An investigation of hormesis of trichloroethylene in L-02 liver cells by differential proteomic analysis.

Hormesis is the dose-response pattern of the biological responses to toxic chemicals, characterized by low-dose stimulation and high-dose inhibition. Although it is known that some cell types exhibit an adaptive response to low levels of cytotoxic agents, its molecular mechanism is still unclear and it has yet to be established whether this is a universal phenomenon that occurs in all cell types in response to exposure to every chemical. Trichloroethylene (TCE) is an organic solvent widely used and is released into the atmosphere from industrial degreasing operations. Acute (short-term) and ch

Hormesis in biomedical and toxicological models: A generalizable phenomenon induced by per- and polyfluoroalkyl agents.

This paper provides the first critical review of the capacity of per- and polyfluoroalkyl substances (PFAS) agents to induce hormetic effects in mammalian toxicological and biomedical cellular and animal models. PFAS induced hormetic responses were reported in primary cells and cell lines from multiple organ systems (e.g., liver, lung, colon, breast, prostate, immune, brain) from animal models and humans. While more limited, PFAS induced hormetic dose responses in experimental animals for short-term and chronic studies. Hormetic effects were induced by long and shorter carbon length alternativ

Enhanced reproduction in mallards fed a low level of methylmercury: an apparent case of hormesis.

Breeding pairs of mallards (Anas platyrhynchos) were fed a control diet or a diet containing 0.5 microg/g mercury (Hg) in the form of methylmercury chloride. There were no effects of Hg on adult weights and no overt signs of Hg poisoning in adults. The Hg-containing diet had no effect on fertility of eggs, but hatching success of eggs was significantly higher for females fed 0.5 microg/g Hg (71.8%) than for controls (57.5%). Survival of ducklings through 6 d of age was the same (97.8%) for controls and mallards fed 0.5 microg/g mercury. However, the mean number of ducklings produced per female

Peculiar stimulatory action of high-energy chemicals: Fifty years of explosive-induced hormesis evidence.

Explosives, such as trinitrotoluene (TNT) and cyclic nitramines (cyclotrimethylene trinitramine, RDX, and cyclotetramethylene tetranitramine, HMX), are widespread environmental contaminants. Although high doses are toxic, the effects of low, environmentally relevant concentrations remain poorly characterized in risk assessment. This review synthesizes evidence from five decades of research to determine explosives-induced hormesis, a biphasic dose-response characterized by stimulation at low doses and inhibition at high doses. Hormetic responses to explosives have been documented across a vast

Shape-Restricted Detection of Natural Product Hormesis: A Re-Analysis of Complex Botanical Mixtures.

INTRODUCTION: Botanical supplements are consumed to improve health despite the mystery of their mechanisms. One possible effect is hormesis, defined as a beneficial and stimulatory health effect at a low dose and detrimental effect at a high dose, or vice versa. Hormesis is usually discovered by manually examining the shape of dose-response curves, which is not scalable to the large number of chemical-assay combinations found in high-throughput screening (HTS) results. METHODS: We proposed a semi-automatic statistical testing procedure to quickly evaluate large-scale dose-response data, accom

Hormesis as a Geometric Necessity of Bounded Adaptive Systems: Quantitative Predictions From First Principles.

OBJECTIVES: To determine whether hormesis can be derived from the geometry of bounded adaptive biological endpoints, and whether the derivation yields quantitative predictions independent of curve fitting. METHODS: Bounded endpoints were represented in rapidity coordinates using the arctanh linearisation of the adopted Möbius composition law. Repair activation and damage accumulation were modelled as opposing bounded rapidity increments with independently measurable thresholds. The molecular-to-functional attenuation factor was derived for linear pathways from the metabolic-control summation

Hormesis-like effects of black phosphorus nanosheets on the spread of multiple antibiotic resistance genes.

The production scalability and increasing demand for black phosphorus nanosheets (BPNSs) inevitably lead to environmental leakage. Although BPNSs' ecotoxicological effects have been demonstrated, their indirect health risks, such as inducing increased resistance in pathogenic bacteria, are often overlooked. This study explores the influence of BPNSs on the horizontal gene transfer of antibiotic resistance genes (ARGs) facilitated by the RP4 plasmid, which carries multiple resistance genes. The results indicated that BPNSs exhibited concentration-dependent hormesis-like effects on bacterial con

Low concentrations of FA exhibits the Hormesis effect by affecting cell division and the Warburg effect.

Formaldehyde (FA), a ubiquitous indoor environmental pollutant, has been classified as a carcinogen. There are many studies showed that low levels of FA could promote cell proliferation, however, little is known about the signal pathways. To determine the potential molecular mechanisms, human chronic myeloid leukemia cells (K562 cells) and human bronchial epithelial cells (16HBE cells) were exposed to different concentrations of FA. The data showed that FA at 0-125 μM or 0-60 μM promoted the proliferation of K562 cells or 16HBE cells respectively, indicating that FA did have the Hormesis effec

Predicting Hormesis Effects of GenX in Zebrafish via Interpretable Machine Learning: Insights From SHAP Analysis.

Hexafluoropropylene oxide-dimer acid (GenX), a prominent alternative to legacy per- and polyfluoroalkyl substances (PFAS), poses a significant challenge to traditional linear risk assessment models due to its ability to induce hormesis-a biphasic "low-dose stimulation, high-dose inhibition" response. This study established an interpretable machine learning (ML) framework to identify and predict GenX-induced non-monotonic dose-response (NMDR) relationships in zebrafish (Danio rerio). By integrating 263 independent experimental records across 11 biological categories, we benchmarked 6 mL paradig

Induced mechanism of phosphatase hormesis by Cd ions and rhizosphere metabolites of Trifolium repens L.

Rhizosphere phosphatases can exhibit hormetic effects in response to cadmium (Cd) ion stimulation. However, understanding the mechanisms underlying hormesis effects on soil ecosystems is challenging as studies on hormesis are usually specific to an organism, cell, or organ. To comprehensively investigate the mechanism of phosphatase hormesis, this study utilized in situ zymography and metabolomics to analyze the rhizosphere of Trifolium repens L. (white clover). Zymograms showed that rhizosphere phosphatase displayed a hormetic effect in 10 mg kg-1 Cd contaminated soil, with a hotspot area 1.8

Targeting metabolic syndrome with phytochemicals: Focus on the role of molecular chaperones and hormesis in drug discovery.

Adaptive cellular stress response confers stress tolerance against inflammatory and metabolic disorders. In response to metabolic stress, the key mediator of cellular adaptation and tolerance is a class of molecules called the molecular chaperones (MCs). MCs are highly conserved molecules that play critical role in maintaining protein stability and functionality. Hormesis in this context is a unique adaptation mechanism where a low dose of a stressor (which is toxic at high dose) confers a stress-resistant adaptive cellular phenotype. Hormesis can be observed at different level of biological o

Hormesis-induced neuroplasticity: Targeting neuroinflammation signaling cascades for therapeutic insights in aging and neurodegenerative diseases.

The signaling pathways associated with α-Klotho, glutamate, mediators of the inflammatory response in the central nervous system (CNS) and that related to different isoforms of the Na, K-ATPase (NKA) protein as a pump and receptor for endogenous steroids (ouabain-like hormones) are associated with neuroplasticity and neuroprotection. This neuroadaptive response induced by pharmacologic (Cardiotonic Steroids, Klotho, Resveratrol, Curcumin, and other Phytochemicals), and non-pharmacologic strategies (intermittent fasting and physical exercise) involves glial and neuronal cell crosstalk through a

A new parameter for quantitatively characterizing antibiotic hormesis: QSAR construction and joint toxic action judgment.

Antibiotics usually induce the hormetic effects on bacteria, featured by low-dose stimulation and high-dose inhibition, which challenges the central belief in toxicity assessment and environmental risk assessment of antibiotics. However, there are currently no ideal parameters to quantitatively characterize hormesis. In this study, an effective area in hormesis (AH) was developed to quantify the biphasic dose-responses of single antibiotics (sulfonamides (SAs), sulfonamides potentiators (SAPs), and tetracyclines (TCs)) and binary mixtures (SAs-SAPs, SAs-TCs, and SAs-SAs) to the bioluminescence

Hormesis management of Moringa oleifera with exogenous application of plant growth regulators under saline conditions.

The study investigated the adaptability of Moringa oleifera to saline conditions, focusing on its hormesis behavior. It also examined how various plant growth regulators affected growth, physiological parameters, and bioactive compounds of moringa. In the first phase, different NaCl stress levels (0, 50, 100, 150, 200, and 250 mM) were applied. Notably, significant stimulation was observed at 100 mM stress for growth, total phenolics, total flavonoids and total chlorophyll content while 150 mM stress had a marked inhibitory effect, with survival decreasing at 200 and 250 mM NaCl levels. A 38%

Hormesis and bone marrow stem cells: Enhancing cell proliferation, differentiation and resilience to inflammatory stress.

This paper identifies and provides the first detailed assessment of hormetic dose responses by bone marrow stem cells (BMSCs) from a broad range of animal models and humans with particular emphasis on cell renewal (proliferation), cell differentiation and enhancing resilience to inflammatory stress. Such hormetic dose responses are commonly reported, being induced by a broad range of chemicals, including pharmaceuticals (e.g., caffeine, dexamethasone, nicotine), dietary supplements (e.g., curcumin, Ginkgo biloba, green tea extracts. resveratrol, sulforaphane), endogenous agents (e.g., hydrogen

On an extended understanding of the term "hormesis" for denoting alternating directions of the organism's response to increasing adverse exposures.

The authors propose to consider as hormesis phenomenon not only a realization of the Arndt-Schulze rule but any non-monotonic dose-response relationship for a certain outcome that is characterized by changing direction of a response between adjacent ranges of doses of an initiator of this response, the number of such ranges being two or more. This approach is illustrated with results of several in vitro experiments on different established cell lines exposed to CdS or PbS nanoparticles.

Interpretable machine learning models for predicting hormesis of binary antibiotic mixtures on Vibrio qinghaiensis sp.-Q67.

The hormesis phenomenon of antibiotic mixtures hinders environmental risk assessment owing to the lack of predictive models for chemical mixtures. This study developed a novel machine learning classification approach to predict the hormesis effect of 275 binary antibiotic mixtures on Vibrio qinghaiensis sp.-Q67. Time-dependent toxicities (12, 18, and 24 h) indicate that 81.1 % of mixtures demonstrate hormesis at 24 h (compared with 54.1 % at 12 h), revealing a time-dependent amplification. A machine learning framework incorporating nine algorithms was developed to distinguish between hormesis

Bioaccumulation, biotransformation and hormesis effects of 6PPD-Q in Eisenia fetida: Impacts on metabolism and gut microbiota.

N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine-quinone (6PPD-Q) is an oxidative byproduct of N-(1,3-dimethylbutyl)-N'-phenyl-p-phenylenediamine, a widely used tire antioxidant, and has emerged as an environmental pollutant of increasing concern. However, its environmental behavior and effects in soil ecosystems remain poorly characterized. This study focused on earthworm Eisenia fetida to reveal the bioaccumulation, biotransformation, and hormesis effects of 6PPD-Q. Findings indicated that 6PPD-Q mainly accumulated in intestinal tissues of E. fetida with concentration of 3465.2 ± 470.8 ng/

Evidence of hormesis: Dose-dependent neuroprotective and neurotoxic effects of genistein in diethyl phthalate-exposed zebrafish.

Among the multifaceted etiological factors implicated in neurodegeneration, exposure to environmental toxicants represents a significant contributor. Diethyl phthalate (DEP), long regarded as relatively safe, has been increasingly identified in recent studies as a potent inducer of neurodegenerative processes. DEP is a low-molecular-weight phthalic acid ester that is found in various plastic products, cosmetics, and perfumes, and can be ingested, inhaled, or absorbed through dermal contact. It was found that DEP can easily be metabolised into monoethyl phthalate, which possesses equivalent or

Aspartame induces hormesis-toxicity switch in microalgal‑bacterial granular sludge: Community assembly and functional gene responses.

Based on the distribution of aspartame in anaerobic digestion effluent (ADE) from food waste, this study explored the potential application of microalgae-bacterial granular sludge (MBGS) for treating ADE containing aspartame. While previous studies have focused on toxicity, the core scientific question of this work is to elucidate the adaptive regulatory mechanisms that govern MBGS performance under varying aspartame stresses. Notably, MBGS exhibited broad adaptive tolerance to varying aspartame concentrations, displaying a typical hormetic response: low-level aspartame exposure exerted a stim
