Begomoviruses: what is the secret(s) of their success?

Begomoviruses constitute an extremely successful group of emerging plant viruses transmitted by whiteflies of the Bemisia tabaci complex. Hosts include important vegetable, root, and fiber crops grown in the tropics and subtropics. Factors contributing to the ever-increasing diversity and success of begomoviruses include their predisposition to recombine their genomes, interaction with DNA satellites recruited throughout their evolution, presence of wild plants as a virus reservoir and a source of speciation, and extreme polyphagia and continuous movement of the insect vectors to temperate reg

Salicylic acid-induced differential resistance to the Tomato yellow leaf curl virus among resistant and susceptible tomato cultivars.

BACKGROUND: In higher plants, salicylic acid (SA) plays important roles in inducing resistance to biotic and abiotic stresses. Tomato yellow leaf curl virus (TYLCV) causes a highly devastating viral disease in plants, particularly in tomato. However, the roles of SA in inducing tomato plant resistance to TYLCV remain unclear. RESULTS: In this study, we investigated whether the exogenous application of SA can improve the resistance of tomato plants to TYLCV in two tomato cultivars, resistant 'Zhefen-702' and susceptible 'Jinpeng-1'. The impacts of SA on the accumulation of ascorbic acid (AsA)

Cloning of Begomoviruses and Betasatellites Associated with Chilli Leaf Curl Disease.

Chilli leaf curl disease (ChiLCD), caused by diverse begomoviruses and associated betasatellites, leads to significant yield losses in chilli crops. This chapter outlines an efficient molecular cloning strategy using rolling circle amplification (RCA) for the full-length amplification of circular viral genomes. Subsequent steps include restriction digestion, ligation into suitable plasmid vectors, transformation into E. coli, screening through RFLP analysis, and sequencing of cloned products. Conventional PCR has been used for begomovirus genome characterization, but it typically needs prior s

The Alarming Eastward Front of Cassava Mosaic Disease Reported in Guinea and Sierra Leone Reaches Western Côte d'Ivoire.

Cassava mosaic begomoviruses are a major threat to cassava cultivation in Africa. The virulent Ugandan variant of the East African cassava mosaic virus (EACMV-Ug), which caused substantial damage to cassava production in Uganda in the 1990s and which was previously confined to East and Central Africa, was recently found to be well established in Guinea and Sierra Leone in West Africa. Molecular analysis of cassava leaf samples from a nationwide cassava fields survey conducted in Côte d'Ivoire in 2022 suggested the absence of EACMV-Ug in the country in 2022. Given the proximity of some confirme

7-Hydroxylathyrol defeats geminivirus through targeting C4 and activating host defense.

Tomato yellow leaf curl virus (TYLCV), which belongs to Geminiviruses, is a notorious plant virus that poses a threat to global agriculture. 7-Hydroxylathyrol is a renowned lathyrane diterpenoid with a broad spectrum of pharmacological activities. However, its efficacy against plant pathogens remains unknown. In this study, we investigated the antiviral activity of 7-hydroxylathyrol against TYLCV and explored the molecular mechanisms underlying its antiviral effects. 7-Hydroxylathyrol significantly disrupted TYLCV and inhibited its accumulation in Nicotiana benthamiana. To explore the mechanis

Immunity responses as checkpoints for efficient transmission of begomoviruses by whiteflies.

Begomoviruses are a group of single stranded DNA plant viruses exclusively transmitted by the sweet potato whitefly Bemisia tabaci in a persistent, circulative manner. After acquisition from plant phloem, this group of viruses circulate and are retained within the whitefly, interacting with tissues, cells and molecular pathways for maintaining the safety of the infective intact virions, by exploiting cellular mechanisms and avoiding degradation by the insect immune responses. During retention, the virions are internalized in the midgut cells, exit and spend hours-days in the hemolymph and cros

Comparative analysis of salicylic acid levels and gene expression in resistant, tolerant, and susceptible cassava varieties following whitefly-mediated SLCMV infection.

Sri Lankan cassava mosaic virus (SLCMV), the primary pathogen responsible for cassava mosaic disease in cassava plantations, is transmitted via infected cutting stems and the whitefly vector, Bemisia tabaci. To obtain better insights into the defense mechanism of cassava against SLCMV, whiteflies were used to induce SLCMV infection for activating the salicylic acid (SA) signaling pathway, which triggers the innate immune system. The study aimed to investigate the specific interactions between viruliferous whiteflies and SA accumulation in resistant (C33), tolerant (Kasetsart 50; KU50), and sus

Exploring Tomato Fruit Viromes through Transcriptome Data Analysis.

This study delves into the complex landscape of viral infections in tomatoes (Solanum lycopersicum) using available transcriptome data. We conducted a virome analysis, revealing 219 viral contigs linked to four distinct viruses: tomato chlorosis virus (ToCV), southern tomato virus (STV), tomato yellow leaf curl virus (TYLCV), and cucumber mosaic virus (CMV). Among these, ToCV predominated in contig count, followed by STV, TYLCV, and CMV. A notable finding was the prevalence of coinfections, emphasizing the concurrent presence of multiple viruses in tomato plants. Despite generally low viral le

Role of aminopeptidase N-like in the acquisition of begomoviruses by Bemisia tabaci, the whitefly vector.

Sri Lankan cassava mosaic virus (SLCMV) is a prominent causative agent of cassava mosaic disease in Asia and relies on the whitefly Bemisia tabaci cryptic complex for its transmission. However, the molecular mechanisms involved in SLCMV transmission by B. tabaci have yet to be understood. In this study, we identified an aminopeptidase N-like protein (BtAPN) in B. tabaci Asia II 1, an efficient vector of SLCMV, which is involved in the SLCMV transmission process. Through the use of glutathione S-transferase pull-down assay and LC-MS/MS analysis, we demonstrated the interaction between BtAPN and

A peptide derived from enzymatic digestion of globulins from amaranth shows strong affinity binding to the replication origin of Tomato yellow leaf curl virus reducing viral replication in Nicotiana benthamiana.

Tomato yellow leaf curl virus (TYLCV; genus Begomovirus; family Geminiviridae) infects mainly plants of the family Solanaceae, and the infection induces curling and chlorosis of leaves, dwarfing of the whole plant, and reduced fruit production. Alternatives for direct control of TYLCV and other geminiviruses have been reported, for example, the use of esterified whey proteins, peptide aptamer libraries or artificial zinc finger proteins. The two latter alternatives affect directly the replication of TYLCV as well as of other geminiviruses because the replication structures and sequences are hi

Identification and Functional Characterization of the Mungbean Yellow Mosaic India Virus Resistant Gene VrADH Using a MAGIC Population.

Mungbean is an important legume and protein source, but its productivity is severely limited by the mungbean yellow mosaic India virus (MYMIV), and no MYMIV resistance gene has been cloned and functionally validated in mungbean. This study conducted extensive phenotypic variation research on the resistance of a newly developed mungbean multiparent advanced-generation intercross (MAGIC) population. Through an integrated genome-wide association study (GWAS), transcriptome analysis, and induced expression analysis, the candidate gene for MYMIV resistance was identified as VrADH, encoding alcohol

Airborne β-caryophyllene disrupts virus-vector mutualism by priming tomato defenses.

Whiteflies pose a major threat to crops worldwide, primarily because they transmit begomoviruses with which they have evolved intricate mutualistic relationships. The mutualisms are known to exacerbate whitefly invasions and drive widespread plant virus pandemics. Yet, certain plant genotypes are able to resist both the whiteflies and the viruses and a good understanding of the underlying mechanisms could help to develop more resistant varieties. Here, we show that the viruliferous whitefly Bemisia tabaci induces an early and strong release of the sesquiterpene β-caryophyllene in cultivated to

Stabilisation of Tomato Yellow Leaf Curl China Virus Infectious Clones Through Micro-Homology Mediated End Joining.

Tomato yellow leaf curl China virus (TYLCCNV) is a major agricultural pathogen and primary model for circular single-stranded DNA (cssDNA) virus studies. The infectious clones of TYLCCNV and other cssDNA viruses are usually constructed as two tandem copies of the small viral genomes in Agrobacterium-mediated T-DNA vectors. However, during our experiment, we observed that successive cultivation of the agrobacterial infectious clone of TYLCCNV led to a reduction or complete loss of its virulence in host plants. Further analysis revealed that the instability of this infectious clone is analogous

Defence-mediated phloem restriction of a plant virus facilitates insect transmission.

Plant viruses are causal agents of devastating diseases in crops and pose a threat to food security. Viruses transmitted by phloem-feeding insects are frequently restricted to the phloem; the mechanism determining this tissue tropism and its consequences for the viral cycle have remained elusive. Here we show that phloem restriction of tobacco curly shoot virus (TbCSV) depends on the allele of the C4 gene it carries. The Y35 allele produces a plasma membrane-associated C4 protein; TbCSV(Y35) is phloem-limited. The Y41 allele, however, produces two C4 variants, targeted to the plasma membrane a

Defender or accomplice? Dual roles of plant vesicle trafficking in restricting and enabling geminiviral systemic infection.

The vesicle trafficking system enables multidirectional cargo fluxes between endomembrane compartments. However, vesicle trafficking plays dual roles during pathogen infections. In plants, it mediates autophagic immune responses but can also be hijacked by pathogens to facilitate successful infections. We demonstrate that vesicle trafficking machinery acts as a double-edged sword during infection by the geminivirus tomato yellow leaf curl Sardinia virus (TYLCSaV) in Nicotiana benthamiana. Virus-induced gene silencing of eight genes encoding key vesicle trafficking regulators revealed contrasti

Genome-Wide Screening and Functional Verification of the Tobacco SGR Family Identify Transcription-Factor-Like NtSGRL as a Negative Regulator of Tomato Yellow Leaf Curl Virus Resistance.

Viral infection often induces chlorosis, a symptom closely associated with chlorophyll (Chl) degradation. STAY-GREEN (SGR) proteins are key regulators of Chl breakdown, but their roles in plant virus resistance remain poorly understood. In this study, we identified six NtSGR family genes in Nicotiana tabacum and functionally characterized NtSGRL1 during tomato yellow leaf curl virus (TYLCV) infection. TYLCV drastically upregulates NtSGRL1 (149-fold at 4 days post-inoculation [dpi] vs. control), suggesting its involvement in the tobacco-TYLCV interaction. Subcellular localization revealed that

Enabling biocontained plant virus transmission studies through establishment of an axenic whitefly (Bemisia tabaci) colony on plant tissue culture.

Whiteflies (Bemisia tabaci) and the diseases they transmit are a major detriment to crop yields and a significant contributor to world hunger. The highly evolved interactions of host plant, phloem-feeding insect vector with endosymbionts and persistently transmitted virus represent a tremendous challenge for interdisciplinary study. Presented here is the establishment of a colony of axenic whiteflies on tissue-cultured plants. Efficient colony establishment was achieved by a surface sterilization of eggs laid on axenic phototrophically tissue-cultured plants. The transfer of emerging whiteflie

Bemisia tabaci Q carrying tomato yellow leaf curl virus strongly suppresses host plant defenses.

The concurrence of tomato yellow leaf curl virus (TYLCV) with the spread of its vector Bemisia tabaci Q rather than B in China suggests a more mutualistic relationship between TYLCV and Q. Here, we investigated the hypothesis that viruliferous B and Q have different effects on plant defenses. We found the fecundity of nonviruliferous B, nonviruliferous Q, viruliferous Q and viruliferous B was 11.080, 12.060, 10.760, and 11.220 respectively on plants previously attacked by the other biotype, however, on their respective noninfested control leaves fecundity was 12.000, 10.880, 9.760, and 8.020 r

Novel insights into tomato leaf curl New Delhi virus introduction and evolution in Southeastern France using an advanced long-read sequencing workflow.

The Mediterranean population of tomato leaf curl New Delhi virus (ToLCNDV-ES) is characterized by a high genetic uniformity, distinguishing it from its Asian counterparts. ToLCNDV-ES is thought to have a monophyletic origin, likely resulting from a single recombination event, prior to its spread throughout the Mediterranean region. Following its first detection in southeastern France in 2020, ToLCNDV-ES re-emerged in France in 2022. Our analysis based on advanced long-read sequencing, circular DNA profiling, and phylogeny indicates both local persistence of French ToLCNDV-ES and multiple indep
