Surgical Anatomy of the Third Ventricle Floor: Implications for Endoscopic Third Ventriculostomy.

Neuroendoscopy has made a breakthrough in the field of neurosurgery. Since its first application at the start of the 20th century, it has evolved enormously and become more specialized. Endoscopic Third Ventriculostomy (ETV) is a type of ventriculoscopies where the floor of the third ventricle is opened to divert the CSF from the third ventricle to the subarachnoid space. Nowadays, ETV is the gold-standard treatment for selected cases of hydrocephalus. Its success depends on, among others, the anatomy of the floor of third ventricle. From anterior to posterior, structures related to the floor

An In Vitro Study on Prestin Analog Gene in the Bullfrog Hearing Organs.

The prestin-based active process in the mammalian outer hair cells (OHCs) is believed to play a crucial role in auditory signal amplification in the cochlea. Prestin belongs to an anion transporter family (SLC26A). It is densely expressed in the OHC lateral plasma membrane and functions as a voltage-dependent motor protein. Analog genes can be found in the genome of nonmammalian species, but their functions in hearing are poorly understood. In the present study, we used the gerbil prestin sequence as a template and identified an analog gene in the bullfrog genome. We expressed the gene in a st

Aging Effects on Behavioural Estimates of Suppression with Short Suppressors.

Auditory two-tone suppression is a nearly instantaneous reduction in the response of the basilar membrane to a tone or noise when a second tone or noise is presented simultaneously. Previous behavioural studies provide conflicting evidence on whether suppression changes with increasing age, and aging effects may depend on whether a suppressor above (high-side) or below (low-side) the signal frequency is used. Most previous studies have measured suppression using stimuli long enough to elicit the medial olivocochlear reflex (MOCR), a sound-elicited reflex that reduces cochlear amplification or

Coherent reflection without traveling waves: on the origin of long-latency otoacoustic emissions in lizards.

Lizard ears produce otoacoustic emissions with characteristics often strikingly reminiscent of those measured in mammals. The similarity of their emissions is surprising, given that lizards and mammals manifest major differences in aspects of inner ear morphology and function believed to be relevant to emission generation. For example, lizards such as the gecko evidently lack traveling waves along their basilar membrane. Despite the absence of traveling waves, the phase-gradient delays of gecko stimulus-frequency otoacoustic emissions (SFOAEs) are comparable to those measured in many mammals.

Efferent-induced change in human cochlear compression and its influence on masking of tones.

Several lines of evidence suggest that medial olivocochlear (MOC) efferent neurons modify cochlear output to improve signal detection in noise. In animal models, stimulation of MOC efferents reduces the amount of compression in basilar membrane (BM) growth functions. Linearization of BM growth functions may assist in extending the neural response to the signal above that of noise, leading to a decrease in masking. In order to test this hypothesis, effects of MOC efferent neurons on BM compression were studied indirectly in humans by examining the effects of contralateral noise on distortion-pr

Novel protein transduction method by using 11R: an effective new drug delivery system for the treatment of cerebrovascular diseases.

BACKGROUND AND PURPOSE: A motif of 11 consecutive arginines (11R) is reported to be one of the most effective protein transduction domains for introducing proteins into the cell membrane. We therefore examined the transduction efficiency of 11R in cerebral arteries. METHODS: Basilar arteries (BAs) obtained from rats were incubated with either 11R-enhanced green fluorescent protein (11R-EGFP) or EGFP without 11R. After incubation, expression of 11R-EGFP or EGFP in BA serial sections was observed by fluorescence microscope. In an additional in vivo experiment, 11R-EGFP or EGFP was injected into

Latencies of the 2f1-f2 distortion product otoacoustic emissions measured using a phase-gradient method in young people, in the elderly and in people exposed to noise.

Distortion product otoacoustic emissions (DPOAE) are generally accepted as a good tool for investigating the micromechanics of the cochlea. The 2f1-f2 DPOAE latencies provide significant data regarding travelling waves. In this study the 2f1-f2 DPOAE latencies were measured using a phase-gradient method, with a swept-f2 procedure, for three groups: Group I comprised 60 healthy young people, aged 17-32 years; Group II comprised elderly patients, aged 56-72 years, with presbyacusis; and Group III consisted of 74 miners, aged 19-35 years, who had been exposed to noise for 2-15 years. Gene expression analysis of oxidative stress-related genes in the apical, middle, and basal turns of the cochlea.

It can be observed from aminoglycoside-induced hair cell damage that the cochlea basal turn is more susceptible to trauma than the apex. Drug-induced hearing loss is closely related to oxidative damage. The basilar membrane directly exposed to these ototoxic drugs exhibits differences in damage, indicating that there is an inherent difference in the sensitivity to oxidative damage from the apex to the base of the cochlea. It has been reported that the morphology and characteristics of the cochlea vary from the apex to the base. Therefore, we investigated oxidative stress-related gene expressio

Chronic Lead Exposure Results in Auditory Deficits and Disruption of Hair Cells in Postweaning Rats.

OBJECTIVE: The effects of lead exposure on cognitive function have been studied intensively over the past decade, but less attention has focused on its impact on auditory function. This study is aimed at investigating the effect of lead on the cochlea and the molecular mechanisms responsible for its actions. METHODS: 0.2% lead acetate was administered to rats in drinking water for 30, 60, and 90 days. Brainstem auditory evoked responses (ABR) were recorded, and morphological changes in the hair cells were observed. We also measured glutathione (GSH) and malondialdehyde (MDA) concentrations an

Efferent-mediated reduction in cochlear gain does not alter tuning estimates from stimulus-frequency otoacoustic emission group delays.

The existence of efferent feedback from cortical and subcortical brain centers to the hair cells of the cochlea has been recognized for many years, but the role that efferent neurons play in hearing is not completely known. Stimulation of medial olivocochlear (MOC) efferent neurons suppresses sound-evoked basilar membrane responses and changes the tuning of single auditory nerve fibers in animal models. Both of these effects are linked to a MOC-induced reduction in the gain of the cochlear amplification provided by outer hair cells. To non-invasively examine the link between cochlear suppressi

Temporal profile of potassium channel dysfunction in cerebrovascular smooth muscle after experimental subarachnoid haemorrhage.

The pathogenesis of cerebral vasospasm after subarachnoid haemorrhage (SAH) involves sustained contraction of arterial smooth muscle cells that is maximal 6-8 days after SAH. We reported that function of voltage-gated K+ (KV) channels was significantly decreased during vasospasm 7 days after SAH in dogs. Since arterial constriction is regulated by membrane potential that in turn is determined predominately by K+ conductance, the compromised K+ channel dysfunction may cause vasospasm. Additional support for this hypothesis would be demonstration that K+ channel dysfunction is temporally coincid

Relationship between cytosolic Ca2+ level and contractile tension in canine basilar artery of chronic vasospasm.

In order to study the role of the Ca2+/calmodulin/myosin light chain kinase system in the development of chronic vasospasm caused by subarachnoid hemorrhage, the cytosolic Ca2+ level ([Ca2+]i), measured with fura-2, a fluorescent Ca2+ indicator, and contractile tension were measured simultaneously, and their quantitative correlation was examined in basilar arterial tissue obtained from the canine "two-hemorrhage" model. Sixteen adult mongrel dogs were divided into two groups, control (n = 8) and vasospasm (n = 8), and were killed 7 days after the first experimental subarachnoid hemorrhage. In

Conflicting electrophysiological and anatomical data from drug-impaired guinea pig cochleas.

In guinea pigs that had been treated with very large doses of the aminoglycoside amikacin (14 x 450 mg/kg/day, i.m.) clear, short-latency responses to various click stimuli could be recorded at the round window. When the same cochleas were examined histologically, no outer or inner hair cells could be found along the entire length of the basilar membrane, save for a very few outer hair cells remaining at the apex. The response patterns resembled that of the compound action potential, and various characteristics suggest that they were of neural origin. Vestibular function, investigated by elect

Effect of stimulus level on speech masking by multitalker noise in a cochlear model.

Previous research shows that intelligibility of speech in noise (SIN) with a constant speech-to-noise ratio (SNR) changes in dependence on overall stimulus level. It has been hypothesized that these changes were associated with nonlinear responses of the cochlear basilar membrane, however, the exact role of auditory periphery in this behavior is not clear. We explore this phenomenon using a nonlinear cochlear model with adjustable gain to examine how multi-talker (babble) noise affects the speech signal at the output of the auditory periphery. The amount of distortion at various intensities ca

Cobalt-induced apoptosis of cochlear organotypic cultures and HEI-OC1 cells is mediated by Dicer.

Cobalt is widely used in the medical industry, mainly including cobalt alloy joint implants and cobalt-chromium porcelain crowns. However, unexplained ototoxicity and neurotoxicity often occur in the clinical use of cobalt agents at present, which limits the development of the cobalt industry. In this study, based on the clinical problem of cobalt ototoxicity, we first conducted an extensive search and collation of related theories, and on this basis, prepared an HEI-OC1 cell model and basilar membrane organotypic cultures after cobalt treatment. We used immunofluorescence staining, western bl

Simple and efficient rat model for studying delayed cerebral ischemia after subarachnoid hemorrhage.

BACKGROUND: Delayed cerebral ischemia (DCI) is a late phase of consequences of subarachnoid hemorrhage (SAH) that causes poor outcome and has become the focus of current research. The aim of this study was to characterize an experimental SAH technique for studying DCI after SAH. NEW METHOD: A double injection SAH rat model with a tiny incision was introduced. At 7 days post-SAH induction, the diameter and luminal cross-sectional area (CSA) of the basilar artery (BA) were measured. In vivo fluorescence microscopy and magnetic resonance perfusion-weighted imaging (MRPWI) were used to evaluate t

Inner ear contribution to bone conduction hearing in the human.

Bone conduction (BC) hearing relies on sound vibration transmission in the skull bone. Several clinical findings indicate that in the human, the skull vibration of the inner ear dominates the response for BC sound. Two phenomena transform the vibrations of the skull surrounding the inner ear to an excitation of the basilar membrane, (1) inertia of the inner ear fluid and (2) compression and expansion of the inner ear space. The relative importance of these two contributors were investigated using an impedance lumped element model. By dividing the motion of the inner ear boundary in common and

Fully implantable hearing device with transducer on the round window as a treatment of mixed hearing loss.

OBJECTIVE: To demonstrate the safety and efficacy of the Otologics Carina Middle Ear Transducer for treatment of mixed hearing loss through a case report. METHODS: A Carina fully implantable device with the MET transducer for conductive applications was implanted in a 48-year-old woman suffering from right mixed hearing loss (mean PTA loss: 80 dB). A facial recess approach was used to access the middle ear. Sclerotic tissue obliterated the stapes footplate so the approach selected was to place the transducer directly on the round window. The mounting bracket was placed on the mastoid and the
