In vivo performance of a bilayer wrap to prevent abdominal adhesions.

There is a high prevalence of intra-abdominal adhesions following bowel resection, which can result in chronic pain, bowel obstruction, and morbidity. Although commercial adhesion barriers have been widely utilized for colonic resections, these barriers do not prevent anastomotic leakage resulting from reduced healing of the anastomosis, which can result in long-term health problems. To address this limitation, we have developed an adhesive bilayer wrap with selective bioactivity to simultaneously prevent intra-abdominal adhesion formation and promote anastomotic healing. [Endoscopic treatment of anastomosis insufficiency and perforation in the esophagus with fibrin glue].

The incidence of anastomotic dehiscence following oesophageal or gastric resection reported in European studies varies between 3.5 and 25%. The outcome of these patients is usually not explained nor is there a relevant number of studies concerning the treatment of this situation. Oesophageal perforation is rare but is commonly combined with high morbidity and mortality. Operative treatment mostly means major surgical procedures. In our institution endoscopic treatment of these defects has been established since 1985. Following the endoscopic diagnosis, the extent of the defect becomes radiolog

The effect of fibrin formulation on cell migration in an in vitro tendon repair model.

BACKGROUND: The purpose of this study was to determine the effect of fibrinogen concentration on cell viability and migration in a tissue culture tendon healing model. METHODS: Forty-eight canine flexor digitorum profundus tendons were randomly divided into three groups. In each group the tendons were lacerated and repaired augmented with a canine bone marrow stromal cell seeded fibrin interposition patch using either 5 mg/ml fibrinogen and 25 U/ml thrombin (physiological as a control), 40 mg/ml fibrinogen and 250 U/ml thrombin (low adhesive), or 80 mg/ml fibrinogen and 250 U/ml thrombin (hig

[Fibrin sealing, a concept for early elective endoscopic therapy].

The technique of fibrin adhesion is a standard haemostatic procedure which uses a two-component adhesive consisting of highly concentrated fibrinogen and thrombin, which when mixed, form a fibrin clot via the third phase of the blood coagulation cascade, and thereby induce tissue-reparative mechanisms. In a new endoscopic procedure for the treatment of bleeding ulcer, the components of the fibrin adhesive are simultaneously injected into the tissue beneath and around the lesion where they form a mechanically resilient and stable fibrin clot. This clot is firmly anchored into the tissue. In con

Complication caused by use of fibrin glue in vessel transposition for trigeminal neuralgia.

A 64-year-old man underwent microvascular decompression of the left superior cerebellar artery (SCA) for left trigeminal neuralgia (TN) using a sling of Teflon tape fixed to the tentorium with fibrin glue. The TN disappeared immediately after surgery, but recurred unusually rapidly at 2 weeks later at the same intensity as before. Second surgery revealed the SCA was suspended from the tentorium, but the trigeminal nerve was stretched and displaced superolaterally because of adhesion to the superior petrosal vein. The adhesion was thought to involve the fibrin glue used during the sling retract

Sticking to the facts: a systematic review of fibrin glue for pilonidal disease.

BACKGROUND: Pilonidal disease occurs when hair invading the natal cleft causes inflammation and abscess formation. Opinions vary on best practice, and most procedures have considerable morbidity and high recurrence rates of 6-40%. OBJECTIVES: This study systematically reviews the use of fibrin glue in the treatment of pilonidal disease. Outcomes measured were healing time and recurrence rate. DATA SOURCE: ScienceDirect and PubMed databases were searched for relevant literature, yielding seven papers including five small trials. Effect of dextran and hyaluronic acid on the development of postoperative peritoneal adhesions in experimental animals.

The development of postoperative peritoneal adhesions was studied in rats and rabbits, the frequency of adhesions in the experimental model used being very high. In the development of an adhesion, fibrin seems to be an important contributor to the bridge between different tissues. Dextran, which modifies the fibrin network and makes it more susceptible to lysis, was used as a possible prophylactic agent, but we found no difference between treated and control groups. It is concluded that the stimulus for fibrin formation in the peritoneal adhesions using this atraumatic model was too strong to

Development of biodegradable hyper-branched tissue adhesives for the repair of meniscus tears.

UNLABELLED: Meniscus tears are one of the most commonly occurring injuries of the knee joint. Current meniscus repair techniques are challenging and do not bring fully satisfactory results. Tissue adhesives are a promising alternative, since they are easy to apply and cause minimal tissue trauma. In this study, a series of amphiphilic copolymers based on polyethylene glycol, trimethylene carbonate and citric acid were synthesized and subsequently end-functionalized with hexamethylene diisocyanate to form reactive adhesive materials. The shear adhesive strength of the networks to bovine meniscu

Snake venom-defined fibrin architecture dictates fibroblast survival and differentiation.

Fibrin is the provisional matrix formed after injury, setting the trajectory for the subsequent stages of wound healing. It is commonly used as a wound sealant and a natural hydrogel for three-dimensional (3D) biophysical studies. However, the traditional thrombin-driven fibrin systems are poorly controlled. Therefore, the precise roles of fibrin's biophysical properties on fibroblast functions, which underlie healing outcomes, are unknown. Here, we establish a snake venom-controlled fibrin system with precisely and independently tuned architectural and mechanical properties. Employing this de

The effect of fibrin glue patch in an in vitro model of postdural puncture leakage.

UNLABELLED: We studied the possibility of stopping a continuing transdural leakage with fibrin glue, a biologic adhesive, in an in vitro model. The model was made by sealing the bottom of a tube filled with saline to a height of 50 cm with a human lyophilized dural specimen. Dural punctures were performed with a 17-gauge Tuohy needle. The needle was then withdrawn, and 0.8 mL of fibrin glue was injected through the same needle to seal the defect. The column was refilled 3 min after sealing. The pressure in the intrathecal chamber was measured during the procedure. Macroscopic and microscopic h

Advances in the use of adhesives in ophthalmology.

PURPOSE OF REVIEW: To summarize the known uses of available medical tissue adhesives in the management of diseases of the anterior segment, highlighting recent developments in the field. RECENT FINDINGS: Human fibrin glues may be used in place of cyanoacrylate tissue adhesives in the treatment of progressive corneal thinning and small perforations, potentially resulting in less corneal and conjunctival inflammatory reaction. Additional currently proposed uses of fibrin glues in ophthalmic surgery include minimizing sutures in recurrent pterygium surgery, forniceal reconstruction, amniotic mem

[Operation technic and after care in lens injuries].

Extensive experimental research proved that the healing of lens capsule wounds can be supported effectively by application of a fibrin tissue adhesive. The result is a circumscribed scar in an otherwise clear lens. The clinical application of the method was successful as well. Preliminary results have been published and a review covering all patients treated is in preparation. This paper is devoted to the indications, techniques and postoperative treatment. The application of the fibrinogen tissue adhesive is indicated if spontaneous healing cannot be expected or has failed and the major parts

Characterization of mechanics and cytocompatibility of fibrin-genipin annulus fibrosus sealant with the addition of cell adhesion molecules.

There is an unmet clinical need for a biomaterial sealant capable of repairing small annulus fibrosus (AF) defects. Causes of these defects include painful intervertebral disc herniations, microdiscectomy procedures, morbidity associated with needle puncture injury from discography, and future nucleus replacement procedures. This study describes the enhancements of a fibrin gel through genipin crosslinking (FibGen) and the addition of the cell adhesion molecules (CAMs), fibronectin and collagen. The gel's performance as a potential AF sealant is assessed using a series of in vitro tests. FibGe

In vitro evaluation of decontamination effects on mechanical properties of fibrin membrane.

Background: Tissue engineering has been investigated as a potential method for healing traumatized tissues. Biomaterials are material devices or implants used to repair or replace native body tissues and organs. The present study was conducted to evaluate the effects of decontamination methods on biological/mechanical properties and degradation/adhesion test of the platelet-rich fibrin (PRF) membranes to compare these properties with intact membranes as a biological biomaterial. Methods: The in vitro degradation tests were conducted by placing the equal sizes of (i) intact PRF membrane, (ii) P

Histopathological effects of fibrin glue on penile fracture in a rat model.

PURPOSE: To evaluate both histopathological effects and potential clinical application of fibrin glue on the penile cavernosal tissue. METHODS: Experimental penile fracture was formed by incising from the proximal dorsal side of the penis in 32 Wistar Albino rats. The rats were randomly assigned to four main groups of eight animals each. In the control group, the incision was not repaired and it was left to secondary healing. In the primary repair group, the incision was primarily repaired. In the fibrin glue group, glue was applied only to the incision. In the final group, fibrin glue was ap
