Metabolism of methionine and biosynthesis of caffeine in the tea plant (Camellia sinensis L.).

1. Caffeine biosynthesis was studied by following the incorporation of 14C into the products of L-[Me-14C]methionine metabolism in tea shoot tips. 2. After administration of a 'pulse' of L-[Me-14C]methionine, almost all of the L-[Me-14C]methionine supplied disappeared within 1 h, and 14C-labelled caffeine synthesis increased throughout the experimental periods, whereas the radioactivities of an unknown compound and theobromine were highest at 3 h after the uptake of L-[Me-14C]methionine, followed by a steady decrease. There was also slight incorporation of the label into 7-methylxanthine, seri

Antioxidant and anti-aging effects of polysaccharides extracted from Camellia oleifera branches.

BACKGROUND: Camellia oleifera Abel is extensively cultivated for edible oil, generating abundant leaves, shells, and branches as by-products. However, the bioactivity of polysaccharides from these different tissues remains poorly characterized. To valorize these agricultural wastes, six crude polysaccharides were extracted from the leaves, shells, and branches of C. oleifera using hot water extraction (HWE) and ultrasound-assisted hot water extraction (USHWE). Their physicochemical properties were compared, and in vitro anti-inflammatory effects were evaluated to identify the most promising fr

Biochemical responses of Camellia sinensis (L.) O. Kuntze to heavy metal stress.

Three heavy metals-mercury (II), copper (II) and nickel (II), each at a concentration of 10 and 100 micrograms/ml, were tested for their effects on various biochemical constituents of tea leaves. Both NI (II) and Hg (II) decreased the phenolic contents, while Cu (II) increased it to some extent. The metal treatments enhanced the activity of phenyl alanine ammonia lyase (PAL), while the activity of poly phenol oxidase (PPO) showed a decline. Heavy metal stress also decreased the chlorophyll content of the leaves, along with a significant reduction in Hill activity. Proline content increased sig

Microwave hydrodiffusion and gravity pretreatment of Camellia japonica flowers for the extraction of bioactives.

The valorization of Camellia japonica flower varieties through the green extraction of their bioactive components is addressed. Microwave hydrodiffusion and gravity (MHG) can be an efficient and rapid pretreatment to dry the petals; for example, in the Royal Velvet variety, the energy consumption was 16 times lower compared to air drying. The hydrolates obtained in this stage represented only 0.5 mg gallic acid equivalents/g flower, but some of the fractions contained up to 10 times higher and a Trolox equivalent antioxidant capacity (TEAC) value equivalent to 0.4 g Trolox/g extract. The dried

miR828a-CsMYB114 Module Negatively Regulates the Biosynthesis of Theobromine in Camellia sinensis.

Theobromine is an important quality component in tea plants (Camellia sinensis), which is produced from 7-methylxanthine by theobromine synthase (CsTbS), the key rate-limiting enzyme in theobromine biosynthetic pathway. Our transcriptomics and widely targeted metabolomics analyses suggested that CsMYB114 acted as a potential hub gene involved in the regulation of theobromine biosynthesis. The inhibition of CsMYB114 expression using antisense oligonucleotides (ASO) led to a 70.21% reduction of theobromine level in leaves of the tea plant, which verified the involvement of CsMYB114 in theobromin

Effect of fluoride treatment on gene expression in tea plant (Camellia sinensis).

Tea plant is a typical fluorine (F) accumulator. F concentration in mature tea leaves is several hundred times higher than that in normal field crops. Long-term consumption of teas with high level F will increase the risks of dental and skeletal fluorosis. The mechanism of F accumulation in tea stands unclear. RNA-Seq and digital gene expression (DGE) techniques were used to investigate the effect of F on the differential expressions of transcriptome in tea plant. The results showed that F content in mature tea leaves was increased with increase in F concentration of cultural solution and dura

Molluscicidal Activity of Camellia Sinensis (Green Tea) and Camellia sinensis var. Assamica (Purple Tea) Extracts Against Biomphalaria pfeifferi, the Major Vector Snail of Human Schistosomiasis in Sub-Saharan Africa.

Freshwater pulmonate snails are prevalent across Kenya and act as intermediate hosts for trematode parasites, some of which are snail vectors for human schistosomiasis. Chemical molluscicides have rarely been used routinely in Kenya to control snails due to high costs of manufacture and the subsequent environmental concerns associated with their use. This study tested extracts from green tea and purple tea plants, Camellia sinensis, which are widely grown in Kenyan highland areas, against Biomphalaria pfeifferi, the obligate intermediate host for Schistosoma mansoni. Snails were exposed to fiv
