Etoposide and secondary haematological malignancies: coincidence or causality?

BACKGROUND: Secondary haematological malignancies (SHM) induced by alkylating agents show cytogenetic abnormalities involving mainly chromosomes 5 and 7. Etoposide and other topoisomerase II inhibitors including anthracyclines induce secondary leukaemias with a short latency period, briefly preceded by a myelodysplastic phase, and frequently associated with balanced translocation characteristically involving the long arms of chromosomes 11 and 21. PATIENTS AND METHODS: Etoposide with doxorubicin containing chemotherapy was administered to 26 patients with Hodgkin's disease (HD), eight of whom

Dissecting Spectral Granger Causality Through Partial Information Decomposition.

OBJECTIVE: Granger causality (GC), a popular statistical method for the inference of directional influences between time series measured from a complex network, is sensitive to high-order (non-pairwise) interactions which fundamentally shape the collective network dynamics. This work introduces Partial Decomposition of Granger Causality (PDGC), a tool to elicit redundant and synergistic causal interactions among the subsystems of physiological networks. METHODS: We use the framework of partial information decomposition to dissect the multivariate GC from a set of driver random processes to a

Deciphering interventional dynamical causality via Kolmogorov-Arnold networks.

Identifying causal relationships from observational time-series data is a central challenge in understanding complex dynamical systems. However, few existing methods attempt to capture the intuitive human understanding of causality: that an intervention on one variable leads to a subsequent change in another. To bridge this gap, this paper introduces a causal discovery method named Intervened TaylorKAN (ITK), which simulates controlled experiments within reconstructed systems. Specifically, we leverage the recently emerging Kolmogorov-Arnold Networks (KANs) with Taylor series as activation fun

Causality between diabetes and membranous nephropathy: Mendelian randomization.

BACKGROUND: Membranous nephropathy (MN) has not yet been fully elucidated regarding its relationship with Type I and II Diabetes. This study aims to evaluate the causal effect of multiple types of diabetes and MN by summarizing the evidence from the Mendelian randomization (MR) study. METHODS: The statistical data for MN was obtained from a GWAS study encompassing 7979 individuals. Regarding diabetes, fasting glucose, fasting insulin, and HbA1C data, we accessed the UK-Biobank, within family GWAS consortium, MAGIC, FinnGen database, MRC-IEU, and Neale Lab, which provided sample sizes ranging

Microspherophakia associated with congenital miosis: Coincidence or causality?

Congenital miosis and microspherophakia are rare congenital disorders of the anterior chamber that are associated with multiple other ophthalmological and systemic pathologies or disorders. Nevertheless, the association of both entities have not been described yet. We present the case of a 2-years-old patient without other relevant medical history that presented with high myopia secondary to microspherophakia and associated with congenital miosis.

Zenker's Diverticulum in Forestier Disease: Chance or Causality?

INTRODUCTION: Zenker's diverticulum is an acquired sac-like outpouching of the mucosa and submucosa layers originating at the pharyngoesophageal junction. The predominant symptom of Zenker's diverticulum is dysphagia. Videofluoroscopy confirms the diagnosis. Forestier disease is a clinical entity characterized by ossification of anterolateral vertebral ligament and anterior osteophyte formation along the anterolateral spinal column. Its etiopathogenesis remains unknown and common symptoms are dysphagia, dysphonia and airway obstruction. The objective of this study is to identify a pathophysiol

Comorbidities in gout and hyperuricemia: causality or epiphenomena?

PURPOSE OF REVIEW: To review advances in the understanding of potentially causal relationships between gout, hyperuricemia and comorbidities. RECENT FINDINGS: Observational studies reveal 4-5 comorbidity clusters in gout patients. There tend to be gout alone, gout with chronic kidney disease and gout with other metabolic comorbidities. However, heterogeneous study populations and confounding make inference difficult for causal relationships. Mendelian randomization leverages genetic information as an instrumental variable to indicate putatively causal relationships between traits of epidemiol

A causality dilemma: ARFID, malnutrition, psychosis, and hypomagnesemia.

We present a novel case of a woman with coincident occurrence of auditory and visual hallucinations, electrolyte disturbances, chloride unresponsive alkalosis, and an eating disorder. The patient was ultimately diagnosed with Gitelman syndrome comorbid with schizophreniform disorder and avoidant restrictive food intake disorder. Eating disorders are often associated with electrolyte abnormalities which, in turn, can cause or contribute to other neuropsychiatric symptoms. At the same time, psychotic disorders can lead to food intake aversions or overconsumption of fluids with associated effects

Cortical Dynamic Causality Network for Auditory-Motor Tasks.

Motor preparation and execution require the interactions of a large-scale brain network, while the study of the dynamic changes of their interactions could uncover the underlying neural mechanism of the corresponding information processing. This dynamic analysis requires high temporal resolution of the recorded signals. Electroencephalogram (EEG) with high temporal resolution has been widely used in related studies. However, studies based on scalp EEG always lead to distorted results, due to scalp volume conduction, compared with that of cortically recorded signals. In the current study, the d

Readmission after pancreatic resection: causes and causality pattern.

BACKGROUND: Readmission rates have been targeted for cost/reimbursement control. Our goal was to identify causes for readmission and delineate the pattern of early and late readmission. METHODS: Between 2011 and 2012, a total of 490 patients underwent pancreaticoduodenectomy, distal pancreatectomy or central pancreatectomy. Logistic regression was used to identify predictors of readmission. K-medoids clustering was performed to identify the major readmission subgroups. RESULTS: Median postoperative length of stay (LOS) was 7 days, and the 30- and 90-day readmission rates were 23 and 29 %, re

Granger causality--statistical analysis under a configural perspective.

The concept of Granger causality can be used to examine putative causal relations between two series of scores. Based on regression models, it is asked whether one series can be considered the cause for the second series. In this article, we propose extending the pool of methods available for testing hypotheses that are compatible with Granger causation by adopting a configural perspective. This perspective allows researchers to assume that effects exist for specific categories only or for specific sectors of the data space, but not for other categories or sectors. Causality and gastrointestinal infections: Koch, Hill, and Crohn's.

The gastrointestinal tract contains a complex mix of microorganisms. Therefore, the finding of a particular microbe in the gastrointestinal tract or stools of a patient with intestinal disease does not necessarily indicate that the clinical state resulted from the presence of this organism. For this reason, extending from epidemiological association to causality is particularly challenging for gastrointestinal diseases. Most established agents of acute bacterial gastroenteritis (eg, Escherichia coli, Campylobacter jejuni) have been shown to fulfil Koch's postulates of causality in human volunt

Pemphigus following hepatitis B vaccination--coincidence or causality?

Pemphigus is an autoimmune blistering disease caused by autoantibodies against epithelial intercellular components. Its etiology is unknown, and neoplasms, antecedent infections or medications are considered possible triggering factors for the disease in some cases. We describe the first case of pemphigus following a hepatitis B virus vaccination. We suggest that in some cases vaccination may be the triggering factor for pemphigus in genetically predisposed individuals and physicians should be aware of this possible association.

Causality-integrated graph learning for multi-endpoint toxicity prediction.

Toxicity prediction remains a longstanding challenge in the safety assessment of numerous artificial chemicals. While conventional single-endpoint models are effective for predicting many molecular properties, they fail to capture the complex mechanisms underlying toxicity, which involve multiple molecular targets, interconnected pathways, and diverse outcomes. We present a causality-integrated graph learning framework that embeds toxicological mechanisms extracted from large-scale literature mining as directed graphs within deep learning (DL) models. By using chemical structure as input, the

Atorvastatin is a potent lipid-lowering agent whose potential hepatotoxicity, anecdotally delayed, has been well documented. We present a case of cholestatic hepatitis due to atorvastatin with an unusually prolonged latency period, classically counterintuitive from a causality standpoint.

Depression and dementia: interrogating the causality of the relationship.

BACKGROUND: Depression is often cited as a major modifiable risk factor for dementia, though the relative contributions of a true causal relationship, reverse causality and confounding factors remain unclear. This study applied a subset of the Bradford Hill criteria for causation to depression and dementia including strength of effect, specificity, temporality, biological gradient and coherence. METHODS: A total of 491 557 participants in UK Biobank aged between 40 and 69 at enrolment and followed up for a mean duration of 12.4 years were studied. Diagnoses of depression and dementia were asc

Lipoprotein(a) and Venous Thromboembolism: Association, Causality, and Medications.

Lipoprotein(a) [Lp(a)] is a circulating plasma lipoprotein with structural similarities to low-density lipoprotein (LDL), distinguished by the addition of apolipoprotein(a) to the LDL structure. Lp(a) levels are approximately 80% genetically determined, and distinct components of this complex particle are thought to confer atherogenic, inflammatory, and antifibrinolytic properties contributing to cardiovascular risk. A growing body of evidence has shown a causal association between elevated Lp(a) levels and both atherosclerotic cardiovascular disease (ASCVD) and valvular aortic stenosis. Howev
