A Review of the Role of Food and the Food System in the Transmission and Spread of Ebolavirus.

The current outbreak of Ebola virus disease (EVD) centered in West Africa is the largest in history, with nearly ten times more individuals contracting the disease than all previous outbreaks combined. The details of human-to-human and zoonotic ebolavirus transmission have justifiably received the largest share of research attention, and much information exists on these topics. However, although food processing-in the form of slaughtering and preparing wildlife for consumption (referred to as bushmeat)-has been implicated in EVD outbreaks, the full role of food in EVD spread is poorly understo

Bundibugyo Virus Disease: Diagnostics and Medical Countermeasures for a Neglected Ebolavirus.

Bundibugyo virus disease, caused by Bundibugyo virus (Orthoebolavirus bundibugyoense), is a severe human Ebola disease with substantial mortality, unresolved reservoir ecology, limited diagnostic implementation, and no licensed vaccines or therapeutics specifically approved for this Orthoebolavirus species. The May 2026 public health emergency in the Democratic Republic of the Congo and Uganda renewed the need for a focused synthesis of Bundibugyo virus-specific diagnostics and medical countermeasures. This review synthesizes the peer-reviewed literature, preprints, and official public-health

A Small-Particle Aerosol Model of Ebolavirus Zaire Infection in Ferrets.

The Ebola virus (EBOV) causes severe disease in humans, and animal models are needed to evaluate the efficacy of vaccines and therapeutics. While non-human primate (NHP) and rodent EBOV infection models have been well characterized, there is a growing need for an intermediate model. Here, we provide the first report of a small-particle aerosol (AE) EBOV ferret model and disease progression compared with the intramuscular (IM) EBOV ferret model. EBOV infection of ferrets by either route resulted in uniform lethality in 5-6.5 days post infection (dpi) in a dose-dependent manner, with IM-infected

The crux and crust of ebolavirus: Analysis of genome sequences and glycoprotein gene.

The recent 2013-15 epidemic of Ebola virus disease (EVD) has initiated extensive sequencing and analysis of ebolavirus genomes. All ebolavirus genomes available until December 2014 have been collated and analyzed in this study to obtain phylogenetic relationship and uncover the variations amongst them. The terminal 'leader' and 'trailer' nucleotide sequences of the genomes were omitted and analysis of the intermediate region accommodating the sole seven genes (hepta-CDS region) of the virus showed relative stability of the genome, including the ones isolated from the current epidemic. The geno

AMP-activated protein kinase is required for the macropinocytic internalization of ebolavirus.

Identification of host factors that are needed for Zaire Ebolavirus (EBOV) entry provides insights into the mechanism(s) of filovirus uptake, and these factors may serve as potential antiviral targets. In order to identify novel host genes and pathways involved in EBOV entry, gene array findings in the National Cancer Institute's NCI-60 panel of human tumor cell lines were correlated with permissivity for EBOV glycoprotein (GP)-mediated entry. We found that the gene encoding the γ2 subunit of AMP-activated protein kinase (AMPK) strongly correlated with EBOV transduction in the tumor panel. The

Epidemiology of Sudan ebolavirus disease cases in Uganda, January-April, 2025.

OBJECTIVES: On January 30, 2025, the Ministry of Health, Uganda (MoH), announced an outbreak of Sudan ebolavirus disease (SVD). We describe the epidemiological characteristics and transmission dynamics of the SVD outbreak in Uganda during January-April, 2025. METHODS: SVD cases were identified using MoH case definitions and were classified as confirmed if positive on RT-qPCR or probable if an SVD-related death was reported with epidemiological linkage but without laboratory confirmation. Cases were identified through contact tracing, community and health facility alerts, and active search. Da

Comparison of Zaire ebolavirus realtime RT-PCRs targeting the nucleoprotein gene.

In last five years, the Africa has faced two outbreaks of Zaire ebolavirus. These outbreaks have been the largest so far, and latest outbreak is still ongoing and affecting the Democratic Republic of the Congo. We tested in parallel three different Zaire ebolavirus (EBOV) realtime RT-PCRs targeting the nucleoprotein gene (EBOV NP-RT-qPCRs) described by Trombley et al. (2010); Huang et al. (2012) and Weidmann et al. (2004). These assays are used regularly in diagnostic laboratories. The limit of detection (LOD), intra-assay repeatability using different matrixes, sensitivity and specificity wer

Protocol for quantification of intercellular connection transmission in ebolavirus infections using ImageJ.

Previous work demonstrates that ebolaviruses can spread to neighboring cells through intercellular connections. Here, we present a protocol to quantify the intercellular spread of ebolaviruses via immunofluorescence. We describe steps for cell plating, Bundibugyo virus infection, and adding a neutralizing antibody. We detail procedures for quantitative microscopy assay using ebolavirus immunodetection. Strong virus accumulation around the plasma membrane leads to high fluorescence signal preventing quantification of viral spread based on signal intensity. This protocol minimizes the impact of

Comparison and Evaluation of Real-Time Taqman PCR for Detection and Quantification of Ebolavirus.

Given that ebolavirus causes severe and frequently lethal disease, its rapid and accurate detection using available and validated methods is essential for controlling infection. Real-time reverse-transcription PCR (RT-PCR) has proven to be an invaluable tool for ebolaviruses diagnostics. Many assays with different targets have been developed, but they have not been externally compared or validated, and limits of detection are not uniformly reported. Here we compared and evaluated the sensitivity, reproducibility and specificity of 23 in-house assays under the same conditions. Our results showe

Ebolavirus: Comparison of Survivor Immunology and Animal Models in the Search for a Correlate of Protection.

Ebola viruses are enveloped, single-stranded RNA viruses belonging to the Filoviridae family and can cause Ebola virus disease (EVD), a serious haemorrhagic illness with up to 90% mortality. The disease was first detected in Zaire (currently the Democratic Republic of Congo) in 1976. Since its discovery, Ebola virus has caused sporadic outbreaks in Africa and was responsible for the largest 2013-2016 EVD epidemic in West Africa, which resulted in more than 28,600 cases and over 11,300 deaths. This epidemic strengthened international scientific efforts to contain the virus and develop therapeut

Bats as putative Zaire ebolavirus reservoir hosts and their habitat suitability in Africa.

The genus Ebolavirus comprises some of the deadliest viruses for primates and humans and associated disease outbreaks are increasing in Africa. Different evidence suggests that bats are putative reservoir hosts and play a major role in the transmission cycle of these filoviruses. Thus, detailed knowledge about their distribution might improve risk estimations of where future disease outbreaks might occur. A MaxEnt niche modelling approach based on climatic variables and land cover was used to investigate the potential distribution of 9 bat species associated to the Zaire ebolavirus. This viral

Modeling the Disease Course of Zaire ebolavirus Infection in the Outbred Guinea Pig.

BACKGROUND: Rodent models that accurately reflect human filovirus infection are needed as early screens for medical countermeasures. Prior work in rodents with the Zaire species of Ebola virus (ZEBOV) primarily used inbred mice and guinea pigs to model disease. However, these inbred species do not show some of the important features of primate ZEBOV infection, most notably, coagulation abnormalities. METHODS: Thirty-six outbred guinea pigs were infected with guinea pig-adapted ZEBOV and examined sequentially over an 8-day period to investigate the pathologic events that lead to death. RESULT

Discovery of three small-molecule inhibitors targeting Ebolavirus genome replication and transcription.

The 2013 Ebola virus (EBOV) outbreak was the largest in history. Despite recent advances in both vaccines and monoclonal antibody therapies, 12 years later, EBOV still poses a substantial threat. Previously, we published a ligand discovery pipeline combining in silico screening of compounds with a robust and rapid EBOV minigenome assay for early-stage inhibitor validation at Biological Safety Level 2. Here, we present the further use of this pipeline to identify three compounds that also inhibit EBOV minigenome transcription and replication. They are efficacious in the nM range, exhibited low

Ebola disease stigma: mixed methods insights from the 2022 Sudan ebolavirus outbreak.

OBJECTIVES: Ebola disease stigma hinders outbreak control and recovery by deterring care-seeking and driving social exclusion. Although this phenomenon is well recognised, gaps remain in understanding how stigma emerges and operates in outbreak settings, limiting the development of effective reduction strategies. The objective of this study was to examine the drivers, manifestations and public health impacts of stigma following the 2022-2023 Sudan ebolavirus outbreak in central Uganda. DESIGN: We conducted a cross-sectional, mixed-methods survey to assess Ebola disease stigma in June 2024. S

Chimpanzee adenovirus vaccine generates acute and durable protective immunity against ebolavirus challenge.

Ebolavirus disease causes high mortality, and the current outbreak has spread unabated through West Africa. Human adenovirus type 5 vectors (rAd5) encoding ebolavirus glycoprotein (GP) generate protective immunity against acute lethal Zaire ebolavirus (EBOV) challenge in macaques, but fail to protect animals immune to Ad5, suggesting natural Ad5 exposure may limit vaccine efficacy in humans. Here we show that a chimpanzee-derived replication-defective adenovirus (ChAd) vaccine also rapidly induced uniform protection against acute lethal EBOV challenge in macaques. Because protection waned over

Design and characterization of protective pan-ebolavirus and pan-filovirus bispecific antibodies.

Monoclonal antibodies (mAbs) are an important class of antiviral therapeutics. MAbs are highly selective, well tolerated, and have long in vivo half-life as well as the capacity to induce immune-mediated virus clearance. Their activities can be further enhanced by integration of their variable fragments (Fvs) into bispecific antibodies (bsAbs), affording simultaneous targeting of multiple epitopes to improve potency and breadth and/or to mitigate against viral escape by a single mutation. Here, we explore a bsAb strategy for generation of pan-ebolavirus and pan-filovirus immunotherapeutics. Fi

Serologic Markers for Ebolavirus Among Healthcare Workers in the Democratic Republic of the Congo.

Healthcare settings have played a major role in propagation of Ebola virus (EBOV) outbreaks. Healthcare workers (HCWs) have elevated risk of contact with EBOV-infected patients, particularly if safety precautions are not rigorously practiced. We conducted a serosurvey to determine seroprevalence against multiple EBOV antigens among HCWs of Boende Health Zone, Democratic Republic of the Congo, the site of a 2014 EBOV outbreak. Interviews and specimens were collected from 565 consenting HCWs. Overall, 234 (41.4%) of enrolled HCWs were reactive to at least 1 EBOV protein: 159 (28.1%) were serorea

Design of a multiepitopic Zaire ebolavirus protein and its expression in plant cells.

The recent Ebola virus disease (EVD) outbreaks make the development of efficacious and low cost vaccines against Ebola virus (EBOV) an urgent goal. Multiepitopic vaccines allow a rational design rendering vaccines able to induce proper immune responses in terms of polarization and potency. In addition, the pathogen variants can be easily covered by including epitopes conserved among relevant isolates. Other important aspects to consider in vaccination are the costs associated to production, distribution, and administration of the vaccine. Plants provide an advantageous platform for this purpos

Engineered bispecific antibodies achieve broad and potent protection against multiple ebolavirus species.

Ebolaviruses, including EBOV, SUDV, and BDBV, cause severe hemorrhagic fever, yet currently licensed monoclonal antibody (mAb) therapies against EBOV lack cross-species efficacy. While mAbs offer high specificity, favourable safety profiles, and durable serum persistence, their susceptibility to viral escape highlights the need for broader, more resilient antibody strategies. Bispecific antibodies (bsAbs), which concurrently target non-overlapping epitopes, have the potential to enhance neutralization potency, expand strain coverage, and mitigate mutation-driven resistance. In this study, we e
