Under lock and key: spatiotemporal regulation of WASP family proteins coordinates separate dynamic cellular processes.

WASP family proteins are nucleation promoting factors that bind to and activate the Arp2/3 complex in order to stimulate nucleation of branched actin filaments. The WASP family consists of WASP, N-WASP, WAVE1-3, WASH, and the novel family members WHAMM and JMY. Each of the family members contains a C-terminus responsible for their nucleation promoting activity and unique N-termini that allow for them to be regulated in a spatiotemporal manner. Upon activation they reorganize the cytoskeleton for different cellular functions depending on their subcellular localization and regulatory protein int

Mother-child separations among homeless and housed families receiving public assistance in New York City.

We examined the incidence, characteristics, and predictors of separations of children from mothers in 543 poor families receiving public assistance, 251 of whom had experienced homelessness during the previous 5 years. Forty-four percent of the homeless mothers and 8% of housed mothers were separated from one or more children. A total of 249 children were separated from 110 homeless families and 34 children from 23 housed families. Children were placed with relatives and in foster care but were rarely returned to their mothers. Maternal drug dependence, domestic violence, and institutionalizat

Preschool sibling interaction in separated and married families: are same-sex pairs or older sisters more sociable?

Two factors have been associated with the quality of sibling relationships: the sex of the children and their family circumstances. Yet the data on each issue are complex. This study examines the sex constellations of sibling pairs in two groups, one of which had experienced a major family disruption (parental separation), with the aim of assessing both influences more fully. Two 1-hour home observations were conducted on 20 preschool sibling dyads of separated parents and 24 pairs in which the parents were married. In both groups the sex configuration of the sibling pairs was important--same-

The ongoing legacy of Indigenous family separation: Long-term outcomes of child welfare involvement among American Indian and First Nations youth.

BACKGROUND: Despite policies designed to preserve family structures, Indigenous children in the U.S. and Canada remain disproportionately removed from their homes and communities. This systemic removal represents an ongoing form of historical trauma with intergenerational consequences, yet research on its long-term psychosocial effects remains limited. OBJECTIVES: To examine the prevalence of foster care and adoption among a sample of Indigenous youth and their associations with psychosocial outcomes in early adulthood. PARTICIPANTS AND SETTING: Data come from a community-based participatory

Forced immigration-related family separation and adverse childhood experiences: effects on mental health among Latinx immigrant youth in the United States.

INTRODUCTION: First-generation Latinx immigrant youth face risks for experiencing unique immigration-related traumas, which could exacerbate their mental health. This study aimed to investigate the potential influence of forced immigration-related family separation on the association between traditional adverse childhood experiences (ACEs) and mental health (i.e., depression, anxiety, and PTSD symptoms) among Latinx immigrant youth from the Northern Triangle (NT; El Salvador, Guatemala, and Honduras). METHODS: Primary surveys assessing forced immigration-related family separation, ACEs, and m

KIF11 prevents retinal endothelial ferroptosis in familial exudative vitreoretinopathy by inhibiting phosphorylation-driven PRDX1 phase separation.

Familial exudative vitreoretinopathy is a hereditary disorder predominantly affecting infants and young children, often leading to severe vision loss. Approximately 40% of patients carry mutations in Norrin/β-catenin pathway genes. Nevertheless, the downstream pathogenic mechanisms remain unclear. Here, by using bulk RNA sequencing and single-cell RNA sequencing analyses, we identify KIF11 as a key downstream effector in retinal endothelial cells. Lentivirus-mediated KIF11 overexpression partially restores vascular defects in endothelial cell-specific Ctnnb1 knockout mice. Functional and multi

Relational Family Therapy as an Aid Toward Resolving the Trauma of Sexual Abuse in Childhood in the Process of Separation in the Couple Relationship.

Relational family therapy (RFT) is a psycho-organic therapeutic approach that combines intrapsychic dynamics with interpersonal mechanisms and a systemic perspective. This integrative model is based on the assumption that the repetitive patterns of relationships in early childhood repeat in adulthood. It often happens that a sexually abused individual will later unconsciously be attracted to someone with whom they will relive the abusive atmosphere and dynamics from childhood. The purpose of this contribution is to present the RFT approach for dealing with sexual abuse. We will present a case

A family study of manic-depressive (bipolar I) disease. Is it a distinct illness separable from primary unipolar depression?

OBJECTIVE: To determine whether bipolar I illness is autonomous or part of a multifactorial continuum with unipolar depression. In this study, we compare familial bipolar I illness and depression among three groups: probands with bipolar I disorder; probands with primary unipolar disorder; and controls. We also examine a continuum of severity between psychotic and nonpsychotic patients with bipolar I disorder. Considerable data suggest that bipolar I illness is distinct from unipolar illness as regards epidemiology, familial psychiatric illness, course, response to treatment, and biologic find

Dynamic action potential clamp predicts functional separation in mild familial and severe de novo forms of SCN2A epilepsy.

De novo variants in SCN2A developmental and epileptic encephalopathy (DEE) show distinctive genotype-phenotype correlations. The two most recurrent SCN2A variants in DEE, R1882Q and R853Q, are associated with different ages and seizure types at onset. R1882Q presents on day 1 of life with focal seizures, while infantile spasms is the dominant seizure type seen in R853Q cases, presenting at a median age of 8 months. Voltage clamp, which characterizes the functional properties of ion channels, predicted gain-of-function for R1882Q and loss-of-function for R853Q. Dynamic action potential clamp, t

Mental Health of Early and Prolonged Parent-Child Separated Children in Chinese Immigrant Families: A Qualitative Study With Parent-Child Dyads.

Parent-child separation is a global crisis that has affected child and family well-being. Many low-income Chinese immigrant families in the US practice early and prolonged separation, sending US-born infants ("satellite babies") to China for extended family care and reuniting them around school age. However, little research has explored child mental health post-reunification, especially based on parent-child dyad reports. The present qualitative study explored child mental health based on key informative interviews from affected families (N= 24 parent-child dyads; N girls=12; Mean child age= 1

Effects of Historical Coercive Control, Historical Violence, and Lawyer Representation on Post-Separation Male Partner Violence Against Mother Litigants Who Participated in Adversarial Family Court Proceedings.

The primary objective of this study was to test the effects of historical male partner violence and lawyer representation on post-separation male partner violence and coercive control against mother litigants participating in adversarial family court proceedings. Toward this end, staff at two women's shelters administered a questionnaire to 40 former residents who met the sample selection criteria. Two findings are noteworthy. First, there was a decrease in mother litigant reports of post-separation physical violence requiring a visit to a hospital. Second, post-separation male partner coerciv

Mutations in the seipin and AGPAT2 genes clustering in consanguineous families with Berardinelli-Seip congenital lipodystrophy from two separate geographical regions of Brazil.

Berardinelli-Seip congenital lipodystrophy (BSCL) is characterized by a near total congenital absence of fat and predisposition to develop diabetes mellitus. In this study, we investigated the presence of mutations in the Seipin and 1-acylglycerol phosphate acyltransferase 2 (AGPAT2) genes in 32 affected subjects with BSCL from 17 consanguineous pedigrees living in two separate geographical regions, the northeastern and southeastern regions, of Brazil. All, except one, of the 22 BSCL subjects from 15 families living in the northeastern region were found to have a homozygous 669insA mutation in

Three-year change in the wellbeing of orphaned and separated children in institutional and family-based care settings in five low- and middle-income countries.

BACKGROUND: With more than 2 million children living in group homes, or "institutions", worldwide, the extent to which institution-based caregiving negatively affects development and wellbeing is a central question for international policymakers. METHODS: A two-stage random sampling methodology identified community representative samples of 1,357 institution-dwelling orphaned and separated children (OSC) and 1,480 family-dwelling OSC aged 6-12 from 5 low and middle income countries. Data were collected from children and their primary caregivers. Survey-analytic techniques and linear mixed eff

Congenital central hypoventilation syndrome: Severe disease caused by co-occurrence of two PHOX2B variants inherited separately from asymptomatic family members.

Congenital Central Hypoventilation Syndrome (CCHS) is a rare disease characterized by autonomic nervous system dysregulation. Central hypoventilation is the most prominent and clinically important presentation. CCHS is caused by mutations in paired-like homeobox 2b (PHOX2B) and is inherited in an autosomal dominant pattern. A co-occurrence of two asymptomatic PHOX2B variants with a classical CCHS presentation highlights the importance of clinical PHOX2B testing in parents and family members of all CCHS probands. Despite being an autosomal dominant disease, once a polyalanine repeat expansion m

Associations of parental alcohol use disorders and parental separation with offspring initiation of alcohol, cigarette and cannabis use and sexual debut in high-risk families.

BACKGROUND AND AIMS: Parental alcohol use disorders (AUDs) and parental separation are associated with increased risk for early use of alcohol in offspring, but whether they increase risks for early use of other substances and for early sexual debut is under-studied. We focused on associations of parental AUDs and parental separation with substance initiation and sexual debut to (1) test the strength of the associations of parental AUDs and parental separation with time to initiation (age in years) of alcohol, tobacco and cannabis use and sexual debut and (2) compare the strength of associatio

Separating Family-Level and Direct Exposure Effects of Smoking During Pregnancy on Offspring Externalizing Symptoms: Bridging the Behavior Genetic and Behavior Teratologic Divide.

Maternal smoking during pregnancy (MSDP) has been robustly associated with externalizing problems and their developmental precursors in offspring in studies using behavioral teratologic designs (Wakschlag et al., Am J Public Health 92(6):966-974, 2002; Espy et al., Dev Psychol 47(1):153-169, 2011). In contrast, the use of behavior genetic approaches has shown that the effects commonly attributed to MSDP can be explained by family-level variables (D'Onofrio et al., Dev Psychopathol 20(01):139-164, 2008). Reconciling these conflicting findings requires integration of these study designs. We util

Targeted Next-Generation Sequencing Identifies Separate Causes of Hearing Loss in One Deaf Family and Variable Clinical Manifestations for the p.R161C Mutation in SOX10.

Hearing loss is the most common sensory deficit in humans. Identifying the genetic cause and genotype-phenotype correlation of hearing loss is sometimes challenging due to extensive clinical and genetic heterogeneity. In this study, we applied targeted next-generation sequencing (NGS) to resolve the genetic etiology of hearing loss in a Chinese Han family with multiple affected family members. Targeted sequencing of 415 deafness-related genes identified the heterozygous c.481C>T (p.R161C) mutation in SOX10 and the homozygous c.235delC (p.L79Cfs∗3) mutation in GJB2 as separate pathogenic mutati
