Increasing evidence suggests modulating effects of cannabinoids on time of onset, severity, and outcome of schizophrenia. Efforts to discover the underlying pathomechanism have led to the assumption of gene x environment interactions, including premorbid genetical vulnerability and worsening effects of continuing cannabis use. The objective of this cross-sectional study is to investigate the relationship between delta-9-tetrahydrocannabinol intake and niacin sensitivity in schizophrenia patients and healthy controls. Intensity of niacin skin flushing, indicating disturbed prostaglandin-mediate

Arachidonic acid (AA) metabolites have been implicated in neonatal pathologic states such as respiratory distress syndrome (RDS). Since free (nonprotein bound) AA is the substrate for synthesis of these compounds, a decreased capacity to bind AA in neonatal plasma could contribute to these disorders. AA binding was assayed by equilibrium dialysis in plasma samples from healthy adults and various infant groups. Plasma from these infant groups bound significantly less AA than adult plasma. Premature infants with RDS and premature infants receiving intralipid had the lowest capacity to bind AA. T

Arachidonic acid (AA) is an essential polyunsaturated fatty acid in humans and a key component of the phospholipid bilayer in cell membranes. Under various stimuli, AA is released from phospholipids; however, elevated levels of AA can act as inflammatory mediators, triggering inflammatory responses. During pregnancy, the placenta synthesizes large amounts of AA to play a critical role in fetal development, preparation for childbirth, and maternal-fetal adaptation. Here, we systematically review the biological mechanisms of AA during pregnancy and its potential involvement in related disorders.

The arachidonic acid (AA) pathway promotes tumor progression by modulating the complex interactions between cancer and immune cells within the microenvironment. In this Review, we summarize the knowledge acquired thus far concerning the intricate mechanisms through which eicosanoids either promote or suppress the antitumor immune response. In addition, we will discuss the impact of eicosanoids on immune cells and how they affect responsiveness to immunotherapy, as well as potential strategies for manipulating the AA pathway to improve anticancer immunotherapy. Understanding the molecular pathw

Blood-dwelling schistosomes and cancer severely impact human health, welfare, and survival, essentially because of their ability to evade host immune cells and humoral effectors. The host immune defenses interact with the surface membrane of the worm and tumor cell. Schistosomes use their sphingomyelin (SM)-rich outer lipid bilayer to form with the surrounding water molecules a tight hydrogen bond barrier in which they open, at will, pores that allow entry of nutrients but not lethal complement components and antibodies. In parasite-free humans, impaired expression of one or more cellular gene

To initiate studies on the significance of filarial arachidonic acid metabolism in the immunopathogenesis of human filariasis, we evaluated the ability of microfilariae of the human filarial parasite Brugia malayi to take up and incorporate exogenous arachidonate. When 4 X 10(5) microfilariae were incubated in vitro with 3 nM [3H]arachidonic acid for 0.2, 24, and 48 hr, 23%, 70%, and 75% of tritium activity were associated with microfilariae, respectively. [3H]arachidonic acid was taken up by viable but not by killed microfilariae. Electron microscopic autoradiographic examination of living mi

This review is about the role of arachidonic acid (ArA) in foetal and early growth and development. In 1975 and '76, we reported the preferential incorporation of ArA into the developing brain of rat pups, its conservation as a principal component in the brains of 32 mammalian species and the high proportion delivered by the human placenta for foetal nutrition, compared to its parent linoleic acid (LA). ArA is quantitatively the principal acyl component of membrane lipids from foetal red cells, mononuclear cells, astrocytes, endothelium, and placenta. Functionally, we present evidence that ArA

OBJECTIVE: Schizophrenia is associated with a blunted flush response to niacin. Since niacin-induced skin flushing is mediated by vasodilators derived from arachidonic acid (AA), we tested whether the blunted flush response to niacin is a marker of AA deficiency.
METHODS: Eight concentrations of methylnicotinate were applied to the forearms of 20 adults with schizophrenia and 20 controls. Laser Doppler measurement of blood flow responses was used to derive values for niacin sensitivity (defined as the concentration eliciting half-maximal response, i.e., EC(50) value) and efficacy (defined as

Cyclic AMP (cAMP)is a natural chemoattractant of the social amoeba Dictyostelium discoideum. It is detected by cell surface cAMP receptors. Besides a signalling cascade involving phosphatidylinositol 3,4,5-trisphosphate (PIP3), Ca2+ signalling has been shown to have a major role in chemotaxis. Previously, we have shown that arachidonic acid (AA) induces an increase in the cytosolic Ca2+ concentration by causing the release of Ca2+ from intracellular stores and activating influx of extracellular Ca2+. Here we report that AA is a chemoattractant for D. discoideum cells differentiated for 8-9 h.

A new radiolabeled metabolite was released into the extracellular fluid by normal human skin fibroblasts that were labeled with [5,6,8,9,11,12,14,15-3H] arachidonic acid. This product continued to accumulate during a 24 h incubation, and its formation was not saturated at arachidonic acid concentrations up to 15 mumol/L. The compound, identified as hexadecatrienoic acid, was not produced by Zellweger fibroblasts which are deficient in peroxisomal fatty acid beta-oxidation. By contrast, radiolabeled hexadecatrienoic acid was produced by mutant fibroblasts having other peroxisomal defects, inclu

The AA cascade in addition to producing various oxygenation products concomitantly yields an oxygen radical. This oxidant selectively deactivates the cyclooxygenase, peroxidase and PGI2 synthetase components whereas TXA2 synthetase is resistant to this oxidant. On the basis of available data it is not possible to assign an inflammatory inducing role to this oxidant on the basis of these actions. Other loci of action must be involved. Among the possibilities are the elements involved in the release of arachidonic acid. Studies relating to this possibility are examined in this report.

We tested the hypothesis that cyclooxygenase-independent vasodilation produced by arachidonic acid (AA) is mediated by epoxyeicosatrienoic acids (EETs) and is blunted in the spontaneously hypertensive rat (SHR). At normal perfusion pressure (PP; 70 to 90 mm Hg), AA constricted the renal vasculature in both SHR and normotensive Wistar-Kyoto rats, an effect abolished by cyclooxygenase inhibition, and converted to vasodilation when PP was raised to approximately 200 mm Hg. Unexpectedly, renal vasodilation elicited by AA was greater in the SHR at high PP; for example, 2.5, 5, and 10 microg of AA p

Oxygen free radicals oxidize arachidonic acid to a complex mixture of metabolites termed isoeicosanoids that share structural similarity to enzymatically derived eicosanoids. However, little is known about oxidations of arachidonic acid mediated by reactive radical nitrogen oxides. We have studied the reaction of arachidonic acid with NO2, a free radical generated by nitric oxide and nitrite oxidations. A major group of products appeared to be a mixture of arachidonic acid isomers having one trans-bond and three cis-double bonds. We have termed these new products trans-arachidonic acids. These

Selenium (Se) and glutathione peroxidase 4 (GPX4) are essential for male reproduction, regulating spermatogenesis, and resisting peroxidative damage. Arachidonic acid (AA) is closely related to male fertility, but excessive intake adversely affects sperm quality. However, how the Se-GPX4 axis ameliorates AA-induced testicular and sperm injury remains unclear. In this study, an AA-induced sperm injury model was established. Combined with Se supplementation and testicular-specific GPX4 knockout, we measured testicular injury and sperm quality phenotypes and explored the potential mechanism via o

Syntaxin regulates pancreatic β cell mass and participates in insulin secretion by regulating insulin exocytosis. In addition, syntaxin 4 reduces IFNγ and TNF-α signaling via NF-ĸB in islet β-cells that facilitates plasma glucose sensing and appropriate insulin secretion. Arachidonic acid (AA) has potent anti-inflammatory actions and prevents the cytotoxic actions of alloxan and streptozotocin (STZ) against pancreatic β cells and thus, prevents the development of type 1 diabetes mellitus (induced by alloxan and STZ) and by virtue of its anti-inflammatory actions protects against the developmen

Hexaclorobenzene (HCB), one of the most persistent environmental pollutants, can cause a wide range of toxic effects including cancer in animals, and hepatotoxicity and porphyria both in humans and animals. In the present study, liver microsomal cytochrome P450 (CYP)-dependent arachidonic acid (AA) metabolism, hepatic PGE production, and cytosolic phospholipase A2 (cPLA2) activity were investigated in an experimental model of porphyria cutanea tarda induced by HCB. Female Wistar rats were treated with a single daily dose of HCB (100 mg kg(-1) body weight) for 5 days and were sacrificed 3, 10,

Arachidonic acid (AA) is a precursor of eicosanoids, which influence human health and the in vitro activity of immune cells. We therefore examined the effects of dietary AA on the immune response (IR) of 10 healthy men living at our metabolic suite for 130 d. All subjects were fed a basal diet containing 27 energy percentage (en%) fat, 57 en% carbohydrate, and 16 en% protein (AA, 200 mg/d) for the first and last 15 d of the study. Additional AA (1.5 g/d) was incorporated into the diet of six men from day 16 to 65 while the remaining four subjects continued to eat the basal diet. The diets of t

The objective of this study was to test the hypothesis that, in human myometrial cells (HMC), PGF2 alpha and oxytocin promote the release of arachidonic acid (AA) which, in turn, acts to mobilize intracellular Ca2+. Primary monolayer cultures of HMC were labeled with [3H]arachidonic acid ([3H]AA) to isotopic equilibrium before exposure to PGF2 alpha or oxytocin. Radiolabeled phospholipids were separated on thin layer chromatography and quantitated by scintillation counting. Prostanoids were analyzed by high performance liquid chromatography. Calcium release was quantitated in digitonin-permeab

The oxidation of arachidonic acid in a superoxide-generating environment results in the formation of a potent chemoattractant that appears to be identical to a chemotactic material generated by hepatocytes when they metabolize alcohol. The product was extracted, chromatographed and characterized by physical methods including GC/MS. The physical properties are consistant with the parent structure: 19-hydroperoxy, 20-hydroxyarachidic acid. This novel saturated 20 carbon product, derived from arachidonic acid by free radical-generating reactions, may play a role in the neutrophilic infiltration o
