Mechanisms of artemether toxicity on single cardiomyocytes and protective effect of nanoencapsulation.

BACKGROUND AND PURPOSE: The artemisinin derivative, artemether, has antimalarial activity with potential neurotoxic and cardiotoxic effects. Artemether in nanocapsules (NC-ATM) is more efficient than free artemether for reducing parasitaemia and increasing survival of Plasmodium berghei-infected mice. NCs also prevent prolongation of the QT interval of the ECG. Here, we assessed cellular cardiotoxicity of artemether and how this toxicity was prevented by nanoencapsulation. EXPERIMENTAL APPROACH: Mice were treated with NC-ATM orally (120 mg·kg-1 twice daily) for 4 days. Other mice received fre

Identification of the Schistosoma mansoni molecular target for the antimalarial drug artemether.

Plasmodium falciparum and Schistosoma mansonii are the parasites responsible for most of the malaria and schistosomiasis cases in the world. Notwithstanding their many differences, the two agents have striking similarities in that they both are blood feeders and are targets of an overlapping set of drugs, including the well-known artemether molecule. Here we explore the possibility of using the known information about the mode of action of artemether in Plasmodium to identify the molecular target of the drug in Schistosoma and provide evidence that artemether binds to SmSERCA, a putative Ca²⁺-

Artemether as adjuvant therapy to praziquantel in murine Egyptian schistosomiasis mansoni.

We investigated the activity of artemether (ART) against different developmental stages of schistosomes alone and in addition to praziquantel (PZQ). ART was administered orally (400 mg/kg) 4 and 6 wk postinfection (PI), 4 and 5 wk PI, or 4 or 6 wk PI alone and in addition to oral PZQ (500 x 2 mg/kg) 6 wk PI. Mice were killed in parallel to infected untreated controls 8 wk PI. Parasitological parameters and histological changes in the liver were studied. ART given 4 and 6 wk PI reduced worm burdens by 59 and 55% and tissue egg load by 96 and 90%, respectively. Moreover, eggs in different develo

Artemether in the treatment of falciparum malaria during pregnancy in eastern Sudan.

This study was carried in New Halfa Hospital, eastern Sudan from October 1997 to February 2001. Twenty-eight pregnant Sudanese women infected with Plasmodium falciparum were treated with intramuscular artemether (six injections, 480 mg) after failure of chloroquine and quinine therapy. The patients were followed-up until delivery; the babies were followed-up until the age of 1 year. Artemether was given to one patient in the tenth week of gestation, to 12 during the second trimester, and to 15 during the third trimester. It was well tolerated, the parasitaemia was cleared and the patients were

Preventive effect of artemether in experimental animals infected with Schistosoma mansoni.

The effect of artemether, an antimalarial drug developed from the plant Artemisia annua, has been tested against the larval stages of Schistosoma mansoni covering the time from skin penetration to the early adult liver-stage. The results show that the experimental animals used (hamster and mice) do not develop schistosomiasis mansoni if treated with artemether during the first month after infection. The parasite was found to be especially susceptible between the 3rd and 4th week after infection, resulting in worm reductions of 75.3-82.0% compared to non-treated controls. This level was boosted

Effect of artesunate and artemether against Opisthorchis felineus in rodent model.

A comparative in vivo assessment of the artemisinin derivatives artesunate and artemether (150 and 300 mg/kg) and praziquantel (400 mg/kg) was conducted in a hamster model of opisthorchiasis caused by Opisthorchis felineus. Physiological, biochemical, and hematological parameters, including complete blood counts and peripheral blood cell composition, were evaluated in all experimental groups. The animals were infected with 100 metacercariae of O. felineus and 1.5 months post-infection received a single oral dose of the tested compounds. Food consumption was monitored before the experiment ende

Antimalarial Drug Repurposing of Epirubicin and Pelitinib in Combination with Artemether and Lumefantrine.

Background: Drug therapy remains the principal management strategy for malaria but is increasingly challenged by the emergence of drug-resistant malaria parasites. The need for new antimalarial drugs is urgent, yet drug discovery and development are hindered by high costs, long durations, and safety concerns that prevent approval. The current study aimed to determine antiplasmodial activities of approved drugs in combination with artemether (ART) and lumefantrine (LU). Methods: Using the SYBR Green I assay test, this study investigated the efficacy of epirubicin (EPI) and pelitinib (PEL) combi

The emerging threat of artemisinin resistance in malaria: focus on artemether-lumefantrine.

The development of artemisinin resistance in the Greater Mekong Subregion poses a significant threat to malaria elimination. Artemisinin-based combination therapies including artemether-lumefantrine (AL) are recommended by WHO as first-line treatment for uncomplicated Plasmodium falciparum malaria. This article provides a comprehensive review of the existing and latest data as a basis for interpretation of observed variability in parasite sensitivity to AL over the last 5 years. Clinical efficacy and preclinical data from a range of endemic countries are summarized, including potential molecul

The efficacy of artemisinin, artemether, and lumefantrine against Babesia gibsoni in vitro.

Artemisinin has many derivatives, and it is effective against Plasmodium spp. However, only a limited number of reports have confirmed the efficacy of artemisinin derivatives against Babesia spp. In this study, whether artemisinin and artemether could inhibit the growth of Babesia gibsoni was evaluated in vitro. In addition, the interaction between artemether and lumefantrine was evaluated. These drugs inhibited the growth of B. gibsoni, but artemisinin and artemether showed lower sensitivity against atovaquone-resistant B. gibsoni than against wild-type B. gibsoni. The interaction between art

A review of the effects of artemether-lumefantrine on gametocyte carriage and disease transmission.

While significant advances have been made in the prevention and treatment of malaria in recent years, these successes continue to fall short of the World Health Organization (WHO) goals for malaria control and elimination. For elimination strategies to be effective, limited disease transmission, achieved through rapid reduction in the infectious parasite reservoir and decreased gametocyte carriage, will be critical. Artemisinin-based combination therapy (ACT) forms the cornerstone of WHO-recommended treatment for uncomplicated Plasmodium falciparum malaria, and in combination with other effect

An experimental trial of artemether in treatment of Pneumocystis carinii in immunosuppressed rats.

An immunosuppressed rat model was established by injecting cortisone acetate 25 mg/rat twice a week for 4 weeks and 12.5 mg/rat for another 2 weeks subcutaneously. A development of Pneumocystis carinii pneumonia (PCP) was found at the end of the 6th week in all rats. These rats were injected intramuscularly with artemether at 100 mg/kg once a day for 5 consecutive days. All rats were necropsied at the end of the 8th week. The lung impression smears were stained with Gomori's stain, and Pneumocystis carinii cysts were counted. The ultrastructural changes of trophozoite, precyst and cyst were in

Rapid coma resolution with artemether in Malawian children with cerebral malaria.

Artemisinin compounds clear parasitaemia more rapidly than other drugs do in both mild and severe malaria, but no advantage in clinical efficacy has been shown. We have compared artemether treatment with standard quinine treatment in Malawian children with cerebral malaria. 65 unconscious children were randomly allocated to intravenous quinine (n = 37) or intramuscular artemether (n = 28) treatment. The two groups were well matched for various prognostic features. Median parasite clearance times were shorter in the artemether group (28 [interquartile range 18-34] vs 40 [36-44] h in the quinine

Parasite Clearance and Artemether Pharmacokinetics Parameters Over the Course of Artemether-Lumefantrine Treatment for Malaria in Human Immunodeficiency Virus (HIV)-Infected and HIV-Uninfected Ugandan Children.

BACKGROUND: Artemisinins are primarily responsible for initial parasite clearance. Antimalarial pharmacokinetics (PK), human immunodeficiency virus (HIV) infection, and antiretroviral therapy have been shown to impact treatment outcomes, although their impact on early parasite clearance in children has not been well characterized. METHODS: Parasite clearance parameters were generated from twice-daily blood smears in HIV-infected and HIV-uninfected Ugandan children treated with artemether-lumefantrine (AL). Artemether and dihydroartemisinin (DHA) area-under-the-curve from 0-8 hours (AUC0-8hr)

Artemether induces JNK1/FOXO1/GPX4-mediated ferroptosis to suppress colorectal cancer.

BACKGROUND: Colorectal cancer (CRC) remains a leading cause of cancer-related mortality worldwide, necessitating novel therapeutic strategies. Ferroptosis, an iron‑dependent, lipid-peroxidation-driven form of cell death, has emerged as a potential tumor‑suppressive mechanism. Artemether, a derivative of Artemisia annua L., exhibits antitumor activity in various cancers, but its role in CRC and its link to ferroptosis remain unclear. METHODS: Using an HCT116 xenograft model and CRC cell cultures, we evaluated artemether's effects on viability, migration, invasion, and histopathology. Inflammat

Modelling the optimal dosing schedule for artemether-lumefantrine chemoprophylaxis against malaria.

OBJECTIVE: Antimalarial chemoprophylaxis for high risk groups in endemic areas of Southeast Asia has the potential to reduce malaria transmission and accelerate elimination. However, the optimal choice of medication and dosing for many potential candidates is not clear. For a planned randomised controlled trial of prophylaxis for forest goers in Cambodia, artemether-lumefantrine (AL) was selected because of its ongoing efficacy and excellent tolerability and safety. As AL had not been used before for this purpose, a previously published pooled pharmacometric meta-model was used to determine th

Hybrid Gold(I) NHC-Artemether Complexes to Target Falciparum Malaria Parasites.

The emergence of Plasmodium falciparum parasites, responsible for malaria disease, resistant to antiplasmodial drugs including the artemisinins, represents a major threat to public health. Therefore, the development of new antimalarial drugs or combinations is urgently required. In this context, several hybrid molecules combining a dihydroartemisinin derivative and gold(I) N-heterocyclic carbene (NHC) complexes have been synthesized based on the different modes of action of the two compounds. The antiplasmodial activity of these molecules was assessed in vitro as well as their cytotoxicity aga

Recurrent Plasmodium falciparum Malaria after Delayed Hemolysis Induced by Artemether-Lumefantrine.

Although artemether-lumefantrine is a first-line treatment for uncomplicated Plasmodium falciparum (P. falciparum) malaria, delayed hemolysis may occur after therapy. A previously healthy 39-year-old Japanese man developed P. falciparum malaria after traveling to Togo and was treated with artemether-lumefantrine in the Philippines. Because fever and fatigue persisted after treatment, he presented to the study hospital and was diagnosed with delayed hemolysis induced by artemether-lumefantrine. Supportive therapy, including blood transfusion, led to symptomatic improvement, and he was discharge
