Gnathostoma spinigerum infection is endemic in Thailand and many Asian countries. Current diagnosis is the skin test and enzyme-linked immunosorbent assay (ELISA) for IgG antibody against the G. spinigerum third-stage larvae (L3), but cross-reactivity is common. We evaluated the sensitivity and specificity of anti-G. spinigerum L3 IgG subclass antibodies for diagnosis of 43 patients with gnathostomiasis. The majority of patients with gnathostomiasis (91%) had eosinophilia. While the anti-G. spinigerum L3 IgG1 antibody provided the highest sensitivity (98%), the anti-G. spinigerum L3 IgG2 antib

Gnathostomiasis is a zoonotic parasitosis endemic in many Asian and some Latin American countries. Most human infections are caused by Gnathostoma spinigerum in Asia and Gnathostoma binucleatum in the Americas, and recently, imported cases have been increasing among travelers returning from endemic regions. Confirmation of the clinical diagnosis relies largely on serologic tests, with a G. spinigerum-antigen-based immunoblot currently being the diagnostic method of choice. However, we repeatedly experienced that sera from patients with clinically suspected American gnathostomiasis gave negativ

Human gnathostomiasis is a food-born parasitic disease of relative importance in many countries in Southeast Asia. It is caused by several species of nematodes of the genus Gnathostoma. In Mexico is an emerging public health problem since 1970, when first cases were reported. Until today, larval morphometric characters that have been proposed to differentiate between the three species of Gnathostoma present in this country, are not satisfactory. Recently, the presence of advanced third-stage larvae AdvL3 (infective form for humans) in freshwater fishes from Pantanos de Centla, Tabasco. was rec

We encountered a case of gnathostomiasis in a pregnant woman; she was successfully treated using ivermectin, and the baby was delivered without complications. Administration of ivermectin during pregnancy may be considered after evaluation of the number of weeks of pregnancy and the risk to mother and child.

Neurognathostomiasis is a severe form of human gnathostomiasis which can lead to disease and death. Diagnosis of neurognathostomiasis is made presumptively by using clinical manifestations. Immunoblotting, which recognizes antigenic components of molecular mass 21 kDa and 24 kDa in larval extracts of Gnathostoma spinigerum (Gs 21/24), has high sensitivity and specificity for diagnosis of neurognathostomiasis. However, only very small amounts of the Gs 21/24 antigens can be prepared from parasites harvested from natural or experimental animals. To overcome this problem, we recently produced a r

Aquatic snails, Pila ampullacea and Pomacea canaliculata were experimentally found to be suitable paratenic hosts for advanced third-stage larvae (L3) of the nematode Gnathostoma spinigerum, the causative parasite of gnathostomiasis in humans. G. spinigerum (L3) were found to be encapsulated in the tissue of the snail's foot and its internal organs. The infection, intensity and survival of third-stage larvae of G. spinigerum in both species of aquatic snails are described. This is the first evidence to reveal that not only vertebrates but also invertebrates (snails) can serve as paratenic host

BACKGROUND: Live intraocular nematode is a rare occurrence that is mostly reported in Southeast Asian countries. Common nematodes that are seen live in the eye are microfilaria, Gnathostoma, and Angiostrongylus. Approximately 12 cases of intraocular gnathostomiasis have been reported in the literature.
METHOD: Two cases of intraocular gnathostoma, removed by vitrectomy in the first case and by paracentesis in the second case, are reported. Morphologic study of the parasites in wet preparation was performed under dissecting microscope and fixed in Karnovosky's fixative. Light microscopic and s

The present study is concerned with a survey of the epidemiology and infectivity of Gnathostoma hispidum larvae in animals. A total or 3,478 pigs were examined in Hongze-Lake, Jiangsu; Po-Lake, Anhui; and Jiujiang, in Jiangxi Province. The infection rate of G. hispidum in pigs was 14.9%. The prevalence in pigs in the lake districts (26.4%) was considerably higher than in the mountainous districts (5.1%). The results of field surveys and experimental studies revealed that in China, 38 animal species served as the first and the second intermediate hosts and paratenic hosts of G. hispidum. Among

Since the 1908s gnathostomiasis has occurred sporadically in Japanese who have eaten loaches, fresh-water fish imported from southeast Asia. Symptoms include creeping skin eruptions and eosinophilia. The symptoms disappeared in most patients within three months after the onset and the patients then remained symptom-free. To elucidate the causative factors, we fed one monkey with 80 gnathostome larvae collected from loaches and autopsied the animal 466 days later. Fifty-one viable larvae (64%) were recovered from muscle; these were in the advanced third stage and were encapsulated.

Indirect fluorescent antibody test (IFAT) was performed on sections of Gnathostoma spinigerum advanced third-stage larva with gnathostomiasis, angiostrongyliasis, trichinosis, strongyloidiasis and cysticercosis sera. Positive fluorescence was observed with the first three sera. Fluorescence was associated with the anterior part of the esophagus, surface of the cuticle and cytoplasmic granules of the intestine. Absorption of sera with gnathostome antigen did not elicit fluorescence. The results suggest that substances secreted from the esophagus and intestine constitute antigens in excretory-se

In this work, the life cycle of Gnathostoma turgidum was studied both in natural temporary water bodies and under experimental conditions, with materials collected from the municipality of San Francisco Ixhuatán, Oaxaca. Adult nematodes inhabit the interstitial layers of the stomach in the definitive host (Didelphis virginiana) and release their eggs into the environment via faeces, typically in a dry environment. Under these conditions, the eggs remain quiescent until the onset of the rainy season. Development of early third-stage larvae occurs in cyclopoid copepods. Fry acts as transport and

BACKGROUND: Gnathostomiasis is an important zoonosis in tropical areas that is mainly caused by third-stage Gnathostoma spinigerum larvae (G. spinigerum L3).
OBJECTIVES: This study aimed to prove whether G. spinigerum L3 produces extracellular vesicles (EVs) and investigate human gene profiles related to the immune response against the larvae.
METHODS: We created an immune cell model using normal human peripheral blood mononuclear cells (PBMCs) co-cultured with the larvae for 1 and 3 days, respectively. The PBMCs were harvested for transcriptome sequencing analysis. The EV ultrastructure was

Human gnathostomiasis is a food-borne zoonotic helminthic infection widely reported in Latin America, Asia and Southeast Asia, particularly in Thailand. There are increasing reports of the parasite in countries where it is not endemic. A study of the survival drug-treated immature stage (STIM) of Gnathostoma spinigerum recovered from infected patients focused on their integument surface using scanning electron microscopy (SEM). STIM displayed a specific, characteristic head bulb, with a pair of large thick equal-sized trilobulated lips in the centre. Cephalic spines had eight transverse rows o

Gnathostomiasis is a helminthic infection caused by the third-stage larvae of nematodes of the genus Gnathostoma. The life cycle in humans starts with an enteric phase, with the worm perforating the gastric or intestinal mucosa to reach the peritoneal cavity and migrating through the human body. Subsequent penetration through the diaphragm may produce pleuropulmonary symptoms. We herein present a previously healthy 56-year-old Thai man from Southern Thailand who was an ex-smoker presented with chronic dry cough progressing to hemoptysis after consuming grilled swamp eels and freshwater fish. This study aims to describe some of the unknown pathological and parasitological traits of experimental feline gnathostomosis. Thirteen female cats were orally inoculated with 30 advanced third-stage Gnathostoma binucleatum larvae and were euthanized at various post-infection (p.i.) periods. Clinically, the cats presented with nausea, vomiting, abdominal pain and other nonspecific signs. None of the cats shed eggs in their fecal matter. One cat, euthanized at 6 months p.i., developed a fibrous vascular nodule 2-3 cm in diameter within its gastric wall. The nodule contained caverns filled with

Gnathostoma spinigerum is a causative agent of human gnathostomiasis, a common parasitic disease involving skin and visceral organs, especially the central nervous system. In this study, we identified a cDNA encoding a cathepsin L-like cysteine protease (GsCL1) from the lambdaZAP cDNA library of G. spinigerum advanced third-stage larva (aL3) and characterized the biochemical properties of the recombinant enzyme. The cloned cDNA of 1484bp encoded 398 amino acids which contained a typical signal peptide sequence (23 amino acids), a pro-domain (156 amino acids), and a mature domain (219 amino aci

ELISA was developed for the detection of IgG antibody in sera obtained from patients in Japan and in a foreign country. Gnathostoma doloresi adult antigen was less specific than G. spinigerum advanced third-stage larval antigen but their sensitivity were similar. Cross reactivity was observed in Toxocara canis-, Anisakis-, Paragonimus westermani- and Fasciola-infected sera when G. doloresi adult worms but not G. spinigerum advanced third-stage larvae were used as antigens.

Eosinophilic meningitis is defined as the presence of >10 eosinophils/μL in cerebrospinal fluid (CSF) or at least 10% eosinophils in the total CSF leukocyte count. Eosinophilic meningitis has been reported in two case series and two case reports in India till date and has not been reported in children below 15 years of age. We present two children with eosinophilic meningitis with peripheral eosinophilia and the proposed etiologic agents based on the clinical setting and their response to antihelminthic agents.
