Glycosphingolipids are located in cell membranes and the brain is especially enriched. We speculated that the subcellular location of glycosphingolipids depends on their fatty acid chain length because their sugar residues are constant, whereas fatty acid chain length can vary within the same molecule. To test this hypothesis we analysed the glycosphingolipid sulfatide, which is highly abundant in myelin and has mostly long fatty acids. We used a negative ion electrospray tandem mass spectrometry precursor ion scan to analyse the molecular species of sulfatide in cultured astrocytes and a mous

Glycosphingolipids from brain, liver, and spleen of a patient with mucopolysaccharidosis type III B were quantitatively analyzed. Neutral glycosphingolipids containing glucosylceramide, lactosylceramide, globotriaosylceramide, globotetraosylceramide, and gangliotriaosylceramide were increased in the brain, while the contents of galactosylceramide and galactosylceramide I3-sulfate were decreased. The total ganglioside levels were low in the grey matter (522 micrograms N-acetylneuraminic acid/g) and high in the white matter (342 micrograms N-acetylneuraminic acid/g), when compared with the norma

Transport of biologically active molecules across tight epithelial barriers is a major challenge preventing therapeutic peptides from oral drug delivery. Here, we identify a set of synthetic glycosphingolipids that harness the endogenous process of intracellular lipid-sorting to enable mucosal absorption of the incretin hormone GLP-1. Peptide cargoes covalently fused to glycosphingolipids with ceramide domains containing C6:0 or smaller fatty acids were transported with 20-100-fold greater efficiency across epithelial barriers in vitro and in vivo. This was explained by structure-function of t

Detailed lipid analyses of 1 liver and 4 endoneurial samples obtained from patients with hereditary motor and sensory neuropathy, type III (Déjerine-Sottas disease) were compared with controls. In parallel with the severe decrease in myelin, there was a substantial alteration of lipid composition including: (1) a decrease in endoneurial content of free cholesterol, glycosphingolipid and phospholipid; (2) a selective reduction of endoneurial phosphatidylserine; (3) a high proportion of 18:0 in endoneurial triacylglycerol and phospholipids; and (4) a striking diminution of long chain fatty acids

Several neuropathies, such as Guillain-Barré syndrome and myelin-associated glycoprotein neuropathy (MAG), are caused by antibodies targeting glycosphingolipids. Several studies have indicated that MAG arises from pathogenic IgM autoantibodies targeting sulfoglucuronyl (HNK-1)-containing glycosphingolipids. The exact mechanism by which IgM neuropathy occurs has not been fully elucidated. Furthermore, no appropriate diagnostic tools are available for MAG using sulfoglucuronyl-containing glycosphingolipids. To address these limitations, we describe here a synthetic strategy that makes it possibl

Glycosphingolipids are cell-type-specific components of the outer leaflet of mammalian plasma membranes. Gangliosides, sialic acid-containing glycosphingolipids, are especially enriched on neuronal surfaces. As amphi-philic molecules, they comprise a hydrophilic oligosaccharide chain attached to a hydrophobic membrane anchor, ceramide. Whereas glycosphingolipid formation is catalyzed by membrane-bound enzymes along the secretory pathway, degradation takes place at the surface of intralysosomal vesicles of late endosomes and lysosomes catalyzed in a stepwise fashion by soluble hydrolases and as

Spatially resolved profiling of lipid carbon-carbon double bond (C═C) isomers, especially for poorly ionizable glycosphingolipids, remains a significant challenge in mass spectrometry imaging (MSI). Herein, we report a novel MSI method that leverages a diselenide-functionalized covalent organic framework (COF-Se2) by integrating its heterogeneous catalysis and selective surface capture capabilities for on-tissue lipid capture-epoxidation, enabling the differentiation of C═C position isomers with significantly enhanced coverage for glycosphingolipids. The structures of neutral and acidic glycosphingolipids from both normal colorectal epithelial cells and colorectal cancer cells, which were highly purified with the epithelial cell marker CD326, have been analyzed. The analysis was performed on samples from 16 patients. The carbohydrate moieties from glycosphingolipids were released by endoglycoceramidase II, labeled by pyridylamination, and identified using two-dimensional mapping and mass spectrometry. The structures from normal colorectal epithelial cells are characterized by dominant expression of neutral type-1 chain oligosaccharides. Th

Glycosphingolipids from the liver, kidney, and spleen of a patient with type 1 II3-N-acetylneuraminosylgangliotetraosylceramide (GM1)-gangliosidosis were quantitatively analyzed. It was noted that large amounts of unusual glycosphingolipids other than GM1 ganglioside or gangliotetrasylceramide accumulated in the liver of the patient. Particularly, the prominent accumulation of III3-alpha-fucosylneolactotetraosylceramide, galactosylceramide I3-sulfate and cholesterol sulfate was observed in addition to a small but significant increase of galabiosylceramide and neolacto-or lactotetraosylceramide

Novel neutral glycosphingolipids (NGSLs) containing Gal-alpha1-->6Gal, previously found in the Zygomycetes species Mucor hiemalis, were synthesized. The structures of these compounds are different from those of other fungal GSLs, and they are expected to be involved in host-parasite interactions. A key step in their synthesis is direct 1,2-cis alpha-selective galactosylation of 4,6-diol tri- and tetrasaccharide acceptors with a galactosyl donor in the presence of N-iodosuccinimide (NIS)/trifluoromethanesulfonic acid (TfOH). The fully protected glycosides were deprotected to give the two target

Previous studies have shown that the glycoprotein oligosaccharides synthesized by adult Schistosoma mansoni, the organism responsible for human schistosomiasis, are unusual in that they contain terminal beta-GalNAc residues and lack sialic acid. These observations and other studies indicating that schistosome glycoproteins and glycolipids are antigenic in infected animals led us to investigate the structures of the glycosphingolipids synthesized by these organisms and to determine whether they are structurally related to those synthesized by their vertebrate hosts. For our studies, adult schis

Gangliosides are a class of sialic acid-containing glycosphingolipids involved in a wide range of biological processes. The terminal sialic acid of gangliosides can be O-acetylated at C7 and/or C9 hydroxyl, contributing to ganglioside structural complexity and function. It has been difficult to obtain panels of structurally well-defined O-acetylated gangliosides for binding and functional studies. We describe here a chemoenzymatic strategy that can provide, for the first time, 7-O-, 9-O-, and 7,9-di-O-acetylated GD3 gangliosides. It is based on the chemical assembly of a common tetrasaccharide

The glycosphingolipids of human thyroid were isolated and characterized by gas-liquid chromatography and sequential enzymic hydrolysis. The major purified components were identified as glucosyl- and galactosyl-ceramides, lactosyl- and galabiosylceramides, globotriaosyl- and globotetraosylceramides. The long-chain base analyses showed a high proportion of phytosphingosine in glycosylceramide and galabiosylceramide. Fatty acids in 22:0, 24:0, 24:1 prevailed, especially in the cerebroside fraction, with a significant content of alpha-hydroxylated species in galactosylceramide. Female thyroid had

The purpose of this study was to analyze the profile of glycosphingolipids (GSLs) in periradicular lesions refractory to endodontic treatment. Sixteen periapical lesions were removed surgically from patients (experimental group) and compared with 10 samples of periodontal ligament removed from extracted intact third molars (control group). After the GSLs extraction and purification procedures were performed the neutral and acidic GSL fractions were analyzed by high-performance thin-layer chromatography and quantified by densitometry. Data reported herein show that: (i) tissues in the experimen

CONTEXT: Complex glycosphingolipids, in majority the ganglioside GM3, surround the insulin receptor in a special membrane compartment (raft) and modulate signaling through this receptor. Increased levels of GM3 in rafts impair insulin signaling, resulting in insulin resistance. Gaucher disease is a lysosomal storage disorder in which impaired breakdown of glucosylceramide leads to its accumulation in macrophages. Secondary to this defect, GM3 concentrations, for which glucosylceramide is the precursor, in plasma and several cell types are elevated.

Ceramide, sphingomyelin, and glycosphingolipids (both neutral and acidic) are characterized by the presence in the lipid moiety of an aliphatic base known as sphingosine. Altogether, they are called sphingolipids and are particularly abundant in neuronal plasma membranes, where, via interactions with the other membrane lipids and membrane proteins, they play a specific role in modulating the cell signaling processes. The metabolic pathways determining the plasma membrane sphingolipid composition are thus the key point for functional changes of the cell properties. Unnatural changes of the neur

Glycosphingolipids extracted from Echinococcus multilocularis metacestodes were tested against sera from patients with alveolar hydatid disease (AHD). The tests were performed first by ELISA then by immunostaining on thin layer chromatography. A binding between the parasite glycolipids and antibodies of AHD human sera was observed by ELISA. The glycolipids related to this binding were specifically parasite neutral glycosphingolipids. No reaction was observed with parasite acid glycolipids nor with neutral glycolipid extracts from human or mongolian gerbil erythrocytes. Comparison between absor

The glycosphingolipid neurosporaside (α-D-Glcp-(1 → 2)-β-D-Galp-(1 → 6)-β-D-Galp-(1 → 6)-β-D-Galp-(1 →)-Cer) occurs in Neurospora crassa. We attempted to synthesize neurosporaside by block synthesis (route A) and linear synthesis (route B). Oligosaccharide derivatives were synthesized using trimethylsilyltrifluoromethanesulfonate and N-iodosuccinimide/trifluoromethane sulfonic acid as promoters. The target tetrasaccharide could not be attained via route A, but route B showed potential: glycosidic bonds (β-D-Galp-(1 → 6)-β-D-Galp-(1 → 6)-β-D-Galp) were formed stereoselectively, leading to the s

Gaucher disease is an inherited autosomal recessive disease caused by mutations of acid β-glucosidase, a lysosomal hydrolase specific for degradation of glucosylceramide and glucosylsphingosine in the glycosphingolipid metabolic pathway. Clinically, Gaucher disease is classified into three types: type 1 is a visceral disease, whereas types 2 and 3 are acute and chronic neuronopathic variants, respectively. In types 2 and 3, the CNS pathology displays neuronal inclusions and neuron death. The underlying mechanism(s) by which the glycosphingolipid storage leads to this pathology is not fully und

Sandhoff disease, one of several related lysosomal storage disorders, results from the build up of N-acetyl-containing glycosphingolipids in the brain and is caused by mutations in the genes encoding the hexosaminidase beta-subunit. Affected individuals undergo progressive neurodegeneration in response to the glycosphingolipid storage. (1)H magnetic resonance spectra of perchloric acid extracts of Sandhoff mouse brain exhibited several resonances ca 2.07 ppm that were not present in the corresponding spectra from extracts of wild-type mouse brain.
