BACKGROUND/PURPOSE: Alveolar bone resorption in periodontitis critically impairs masticatory function and tooth stability, yet the mechanistic drivers linking immune dysregulation to osteolytic pathology remain incompletely characterized. This study elucidates how ferritin-an inflammation responsive iron chaperone secreted by activated monocytes-orchestrates monocyte-to-osteoclast differentiation to exacerbate bone destruction in periodontitis.
MATERIALS AND METHODS: Using a multidisciplinary approach, we combined: Immunohistochemical profiling of cluster of differentiation 68-positive (CD68+

The absence of complement receptor 1 (CR1) related gene/protein y (Crry) leads to embryonic lethality as a result of unrestricted complement activation and concomitant neutrophil infiltration. Here we used Crry(-/-)C3(+/-) mice to investigate the role of Crry in the pathogenesis of immune complex glomerulonephritis (GN). After 3 weeks of immunization with horse spleen apoferritin, six of nine Crry(-/-) C3(+/-) mice and none of the six control C3(+/-) mice developed proliferative GN (P = 0.010). After 5 weeks of immunization, GN scores in Crry(-/-) C3(+/-) mice were 0.67 +/- 0.22 mean +/- stand

Cryo-electron microscopy (cryo-EM) has revolutionized structural biology by enabling imaging of macromolecular complexes at near-atomic resolution, but its success depends critically on specimen quality. Standard plunge freezing often produces nonuniform particle distributions, resulting in poor particle statistics and reduced imaging quality, particularly for vesicular structures and low-concentration, labile macromolecular complexes. Here, we introduce acoustofluidic cryo-EM (ACE), a noncontact specimen-preparation method that integrates acoustofluidic manipulation into a conventional plunge

Contact with the air-water interface can bias the orientation of macromolecules during cryo-electron microscopy sample preparation, leading to uneven sample distribution, preferred orientation and damage to the molecules of interest. To prevent this, we describe a method to encapsulate target proteins within highly hydrophilic, structurally homogeneous and stable protein shells, which we refer to as 'nanocrates' for this purpose. Here we describe the packaging, data acquisition and reconstruction of three proof-of-principle examples, each illuminating a different aspect of the method: apoferri

The effect of intravenously infused iron-dextran (Imferon) on the progression of antigen induced monarticular arthritis in rabbits was studied. A rapid deposition of iron and apoferritin in the synovia of arthritis joints occurred after infusion of iron-dextran during either the acute or chronic phases of the disease. This coincided with the appearance of catalytic (bleomycin reactive) iron in the synovial fluid. There was no evidence, however, for an exacerbation of the antigen induced arthritis as a result of the iron-dextran, and synovial and bleomycin reactive iron concentrations decreased

Nanoparticles are used as carriers for the delivery of drugs and imaging agents. Proteins are safer than synthetic nanocarriers due to their greater biocompatibility and the absence of toxic degradation products. In this context, ferritin has the additional benefit of inherently targeting the membrane receptor transferrin 1, which is overexpressed by most cancer cells. Furthermore, this self-assembling multimeric protein can be loaded with more than 2000 iron atoms, as well as drugs, contrast agents, and other cargos. However, recombinant ferritin currently costs ~3.5 million € g-1 , presumabl

The throughput of cryo-electron microscopy (cryo-EM) can be improved by employing a procedure that collects beam-image shift data. However, this procedure inadvertently induces a beam tilt, thus decreasing the resolution of the reconstruction. Here, we report an automatic calibration procedure for correcting the beam tilt and a large unexpected astigmatism in the beam-image shift data collection. In this procedure, the changes of the beam tilt and the astigmatism against the beam shift are measured and calibrated. The beam tilt and the astigmatism are corrected by changing the setting of the m

Monolayer graphene has great potential for a range of technological and biomedical applications. To achieve this potential, however, selective functionalization of the surface using mild chemical methods that yield spatially and chemically well-defined functional groups is needed. Here, we describe a method to densely and uniformly modify the surface of graphene using a diazonium salt containing a tetrazine moiety, a functional group amenable to bioorthogonal chemistry. The resulting functionalized graphene efficiently undergoes inverse electron demand Diels-Alder reactions with trans-cyclooct

BACKGROUND: An iron rich layer on the labial surface is characteristic of the enamel of rodent incisors. In order to address a role for iron content in continuously growing incisors during odontogenesis, we studied iron deposition patterns in enamel and dentine using Perls' blue staining and ferritin heavy chain (Fth) immunolocalization. Fth expression is regulated by iron level; therefore its localization can be used as a sensitive indicator for iron deposition.
RESULTS: Sagittal sections of 4-week old rat incisors showed a gradual increase in iron level in the enamel organ from secretory to

The brain and liver are both critical organs involved in the pathogenesis of Alzheimer's disease (AD), particularly in the modulation of amyloid-beta (Aβ) metabolism and neuroinflammation. Based on this, a multifunctional nanodrug delivery system, termed OAF, was developed by encapsulating oridonin (ORI) into apoferritin (ApoFn), enabling simultaneous targeting of both brain and the liver through transferrin receptor 1 (TfR1). OAF upregulated the expression of low-density lipoprotein receptor-related protein 1 (LRP1) in cerebral capillary endothelial cells and hepatic parenchymal cells to prom

The physical presence of the antigen used to stimulate a secondary antibody response was demonstrated in the cells of popliteal lymph nodes. Rabbits previously injected with apoferritin (containing no iron), which was prepared from recrystallized horse ferritin, were given an injection of ferritin 5 weeks later. The antigen was traced by means of the Prussian blue reaction, by specific fluorescent antibody, and by electron microscopy. Antiferritin antibody was localized by immunofluorescence, though it was not possible to test cells simultaneously for antigen and antibody. Horse ferritin induc

Ferritin dysregulation is implicated in numerous pathological conditions; however, its role in autosomal dominant polycystic kidney disease (PKD) remains poorly understood. Ferritin expression is increased in cyst-lining epithelial cells and macrophages in both PKD mouse and human kidneys. To investigate the functional significance of ferritin/iron homeostasis in disease progression, we generated conditional knockout of ferritin heavy chain (FTH) in collecting duct or myeloid lineage cells of PKD mice. FTH deletion in either cell type did not impact renal cyst growth. Notably, loss of FTH expr

BACKGROUND: Ferritin exhibits complex behavior in the ultracentrifuge due to variability in iron core size among molecules. A comprehensive study was undertaken to develop procedures for obtaining more uniform cores and assessing their homogeneity.
METHODS: Analytical ultracentrifugation was used to measure the mineral core size distributions obtained by adding iron under high- and low-flux conditions to horse spleen (apoHoSF) and human H-chain (apoHuHF) apoferritins.
RESULTS: More uniform core sizes are obtained with the homopolymer human H-chain ferritin than with the heteropolymer horse s

BACKGROUND: The brain requires iron for a number of processes, including energy production. Inadequate or excessive amounts of iron can be detrimental and lead to a number of neurological disorders. As such, regulation of brain iron uptake is required for proper functioning. Understanding both the movement of iron into the brain and how this process is regulated is crucial to both address dysfunctions with brain iron uptake in disease and successfully use the transferrin receptor uptake system for drug delivery.
METHODS: Using in vivo steady state infusions of apo- and holo-transferrin into t

Visualizing the structure of the protein-inorganic interface is critically important for a more complete understanding of biomineralization. Unfortunately, there are limited approaches for the direct and detailed study of biomolecules that interact with inorganic materials. Here, we use single-particle cryo-electron microscopy (cryo-EM) to study the protein-nanoparticle (NP) interactions of human light chain ferritin and visualize the high-resolution details of the protein-inorganic interface. In this work, we determined the 2.85 Å structure of human light chain ferritin bound to its native ir

Ferritins are ubiquitous proteins that function in iron storage/detoxification by catalyzing the oxidation of Fe2+ ions and solubilizing the resulting Fe3+-oxo mineral. Mammalian tissues that are metabolically highly active contain, in addition to the widespread cytosolic ferritin, a ferritin that is localized to mitochondria. Mitochondrial ferritin (FtMt) protects against oxidative stress and is found at higher levels in diseases associated with abnormal iron accumulation, including Alzheimer's and Parkinson's. Here we demonstrate that, despite 80% sequence identity with cytosolic human H-cha

Ferritins are highly conserved supramolecular protein nanostructures that play a key role in iron homeostasis. Thousands of iron atoms can be stored inside their hollow cavity as a hydrated ferric oxyhydroxide mineral. Although phosphate associates with the ferritin iron nanoparticles, the effect of physiological concentrations on the kinetics, structure, and reactivity of ferritin iron cores has not yet been explored. Here, the iron loading and mobilization kinetics were studied in the presence of 1-10 mM phosphate using homopolymer and heteropolymer ferritins having different H to L subunit

Ferritin is a major iron storage protein essential not only in the infectious process, but also in any circumstance generating oxidative stress. In this study, the cDNA coding sequence of ferritin-H was obtained from the sub-Antarctic Notothenioid fish Eleginops maclovinus through transcriptomic analysis of the head kidney. This sequence contained a 534 bp open reading frame that coded for a 177 amino acid protein with a molecular weight of 20,786.2 Da and a theoretical pI of 5.56. The protein displayed a region of iron putative response elements in the 5'UTR, two putative ferritin iron-bindin
