68Ga radiolabeling strategies in Pt(IV)-deferoxamine scaffolds for potential theranostic application.

Since the discovery of cisplatin's anticancer activity and its clinical approval in 1978, substantial efforts have focused on improving its physiological stability and minimizing off-target toxicity. One promising strategy has been the development of Pt(IV) complexes, which act as redox-activated prodrugs with improved pharmacological profiles compared to Pt(II) drugs. In this study, we present the synthesis of three novel Pt(IV) complexes bearing a pendant deferoxamine (DFO) chelator, designed for theranostic application, combining therapeutic activity with radiometal labeling for positron em

Evaluation of 9-Minute Dual-Energy Peak Time-Reduced 360° CZT Total-Body [177Lu]Lu-PSMA SPECT/CT Imaging with Comparisons to [68Ga]Ga-PSMA PET.

Eighteen-minute 360° cadmium zinc telluride (CZT) total-body 177Lu-labeled SPECT acquisitions on the single 113-keV 177Lu photopeak (SP-18) have been reported to be suitable for monitoring [177Lu]Lu-PSMA therapy. This study evaluated dual-energy 113- and 208-keV acquisitions with a more than 2-fold increase in acquired counts, allowing acquisition time to be reduced to 9 min (DP-9). Methods: In 20 patients with prostate cancer, 360° CZT total-body 177Lu-labeled SPECT acquisitions performed after the first [177Lu]Lu-PSMA therapeutic injection were reconstructed as SP-18, DP-9, and dual-energy 1

Does PARP1 up-regulation correlate with PSMA expression in patients with metastatic castration-resistant prostate cancer studied with [18F]PARPi and [68Ga]PSMA PET/CT?

PURPOSE: [18F] Poly-ADP-ribose polymerase inhibitors (PARPi), a novel radiotracer, enables visualization of PARP1 upregulation by PET imaging. Here, we aimed to quantify PARPi uptake in tumor lesions of metastatic castration-resistant PCa (mCRPC) patients and perform a comparison with prostate specific membrane antigen (PSMA) expression using PET/CT scans. METHODS: Data from 22 male patients with mCRPC, who underwent [18F]PARPi and [68Ga]Ga-PSMA-11 PET/CT scans, were retrospectively quantified. Lesions with relevant PARPi uptake (higher than background) were delineated and correlated with the

Real-World Treatment Outcomes in Patients With Ga68-PSMA-PET Positive Metastatic Hormone-Sensitive Prostate Cancer With or Without Conventional Imaging Correlates.

BACKGROUND: Prostate-specific membrane antigen (PSMA) positron emission tomography (PET) imaging is highly sensitive, enabling detection of disease not visualized on conventional imaging, and leading to a new disease state: PSMA-avid/CT-negative metastatic prostate cancer. The optimal management and treatment outcomes for this group remain poorly defined. We investigated whether treatment intensification with androgen receptor signaling inhibitors (ARSI) provides additional benefit for these patients. METHODS: We conducted a retrospective study of patients with metastatic hormone-sensitive pr

Rapid chelator-free radiolabeling of quantum dots for in vivo imaging.

Most current nanoparticle-based PET tracers are radiolabeled through metal chelators conjugated on the nanoparticle surface. Metal chelation usually requires sophisticated optimization and may impact the physical or chemical properties of nanoparticles, which leads to the changes in their distribution and pharmacokinetics in vivo. A chelator-free radiolabeling approach is thus highly desirable. Here, we report that zinc sulfide (ZnS) quantum dots (QDs) can be rapidly radiolabeled with 68Ga or 64Cu through cation exchange without chelators. The radiolabeling was accomplished in times as short a

Long-term sequelae of gastrectomy.

The sequelae of gastrectomy are well-known. This paper describes a complicated clinical course which occurred eight years after partial gastrectomy. Initial features included marked weight loss, anaemia, multiple lung and liver abscesses, and cholestatic jaundice. Acalculous cholecystitis with a mucocele of the gallbladder developed later. We emphasise the value of total parenteral nutrition in supporting such critically ill patients.

Biodistribution and toxicity of innate defense regulator 1018 (IDR-1018).

Innate defense regulators (IDRs) are synthetic host-defense peptides (HDPs) with broad-spectrum anti-infective properties, including immunomodulatory, anti-biofilm and direct antimicrobial activities. A lack of pharmacokinetic data about these peptides hinders their development and makes it challenging to fully understand how they work in vivo since their mechanism of action is dependent on tissue concentrations of the peptide. Here, we set out to define in detail the pharmacokinetics of a well-characterized IDR molecule, IDR-1018. To make the peptide traceable, it was radiolabeled with the lo

Synthesis and evaluation of a novel 68Ga-chelate-conjugated bisphosphonate as a bone-seeking agent for PET imaging.

INTRODUCTION: (68)Ga is a positron-emitting nuclide that has significant imaging potential given that, unlike cyclotron-produced (18)F, the isotope can be produced on-site utilizing a (68)Ge/(68)Ga generator. We recently synthesized a novel bone-seeking agent by coupling a bisphosphonate with the (68)Ga chelator 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA). This study presents a first report on the potential of this (68)Ga bone-seeking radiopharmaceutical in the detection of bone metastases. METHODS: 4-Amino-1-hydroxybutylidene-1,1-bisphosphonate was conjugated with 2-[4,7-di(carboxyme

Triaza-based amphiphilic chelators: synthetic route, in vitro characterization and in vivo studies of their Ga(III) and Al(III) chelates.

Radiogallium chelates are important for diagnostic imaging in nuclear medicine (PET (positron emission tomography) and gamma-scintigraphy). Micelles are adequate colloidal vehicles for the delivery of therapeutic and diagnostic agents to organs and tissues. In this paper we describe the synthesis and in vitro and in vivo studies of a series of micelles-forming Ga(III) chelates targeted for the liver. The amphiphilic ligands are based on NOTA (NOTA=1,4,7-triazacyclonoane-N,N'N''-triacetic acid) and bear a alpha-alkyl chain in one of the pendant acetate arms (the size of the chain changes from f

PSMA-PET follow-up to assess response in patients not receiving PSMA therapy: Is there value beyond localization of disease?

Introduction: Prostate Specific Membrane Antigen Positron Emission Tomography (PSMA-PET) is routinely used for the staging of patients with prostate cancer, but data on response assessment are sparse and primarily stem from metastatic castration-resistant prostate cancer (mCRPC) patients treated with PSMA radioligand therapy. Still, follow-up PSMA-PET is employed in earlier disease stages in case of clinical suspicion of disease persistence, recurrence or progression to decide if localized or systemic treatment is indicated. Therefore, the prognostic value of PSMA-PET derived tumor volumes in
