Evodiamine suppresses capsaicin-induced thermal hyperalgesia through activation and subsequent desensitization of the transient receptor potential V1 channels.

Evodiae fructus (EF), a fruit of Evodia rutaecarpa Bentham, has long been used as an analgesic drug in traditional Chinese and Japanese medicine. However, the underlying molecular mechanism of its pharmacological action is unclear. Here, using calcium imaging, whole-cell patch-clamp recording, and behavioral analysis, we investigated the pharmacological action of EF and its principal compound, evodiamine, on the transient receptor potential (TRP) V1 channels. Dorsal root ganglion (DRG) neurons and TRPV1- or TRPA1-transfected human embryonic kidney-derived (HEK) 293 cells were used for calcium

An evodiamine derivative inhibits osteoclast differentiation and protects against OVX-induced bone loss in mice.

Evodiamine is a biologically active alkaloid extracted from the fruit of the traditional Chinese medicine Evodia rutaecarpa (Juss.) Benth. (Fructus Evodiae, Wuzhuyu). However, due to its lipophilic chemical structure, low water solubility results in poor bio-availability, which limits its broader application. 3-Amino-10-hydroxyl-evodiamine (E2) was a water-soluble derivative of evodiamine with good anti-tumour bioactivity previously developed by our team; however, its anti-osteoporosis activity remains unclear. This study demonstrates that E2 inhibits the maturation of osteoclasts and bone res

Up-regulation of CYP1A1 by rutaecarpine is dependent on aryl hydrocarbon receptor and calcium.

Rutaecarpine is a quinazolinocarboline alkaloid isolated from a traditional Chinese medicinal fruit, Evodia rutaecarpa. In the present study, we investigated the effect of rutaecarpine on CYP1A1 expression mediated by [Ca(2+)] and the AhR pathway in mouse hepatoma Hepa-1c1c7 cells. Rutaecarpine also significantly increased CYP1A1 enzyme activity and mRNA and protein levels. Rutaecarpine markedly induced XRE and AhR binding activity. CH-223191, an AhR antagonist, blocked the rutaecarpine-induced CYP1A1 enzyme activity and mRNA and protein expression. In addition, rutaecarpine remarkably induced

Evodiamine attenuates neurological impairment by inducing PANoptosis in microglia after severe traumatic brain injury.

Severe traumatic brain injury (sTBI) lacks effective therapies despite its high morbidity and mortality in young adults. Here, we demonstrate that evodiamine (EVO), a natural alkaloid from Evodia rutaecarpa, significantly improves neurological outcomes in a murine sTBI model (controlled cortical impact). EVO administration (0.3-3.0 mg/kg, intraperitoneal) attenuated cerebral edema, preserved neuronal ultrastructure, and reduced neuroinflammation by suppressing microglial activation and cytokine release. Mechanistically, while EVO showed no direct neuroprotection against H2O2, CoCl2, or oxygen-

Discovery of Novel Hsp90 Inhibitor-Evodiamine Derivative Conjugates as Potent and Orally Active Antitumor Agents for Colon Cancer Therapy.

Natural products (NPs) and their derivatives have long been important components of antitumor drugs. Nevertheless, traditional natural product derivatives are generally limited by tumor-targeting deficiency and toxicity to normal tissue. Particularly, there remains a lack of effective strategies to improve in vivo antitumor efficacy and reduce the toxicity of natural product derivatives. Herein, a series of heat shock protein 90 (Hsp90) inhibitor-evodiamine (an NP from Evodiae fructus) conjugates were rationally designed, binding extracellular Hsp90 (eHsp90), which could mediate endocytosis to

Evodiamine Alleviates 2,4-Dinitro-1-Chloro-Benzene-Induced Atopic Dermatitis-like Symptoms in BALB/c Mice.

Evodiamine is an alkaloid found in Evodia fruits, a traditional Chinese medicine. Preclinical studies have demonstrated its anti-inflammatory and neuroprotective properties. The 2,4-dinitro-1-chloro-benzene (DNCB) was used to test the effects of evodiamine on a chemically induced atopic dermatitis-like model in BALB/c mice. Evodiamine significantly lowered serum immunoglobulin E levels, which increased as an immune response to the long-term application of DNCB. Several atopic dermatitis-like skin symptoms induced by DNCB, including skin thickening and mast cell accumulation, were suppressed by

Protective effects of calcitonin gene-related peptide-mediated evodiamine on guinea-pig cardiac anaphylaxis.

Previous investigations have indicated that the pharmacological effects of evodiamine, a major alkaloidal component of the dried, unripe fruit of Evodia rutaecarpa Bentham (Rutaceae), are associated with stimulation of calcitonin gene-related peptide (CGRP) release and CGRP prevents cardiac anaphylactic injury. In the present study, the protective effects of evodiamine on cardiac anaphylaxis were examined. Presensitized guinea-pig hearts challenged with specific antigen (bovine serum albumin) caused a marked decrease in coronary flow, left ventricular pressure and its derivatives (+/-dp/dt(max

Evodiamine alleviates cancer cachexia-induced muscle atrophy by inhibiting STAT3 signaling pathway.

Cancer-associated cachexia (CAC) is a multifactorial syndrome characterized by progressive skeletal muscle and adipose tissue wasting, for which effective therapies remain limited. Here, we report that evodiamine (EVO), a bioactive alkaloid derived from Evodia rutaecarpa, markedly alleviates bladder cancer-associated cachexia in both in vivo and in vitro models. In T24 tumor-bearing mice, EVO significantly attenuated body weight loss and preserved skeletal muscle and fat mass without affecting primary tumor growth. In C2C12 myotubes exposed to tumor-conditioned medium, EVO dose-dependently pre
