Does Excessive Tonic Inhibition Cause Unstable Feedback in Parkinson's Disease?

Parkinson's disease is characterized by decreased dopamine in the basal ganglia, which causes excessive tonic inhibition of the thalamus. This excessive inhibition seems to explain decreased motor vigor in Parkinson's disease, but whether it causes tremor remains unclear. This paper explores how inhibition may change closed-loop characteristics to determine how this established pathophysiology could produce tremor. First, single-neuron simulations establish the rate and time characteristics of increased tonic inhibition by adjusting the membrane time constant. These characteristics point towar

Interdependent feedback regulation of breathing by the carotid bodies and the retrotrapezoid nucleus.

The retrotrapezoid nucleus (RTN) regulates breathing in a CO2 - and state-dependent manner. RTN neurons are glutamatergic and innervate principally the respiratory pattern generator; they regulate multiple aspects of breathing, including active expiration, and maintain breathing automaticity during non-REM sleep. RTN neurons encode arterial PCO2 /pH via cell-autonomous and paracrine mechanisms, and via input from other CO2 -responsive neurons. In short, RTN neurons are a pivotal structure for breathing automaticity and arterial PCO2 homeostasis. The carotid bodies stimulate the respiratory pat

A potential overestimation of CO2 physiological effects on evapotranspiration.

It is generally believed that CO2 physiological forcing can partially mitigate land surface drying under global warming by reducing stomatal conductance and evapotranspiration. Most of this type of study focuses on the direct regulation by vegetation physiology, overlooking interactive feedback from the atmosphere. Using fully coupled earth system model simulations, we find that the physiological benefit may have been optimistically overestimated. Vegetation-induced energy change may in turn further affect atmospheric vapor pressure deficit (VPD), exerting extra evapotranspiration demand indir

Biofeedback for treatment of irritable bowel syndrome.

BACKGROUND: Irritable bowel syndrome (IBS) is a prevalent condition that currently lacks highly effective therapies for its management. Biofeedback has been proposed as a therapy that may help individuals learn to exert conscious control over sympatho-vagal balance as an indirect method of symptom management. OBJECTIVES: Our primary objective was to assess the efficacy and safety of biofeedback-based interventions for IBS in adults and children. SEARCH METHODS: We searched the Cochrane Inflammatory Bowel Disease (IBD) Group Specialized Trials Register, Cochrane Central Register of Controlled

An Examination of Cardiac Vagal Control Indices and Cognitive Stress Appraisal in Cigarette Smokers.

Identifying factors that influence how individuals who smoke cigarettes respond to stress is important as stress is a risk factor for smoking and its maintenance. This study examined the modulatory role of cardiac vagal control (CVC), a physiological correlate of self-regulation, on cognitive stress appraisal processes of adults who smoke. Sixty daily cigarette smokers were randomized to receive positive or negative feedback during a modified Trier Social Stress Test. Pre- and post-task stress appraisals were assessed and resting and reactivity CVC measures were computed. Moderated regression

Comparing feedback learning and arousal responses in Down, Fragile X, and Williams syndromes.

Mechanisms underlying feedback learning in intellectual disability (ID) remain poorly understood. Down syndrome (DS), Fragile X syndrome (FXS), and Williams syndrome (WS) are genetic syndromes associated with ID, with distinct attention and arousal regulation profiles. Pupil dilation is a well-established index of feedback processing in typical development; little is known about these processes in non-social learning in these syndromes. Participants with DS (n = 13), FXS (n = 13), WS (n = 27), and typically developing (TD) individuals (n = 56) aged 6-60 years completed a reward contingency rev

The masticatory system--an overview.

Masticatory muscle physiology has been evaluated mostly from electromyographic recordings. However, electromyography coupled with jaw-tracking devices has provided much more information of the correlation between jaw movements and muscle activity. Knowledge of how the mandible moves during mastication has greatly influenced procedures in clinical dentistry. The aim of this overview is to give basic description of the classical studies of the physiology, function and neural control principles of the mastication. Mastication is the action of breaking down of food, preparatory to deglutition. Thi

Mechanisms of emmetropization and what might go wrong in myopia.

Studies in animal models and humans have shown that refractive state is optimized during postnatal development by a closed-loop negative feedback system that uses retinal image defocus as an error signal, a mechanism called emmetropization. The sensor to detect defocus and its sign resides in the retina itself. The retina and/or the retinal pigment epithelium (RPE) presumably releases biochemical messengers to change choroidal thickness and modulate the growth rates of the underlying sclera. A central question arises: if emmetropization operates as a closed-loop system, why does it not stop my

Visual feedback of the tongue influences speech adaptation to a physical modification of the oral cavity.

Studies examining sensorimotor adaptation of speech to changing sensory conditions have demonstrated a central role for both auditory and somatosensory feedback in speech motor learning. The potential influence of visual feedback of oral articulators, which is not typically available during speech production but may nonetheless enhance oral motor control, remains poorly understood. The present study explores the influence of ultrasound visual feedback of the tongue on adaptation of speech production (focusing on the sound /s/) to a physical perturbation of the oral articulators (prosthesis alt

Closed-loop brain-machine-body interfaces for noninvasive rehabilitation of movement disorders.

Traditional approaches for neurological rehabilitation of patients affected with movement disorders, such as Parkinson's disease (PD), dystonia, and essential tremor (ET) consist mainly of oral medication, physical therapy, and botulinum toxin injections. Recently, the more invasive method of deep brain stimulation (DBS) showed significant improvement of the physical symptoms associated with these disorders. In the past several years, the adoption of feedback control theory helped DBS protocols to take into account the progressive and dynamic nature of these neurological movement disorders tha

Real time BOLD functional MRI neuro-feedback affects functional connectivity.

Functional connectivity in the resting state based on BOLD functional MRI (fMRI) has been used mainly to observe brain networks while subjects 'do nothing'. The same principle, however, can be used for any other steady-state brain condition. In this study, we compared the connectivity of the motor area during simple finger tapping with and without real time neuro-feedback of the activation in the hand motor area. The presence of the neuro-feedback not only induces correlations between the visual and motor areas, but also increases basal ganglia involvement and bilateral motor cortex connectivi

Origin of threshold behaviour in psoriatic skin.

The mechanisms that lead to the psoriatic morphology are not fully elucidated. Several lines of evidence suggest that the positive feedback between keratinocytes and immunocytes plays a key role in the development of the lesions. On the other hand, little information is available on the negative regulatory controls that maintain a new homoeostasis level in psoriatic skin. We suggest here that the interplay of these two contrary feedbacks is likely to entail a hysteresis effect and that psoriasis is likely to be interpreted as a critical phenomenon characterized by a catastrophic shift of the s

Divergent effects of hyperandrogenemia on the female reproductive neuroendocrine axis under different physiological conditions.

The hypothalamic-pituitary-gonadal (HPG) axis coordinates reproductive function and incorporates gonadal sex steroid feedback to regulate gonadotropin secretion in a homeostatic manner. In females, ovarian estrogens and progestins act as the primary negative feedback signals to suppress luteinizing hormone (LH) release, whereas in males, androgens, such as testosterone (T), typically provide this feedback. However, androgens can also exert feedback effects on the female HPG axis, particularly in conditions where circulating T levels exceed that of the normal female physiological range. Three c

Positive Feedback Mechanisms in Pain Modulation and Chronic Pain.

Feedback controls play important roles in biological systems, from the cellular level to systemic levels. Positive and negative feedback are two major feedback mechanisms. In this review, I will mainly investigate the positive feedback system involved in pain transmission, modulation and plasticity. At the level of the spinal cord, descending facilitation from different regions of supraspinal structures exert facilitatory effects on spinal nociceptive transmission. At the level of the cortex, positive feedback at synaptic and cellular levels contribute to long-term potentiation (LTP) of synapt

Central neural control of the cardiovascular system: current perspectives.

This brief review, which is based on a lecture presented at the American Physiological Society Teaching Refresher Course on the Brain and Systems Control as part of the Experimental Biology meeting in 2015, aims to summarize current concepts of the principal mechanisms in the brain that regulate the autonomic outflow to the cardiovascular system. Such cardiovascular regulatory mechanisms do not operate in isolation but are closely coordinated with respiratory and other regulatory mechanisms to maintain homeostasis. The brain regulates the cardiovascular system by two general means: 1) feedforw

Effects of sensorimotor delays and muscle force capacity limits on the performance of feedforward and feedback control in animals of different sizes.

Animals rely on feedforward and feedback control for perturbation responses. When comparing terrestrial mammals of different sizes, we generally find that several features that affect perturbation responses change-larger animals have longer sensorimotor time delays, heavier body segments, and proportionally weaker muscles. We used simple computational models to study how control of fast perturbation responses is constrained by two limitations-sensorimotor delays and muscle force capacity-as a function of animal size. We developed two tasks representing common perturbation response scenarios in
