Part XIV. Antiviral agents other than antiretrovirals: therapy for herpesvirus infections, viral hepatitis, and influenza.

The incidences of viral infectious diseases are increasing at an alarming rate in the US and worldwide. Antiviral therapy is challenging because viruses subsume normal host cellular mechanisms for many functions, have rapid replication rates, have poor error scanning when reading genetic code, and undergo frequent drug target mutations. This article will focus on antiviral drugs and principles of treatment for infections due to herpes simplex viruses (HSV1 and HSV2), varicella zoster virus (VZV), cytomegalovirus (CMV), hepatitis B and hepatitis C viruses, and influenza virus. Therapy for human

Disruption of FBN1 by an Alu element insertion: A novel genetic cause of Marfan syndrome.

Alu elements are retrotransposons with ubiquitous presence in the human genome that have contributed to human genomic diversity and health. These approximately 300-bp sequences can cause or mediate disease by disrupting coding/splicing regions in the germline, by insertional mutagenesis in somatic cells, and in promoting formation of copy-number variants. Alu elements may also disrupt epigenetic regulation by affecting non-coding regulatory regions. There are increasing reports of apparently sporadic and inherited genetic disorders caused by Alu-related gene disruption, but Marfan syndrome res

Genetic analysis of West Nile virus isolates from US blood donors during 2002-2005.

BACKGROUND: Since its introduction into North America in 1999, West Nile virus (WNV) has spread rapidly across United States (US). OBJECTIVE: To genetically analyze WNV isolates from US blood donors during 2002-2005. STUDY DESIGN: Full-length nucleotide (NT) sequences of WNV isolates from 23 US volunteer blood donors of different geographic areas from 2002 to 2005 were determined and analyzed. RESULTS: Results indicated an overall lack of geographic pattern to WNV in US. Analyses of the viral genetic diversity demonstrated that the WNV evolved at approximately five NT substitutions and 0.8

Deus ex Candida genetics: overcoming the hurdles for the development of a molecular toolbox in the CTG clade.

Dominant selectable markers, reporter genes and regulatable systems remain powerful molecular tools for genetic and cell biology studies in fungi. Among Saccharomycotina, it is currently accepted that most species belonging to the genus Candida have adopted a specific codon usage, whereby the CTG codon encodes serine instead of leucine. This group is now widely referred to as the CTG clade. For a long time, this uncommon genetic code has precluded the use of the available Saccharomyces or bacterial markers and reporter systems for genetic studies in Candida species. Over the last 15 years, inc

Craniofacial and Dental Anomalies of a Patient Carrying Two MicroRNA Variants: A Proof-Of-Concept Case Report.

Craniofacial and dentofacial anomalies often result from disruptions in embryogenetic processes driven by genetic alterations. Dental development involves complex interactions between coding and non-coding genes, orchestrated by a network of signaling pathways. Next Generation Sequencing (NGS) has identified genes, particularly in the WNT signaling pathway, associated with dental anomalies. MicroRNAs (miRNAs), small non-coding RNA molecules, play a crucial role in post-transcriptional regulation. Variants in miRNAs, such as in MIR146A, have been linked to various craniofacial pathologies. A 10

[Mutations in the genes coding for hemagglutinin and neuraminidase in cold-adapted variants of the influenza virus A/Leningrad/134/57 (H2N2)].

An analysis of ts-mutations in the genomes of native and cold-adapted variants of influenza A/Leningrad/134/57 (H2N2) virus based on the use of fowl plague virus ts mutants was carried out. The recombination test was done by the conventional method in chick embryo fibroblast culture (genes PB2, PB1, PA, NP, NA, M and NS) or cell systems permissive for reproduction of human influenza virus (gene HA). The cold-adapted strain A/Len/17 used for preparation of live influenza vaccine (LIV) for adults was shown to have 4 ts mutations: three in "internal" genes (PB2, NP, and M) and one in gene 4 codin

A novel method for the detection of point mutation in DNA using single-base-coded CdS nanoprobes.

A novel method for DNA point mutation detection using single-base-coded CdS nanoparticle probes is proposed. Target DNA was immobilized on core/shell Fe(3)O(4)/Au magnetic nanoparticles. Single-base-coded CdS nanoparticles, such as guanosine coded CdS (G-CdS), cytidine coded CdS (C-CdS), thymidine coded CdS (T-CdS) and adenosine coded CdS (A-CdS) were used as the probes to identify the mutation sites in DNA strand. The hybridization process of single-base-coded CdS nanoparticle probes with the mutation sites in DNA was monitored using piezoelectric quartz crystal microbalance (QCM). The hybrid

UGT1A1 sequence variants and bilirubin levels in early postnatal life: a quantitative approach.

BACKGROUND: Fundamental to definitively identifying neonates at risk of developing significant hyperbilirubinemia is a better understanding of the genetic factors associated with early bilirubin rise. Previous genetic studies have focused on the UGT1A1 gene, associating common variation in the coding or promoter regions with qualitative assessments of bilirubin (i.e. significantly elevated or not). These studies have had conflicting results and limited success. We chose to approach the problem by focusing on the quantitative (absolute) change in bilirubin levels early in post-natal life. We ap

The Global ALPL gene variant classification project: Dedicated to deciphering variants.

BACKGROUND: Hypophosphatasia (HPP) is an inherited multisystem disorder predominantly affecting the mineralization of bones and teeth. HPP is caused by pathogenic variants in ALPL, which encodes tissue non-specific alkaline phosphatase (TNSALP). Variants of uncertain significance (VUS) cause diagnostic delay and uncertainty amongst patients and health care providers. RESULTS: The ALPL gene variant database (https://alplmutationdatabase.jku.at/) is an open-access archive for interpretation of the clinical significance of variants reported in ALPL. The database contains coding and non-coding va

Cell-Free Fetal DNA Testing for Single Gene Conditions (Scientific Impact Paper No. 80).

The DNA code represents a genetic instruction as to how the human body develops and works. A 'spelling mistake' in the DNA code of the developing fetus can lead to a single gene condition. Examples of single-gene conditions include cystic fibrosis, sickle cell disease and Huntington's disease. Single gene conditions collectively affect 1 in 100 births and can be inherited from one or both parents, or can occur without any family history, arising spontaneously out of a one-off change (pathogenic variant) in a known gene. Sometimes there can be specific anomalies seen on the ultrasound scan that

Nexilin in cardiomyopathy: unveiling its diverse roles with special focus on endocardial fibroelastosis.

Cardiac disorders exhibit considerable heterogeneity, and understanding their genetic foundations is crucial for their diagnosis and treatment. Recent genetic analyses involving a growing number of participants have uncovered novel mutations within both coding and non-coding regions of DNA, contributing to the onset of cardiac conditions. The NEXN gene, encoding the Nexilin protein, an actin filament-binding protein, is integral to normal cardiac function. Mutations in this gene have been linked to cardiomyopathies, cardiovascular disorders, and sudden deaths. Heterozygous or homozygous varian

Optimized phenotype definitions boost GWAS power.

Complex diseases are a major challenge, and genetics underlie a large fraction of the risk for these diseases. Observational data are helpful for this research due to large scale, cost-effectiveness, information on many different conditions, and future scalability, but they reflect factors such as healthcare processes, access to care, and broader societal effects like systemic biases. Here, we introduce MaxGCP, a phenotyping method designed to purify the genetic signal in observational data. MaxGCP optimizes a phenotype definition to maximize its coheritability-the genetic covariance between t

Neurological disorders caused by novel non-coding repeat expansions: clinical features and differential diagnosis.

Nucleotide repeat expansions in the human genome are a well-known cause of neurological disease. In the past decade, advances in DNA sequencing technologies have led to a better understanding of the role of non-coding DNA, that is, the DNA that is not transcribed into proteins. These techniques have also enabled the identification of pathogenic non-coding repeat expansions that cause neurological disorders. Mounting evidence shows that adult patients with familial or sporadic presentations of epilepsy, cognitive dysfunction, myopathy, neuropathy, ataxia, or movement disorders can be carriers o

Immunogenetic factors in beryllium sensitization and chronic beryllium disease.

Exposure to beryllium in the workplace can cause beryllium sensitization and chronic beryllium disease. Sensitization to beryllium can be detected in the laboratory using the beryllium lymphocyte proliferation test. It was shown that anti-HLA antibodies could block the beryllium-specific response in the beryllium lymphocyte proliferation test, thereby implicating HLA genes in chronic beryllium disease. A supratypic genetic marker, HLA-DPB1*E69, was found to be strongly associated with immunologic sensitization to beryllium and chronic beryllium disease in beryllium workers. However, there are
