Self-assembled elastin-like polypeptide fusion protein coacervates as competitive inhibitors of advanced glycation end-products enhance diabetic wound healing.

Chronic and non-healing skin wounds are some of the most significant complications in patients with advanced diabetes. A contributing mechanism to this pathology is the non-enzymatic glycation of proteins due to hyperglycemia, leading to the formation of advanced glycation end products (AGEs). AGEs bind to the receptor for AGEs (RAGE), which triggers pro-inflammatory signals that may inhibit the proliferative phase of wound healing. Soluble forms of RAGE (sRAGE) may be used as a competitive inhibitor of AGE-mediated signaling; however, sRAGE is short-lived in the highly proteolytic wound envir

Development of an Elastin-like Polypeptide-Based Nucleic Acid Delivery System Targeted to EGFR+ Bladder Cancer Cells Using a Layer-by-Layer Approach.

Nucleic acid (NA)-based therapies are revolutionizing biomedical research through their ability to control cellular functions at the genetic level. This work demonstrates a versatile elastin-like polypeptide (ELP) carrier system using a layer-by-layer (LbL) formulation approach that delivers NA cargos ranging in size from siRNA to plasmids. The components of the system can be reconfigured to modulate the biochemical and biophysical characteristics of the carrier for engaging the unique features of the biological target. We show the physical characterization and biological performance of LbL EL

Engineering of dual-responsive elastin-like polypeptide (ELP) systems for precision delivery of calcitonin gene-related peptide (CGRP): Impact of chain length on performanc.

BACKGROUND: Calcitonin gene-related peptide (CGRP) holds significant potential for tissue regeneration and bone repair; however, its clinical application is limited by a rapid systemic half-life and the difficulty of maintaining localized therapeutic concentrations. This study aimed to design and characterize a dual-responsive fusion protein system, utilizing Elastin-Like Polypeptides (ELPs) as a smart delivery vehicle to achieve spatiotemporal control of CGRP release. METHODS: We engineered four fusion protein variants ELPn-CGRP, where n = 20, 40, 60, 80) by modulating the ELP chain length t

Unlocking the potential of Extensin Signal peptide and Elastin-like polypeptide tag fused to Shigella dysenteriae's IpaDSTxB to improve protein expression and purification in Nicotiana tabacum and Medicagosativa.

Plants are often seen as a potent tool in the recombinant protein production industry. However, unlike bacterial expression, it is not a popular method due to the low yield and difficulty of protein extraction and purification. Therefore, developing a new high efficient and easy to purify platform is crucial. One of the best approaches to make extraction easier is to utilize the Extensin Signal peptide (EXT) to translocate the recombinant protein to the outside of the cell, along with incorporating an Elastin-like polypeptide tag (ELP) to enhance purification and accumulation rates. In this re

An Elastin-Like Protein for Injectable Tissue-Like Network.

Protein-based tissue fillers are promising for adipose regeneration due to their biocompatibility and tunable properties. Inspired by elastin's structure, we designed a low-immunogenicity "multiple-domain-collective elastin-like" (MDCE) protein for injectable tissue-like networks. The MDCE protein was composed of thermal-responsive elastin-like peptides (disordered domains), polyalanine enrichment (functional ordered domains), and zwitterionic polypeptides (hydrophilic domains). This design endows the MDCE protein hydrogel with in situ injectability and antifouling properties, resulting in a t

Enhanced yields and purity: a robust optimization framework for the production of multifunctional elastin-like proteins.

Elastin-Like Proteins (ELP) are highly versatile biomaterials with significant potential in tissue-engineering, due to their design flexibility, mechanical properties and tunable stimuli responsiveness. However, expression conditions reported in literature do not always translate to new ELP fusion proteins. This study establishes a methodology to optimize expression conditions for modular ELP constructs with intermittent peptide domains. Response Surface Methodology is employed to systematically optimize protein expression of three distinct ELP constructs (containing either RGD domains, hepari

AI-validated fusion proteins for local inhibition of interleukin-17A.

IL-17A is a potent immunomodulatory cytokine that is upregulated in many disorders characterized by chronic inflammation, including autoimmune-mediated dry eye observed in patients with Sjögren's Disease. Existing IL-17A inhibiting therapies are delivered systemically and have been associated with increased infection rates in patients. Inhibiting IL-17A signaling locally, only in affected tissues, may provide therapeutic benefits without compromising systemic immune functions. To address this unmet need, we have developed a local IL-17A inhibiting therapy by fusing HAP, a peptide which binds a

Elastin-Derived Peptides in the Central Nervous System: Friend or Foe.

Elastin is one of the main structural matrix proteins of the arteries, lung, cartilage, elastic ligaments, brain vessels, and skin. These elastin fibers display incredible resilience and structural stability with long half-life. However, during some physiological and pathophysiological conditions, elastin is prone to proteolytic degradation and, due to the extremely low turnover rate, its degradation is practically an irreversible and irreparable phenomenon. As a result of elastin degradation, new peptides called elastin-derived peptides (EDPs) are formed. A growing body of evidence suggests t

Leech bionic Hirudin fusion protein prodrug loaded microneedles for long-term inhibition of thrombosis.

The clinical use of anticoagulant drugs for thrombosis is limited by short half-life and bleeding risk, requiring frequent administration and close monitoring. Here, inspired by leech mouthparts, we present a hirudin-based fusion protein prodrug incorporated with microneedles (MNs) for long-term antithrombotic therapy to achieve a simplified dosing regimen, continuous protection, and on-demand antithrombotic bioactivity recovery. Genetic-engineered anticoagulant prodrugs are precisely customized by fusing hirudin to elastin-like polypeptides with adjustable assembly characteristics. The introd

Actomyosin Fluctuations Couple Temperature to Intracellular Transport and ELPV Phase Separation.

How living cells transduce thermal perturbations into altered intracellular organization remains a fundamental biophysical question. Here, using single-particle tracking of quantum dot (QD) diffusion and endocytic vesicle transport, we show that temperature-enhanced transport is not a simple passive solvent effect; instead, heating enhances active cytoplasmic dynamics mediated by myosin-II-dependent actomyosin activity. While active fluctuations enhance random QD diffusion, they also accelerate active vesicular transport by reducing its mechanical resistance. Using thermoresponsive elastin-lik

Elastin: relation of protein and gene structure to disease.

The elastic properties of many tissues such as the lung, dermis, and large blood vessels are due to the presence of elastic fibers in the extracellular space. These fibers have been shown by biochemical and ultrastructural analysis to be comprised of two distinct components, a more abundant amorphous component and the microfibrillar component. The microfibrillar component is found in 10- to 12-nm fibrils which are located primarily around the periphery of the amorphous component but, to some extent, interspersed within it. The protein, elastin, makes up the highly insoluble amorphous component

Temperature-Triggered Nanoscale Morphological Transitions in a Synthetic Intrinsically Disordered Protein.

Peptide self-assembly and liquid-liquid phase separation (LLPS), often mediated by intrinsically disordered regions (IDRs), are natural mechanisms that translate protein molecular features into complex nano- and mesoscale architectures. Although the thermodynamics and kinetics of these processes are well understood, synthetic materials integrating both functionalities remain rare. Inspired by the conserved IDR-assembly domain (AD) architecture of amyloidogenic proteins, we hypothesized that modular recombinant constructs combining LLPS-capable IDRs with β-sheet-forming ADs could generate mater

Design and evaluation of a bifunctional fusion polypeptide for targeted delivery of tigecycline against drug-resistant Klebsiella pneumoniae.

The overuse of antibiotics and the continuous evolution of drug-resistant bacteria present substantial obstacles to the clinical treatment of Carbapenem-resistant Klebsiella pneumoniae (CRKP) infections. This study reports the rational design of a bifunctional fusion polypeptide, S-thanatin (Ts)-functionalized elastin-like carrier (Ts-ELP), which integrates the antimicrobial peptide (AMP) Ts with a human-derived elastin‑like polypeptides (ELPs). This designer polypeptide serves as a smart carrier, capable of efficiently encapsulating the broad-spectrum antibiotic tigecycline (Tig) via reversib

Single nucleotide polymorphisms and domain/splice variants modulate assembly and elastomeric properties of human elastin. Implications for tissue specificity and durability of elastic tissue.

Polymeric elastin provides the physiologically essential properties of extensibility and elastic recoil to large arteries, heart valves, lungs, skin and other tissues. Although the detailed relationship between sequence, structure and mechanical properties of elastin remains a matter of investigation, data from both the full-length monomer, tropoelastin, and smaller elastin-like polypeptides have demonstrated that variations in protein sequence can affect both polymeric assembly and tensile mechanical properties. Here we model known splice variants of human tropoelastin (hTE), assessing effect

Nano-Enabled Therapeutics: Novel Strategies for Preeclampsia Treatment.

Preeclampsia (PE), a pregnancy-specific disorder characterized by hypertension and placental dysfunction, remains a leading cause of maternal and fetal morbidities worldwide. Recent advances in nanomedicine offer promising therapeutic strategies by targeting placental pathologies. Studies have demonstrated that in PE mouse models, the regulation of key disease-related genes (such as sFlt1 and VEGF) using siRNA- or mRNA-loaded carriers (eg, lipid nanoparticles, exosomes, or elastin-like polypeptides) can effectively alleviate PE symptoms. This review summarizes the progress in nanoparticle-base

Engineering Elastin-Like Peptide-Based Nanoparticles displaying Variable Domain of the Heavy Chain of Heavy-Chain-Only Antibodies for SARS-CoV-2 Neutralization.

Elastin-like polypeptides (ELPs) are self-assembling recombinant biopolymers that can be precisely engineered to display functional targeting ligands. In this study, we developed ELP-based nanoparticles (NPs) displaying the variable domain of the heavy chain of heavy-chain-only antibodies (VHHs) targeting the SARS-CoV-2 spike protein. By tuning VHH selection, multivalency, and surface display density, we created targeted ELP NPs capable of blocking entry of spike-protein-presenting virus-like particles (VLPs) and live viruses, with subnanomolar IC50 values, significantly outperforming the mono

Optimizing delivery of an anti-cytomegalovirus inhibitory peptide using a cell-penetrating peptide.

Cytomegalovirus (CMV) is a beta herpesvirus that persists quiescently in healthy individuals but causes severe disease in immunocompromised patients and congenitally infected newborns. Side effects posed by the currently available antivirals necessitate the development of new antivirals with improved safety profiles. This study aims to optimize the potency of an anti-CMV peptide (P10) mimicking conserved region 2 of tegument pp150, fused with an elastin-like polypeptide (ELP). Our previous study has demonstrated that ELP-P10 inhibits murine cytomegalovirus (MCMV) growth in vitro and in vivo wi
