BACKGROUND: Phegopteris connectilis is a perennial fern distributed throughout the Northern Hemisphere, including Eurasia and North America. Although Phegopteris connectilis is widely distributed, local populations occurring in the Western Suwałki Lakeland region of North Eastern Poland are considered critically endangered, highlighting the need for efficient propagation methods to support conservation efforts. Because this species reproduces apogamously, the development of a reliable in vitro propagation system is important for both conservation and research. This study aimed to determine opt

Phegopteris decursivepinnata includes diploids, tetraploids, and triploid hybrids based on x = 30. We obtained polyploid progeny from triploid hybrids through selfing and crossing experiments. Triploids occasionally formed well-filled spores. The mean occurrence frequencies of well-filled and germinated spores were 2.8% and 0.8%, respectively. Viable spores that succeeded in germinating were regarded as unreduced, triploid spores, because the resulting gametophytes yielded triploid (2n = 86-92) and hexaploid (2n = 170-184) progeny in both isolated and mixed cultures of gametophytes. The triplo

BACKGROUND: To examine whether the previously observed excess risk of male germ cell cancer in a cohort of car-manufacturing workers can be attributed to occupational activities inside and/or outside the car industry.
METHODS: A nested case-control study among workers in six plants included 205 cases of germ cell cancer and 1,105 controls, individually matched by year of birth (±2 years). Job periods of the individual occupational histories were coded based on the International Standard Classification of Occupations (ISCO) and the industrial classification of economic activities (NACE). Odds

BACKGROUND: Wheat germ, one of the byproducts of flour milling, contains abundant physiologically active components. Globulins in wheat germ are a class of high-quality functional proteins and have received widespread attention. However, the composition of wheat germ globulin( WGG)and the structure of the typical proteins have not yet been proved. The immunological activities and immune mechanisms of the WGG have not yet been revealed in vivo.
OBJECTIVES: The proteomic analysis of WGG and the structure simulation of typical proteins were studied. The immunoregulatory effects of WGG on immunos

Lichen secondary metabolites can function as allelochemicals and affect the development and growth of neighboring bryophytes, fungi, vascular plants, microorganisms, and even other lichens. Lichen overgrowth on bryophytes is frequently observed in nature even though mosses grow faster than lichens, but there is still little information on the interactions between lichens and bryophytes. In the present study, we used extracts from six lichen thalli containing secondary metabolites like usnic acid, protocetraric acid, atranorin, lecanoric acid, nortistic acid, and thamnolic acid. Meiosis is of crucial importance in agriculture because it provides the genetic variation required for breeding programs. Many cytological techniques have thus been developed to study meiosis in plants. Maize provides a powerful system for cytogenetic analysis due to its large conspicuous chromosomes. This protocol outlines a method for the simultaneous visualization of specific chromosomes or their segments, alongside proteins involved in meiosis in maize germ cells. The workflow combines fluorescence in situ hybridization (FISH) of oligonucleotide probe cocktails (oligo-FISH) covering entire

This paper studies the effect of plant peptides of thionine Ns-W2 extracted from seeds of fennel flower (Nigella sativa) and β-purothionine from wheat germs (Triticum kiharae), as well as a synthetic antimutagen (crown-compound), on the expression of several genes involved in the.control of cellular homeostasis, processes of carcinogenesis, and radiation response in human rhabdomyosarcoma cells (RD cells), T-lymphoblastoid cell line Jurkat, and blood cells. All of these agents acted as antimutagens-anticarcinogens, reducing the expression of genes involved in carcinogenesis (genes of families

Effective management of microbial biofilms holds significance within food and medical environments. Candida albicans, an opportunistic fungus, forms mucosal biofilms closely linked to candidiasis and drug-resistant infections due to their drug tolerance. Morphologic change from yeast to filamentous cells is a key virulence factor and a prerequisite for biofilm development. This study investigated the anti-fungal and antibiofilm activities of 20 flavonoids against C. albicans. With their known antioxidant capabilities, flavonoids hold promise in combating infections associated with biofilms. Am

Kelp, brown macroalgae in the order Laminariales, provide ecosystem services vital to ocean biodiversity. However, kelp forests worldwide are declining due to abiotic stressors such as ocean warming. In this study, we present results from high-resolution confocal microscopy and in vivo imaging system imaging using protocols developed to visualize kelp gametophyte cells exposed to heat-stress treatments. Imaging revealed chloroplast mislocalization, fragmentation, and subsequent loss of chloroplasts in heat-stressed gametophyte cells. Additionally, nuclei exhibited fragmentation and a progressi

Humans develop immunoglobulin G (IgG) antibodies to the foods they consume. In the context of food allergy, allergen-specific IgG antibodies can sequentially class-switch to pathogenic IgE. However, the mechanism underlying the antigenicity of food proteins remains uncharacterized. Here, we identified convergent antibodies arising from different antibody gene rearrangements that bind to the immunodominant peanut allergen Ara h 2 and characterized allelic and junctional constraints on germline antibody specificity. Structurally, we found similar epitope-paratope interactions across multiple gen

Plants respond to insect feeding with a number of defense mechanisms. Using maize genotypes derived from Antiquan germ plasm that are resistant to Lepidoptera, we have demonstrated that a unique 33-kD cysteine proteinase accumulates in the whorl in response to larval feeding. The abundance of the proteinase increased dramatically at the site of larval feeding after 1 hr of infestation and continued to accumulate for as long as 7 days. The 33-kD cysteine proteinase was most abundant in the yellow-green portion of the whorl-the normal site of larval feeding and the tissue that has the greatest i

Protein and glycan antigens synthesized by gut microbes stimulate circulating and secreted antibody production. Despite continuous exposure of hosts to the plant glycans that constitute dietary fiber, it remains unclear whether these foreign structures induce mucosal immune responses. We report that humans and mice generate antibodies specific for common glycans in plant foods. Oral exposure to individual fiber types induced T cell-independent, glycan-specific IgM and IgA. The induction of anti-fiber antibodies required colonization by particular microbes, since germ-free mice and mice harbori

Bioactive peptides from plant sources are promising alternatives to conventional antifungal therapies. Here, we report the isolation and partial characterisation of a low-molecular-weight antifungal peptide (∼2.5 kDa) from Curcuma longa rhizomes. Bioactivity-guided purification yielded a fraction with potent activity against Candida albicans (MIC = 0.7 μg/mL). In silico analysis identified candidate peptides, including fragments from curcumin synthase 1, consistent with the observed mass.1³C NMR revealed signals compatible with carbon environments associated with residues such as tryptophan an

During natural infection malaria parasites are injected into the bloodstream of a human host by the bite of an infected female Anopheles mosquito. Both asexual and mature sexual stages of Plasmodium circulate in the blood. Asexual forms are responsible for clinical malaria while sexual stages are responsible for continued transmission via the mosquitoes. Immune responses generated against various life cycle stages of the parasite have important roles in resistance to malaria and in reducing malaria transmission. Phagocytosis of free merozoites and erythrocytic asexual stages has been well stud

BACKGROUND: Candida albicans is one of the most common causative of opportunistic infections. Treatment of candidiasis is challenging considering the few antifungal drugs available and the increase in resistance. Antimicrobial photodynamic therapy (aPDT) is a recently developed therapeutic option that combines a non-toxic photosensitizer (PS) and light to kill the microbial pathogens. Targeting virulence, defined as the ability of a pathogen to cause overt disease, represents another attractive target for the development of novel antifungal agents. Thymophylla pentachaeta (DC.) Small var. bele

Diploid gametes are usually applied to produce triploids of Populus [originating from first-division restitution (FDR), second-division restitution (SDR), and postmeiotic restitution (PMR) 2n eggs]. Three types of 2n gametes transmitted different parental heterozygosities in Populus. Failed spindle formation and no chromosomal separation to opposite poles during meiosis I mean that FDR 2n gametes carry nonsister chromatids that are potentially heterozygous. By contrast, SDR 2n gametes result from failed sister chromatid separation in meiosis II, and therefore, they carry sister chromatid that

BACKGROUND: Fungal phytopathogens pose a significant threat to global crop yields. The overuse of pesticides leads to pesticide-resistant pathogens, environmental contamination and risks to human health. Consequently, there is an urgent need for effective and ecofriendly antifungal strategies to address these challenges.
RESULTS: Herein, ZnO-ZIF-8 is synthesized via an in situ growth method to integrate the antifungal property of zinc oxide (ZnO) and the control release property of zeolitic imidazolate framework (ZIF)-8. Results suggest that the inhibition rates of ZnO, ZIF-8 and ZnO-ZIF-8 ag

Life cycles with a prolonged haploid phase are thought to be correlated with greater rates of selfing and asexual reproduction. In red algae, recent population genetic studies have aimed to test this prediction but have mostly focused on marine species with separate sexes. We characterized the reproductive system of the obligately monoicous (i.e., hermaphroditic) freshwater red alga Batrachospermum gelatinosum and predicted that we would find genetic signatures of uniparental reproduction because of its haploid-diploid life cycle. We sampled 18 sites and genotyped 311 gametophytes with 10 poly
