatives correlate with their capacity to inhibit 3',5'-cAMP phosphodiestera

Gas Phase Studies of Purine Base Analogs: Intrinsic Properties and Hypoxanthine-Guanine-(Xanthine) Phosphoribosyltransferase (HG(X)PRT).

The gas-phase acidity and proton affinity of purine nucleobases that are found to be inactive for the enzyme Plasmodium falciparum hypoxanthine-guanine-(xanthine) phosphoribosyltransferase (Pf HG(X)PRT) have been examined using both computational and experimental methods. These thermochemical values have not heretofore been measured and provide experimental data to benchmark the theoretical results. Pf HG(X)PRT is an antimalarial target; our gas-phase results lend insight into the Pf HG(X)PRT mechanism, providing a potential means of predicting enzyme substrate activity.

Design, synthesis, and biological evaluation of benzoisonicotinamide derivatives as novel xanthine oxidase inhibitors guided by metabolic stability.

In our previous work, we identified TS10, an isonicotinamide-based xanthine oxidase (XO) inhibitor with excellent in vitro enzymatic activity. However, its poor metabolic stability, primarily caused by the metabolically labile phenyl benzyl ether moiety, may have contributed to its limited in vivo efficacy. In the present study, two optimization strategies, including all-carbon scaffold replacement and phenyl-O-piperidine/tetrahydropyrrole reconstruction, were employed to improve the metabolic stability and overall drug-like properties of TS10. Among the synthesized derivatives, S40 exhibited

Stereoisomer-specific allosteric activation of xanthine oxidoreductase by shikonin and alkannin: mechanistic insights and hyperuricemia risk assessment.

Xanthine oxidoreductase (XOR), the rate-limiting enzyme in uric acid metabolism, reversibly interconverts between dehydrogenase (XDH) and oxidase (XO) forms. Our previous study revealed allosteric activation of XOR by 1,4-naphthoquinones, highlighting their potential risk in food industry. Herein, we employed the isomeric 1,4-naphthoquinone probes shikonin (SHI) and alkannin (ALK) to elucidate the underlying mechanism. Site-directed mutagenesis demonstrated that SHI and ALK activate XOR by binding a Trp336-centered allosteric pocket, independent of classic Cys536-Cys993 bond. This binding dest

Exploring the role of xanthine oxidase and aldehyde oxidase in metabolic dysfunction-associated steatotic liver disease (MASLD).

Globally, metabolic dysfunction-associated steatotic liver disease (MASLD), formerly termed as non-alcoholic fatty liver disease (NAFLD), has been affecting millions of people. It includes a spectrum of liver conditions, ranging from simple fat accumulation (steatosis) to advanced stages such as fibrosis, cirrhosis and hepatocellular carcinoma (HCC). Oxidative stress has been identified as a major contributing factor in driving disease progression. Enzymes such as xanthine oxidase (XO) and aldehyde oxidase (AO) are significant contributors of reactive oxygen species (ROS), which in turn promot

Engineering oxypurinol-responsive riboswitches based on bacterial xanthine aptamers for gene expression control in mammalian cell culture.

Riboswitch-mediated control of gene expression without the interference of potentially immunogenic proteins is a promising approach for the development of tailor-made tools for biological research and the advancement of gene therapies. However, the current selection of applicable ligands for synthetic riboswitches is limited and strategies have mostly relied on de novo selection of aptamers. Here, we show that the bacterial xanthine I riboswitch aptamer recognizes oxypurinol, the active metabolite of the widely prescribed anti-gout drug allopurinol (Zyloprim®). We have characterized the aptame

Serum xanthine oxidoreductase and oxidative stress are associated with bladder cancer: a case-control study from Jordan.

Xanthine oxidoreductase (XOR) is an oxidant enzyme that exists mainly in two distinct forms: the dehydrogenase form [xanthine dehydrogenase (XDH)] and the oxidized form [xanthine oxidase (XO)]. XO might contribute to tumorigenesis through direct metabolic activation of carcinogens and indirect generation of free radicals. Oxidative stress is one of the leading causes of bladder cancer (BC). Smoking and genetic susceptibility are also linked to oxidative stress and BC. This study investigated the association between XO serum levels and XOR genetic polymorphisms with BC. A case-control study was

Bifunctional Fe-CDs@MOF composite nanozyme based ratiometric fluorescent probe for the detection of xanthine in fish.

A novel bifunctional Fe-CDs@MOF composite nanozyme was synthesized successfully by loading Fe doped carbon dots (Fe-CDs) onto metal-organic framework (MOF) MIL-101 (Fe). The introduction of Fe-CDs enhanced the peroxidase (POD)-like activity of MOF and endowed it with excellent fluorescence properties. Considering the dual functions of Fe-CDs@MOF, we combined it with o-phenylenediamine (OPD)/xanthine oxidase (XOD) to construct a ratiomeric fluorescence probe for xanthine (XAN) detection. XAN was first oxidized by XOD to produce H2O2, then Fe-CDs@MOF exerted its POD-like activity to catalyze the

Impact of adding urine alkalization therapy to xanthine oxidase inhibitor in gout management: a prospective cohort study.

OBJECTIVES: Gout patients frequently have decreased urine pH, related to metabolic syndrome (MetS) and chronic kidney disease. Here, we aimed to investigate whether the addition of urine alkalization to urate-lowering therapy (ULT) is associated with improvements in albuminuria, gout flares or MetS outcomes in men with gout and low urinary pH (pH < 6.2). METHODS: A prospective cohort study enrolled 385 participants starting ULT with febuxostat (initially 20 mg daily, escalated to 40 mg daily if serum urate ≥360 µmol/l), with or without alkalization based on patient preference using a 3.5 g bi

Sustained xanthine oxidase inhibitor treat to target urate lowering therapy rewires a tight inflammation serum protein interactome.

BACKGROUND: Effective xanthine oxidoreductase inhibition (XOI) urate-lowering treatment (ULT) to target significantly reduces gout flare burden and synovitis between 1-2 years therapy, without clearing all monosodium urate crystal deposits. Paradoxically, treat to target ULT is associated with increased flare activity for at least 1 year in duration on average, before gout flare burden decreases. Since XOI has anti-inflammatory effects, we tested for biomarkers of sustained, effective ULT that alters gouty inflammation. METHODS: We characterized the proteome of febuxostat-treated murine bone

Antioxidant action of xanthine oxidase inhibitor febuxostat protects the liver and blood vasculature in SHRSP5/Dmcr rats.

BACKGROUND: Xanthine oxidase (XO) generates reactive oxygen species during uric acid production. Therefore, XO inhibitors, which suppress oxidative stress, may effectively treat non-alcoholic steatohepatitis (NASH) and atherosclerosis via uric acid reduction. In this study, we examined the antioxidant effect of the XO inhibitor febuxostat on NASH and atherosclerosis in stroke-prone spontaneously hypertensive 5 (SHRSP5/Dmcr) rats. METHODS: SHRSP5/Dmcr rats were divided into three groups: SHRSP5/Dmcr + high-fat and high-cholesterol (HFC) diet [control group, n = 5], SHRSP5/Dmcr + HFC diet + 10%
