Distinct Longitudinal Changes in EEG Measures Reflecting Functional Network Disruption in ALS Cognitive Phenotypes.

Amyotrophic lateral sclerosis (ALS) is characterised primarily by motor system degeneration, with clinical evidence of cognitive and behavioural change in up to 50% of cases. We have shown previously that resting-state EEG captures dysfunction in motor and cognitive networks in ALS. However, the longitudinal development of these dysfunctional patterns, especially in networks linked with cognitive-behavioural functions, remains unclear. Longitudinal studies on non-motor changes in ALS are essential to further develop our understanding of disease progression, improve care and enhance the evaluat

Information-processing dynamics in neural networks of macaque cerebral cortex reflect cognitive state and behavior.

One of the essential functions of biological neural networks is the processing of information. This includes everything from processing sensory information to perceive the environment, up to processing motor information to interact with the environment. Due to methodological limitations, it has been historically unclear how information processing changes during different cognitive or behavioral states and to what extent information is processed within or between the network of neurons in different brain areas. In this study, we leverage recent advances in the calculation of information dynamic

Functional network dynamics revealed by EEG microstates reflect cognitive decline in amyotrophic lateral sclerosis.

Recent electroencephalography (EEG) studies have shown that patterns of brain activity can be used to differentiate amyotrophic lateral sclerosis (ALS) and control groups. These differences can be interrogated by examining EEG microstates, which are distinct, reoccurring topographies of the scalp's electrical potentials. Quantifying the temporal properties of the four canonical microstates can elucidate how the dynamics of functional brain networks are altered in neurological conditions. Here we have analysed the properties of microstates to detect and quantify signal-based abnormality in ALS.

Topographical differences of frontal-midline theta activity reflect functional differences in cognitive control abilities.

Electrophysiological oscillations are assumed to be the core mechanism for large-scale network communication. The specific role of frontal-midline theta oscillations as cognitive control mechanism is under debate. According to the dual mechanisms of control framework, cognitive control processes can be divided into proactive and reactive control. The present study aimed at investigating the role of frontal-midline theta activity by assessing oscillations in two tasks varying in the type of cognitive control needed. More specifically, a delayed match to sample (DMTS) task requiring proactive co

The impact of negative moods on self-enhancing cognitions: the role of reflective versus ruminative mood orientations.

This research program examined how self-focused attention to feelings affects the relation between mood negativity and self-enhancing thought. The primary hypothesis was that the particular manner in which people focus on their moods (reflective vs. ruminative) determines whether they reveal positive (i.e., mood-incongruent) or negative (i.e., mood-congruent) self-relevant thoughts in response to negative moods. Studies 1-4 revealed that social comparisons, temporal comparisons, and other self-enhancing cognitions (i.e., attributions, disidentification, relationship evaluations) are more likel

Object recognition and Morris water maze to detect cognitive impairment from mild hippocampal damage in rats: A reflection based on the literature and experience.

Object recognition (OR) and the Morris water maze (MWM) are classical tasks widely used to assess memory parameters and deficits in rodents. Learning processes in both tasks involve integrity of the hippocampus and associated regions, and prefrontal cortex connections. Here, we highlight the idea that these classical tests can be used to indicate memory deficits caused by models of disease that affect hippocampal function in rats, and identify some practical issues of OR and MWM, based on the literature and our experience. Additionally, we have shown that the performance of both tasks does not

Periventricular diffusivity reflects APOE ε4-modulated amyloid accumulation and cognitive impairment in the Alzheimer's disease continuum.

INTRODUCTION: Altered glymphatic-related fluid dynamics are increasingly recognized as a feature of Alzheimer's disease (AD). We generalized an established diffusion imaging framework to quantify periventricular diffusivity (PVeD), hypothesizing that fast diffusion signals in the periventricular region can reflect amyloid beta (Aβ) deposition across the AD continuum. METHODS: Participants from two multi-site cohorts (n = 440 and 414), comprising cognitively unimpaired individuals, those with mild cognitive impairment, and patients with AD, were included. We tested and validated the associatio

Salivary levels of amyloid beta reflect brain amyloid beta burden in cognitively-normal older adults.

BACKGROUND: Amyloid beta (Aβ) plaque burden, as measured by positron emission tomography (PET), is increasingly being used as a biomarker for Alzheimer's disease (AD) as well as a screening or monitoring tool for clinical trials with amyloid-lowering drugs. However, PET imaging is expensive, invasive and not widely available for all patients, necessitating alternative means to assess brain Aβ accumulation. OBJECTIVES: In this study, we measured levels of Aβ42, Aβ40 and Aβ38 in saliva samples from cognitively unimpaired older adults (n=93; 61.7 % female; mean age = 70.1 ± 6.6 years) using the

A neural index reflecting the amount of cognitive resources available during memory encoding: A model-based approach.

Humans have a limited amount of cognitive resources to process various cognitive operations at a given moment. The Source of Activation Confusion model of episodic memory proposes that resources are consumed during each processing, and once depleted, they need time to recover gradually. This has been supported by a series of behavioral findings in the past. However, the neural substrate of the resources is not known. In the present study, over an existing electroencephalogram data set of a free recall task (Kahana et al., 2022), we provided a neural index reflecting the amount of cognitive res

The Theta Paradox: 4-8 Hz EEG Oscillations Reflect Both Sleep Pressure and Cognitive Control.

Human electroencephalographic (EEG) oscillations characterize specific behavioral and vigilance states. The frequency of these oscillations is typically sufficient to distinguish a given state; however, theta oscillations (4-8 Hz) have instead been found in near-opposite conditions of drowsiness during sleep deprivation and alert cognitive control. While the latter has been extensively studied and is often referred to as "frontal midline theta," (fmTheta) the former has been investigated far less but is considered a marker for sleep pressure during wake. In this study we investigated to what e

Biotypes of deeply phenotyped depressed patients reflect signatures of adverse childhood experience and depressive cognitive biases.

BACKGROUND: Negative Cognitive Styles (NCS) are key features of depression contributing to severe clinical outcomes by sustaining negative affect. However, depression is clinically heterogeneous, reflecting complex neurobiological and environmental interactions. Characterizing heterogeneity using multimodal data could help identify mechanisms mapping onto different phenotypes and discover high-impact biomarkers. METHODS: Using a stability-based relative clustering validation pipeline, 344 depressed patients (135 major depressive disorder, 209 bipolar disorder) were stratified based on multimo

The relationship between sleep quality and depression: Critical roles of reflective and brooding rumination and sleep-related cognition.

There is a complex interplay between sleep problems and depression. This study explored the possible effects of rumination and dysfunctional beliefs about sleep on the relationship between sleep quality and depression. A cross-sectional survey of 1240 Chinese adults was conducted to assess the possible relationships. The results showed a chain mediating effect of reflection rumination and brooding rumination on the relationship between sleep quality and depression, accounting for 38.91% of the total variance. A moderating role of unreasonable attitudes about sleep was also discovered among the
