A comparison of intragastric and intrajejunal feedings in neonatal piglets.

Intrajejunal feedings by continuous infusion of nutrients into the proximal jejunum have been advocated for premature and low-birthweight infants. This technique eliminates the risks of gastric distention and aspiration associated with intragastric feedings. However, intrajejunal feedings disregard the normal physiologic responses to gastric feedings and completely bypass the stomach and duodenum, thus eliminating the important contributions to intestinal function of salivary enzymes, pancreatic secretions, and duodenal osmoregulation and absorption. The objective of this investigation is the

Lysozyme Counteracts β-Lactam Antibiotics by Promoting the Emergence of L-Form Bacteria.

β-lactam antibiotics inhibit bacterial cell wall assembly and, under classical microbiological culture conditions that are generally hypotonic, induce explosive cell death. Here, we show that under more physiological, osmoprotective conditions, for various Gram-positive bacteria, lysis is delayed or abolished, apparently because inhibition of class A penicillin-binding protein leads to a block in autolytic activity. Although these cells still then die by other mechanisms, exogenous lytic enzymes, such as lysozyme, can rescue viability by enabling the escape of cell wall-deficient "L-form" bact

Deciphering transcriptomic insights into climate adaptation in Etroplus suratensis (pearl spot) under combined thermal-saline stress.

Climate change poses significant challenges to aquaculture through rising temperatures and salinity fluctuations, necessitating insights into molecular adaptation mechanisms in resilient species like pearl spot (Etroplus suratensis) which is an euryhaline fish. This study performed de novo transcriptomic profiling of gill, liver, and kidney tissues from juvenile pearl spot exposed to combined temperature (28 °C control, 31 °C intermediate, 34 °C stress) and salinity (0 ppt freshwater, 15 ppt brackishwater) treatments over 90 days in triplicate setups. Gill ionocytes of the Lake Magadi tilapia (Oreochromis Alcolapia grahami), an extremophilic teleost native to a highly alkaline environment.

The Magadi tilapia thrives in arguably the most extreme aquatic environment on earth for fish, the hot springs of Lake Magadi in Kenya with its severe water chemistry: pH 10, alkalinity 300 mEq·L-1. This fish is 100% ureotelic yet has an osmoregulation pattern typical of marine teleosts, although the dominant water anion is HCO3- rather than Cl-. The gills must actively export base equivalents (HCO3- + CO32-) and Na+ against strong electrochemical gradients, yet simultaneously take up Cl-, for which a hypothetical "Laurent model" based on ionocyte structure alone was proposed. This model has b

Early Cellular Responses of Neurospora crassa to Azole Stress: Osmotic Adjustment, Redox Buffering, Sterol-Pathway Feedback, and Cell Wall Remodelling.

Background/Objectives: Azole resistance in fungal pathogens is a growing clinical and environmental concern. However, short-term cellular responses during the first hours of azole exposure remain insufficiently characterised, particularly in filamentous fungi. This study used Neurospora crassa as a genetically tractable model to investigate early cellular responses to azole stress. Methods: Exponentially growing mycelia were exposed to conidium-derived MIC80 reference concentrations of fluconazole, voriconazole, ravuconazole, and ketoconazole. Early responses were assessed by monitoring radial

Untangling mechanisms of crude oil toxicity: Linking gene expression, morphology and PAHs at two developmental stages in a cold-water fish.

Early life stages of fish are highly sensitive to crude oil exposure and thus, short term exposures during critical developmental periods could have detrimental consequences for juvenile survival. Here we administered crude oil to Atlantic haddock (Melanogrammus aeglefinus) in short term (3-day) exposures at two developmental time periods: before first heartbeat, from gastrulation to cardiac cone stage (early), and from first heartbeat to one day before hatching (late). A frequent sampling regime enabled us to determine immediate PAH uptake, metabolite formation and gene expression changes. In

The osmoregulatory effects of rearing Mozambique tilapia in a tidally changing salinity.

The native distribution of Mozambique tilapia, Oreochromis mossambicus, is characterized by estuarine areas subject to salinity variations between fresh water (FW) and seawater (SW) with tidal frequency. Osmoregulation in the face of changing environmental salinity is largely mediated through the neuroendocrine system and involves the activation of ion uptake and extrusion mechanisms in osmoregulatory tissues. We compared plasma osmolality, plasma prolactin (PRL), pituitary PRL mRNA, and mRNA of branchial ion pumps, transporters, channels, and PRL receptors in tilapia reared in FW, SW, brackis

Gonadotropin releasing hormone in the primitive vertebrate family Myxinidae: reproductive neuroanatomy and evolutionary aspects.

The family Myxinidae embraces all hagfish species, and occupies an evolutionary niche intermediate between ancestral vertebrates and the gnathostomes (jawed vertebrates). Gonadotropin releasing hormone (GnRH) modulates neuroendocrine activity in vertebrates and works in the context of the hypothalamic-pituitary (H-P) axis. The appearance of this neuroendocrine axis marks one of the most crucial developmental achievements in vertebrate evolution, because it enabled further diversification in general growth, metabolism, osmoregulation and reproduction as jawed vertebrates evolved. GnRH studies i

Caffeine consumption attenuates neurochemical modifications in the hippocampus of streptozotocin-induced diabetic rats.

Type 1 diabetes can affect hippocampal function triggering cognitive impairment through unknown mechanisms. Caffeine consumption prevents hippocampal degeneration and memory dysfunction upon different insults and is also known to affect peripheral glucose metabolism. Thus we now characterized glucose transport and the neurochemical profile in the hippocampus of streptozotocin-induced diabetic rats using in vivo(1)H NMR spectroscopy and tested the effect of caffeine consumption thereupon. We found that hippocampal glucose content and transport were unaltered in diabetic rats, irrespective of ca

Centrally administered tuberoinfundibular peptide of 39 residues inhibits arginine vasopressin release in conscious rats.

Tuberoinfundibular peptide of 39 residues (TIP39) is a recently discovered neuropeptide identified on the basis of its ability to activate the PTH2 receptor, and it is thought to be the brain PTH2 receptor's endogenous ligand. The PTH2 receptor is highly expressed in the hypothalamus, suggesting a role in the modulation of neuroendocrinological functions. PTHrP, which also belongs to the PTH-related peptides family, stimulates arginine vasopressin (AVP) release. In the present study, therefore, we investigated the effect of centrally administered TIP39 on AVP release in conscious rats. Intrace

Exploring the role of sugar alcohols in enhancing plant resilience to abiotic stresses.

Plants face different types of biotic and abiotic stresses in their natural environments. Abiotic stresses include heat, cold, drought, flood, salinity, and heavy metal toxicity. They affect plant metabolism by imposing several environmental and nutritional constraints. This leads to altered physiological processes hindering the normal growth and development of plants. As a result of natural adaptations, plants have developed several cope-up mechanisms to overcome these stresses. Plants sense changes in osmolarity, salinity, and temperature to identify stress conditions. Since the discovery of

Synthesis of PbCrO4 nanorods-Ti3C2Tx MXene composites: A sensitive photoelectrochemical sensor for the detection of cysteine in human blood serum.

BACKGROUND: l-cysteine plays an important role in protein synthesis, osmoregulation, detoxification, nervous system function, and antioxidant process. Low levels of cysteine are associated with various diseases like cardiovascular diseases, ischemic stroke, neurological disorders, diabetes, cancer like lung and colorectal cancer, renal dysfunction-linked conditions, and vitiligo. Therefore, it is of great significance to develop a rapid and accurate method for the determination of cysteine concentration. RESULTS: A photoelectrochemical (PEC) biosensor based on PbCrO4-MXene composites is propo

Molecular and physiological responses predict acclimation limits in juvenile brook trout (Salvelinus fontinalis).

Understanding the resilience of ectotherms to high temperatures is essential because of the influence of climate change on aquatic ecosystems. The ability of species to acclimate to high temperatures may determine whether populations can persist in their native ranges. We examined physiological and molecular responses of juvenile brook trout (Salvelinus fontinalis) to six acclimation temperatures (5, 10, 15, 20, 23 and 25°C) that span the thermal distribution of the species to predict acclimation limits. Brook trout exhibited an upregulation of stress-related mRNA transcripts (heat shock prote

The Diadenylate Cyclase CdaA Is Critical for Borrelia turicatae Virulence and Physiology.

Relapsing fever (RF), caused by spirochetes of the genus Borrelia, is a globally distributed, vector-borne disease with high prevalence in developing countries. To date, signaling pathways required for infection and virulence of RF Borrelia spirochetes are unknown. Cyclic di-AMP (c-di-AMP), synthesized by diadenylate cyclases (DACs), is a second messenger predominantly found in Gram-positive organisms that is linked to virulence and essential physiological processes. Although Borrelia is Gram-negative, it encodes one DAC (CdaA), and its importance remains undefined. To investigate the contribu

Identification of elevated urea as a severe, ubiquitous metabolic defect in the brain of patients with Huntington's disease.

Huntington's disease (HD) is a neurodegenerative disorder wherein the aetiological defect is a mutation in the Huntington's gene (HTT), which alters the structure of the huntingtin protein through the lengthening of a polyglutamine tract and initiates a cascade that ultimately leads to dementia and premature death. However, neurodegeneration typically manifests in HD only in middle age, and processes linking the causative mutation to brain disease are poorly understood. Here, our objective was to elucidate further the processes that cause neurodegeneration in HD, by measuring levels of metabol

Sex differences in the association between plasma copeptin and incident type 2 diabetes: the Prevention of Renal and Vascular Endstage Disease (PREVEND) study.

AIMS/HYPOTHESIS: Vasopressin plays a role in osmoregulation, glucose homeostasis and inflammation. Therefore, plasma copeptin, the stable C-terminal portion of the precursor of vasopressin, has strong potential as a biomarker for the cardiometabolic syndrome and diabetes. Previous results were contradictory, which may be explained by differences between men and women in responsiveness of the vasopressin system. The aim of this study was to evaluate the usefulness of copeptin for prediction of future type 2 diabetes in men and women separately. METHODS: From the Prevention of Renal and Vascula

Physiological effects of waterborne lead exposure in spiny dogfish (Squalus acanthias).

To broaden our knowledge about the toxicity of metals in marine elasmobranchs, cannulated spiny dogfish (Squalus acanthias) were exposed to 20 μM and 100 μM lead (Pb). Since we wanted to focus on sub lethal ion-osmoregulatory and respiratory disturbances, arterial blood samples were analysed for pH(a), PaO(2), haematocrit and total CO(2) values at several time points. Plasma was used to determine urea, TMAO, lactate and ion concentrations. After 96 h, Pb concentrations were determined in a number of tissues, such as gill, rectal gland, skin and liver. To further investigate ion and osmoregulat

Implications for glycine receptors and astrocytes in ethanol-induced elevation of dopamine levels in the nucleus accumbens.

Elevated dopamine levels are believed to contribute to the rewarding sensation of ethanol (EtOH), and previous research has shown that strychnine-sensitive glycine receptors in the nucleus accumbens (nAc) are involved in regulating dopamine release and in mediating the reinforcing effects of EtOH. Furthermore, the osmoregulator taurine, which is released from astrocytes treated with EtOH, can act as an endogenous ligand for the glycine receptor, and increase extracellular dopamine levels. The aim of this study was to address if EtOH-induced swelling of astrocytes could contribute to elevated d

Salinity effects on the bioavailability of aqueous metals for the estuarine killifish Fundulus heteroclitus.

Estuarine organisms experience varying salinity conditions on a daily and seasonal basis, and these fluctuations could influence the amount of metal accumulated from the aqueous phase. The present study experimentally assessed the role of salinity (0, 2, 6, 12, and 25 ppt) on the uptake of As, Cd, Cr, inorganic Hg [Hg(II)], and methylmercury (MeHg) into the euryhaline killifish (Fundulus heteroclitus) from the aqueous phase using gamma-emitting radioisotopes. Patterns of metal uptake as a function of salinity varied by metal. Chromium showed no relationship with salinity; Cd, which was most af

Basolateral Mg2+/Na+ exchange regulates apical nonselective cation channel in sheep rumen epithelium via cytosolic Mg2+.

High potassium diets lead to an inverse regulation of sodium and magnesium absorption in ruminants, suggesting some form of cross talk. Previous Ussing chamber experiments have demonstrated a divalent sensitive Na(+) conductance in the apical membrane of ruminal epithelium. Using patch-clamped ruminal epithelial cells, we could observe a divalent sensitive, nonselective cation conductance (NSCC) with K(+) permeability > Cs(+) permeability > Na(+) permeability. Conductance increased and rectification decreased when either Mg(2+) or both Ca(2+) and Mg(2+) were removed from the internal or extern
