surA, an Escherichia coli gene essential for survival in stationary phase.

Mutations in genes not required for exponential growth but essential for survival in stationary phase were isolated in an effort to understand the ability of wild-type Escherichia coli cells to remain viable during prolonged periods of nutritional deprivation. The phenotype of these mutations is referred to as Sur- (survival) and the genes are designated sur. The detailed analysis of one of these mutations is presented here. The mutation (surA1) caused by insertion of a mini-Tn10 element defined a new gene located near 1 min on the E. coli chromosome. It was located directly upstream of pdxA a

Localization of the gene for classic Alport syndrome.

The inheritance of Alport syndrome has been controversial for 30 years because no clear diagnostic criteria were established to define a clinically homogeneous group of patients. In this study, 41 families with "classic" Alport syndrome were identified and studied. All the pedigrees are compatible with X-linked inheritance. A formal genetic study confirmed linkage to probe S21 (DXS17), with a maximum LOD score of 4.72 at a recombination frequency of 0.06.

Maize plastid photogenes: mapping and photoregulation of transcript levels during light-induced development.

Positively photoregulated regions that show increased transcript levels upon illumination of dark-grown seedlings are scattered over approximately 19% of the maize plastid chromosome. Some photogenes, i.e., genes within these regions, are transcribed individually, whereas others that are transcribed as polycistronic mRNAs appear to be functionally organized into operons. Multiple light-induced transcripts are complementary to most photogenes; these mRNAs are not present in equimolar amounts during plastid photomorphogenesis, but particular transcripts predominate at specific stages of developm

Plasmid gene organization: naphthalene/salicylate oxidation.

Genes for naphthalene metabolism are localized on nah7, an 83-kilobase (kb) plasmid, in two gene clusters under salicylate control. Polar mutations formed by insertion of the transposon Tn5 permit detection of the transcription direction and the gene organization within two approximately 10-kb DNA segments separated by a approximately 7-kb regulatory gene region. The gene cluster specifying conversion of naphthalene to salicylate lies near the left initiation of a 25-kb DNA fragment A released by EcoRI; that for the salicylate pathway via catechol meta-fission lies near the right terminus with

Hypertension in chronic glomerulonephritis.

In 74 chronic glomerulonephritis (CGN) and 13 hemodialysis patients (HD) blood volume and hemodynamic indices were examined. Significant correlation was observed between MPA and PV in CGN. High PV in high S-Cr group is considered to be correlated with anemia. Salt restriction reduced MAP without the change in BV in the impaired kidney function group in CGN. Volume depletion in HD shifted TPRI to lower level in the group with significant MAP reduction. The results indicate that sodium rather than water will play an important role in this type of hypertension.

Multiple blood pressure parameters and risk of incident diabetes in adults with elevated blood pressure: a cohort study with non-linear analyses.

AIMS: This study aimed to compare the strengths of association and predictive performance of systolic BP (SBP), diastolic BP (DBP), mean arterial pressure (MAP), pulse pressure (PP), pulse pressure index (PPI), and the SBP/DBP ratio for incident diabetes among Chinese adults with elevated BP. METHODS: This retrospective cohort study included 29,377 adults with elevated BP from a large Chinese health examination database. Cox proportional hazards models were used to estimate hazard ratios (HRs), and restricted cubic spline analyses were applied to evaluate non-linear associations. RESULTS: Du

Atypical cortical representation of peripheral visual space in children with an autism spectrum disorder.

A key feature of early visual cortical regions is that they contain discretely organized retinotopic maps. Titration of these maps must occur through experience, and the fidelity of their spatial tuning will depend on the consistency and accuracy of the eye movement system. Anomalies in fixation patterns and the ballistics of eye movements are well documented in autism spectrum disorder (ASD), with off-center fixations a hallmark of the phenotype. We hypothesized that these atypicalities might affect the development of visuo-spatial maps and specifically that peripheral inputs might receive al

Changes in the tonotopic map of the dorsal cochlear nucleus following induction of cochlear lesions by exposure to intense sound.

Hamsters were exposed to intense tones (10 kHz) at levels and durations sufficient to cause stereocilia lesions. The purpose was to determine how the tonotopic map of the dorsal cochlear nucleus (DCN) readjusts to loss of receptor sensitivity. Neural population thresholds and tonotopic organization was mapped over the surface of the DCN in normal unexposed animals and those showing tone-induced lesions. The results indicate that cochlear lesions characterized mainly by loss of stereocilia in a restricted portion of the organ of Corti cause changes in a corresponding region of the tonotopic map

Enhanced Transformation Efficiency in Fusobacterium nucleatum by Bypassing Restriction-Modification Systems: A Methylation-Guided Protocol.

The genetic intractability of Fusobacterium nucleatum has slowed progress in defining mechanisms of fusobacterial pathogenesis. Transformation is inefficient, and broadly applicable genetic tools are still limited. A major obstacle is the presence of endogenous restriction-modification (RM) systems that cleave incoming DNA lacking appropriate methylation. This chapter presents a concise, reproducible protocol to increase transformation efficiency in F. nucleatum by systematically bypassing RM barriers. The workflow combines methylome mapping by single-molecule real-time (SMRT) sequencing, expr

YAC and cosmid contigs spanning the Batten disease (CLN3) region at 16p12.1-p11.2.

A yeast artificial chromosome (YAC) contig has been constructed in 16p12.1-p11.2 that encompasses three loci (D16S288, D16S299, and D16S298) closely linked to the gene causing Batten disease or juvenile-onset neuronal ceroid lipofuscinosis (CLN3). The physical map has been ordered using 42 sequence tagged sites. Four genes, interleukin-4 receptor (IL4R), phenol-preferring phenol sulfotransferase (STP), monoamine-preferring phenol sulfotransferase (STM), and sialophorin (SPN), have been mapped to the YAC contig. A partial genomic restriction map has been constructed to confirm the order and dis

The pseudoautosomal regions of the human sex chromosomes.

In human females, both X chromosomes are equivalent in size and genetic content, and pairing and recombination can theoretically occur anywhere along their entire length. In human males, however, only small regions of sequence identity exist between the sex chromosomes. Recombination and genetic exchange is restricted to these regions of identity, which cover 2.6 and 0.4 Mbp, respectively, and are located at the tips of the short and the long arm of the X and Y chromosome. The unique biology of these regions has attracted considerable interest, and complete long-range restriction maps as well

The LCK gene is involved in the t(1;7)(p34;q34) in the T-cell acute lymphoblastic leukemia derived cell line, HSB-2.

HSB-2 is a cell line derived from a patient who had T-cell acute lymphoblastic leukemia (T-cell ALL) with a t(1;7)(p34;q34). We used a genomic probe from the T-cell receptor beta (TCR beta) locus (7q34) to identify DNA rearrangements in HSB-2. Two rearranged BglII DNA fragments were cloned, and one of these clones was shown to contain the translocation breakpoint on the derivative chromosome I [der(I)]. We used a probe derived from this clone to isolate an unrearranged phage clone encompassing the breakpoint at Ip34. The restriction map of this clone was compared to the published maps of known

Improved topical delivery of curcumin by mussel adhesive protein functionalized ethosomes for effective psoriasis treatment.

Psoriasis is a prevalent inflammatory skin disorder exhibiting a rapidly increasing incidence. Curcumin (Cur) serves as an effective therapeutic agent for psoriasis and is commonly administered through the cutaneous route. Nevertheless, the poor skin permeability and retention of Cur restrict its therapeutic efficacy against psoriasis. In this study, we fabricated mussel adhesion protein (MAP)-modified Cur-loaded ethosomes (Cur-MAP-Es) aimed at enhancing both the permeation and retention of Cur within the skin for improved topical treatment of psoriasis. The average particle size of Cur-MAP-Es

Male-associated polypeptide (MAP) expression in different compartments of the reproductive system of the mussel Mytilus galloprovincialis: immunocytochemical and western blot study.

Mytilus mussels are characterized by annually repeated reproduction which is associated with subsequent growth, morphogenesis, breakdown and redevelopment of the gonad and reproductive tract into mantle mesenchyme. We present a description of the expression of the male-associated polypeptide (MAP; see Mikhailov et al. 1995) in different compartments of the male reproductive system as well as in mantle gonad-supporting tissue. MAP is expressed in both gonad and mantle structures in dynamic patterns that show a substantial overlap in terms of dependence on the stage of gonad development/involuti

Chromosome region 8p11-p21: refined mapping and molecular alterations in breast cancer.

Several genes, most of them unknown, of the short arm of chromosome 8 are involved in malignant diseases. Numerous studies have implicated a portion of the 8p11-p21 region as the location of one or more tumor suppressor genes involved in a variety of human cancers, including breast cancer. We and others have reported linkage analyses suggesting the presence of a putative breast cancer susceptibility gene. Furthermore, several oncogenes of the 8p11-p12 region are involved in reciprocal translocations in myeloproliferative and myelodysplastic disorders and in amplification in breast cancer. To f

Genomic cloning, physical mapping, and expression of human type 2 cystatin genes.

Humans carry one gene encoding cystatin C and six to eight genes with homology to an S-like cystatin hybridization probe. However, the precise composition and organization of the cystatin gene family remains to be established. Further, the pattern of tissue-specific expression has not been fully defined. We have previously shown that the type 2 cystatin genes are clustered together in a ca. 270 kb region (the CST locus). To determine the structure of this region, we have sought to clone the entire CST locus. Our approach has been to isolate cosmid and lambda genomic clones carrying cystatin ge

The genomic structure of Brucella strains isolated from marine mammals gives clues to evolutionary history within the genus.

The genomic structure and the restriction maps were studied in 24 Brucella strains isolated from marine mammals. From SpeI restriction profiles, the strains could be ascribed to three clonal groups, each corresponding to a specific host. Cross contamination between exclusively terrestrial and exclusively marine hosts is unlikely suggesting the divergence of the different species of the genus Brucella which may have taken place 60 million years ago, concomitant with the radiation of their mammalian hosts (Artiodactyla) from other mammalian orders.

Diffusion-Weighted Zonal Oblique Multislice-EPI Enhances the Detection of Small Lesions with Diffusion Restriction in the Brain Stem and Hippocampus: A Clinical Report of Selected Cases.

Diffusion restriction is the morphologic hallmark of acute ischemic infarcts and excitotoxic brain injury in various cerebral pathologies. Diffusion restriction is visible as hyperintensity on DWI and as hypointensity on ADC maps. Due to the vicinity of multiple anatomic structures in the brain stem and hippocampus, very small lesions with diffusion restriction may result in severe clinical symptomatology, but these small lesions easily go undetected on standard cerebral DWI due to insufficient spatial resolution, T2* blurring, and image artifacts caused by susceptibility-related image distort

Dissecting the Immune Response Elicited by WbALT-2, ALT MAP in Clinical Populations and Mouse Model: A Prophylactic Measure Against Lymphatic Filariasis.

BACKGROUND: Abundant Larval Transcript (ALT) is one of the major groups of immune-dominant proteins produced by filarial worms during their larval stage. The major B-cell and T-cell epitopic domains of the ALT-2 antigen were mapped to develop a multiple antigenic peptide (MAP) prophylactic antigen against lymphatic filariasis. METHODS AND RESULTS: ALT MAP was constructed by solid phase peptide synthesis. The reactivity of whole ALT protein and ALT MAP against clinical sera described a high reactivity of endemic normal sera against ALT MAP compared to WbALT-2 protein. The antibody isotype patt
