Cytopathic effects in MDBK cell lines after adhesion and internalization of Corynebacterium pseudotuberculosis biovar ovis.

Caseous lymphadenitis is a contagious disease of chronic evolution affecting mainly sheep, goats, and other domestic animals. The virulence of its etiological agent, Corynebacterium pseudotuberculosis, depends on the activity of enzymes, such as phospholipase D, and its waxy lipid mycolic acid layer, which induces cytotoxicity on the tissue cells of the host. Phospholipase D and mycolic acids contribute to inflammation, edema, and bacterium spread during abscess development. Here, we assessed the adhesion and internalization capacity of six isolates of C. pseudotuberculosis biovar ovis in a no

Spontaneous Pasteurella pneumonia in adult laboratory goats complicated by superinfection with Corynebacterium pseudotuberculosis and Muellerius capillaris.

Three cases of Pasteurella pneumonia in laboratory goats were diagnosed during a 21 month period. Two goats died acutely and a third was euthanized after four weeks of unsuccessful therapy. Pasteurella multocida was isolated from the lung lesions of all three goats. Corynebacterium pseudotuberculosis and Muellerius capillaris also were present in the lungs of the third goat. Pasteurella infections were associated with fibrino-hemorrhagic-necrotizing pleuritis and pneumonia, while lungs superinfected with Corynebacterium also contained microabcesses. The gross and histologic lesions are describ

An integrated structural proteomics approach along the druggable genome of Corynebacterium pseudotuberculosis species for putative druggable targets.

BACKGROUND: The bacterium Corynebacterium pseudotuberculosis (Cp) causes caseous lymphadenitis (CLA), mastitis, ulcerative lymphangitis, and oedema in a number of hosts, comprising ruminants, thereby intimidating economic and dairy industries worldwide. So far there is no effective drug or vaccine available against Cp. Previously, a pan-genomic analysis was performed for both biovar equi and biovar ovis and a Pathogenicity Islands (PAIS) analysis within the strains highlighted a large set of proteins that could be relevant therapeutic targets for controlling the onset of CLA. In the present wo

A novel approach for an immunogen against Corynebacterium pseudotuberculosis infection: An Escherichia coli bacterin expressing phospholipase D.

Corynebacterium pseudotuberculosis is the causative agent of caseous lymphadenitis (CLA) in small ruminants. There is still needed an immunoprophylaxis model, which induces a protective and sustained immune response against the bacteria. In this study, we evaluated a recombinant Escherichia coli bacterin expressing the recombinant phospholipase D (rPLD) protein, the most relevant virulence factor of C. pseudotuberculosis, as a potential vaccine formulation. E. coli BL21 (DE3) Star strain was used for rPLD protein expression and was then inactivated by formaldehyde. Four groups with 10 Balb/c m

Corynebacterium pseudotuberculosis is an important animal pathogen, which is also able to infect humans. An optimal treatment of infections with this pathogen is not available today and consequently, more research is necessary to understand the infection process. Here, we present a combined -omics and bioinformatics approach to characterize C. pseudotuberculosis 12CS0282. The genome sequence of strain 12CS0282 was determined, analyzed in comparison with the available 130 C. pseudotuberculosis sequences and used as a basis for proteome analyses. In a reverse vaccinology approach, putative vacci

Genome sequence of Corynebacterium pseudotuberculosis biovar equi strain 258 and prediction of antigenic targets to improve biotechnological vaccine production.

Corynebacterium pseudotuberculosis is the causative agent of several veterinary diseases in a broad range of economically important hosts, which can vary from caseous lymphadenitis in sheep and goats (biovar ovis) to ulcerative lymphangitis in cattle and horses (biovar equi). Existing vaccines against C. pseudotuberculosis are mainly intended for small ruminants and, even in these hosts, they still present remarkable limitations. In this study, we present the complete genome sequence of C. pseudotuberculosis biovar equi strain 258, isolated from a horse with ulcerative lymphangitis. The genome

Corynebacterium pseudotuberculosis phospholipase D targets mitochondrial sphingomyelin and induces NLRP3-GSDMD axis-mediated pyroptosis in macrophages to promote infection.

Infection by Corynebacterium pseudotuberculosis (Cp), a facultative intracellular bacterium, causes mainly purulent inflammation and chronic granulomas in animals and humans. Pyroptosis is a proinflammatory form of programmed cell death that is important in response to pathogen infection. The role of pyroptosis in Cp infection and the related mechanisms are still unclear. Here, we reveal that Cp infection induces pyroptosis in macrophages. The inhibition of pyroptosis by disulfiram (DSF) or dimethyl fumarate (DMF) decreases the pathogenicity of Cp in mice and prevents the escape of this pathog

Quantitative Proteomic Analysis Reveals Changes in the Benchmark Corynebacterium pseudotuberculosis Biovar Equi Exoproteome after Passage in a Murine Host.

Corynebacterium pseudotuberculosis biovar equi is the etiologic agent of ulcerative lymphangitis. To investigate proteins that could be related to the virulence of this pathogen, we combined an experimental passage process using a murine model and high-throughput proteomics with a mass spectrometry, data-independent acquisition (LC-MSE) approach to identify and quantify the proteins released into the supernatants of strain 258_equi. To our knowledge, this approach allowed characterization of the exoproteome of a C. pseudotuberculosis equi strain for the first time. Interestingly, the recovery

Spatial analysis and molecular typing of Corynebacterium pseudotuberculosis in Korean native goats show a cohesive lineage with distributed multidrug resistance.

Caseous lymphadenitis (CLA), caused by Corynebacterium pseudotuberculosis, is endemic in Korean native goats, but relationships among antimicrobial resistance (AMR), spatial distribution, and population structure remain unclear. We profiled AMR and multidrug resistance (MDR) in 286 pld-positive isolates from CLA lesions of 1177 goats in the Jeolla provinces, South Korea, integrating spatial analysis with five-locus multilocus sequence typing (MLST) and phylogenetics. Susceptibility to 12 antimicrobials was determined by disk diffusion, and MDR was defined as resistance to ≥ 3 antimicrobial cla
