Geographic isolates of atypical Parkinsonism and tauopathy in the tropics: possible synergy of neurotoxins.

Over the last 60 years an abnormally high prevalence of atypical Parkinsonism has been reported in 5 different geographic isolates. It was first described on Guam, later in New Guinea and in the Kii peninsula, on Guadeloupe, and in New Caledonia. We investigated the phenotype of atypical Parkinsonism in three of these foci and observed several similarities with dementia in most and amyotrophic lateral sclerosis in some. This disappearance of this disease in two places--Guam and New Guinea--suggested an environmental origin which has not been clarified before the disease ended. The exposure to

Cooperative effects of natural acetylenic acetogenins fromPorcelia macrocarpaon lipid packing and mechanical stability of Langmuir monolayers as protozoan membrane models.

This study investigates the synergistic interfacial behavior of two natural bioactive acetylenic acetogenins, (2S,3R,4R)-3 hydroxy-4-methyl-2-(n-eicos-11'-yn-19'-enyl)butanolide (1) and (2S,3R,4R)-3-hydroxy-4 methyl-2-(n-eicos-11'-ynyl)butanolide (2), obtained from Brazilian plant Porcelia macrocarpa (Annonaceae) and their interactions with membrane models using Langmuir films. Previous biological studies demonstrated that mixtures of 1 and 2 exhibit enhanced antiparasitic activity compared to the isolated compounds, although the physicochemical basis for this synergism remained unclear. Macroevolutionary dynamics and predictors of tropical rain forest plant diversity.

Tropical rain forest is the most diverse terrestrial biome, yet the drivers of its diversification across space and time remain poorly understood. Although heterogeneous diversification rates, including both ancient and recent radiations, have been reported, these dynamics have rarely been examined within a single lineage across deep evolutionary timescales. Here, we reconstruct a dated phylogenomic tree of the pantropical plant family Annonaceae, sampling 2045 species (∼82% of its diversity) and spanning ∼100 million years of evolutionary history. Diversification dynamics vary markedly across

Chemistry and biology of ent-morphinan alkaloids.

Morphinan alkaloids have attracted constant attention since the isolation of morphine by Sertürner in 1805. However, a group of 45 compounds possessing a complete ent-morphinan backbone can also be found in the literature. These compounds are related to the morphinandienone subgroup and display a substitution pattern which is different from the morphinans. In particular, these alkaloids could be substituted at position C-2 and C-8 either by a hydroxy function or a methoxy moiety. Four groups of ent-morphinan alkaloids can be proposed, the salutaridine, pallidine, cephasugine and erromangine se

Alkaloid and phenolic compounds of Xylopia aromatica inhibits tumor growth by down-regulating matrix metalloproteinase-2 (MMP-2) expression.

Annonacea species have been reported to possess antitumor properties. However, the in vitro and in vivo antitumor activities of Xylopia aromatica (Annonacea) have not yet been elucidated. This study aimed to investigate the effects of Xylopia aromatica leaves hexane fraction (XaHF) on Ehrlich ascites carcinoma cells lines (EAC), both in vitro and in vivo. In vitro assays revealed a significant cytotoxic effect with the two lower XaHF concentrations (62.5 and 32.3mg/mL). EAC (2.5x106 cells) were inoculated in the right flank of Swiss mice, and the animals were treated intraperitoneally with 32.

Chromosome-scale genome assembly of Artabotrys hexapetalus provides insights into its evolution and terpenoid biosynthesis.

Artabotrys hexapetalus is used as traditional Chinese medicine for the treatment of malaria. Its antimalarial activity has been shown to be the result of the sesquiterpene designated as yingzhaosu A (YZSA), which mainly occurs in the roots of A. hexapetalus. Despite its medicinal significance, the genetic basis and molecular mechanisms underlying the biosynthesis of its active terpenoids remain unclear. In this study, a high-quality chromosome-scale genome of A. hexapetalus was first reported. In total, 915.15 Mb of genome sequences were assembled, of which 911.58 Mb was sorted into 8 chromoso

Antiviral amide derivatives from Uvaria siamensis.

A screening of 824 plant extracts from Rutaceae and Annonaceae species against a human alphacoronavirus (HCoV-229E-Luc) identified Uvaria siamensis (Scheff.) L.L.Zhou, Y·C·F.Su & R.M.K.Saunders (Annonaceae) as a promising source of bioactive metabolites. Subsequent bioassay-guided fractionation of its ethyl acetate and methanol leaf extracts led to the isolation of three previously undescribed amide derivatives, melodamides B-D (3-5), together with melodamide A (2), uvariadiamide (6), (+)- and (-)-toussaintine C (1), and five known polyphenols (7-11). Molluscicidal properties of some species of Annona.

In search for plant molluscicides for the vector control of schistosomiasis, ethanolic extracts from different parts of six species of the Annonaceae family were evaluated against adult forms and egg masses of Biomphalaria glabrata. Results from accurate experiments indicate that the majority of analyzed extracts possess properties lethal to Biomphalaria glabrata, some of them with significant LD90 values (< 20 ppm), as showed for Annona crassiflora M: [pulp and seed (LD50 = 13.21), steem (2.34), root bark (3.79)], Annona glabra L. [seed (17.02)], Annona muricata [leaf (8.75)]. Emphasis is hel

Chemical components of the volatile oil from leaves of Cananga odorata and its anti-oxidant activity.

Cananga odorata (Lamk.) Hook. f. et Thoms., belonging to Annonaceae, is an evergreen tree. The oils extracted from its flower are a famous perfume and used in daily chemical and food industry. Although this plant has been widely cultivated in tropical regions of the world, the yield of oils from its flower is very limited. In order to develop the other parts of this plant, the chemical constituents of the volatile oils from the leaves of C. odorata was analyzed by gas chromatography/flame ionization detector (GC-FID) and GC/mass spectrometry (GC-MS). And the volatiles showed nitric oxide (NO)

Antifilarial activity in vitro of polycarpol and 3-O-acetyl aleuritolic acid from cameroonian medicinal plants against Onchocerca gutturosa.

Previous results have revealed the antifilarial activities of crude extracts and pure compounds from some Cameroonian medicinal plants against Onchocerca volvulus and Onchocerca gutturosa. In our efforts to find new filaricidal agents against adult male O. gutturosa worms, we have isolated and screened three compounds: polycarpol and polyveoline from Polyalthia suaveolens (Annonaceae) and 3-O-acetyl aleuritolic acid from Discoglypremna caloneura (Euphorbiaceae). Only polycarpol and 3-O-acetyl aleuritolic acid exhibited significant inhibitory activities on the vitality of adult male worms of O.

Annonacin, a mono-tetrahydrofuran acetogenin, arrests cancer cells at the G1 phase and causes cytotoxicity in a Bax- and caspase-3-related pathway.

Annonaceous acetogenins are a group of potential anti-neoplastic agents isolated from Annonaceae plants. In this study, we purified annonacin, a cytotoxic mono-tetrahydrofuran acetogenin, from the seeds of Annona reticulata and analyzed its biological effects. Herein, we have shown that annonacin caused significant cell death in various cancer cell lines. T24 bladder cancer cells at the S phase were more vulnerable to the cytotoxicity of annonacin. Furthermore, annonacin activated p21 in a p53-independent manner and arrested T24 cells at the G1 phase. It also induced Bax expression, enhanced c

[Active antihelminitic alkaloids: active in vitro against Leishmanic Tropica the protozoa involved in leishmaniasis].

Leishmaniasis caused by protozoan Leishmania ssp., is an endemic parasitic disease in Central and South America. The chemotherapeutic agents against Leishmania ssp. (pentavalent antimony compounds, pentamidine and amphothericine B) are toxic and expensive products. Basing on the Bolivian folk medicine, we tried to find new active principles. Fourteen isoquinoline alkaloids, especially bisbenzylisoquinoline alkaloids extracted from Annonaceae, Berberidaceae, Hernandiaceae and Menispermaceae, demonstrate highly effective activity against this protozoan. Among them gyrocarpine, daphnandrine and o
