TGM2 Aggravates Acute Pancreatitis by Impairing Macrophage Efferocytosis Through Inhibition of the STAT6-GAS6 Axis.

BACKGROUND: Acute pancreatitis (AP) is characterized by dysregulated inflammation, with macrophage dysfunction (impaired efferocytosis, pro/anti-inflammatory phenotype imbalance) exacerbating the disease. Current therapies are mostly supportive, highlighting the critical need for targeted interventions. METHODS: Transglutaminase 2 (TGM2) was identified via public transcriptomic analysis. Its function was validated in caerulein-induced AP mice and in vitro cell models; mechanisms were explored via Co-IP, ChIP, and dual-luciferase assays. A lactoferrin-modified, ROS-responsive LF-LNP system was

Proteomics Analyses of Small Extracellular Vesicles of Aqueous Humor: Identification and Validation of GAS6 and SPP1 as Glaucoma Markers.

Cataracts and glaucoma account for a high percentage of vision loss and blindness worldwide. Small extracellular vesicles (sEVs) are released into different body fluids, including the eye's aqueous humor. Information about their proteome content and characterization in ocular pathologies is not yet well established. In this study, aqueous humor sEVs from healthy individuals, cataracts, and glaucoma patients were studied, and their specific protein profiles were characterized. Moreover, the potential of identified proteins as diagnostic glaucoma biomarkers was evaluated. The protein content of

Axl contributes to efficient migration and invasion of melanoma cells.

Axl, a member of the TAM receptor family has been broadly suggested to play a key role in tumor metastasis. However, the function of Axl in the invasion and metastasis of melanoma, the most lethal skin cancer, remains largely unknown. In the present study, we found that melanoma cell lines present variable protein levels of Axl and Tyro3; interestingly, MerTK is not noted at detectable levels in any of tested MGP (metastatic growth phase) cell lines. Treatment with recombinant human Gas6 significantly activates Akt in the Axl-expressing WM852 and IgR3 lines but just slightly in WM1158. IgR3, W

Vitamin K-Dependent Proteins in Skeletal Development and Disease.

Vitamin K and Vitamin K-dependent proteins (VKDPs) are best known for their pivotal role in blood coagulation. Of the 14 VKPDs identified in humans to date, 6 play also important roles in skeletal biology and disease. Thus, osteocalcin, also termed bone Gla-protein, is the most abundant non-collagenous protein in bone. Matrix Gla protein and Ucma/GRP on the other hand are highly abundant in cartilage. Furthermore, periostin, protein S, and growth arrest specific 6 protein (GAS 6) are expressed in skeletal tissues. The roles for these VKDPs are diverse but include the control of calcification a

Functions of TAM Receptors and Ligands Protein S and Gas6 in Atherosclerosis and Cardiovascular Disease.

Atherosclerosis and cardiovascular disease are associated with high morbidity and mortality in industrialized nations. The Tyro3, Axl, and Mer (TAM) family of receptor tyrosine kinases is involved in the amplification or resolution of atherosclerosis pathology and other cardiovascular pathology. The ligands of these receptors, Protein S (PS) and growth arrest specific protein 6 (Gas6), are essential for TAM receptor functions in the amplification and resolution of atherosclerosis. The Axl-Gas6 interaction has various effects on cardiovascular disease. Mer and PS dampen inflammation, thereby pr

[At the intersection of immunology and oncology: TAM receptors in the regulation of immune responses and tumorigenesis-related processes].

TAM receptor tyrosine kinases (TYRO3, AXL, MER) and their ligands, protein S (PROS1) and growth inhibition-specific protein 6 (GAS6), play a key role in maintaining homeostasis and regulating the immune response through involvement in efferocytosis, i.e., phagocytic removal of apoptotic cells and suppression of the innate immune response. Thus, their dysfunction leads, among others, to the development of autoimmune diseases. In turn, excessive production of TAM receptors correlates with the invasive phenotype of cancer cells, metastasis, drug resistance, and poor prognosis for patients with ca

Plasma extracellular vesicle surface-located GAS6/PROS1 and CD39/CD73 attenuate inflammation.

Herein, we demonstrate that extracellular vesicles (EVs) present in healthy human plasma regulate macrophage activation by triggering MERTK-dependent signaling in a growth arrest-specific 6 (GAS6)- and protein S (PROS1)-dependent manner. Moreover, EVs regulate oxidative burst and degranulation in neutrophils and the PAMP-stimulated secretion of the pro-inflammatory cytokine interleukin (IL)-1β while enhancing the secretion of the anti-inflammatory cytokine transforming growth factor beta (TGF-β). Neutrophil regulation is mediated by the biochemical activity of the EV-exposed extracellular nucl

Expression levels of tam receptors and ligands in the testes of rats exposed to short and middle-term 2100 MHz radiofrequency radiation.

With advances in technology, the emission of radiofrequency radiation (RFR) into the environment, particularly from mobile devices, has become a growing concern. Tyro 3, Axl, and Mer (TAM) receptors and their ligands are essential for spermatogenesis and testosterone production. RFR has been shown to induce testicular cell apoptosis by causing inflammation and disrupting homeostasis. This study aimed to investigate the role of TAM receptors and ligands in the maintenance of homeostasis and elimination of apoptotic cells in the testes (weeks), short-term sham exposure (sham/1 week), and middle-

The therapeutic effect of GAS6 in remyelination is dependent upon Tyro3.

Multiple sclerosis is an autoimmune disease of the central nervous system (CNS) characterized by demyelination, axonal damage and, for the majority of people, a decline in neurological function in the long-term. Remyelination could assist in the protection of axons and their functional recovery, but such therapies are not, as yet, available. The TAM (Tyro3, Axl, and MERTK) receptor ligand GAS6 potentiates myelination in vitro and promotes recovery in pre-clinical models of MS. However, it has remained unclear which TAM receptor is responsible for transducing this effect and whether post-transl

Nano-Biomechanical Investigation of Phosphatidylserine-Mediated Ebola Viral Attachment via Human Gas6 and Axl.

The Ebola virus is a deadly pathogen that has been threatening public health for decades. Recent studies have revealed alternative viral invasion routes where Ebola virus approaches cells via interactions among phosphatidylserine (PS), PS binding ligands such as Gas6, and TAM family receptors such as Axl. In this study, we investigate the interactions among phosphatidylserine on the Ebola viral-like particle (VLP) membrane, human Gas6, and human Axl using atomic force microscope-based single molecule force spectroscopy to compare their binding strength and affinity from a biomechanical perspec

In situ sustained and pH-responsive Gas6-loaded microspheres for alleviating acute gouty arthritis by immunoregulation.

Acute gouty arthritis is characterized by sudden, severe attacks of pain, swelling, redness, and tenderness in joints. Managing acute gout attacks requires timely and effective control of the inflammatory reaction caused by monosodium urate (MSU) crystals. In clinical practice, intra-articular drug injection is a common treatment which partially reduces the side effects of oral administration. However, due to the low retention time and persistence of small-molecule drugs in the joints, the effects are often minimal. Therefore, a sustainable and on-demand drug delivery system is warranted. Corticosterone inhibits GAS6 to govern hair follicle stem-cell quiescence.

Chronic, sustained exposure to stressors can profoundly affect tissue homeostasis, although the mechanisms by which these changes occur are largely unknown. Here we report that the stress hormone corticosterone-which is derived from the adrenal gland and is the rodent equivalent of cortisol in humans-regulates hair follicle stem cell (HFSC) quiescence and hair growth in mice. In the absence of systemic corticosterone, HFSCs enter substantially more rounds of the regeneration cycle throughout life. Conversely, under chronic stress, increased levels of corticosterone prolong HFSC quiescence and

Replication Study of the Association of GAS6 and PROS1 Polymorphisms with Behçet's Disease in a Japanese Population.

PURPOSE: To investigate whether polymorphisms of GAS6 and PROS1, which each encode protein ligands for a family of tyrosine kinase receptors, are associated with Behçet's disease (BD) in a Japanese population. METHODS: We recruited 734 Japanese patients with BD and 1789 Japanese healthy controls. In all participants, we genotyped two single-nucleotide polymorphisms (SNPs) reportedly associated with BD: rs9577873 in GAS6 and rs4857037 in PROS1. RESULTS: We found that GAS6 rs9577873 was not significantly associated with BD. In contrast, PROS1 rs4857037, specifically the A allele, was associate

Specific killing of multiple myeloma cells by (-)-epigallocatechin-3-gallate extracted from green tea: biologic activity and therapeutic implications.

Epigallocatechin-3-gallate (EGCG), a polyphenol extracted from green tea, is an antioxidant with chemopreventive and chemotherapeutic actions. Based on its ability to modulate growth factor-mediated cell proliferation, we evaluated its efficacy in multiple myeloma (MM). EGCG induced both dose- and time-dependent growth arrest and subsequent apoptotic cell death in MM cell lines including IL-6-dependent cells and primary patient cells, without significant effect on the growth of peripheral blood mononuclear cells (PBMCs) and normal fibroblasts. Treatment with EGCG also led to significant apopto

Revealing the mechanism of Tilvestamab in treating cancer from a single-molecule perspective using atomic force microscopy.

BACKGROUND: AXL receptor tyrosine kinase, activated by its ligand GAS6, is a key driver of tumor aggressiveness, metastasis, and therapeutic resistance in cancers. Therapeutic inhibition of AXL signaling has emerged as a promising anti-cancer strategy. Tilvestamab, a monoclonal antibody, competitively blocks GAS6-AXL binding to suppress oncogenic signaling. However, the biomechanical basis of tilvestamab's molecular interactions-including its binding kinetics, affinity relative to GAS6, and functional impact on cellular mechanics-remains uncharacterized at the single-molecule level. Addressing

Variation in virion phosphatidylserine content drives differential GAS6 binding among closely related flaviviruses.

UNLABELLED: Many enveloped viruses engage phosphatidylserine (PS) receptors to enter cells, a phenomenon known as "apoptotic mimicry." We previously reported that Zika virus (ZIKV), but not closely related West Nile virus (WNV) or dengue virus (DENV), utilized AXL to infect cells because only ZIKV could bind the AXL ligand GAS6, a PS-binding protein. In this study, we investigated the mechanisms underlying the differential ability of these viruses to bind GAS6. Although immature virions expose larger patches of the viral membrane than do mature ones, our data show that virion maturity levels d

Gas-6 Scavenges Anionic Phospholipid-Expressing Microparticles and Mitigates TBI-Induced Endotheliopathy and Coagulopathy in Mice.

BACKGROUND: Traumatic brain injury (TBI) results in the release of microparticles from injured brain cells into circulation. These microparticles induce a systemic hypercoagulable state that rapidly transitions into secondary coagulopathy and endotheliopathy. We hypothesize that removing these microparticles from circulation could mitigate the TBI-induced secondary pathologies and improve outcomes. In this study, we investigated the role of Gas-6 (growth arrest-specific 6) as a scavenging factor for microparticles. METHODS: We quantified plasma Gas-6 levels in a mouse model of TBI and adminis
