Evaluation of cell viability and metabolic activity of a 3D cultured human epidermal model using a dynamic autoradiographic technique with a PET radiopharmaceutical.

Quality control of tissues and organs for transplant is important to confirm their safety and effectiveness for regenerative medicine. However, quality evaluation is only carried out using a limited range of inspection criteria, because many of the available evaluation tests are invasive. In order to explore the potential of 2-[18F]fluoro-2-deoxy-D-glucose ([18F]FDG)-bioradiography as a non-invasive test for estimation of the safety, soundness, and effectiveness of tissues for transplantation, [18F]FDG uptake and cell viability or metabolism were investigated using a reconstructed human epider

Effects of acetylcholine and other medullary secretagogues and antagonists on the membrane potential of adrenal chromaffin cells: an analysis employing techniques of tissue culture.

1. A method has been devised for isolating adrenal chromaffin cells (from gerbils) and maintaining them in vitro. Transmembrane potentials of these cells were recorded with intracellular micro-electrodes.2. Acetylcholine depolarized the chromaffin cells and so did various other substances known to evoke catecholamine secretion: nicotine, pilocarpine, histamine, 5-hydroxytryptamine, angiotensin, and bradykinin.3. The depolarizing effect of acetylcholine was partially antagonized by hexamethonium and was blocked completely by hexamethonium in combination with atropine.4. Hexamethonium alone comp

A Comparative Study between Follicular Unit Transplantation and Autologous Non-cultured Non-trypsinized Epidermal Cells Grafting (Jodhpur Technique) in Stable Vitiligo.

INTRODUCTION: Several modalities are currently available for the treatment of vitiligo, but generally they do not result in complete cure of the disease. Despite the limitations and some side effects, surgical modalities appear to be the method of choice in recalcitrant stable vitiligo. Both Jodhpur technique (JT) and follicular unit transplantation (FUT) are simpler and cheaper methods, requiring minimal infrastructure. MATERIALS AND METHODS: Hundred patches in 30 patients were divided into Group A and Group B, that is, hair follicle transplantation and autologous non-cultured non-trypsinize

Circulating megakaryocytic progenitor cells in patients with primary thrombocythaemia and reactive thrombocytosis: results using a serum-deprived culture assay and a positive detection technique.

A serum-free culture method was used to study the growth of megakaryocytic progenitor cells (CFU-Meg) from patients with elevated platelet counts. The culture technique was combined with immunocytochemistry (APAAP, CD61) for the identification of CFU-Meg derived cells in cytopreparations of cells eluted from the culture dishes. Twenty-six patients with primary thrombocythaemia (14 untreated patients, UPT, 12 treated patients, TPT), 14 patients with reactive thrombocytosis (RT) and 9 normal individuals were studied. Unstimulated growth of CD61-positive cells was detected in 8/14 UPT, 8/12 TPT,

Four compartment method as an efficacious and simplified technique for autologous non-cultured epidermal cell suspension preparation in vitiligo surgery: A randomized, active-controlled study.

BACKGROUND: Autologous non-cultured epidermal cell suspension (NCES) is a successful surgical method for repigmentation of stable, refractory vitiligo. The need for laboratory equipment and expertise restricts its use to only a few research centres; hence, there is a requisite to simplify the technique of NCES preparation. OBJECTIVES: To evaluate the efficacy of NCES prepared by four compartment (4C) method compared to the laboratory-based method (lab-NCES). METHODS: Anatomically based matched lesions (41 pairs) in 30 stable vitiligo patients were randomized to receive NCES prepared by 4C me

Cell, tissue and organ culture as in vitro models to study the biology of squamous cell carcinomas of the head and neck.

In vitro models are currently being used to study head and neck squamous cell carcinoma (HNSCC). Several hundred HNSCC cell lines have been established by various investigators and used to study a broad spectrum of questions related to head and neck cancer. The head and neck model with respect to multistage carcinogenesis is now complete. Several techniques exist for the culture of normal epithelial cells from the upper aerodigestive tract (UADT). The biology of these UADT cells (oral cavity, oropharynx, hypopharynx and larynx) is being studied. Successful culture of premalignant lesions (dysp

Development of an Insert Co-culture System of Two Cellular Types in the Absence of Cell-Cell Contact.

The role of secreted soluble factors in the modification of cellular responses is a recurrent theme in the study of all tissues and systems. In an attempt to make straightforward the very complex relationships between the several cellular subtypes that compose multicellular organisms, in vitro techniques have been developed to help researchers acquire a detailed understanding of single cell populations. One of these techniques uses inserts with a permeable membrane allowing secreted soluble factors to diffuse. Thus, a population of cells grown in inserts can be co-cultured in a well or dish co

Impact of mouse tracheal basal cell expansion medium on formation and metabolic characteristics of tracheospheres.

BACKGROUND: Airway basal cells have been shown to play important roles in lung disease, prompting their use in research involving 3-dimensional (3D) organoid culture. Mouse tracheal basal cells (MTBC) represent a key model in understanding the differentiation of airway basal cells in normal development and diseased states. The isolation, expansion, and culture media of basal cells can affect their capacity to maintain stemness and properly differentiate, thereby introducing unknown experimental variables. As MTBC use in 3D organoid culture proceeds, there is an increasing need for comparative

Role of cell and explant culture in the diagnosis and characterization of human pituitary tumours.

The pattern of hormone secretion by human pituitary tumours in cell or explant culture has been shown to be of value in establishing the nature of the tissue. There was complete agreement between the diagnosis reached by conventional immunocytochemical techniques and by examining the secretion of hormones in culture. Culture techniques, however, have some advantages over immunocytochemical analysis. In particular, immunocytochemical techniques can only be used to examine a small, possibly unrepresentative, portion of the pituitary tissue, whereas the in vitro culture systems make use of the wh
