We tested correlations between anti-Helicobacter pylori IgG and IgA levels and the urease test, anti-CagA protein antibody, degree of gastritis, and age. In total, 509 children (0-15 years) were enrolled. Subjects were stratified as 0-4 years (n = 132), 5-9 years (n = 274), and 10-15 years (n = 103) and subjected to the urease test, histopathology, ELISA, and western blot using whole-cell lysates of H. pylori strain 51. The positivity rate in the urease test (P = 0.003), the degree of chronic gastritis (P = 0.021), and H. pylori infiltration (P < 0.001) increased with age. The median titer for

Helicobacter pylori is a spiral, slow growing gram-negative microaerophilic bacterium. It has been shown to be the etiological agent of gastroduodenal diseases, such as chronic gastritis, gastric and duodenal ulcers, and gastric cancer. To address the influence of oxidative stress and its underlying mechanisms, we have compared proliferation, urease activity and protein expression profile of H. pylori incubated under normal microaerophilic (5% O2) and aerobic stress (20% O2) conditions. Oxidative-stress cells displayed coccoid morphology and time-dependent decrease in proliferation. The urease

Helicobacter pylori (formerly Campylobacter pylori) has been recently described as a gastritis-associated bacterium. We examined endoscopic biopsies of 100 patients with dyspepsia and found H. pylori in the gastric antrum of 34 (34%) by either culture, urease tests and/or histology. Thirty-one out of 41 patients (75.6%) confirmed to have chronic active gastritis histologically had H. pylori in their gastric antrum compared to 3 out of 59 patients (5.1%) with dyspepsia but normal histology (p less than 0.01). Histological examination, using gram stain and the Warthin-Starry Silver stain, detect

BACKGROUND: In cirrhotics, Helicobacter pylori infection is the major cause of peptic lesions, which are an important cause of upper intestinal haemorrhage in these patients. However, some diagnostic methods are not accurate for H. pylori detection in cirrhotics.
AIMS: The study assessed the accuracy of different diagnostic methods for H. pylori detection in cirrhotics with and without gastroduodenal lesions.
METHODS: The study population comprised of 53 cirrhotics. All patients underwent upper endoscopy: three biopsies were taken in the antrum and three in the gastric body. Four biopsies we

UNLABELLED: Helicobacter pylori is a gram negative curved bacillus recognized as etiologic agent of chronic gastritis and an important factor in the development of duodenal ulcer and gastric cancer. The purpose of this work is to evaluate three microbiologic methods for diagnosing H. pylori infection. We studied 375 samples from 218 patients, who consulted at the Department of Gastroenterology of the Hospital Vargas and were candidates for endoscopy. Samples of gastric mucous tissue were taken at level of the antrum and each sample was stained with Gram, tested for urease and cultured.

An in vitro stimulation protocol has been established which allows production of IgG-secreting murine hybridomas. This procedure has been examined using jack bean urease and human luteinizing hormone as antigens. Parameters which have been optimized include selection of media and serum supplements, thymocyte-conditioned media, antigen dosage, length of stimulation and the effect of medium changes during stimulation and additions of polyclonal mitogen. Murine spleen cells (1 X 10(8) in 10 ml) were incubated with varying doses of jack bean urease and human luteinizing hormone in a six-well plate

The continuing emergence of drug-resistant Helicobacter pylori (HP) drives the ongoing need for the development of new and effective anti-HP drugs. Urease inhibitor has now gained strong interest as an alternative approach for HP infections. 3-Chlorophenyl-3-hydroxypropionylhydroxamic acid (CPH), a novel urease inhibitor identified in our group, shows impressive potency, which was optically separated for a further exploration. Here, we report in vitro/in vivo pharmacological evaluation of (±)-CPHs and the enantiomers. The raceme and the individual enantiomers significantly suppress gastritis a

A variety of methods exist for determining gastric colonization with Helicobacter pylori, which has been implicated in the development of peptic ulcer disease. The goal of this study was to evaluate four of the current methods available in a clinical surgical practice setting through a prospective evaluation of 40 consecutive patients undergoing upper diagnostic endoscopy. All patients underwent six antral gastric biopsies for use with the following detection methods: histologic demonstration of organisms (hematoxylin and eosin stain), direct detection of urease activity (Remel Selective Rapid

Helicobacter pylori's powerful urease enzyme is essential for colonisation and adaptation to the acid milieu of the stomach. Eradication of infection with "standard triple therapy" abolishes the chronic immunological and inflammatory responses to H. pylori and, thus, cures gastritis and peptic ulcer. In vitro, proton pump inhibitors (PPI) are active against H. pylori with minimum inhibitory concentrations that compare favourably with bismuth salts. PPIs are also potent urease inhibitors, but because PPIs are also active against urease-negative mutant Helicobacter spp., it is unlikely that urea

A novel series of thiadiazole-based Schiff base derivatives (1-16) was synthesized via an efficient multi-step route and characterized using 1H NMR, 13C NMR, and HREI-MS. These compounds were evaluated for their anti-Alzheimer's and anti-urease potential. Among them, compounds 8 and 9 exhibited the most potent inhibition with IC₅₀ values of 6.60 ± 0.40 and 8.10 ± 0.10 μM for AChE, 7.40 ± 0.20 and 8.80 ± 0.40 μM for BChE, and 9.75 ± 1.60 and 10.15 ± 0.50 μM for urease, respectively. Donepezil and thiourea were used as standard inhibitors for comparative evaluation. Molecular docking and pharmac

We have evaluated a 13C-urea breath test for the diagnosis of Helicobacter pylori infection. The 13C-test was analyzed with isotope-selective nondispersive infrared spectrometry and compared with a 14C-urea breath test and the urease test in gastric mucosal biopsies. 46 patients were analyzed with breath tests; 23 patients were negative and 22 patients were positive with both methods. One patient was positive with the 14C-method and negative with the 13C-urea breath test. 61 patients were analyzed with the 13C-urea breath test and the urease test. 30 patients were negative and 30 patients were

Intestinal motility is a function of the enteric nervous system involving secretion of the excitatory neurotransmitter acetylcholine (ACh). Although gut commensal bacteria are key regulators of intestinal physiology, the molecular mechanisms underlying microbial influence on intestinal peristalsis and constipation remain unclear. Here we report a link between microbial nitrogen metabolism and intestinal motility regulation via ammonia production. We observed compensatory elevation of intestinal ammonia levels and urease activity in mouse models of intestinal dysmotility, induced by ACh deficie

The present study aimed to evaluate in vitro cross-strain inhibitory effects of IgY polyclonal antibody on both growth and urease enzyme of four local strains of H. pylori. Leghorn chickens were immunized with whole cells of four different strains of H. pylori, separately. Rising of specific IgY was detected by ELISA. The IgY purified using polyethylene glycol method and the purity was evaluated by SDS-PAGE and Western blotting. Each strain was treated with its own-specific and also other strain-specific IgYs. The strain-specific IgY could inhibit the growth of specific strains by 49-72% and a

The role of the periplasmic alpha-carbonic anhydrase (alpha-CA) (HP1186) in acid acclimation of Helicobacter pylori was investigated. Urease and urea influx through UreI have been shown to be essential for gastric colonization and for acid survival in vitro. Intrabacterial urease generation of NH3 has a major role in regulation of periplasmic pH and inner membrane potential under acidic conditions, allowing adequate bioenergetics for survival and growth. Since alpha-CA catalyzes the conversion of CO2 to HCO3-, the role of CO2 in periplasmic buffering was studied using an alpha-CA deletion muta

The NikR protein is a nickel-dependent regulatory protein which is a member of the ribbon-helix-helix family of transcriptional regulators. The gastric pathogen Helicobacter pylori expresses a NikR ortholog, which was previously shown to mediate regulation of metal metabolism and urease expression, but the mechanism governing the diverse regulatory effects had not been described until now. In this study it is demonstrated that NikR can regulate H. pylori nickel metabolism by directly controlling transcriptional repression of NixA-mediated nickel uptake and transcriptional induction of urease e
