Viruses have long been recognized as significant pathogens, contributing to multiple global pandemics throughout human history. Recent examples include the 2009 influenza pandemic and the COVID-19 pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2) in 2019. Despite ongoing experimental and clinical efforts, the development of effective antiviral treatments and vaccines remains challenging due to the high mutation rates of many human pathogenic viruses including influenza virus and SARS-CoV-2. As an alternative approach, antiviral strategies targeting host factors sh

Environmental viruses in wastewater and sludge are widely recognized for their roles in waterborne diseases. However, previous studies mainly focused on RNA viruses, and little is known about the diversity of DNA viral communities and their driving factors in municipal wastewater treatment environments. Herein, we conducted a pilot study to explore DNA virus profiles in municipal wastewater and recycled sludge by metagenomics method, and track their temporal changes in northern China. Results showed that 467 viral species were co-shared among all the samples. We identified six families of huma

Vaccination against papillomavirus: outstanding issues. Human papillomavirus or HPV are oncogenic viruses involved in many cancers. It is estimated that around 5% of cancers worldwide are linked to HPV infection. Cancers of the cervix, vagina, vulva, anus, penis and throat (tonsil) are virally induced in proportions ranging from 35 to almost 100%. These cancers are not all easily detected and the detection of lesions is not always simple and effective. Prophylactic anti-HPV vaccination is recognized for its effectiveness in the prevention of cervical cancers, its place in the prevention of oth

Whilst papillomaviruses mainly induce epithelial proliferations in man and in different animal species, the association of this group of viruses with malignant tumours has become increasingly evident. Despite the heterogeneity of human papillomavirus (HPV), only a small number are identified within carcinomas, implying a different, oncogenic potential for different HPV types.

The human papillomavirus class of DNA viruses are more than circumstantially related to oral and airway papillomas. Whether they are fully oncogenic, in the malignant sense, without other agents is questionable. Recent advances in molecular virology and the use of genus-specific (common) antigen-antibody reactions have identified papillomavirus in laryngeal and oral papillomas. Laryngeal and oral papillomas could be precancerous lesions, but they show a low-risk and long-time interval leading to malignancy unless significant iatrogenic or host variables--such as radiotherapy or immuno-incompet

This review aimed to assess the prevalence of oncogenic and non-oncogenic viruses in odontogenic cysts and tumours. A comprehensive database search was performed for published articles regarding the presence of viruses in all odontogenic cysts and tumours, following guidelines without date constraints, and was registered with PROSPERO. After screening 164 articles, 24 studies were included for systematic evaluation. A meta-analysis was performed to determine the prevalence of human papillomavirus (HPV), revealing an overall pooled prevalence of HPV in ameloblastomas of 18.21% (95% confidence i

INTRODUCTION: The sexually transmitted infections (STIs) caused by viruses including human T cell leukemia virus type-1 (HTLV-1), human immunodeficiency virus-1 (HIV-1), human simplex virus-2 (HSV-2), hepatitis C virus (HCV), hepatitis B virus (HBV), and human papillomavirus (HPV) are major public health issues. These infections can cause cancer or result in long-term health problems. Due to high prevalence of STIs, a safe and effective vaccine is required to overcome these fatal viruses.
AREAS COVERED: This review includes a comprehensive overview of the literatures relevant to vaccine devel

Sixty-seven proliferous squamous epithelial lesions of the oral cavity were examined for the presence of human papillomavirus whole (structural) virion antigens by a peroxidase-antiperoxidase technique having immunospecificity against genus-specific (common) antigens of the papillomaviruses. A positive reaction for papillomavirus genus specific antigens was found in 18 of 29 verrucae, 2 of 5 multiple papillomas, and 3 of 5 condylomata; common antigens were not detected in 28 keratoacanthomas. The positive reaction was invariably intranuclear in cells having a focal or diffuse distribution in t

Human papillomavirus (HPV) is one of the most common sexually transmitted infectious viruses among men and women of reproductive age throughout the world. Pregnant women are susceptible during pregnancy and most infections of them are self-limiting infections, which can be removed by their autoimmunity, while the persistent infections are associated with precancerous lesions and cancer of the anogenital mucosa in women and men. In addition, HPV infection may also affect reproductive health and fertility. The effect of HPV on female fertility requires further study, but HPV influences sperm par

Minor intron-containing genes (MIGs) account for <2% of all human protein-coding genes and are uniquely dependent on the minor spliceosome for proper excision. Despite their low numbers, we surprisingly found a significant enrichment of MIG-encoded proteins (MIG-Ps) in protein-protein interactomes and host factors of positive-sense RNA viruses, including SARS-CoV-1, SARS-CoV-2, MERS coronavirus, and Zika virus. Similarly, we observed a significant enrichment of MIG-Ps in the interactomes and sets of host factors of negative-sense RNA viruses such as Ebola virus, influenza A virus, and the retr

Human oncogenic viruses contribute significantly to the global health burden and include seven types: Epstein-Barr virus, hepatitis B virus, human T-cell leukemia virus type 1, human papillomavirus, hepatitis C virus, Kaposi's sarcoma-associated herpesvirus, and Merkel cell polyomavirus. While the roles of latent or integrated viral genomes in cancer have been documented, emerging evidence highlights the contribution of defective viruses-those carrying intragenic deletions or loss-of-function mutations-in promoting viral oncogenesis. These altered genomes often lack genes essential for lytic r

Haematological malignancies are one of the most common tumors, with a rising incidence noted over recent decades. Viral infections play significant roles in the pathogenesis of these malignancies globally. This review delves into the contributions of various known viruses-specifically Epstein-Barr virus (EBV), human immunodeficiency virus (HIV), human T-cell leukemia virus type 1 (HTLV-1), Kaposi's sarcoma-associated herpesvirus (KSHV), human cytomegalovirus (HCMV), hepatitis B virus (HBV), hepatitis C virus (HCV), and human papillomavirus (HPV)-in the development of haematological malignancie

There is an association between human papillomavirus (HPV), in particular types 16 and 18, and premalignant and malignant disease of the cervix. At present it is not possible to say that this is a causal association but in vitro transformation studies indicate that these viruses can transform primary mammalian cells in cooperation with Ha-ras. This may mean that these viruses have at least a c-myc like activity which is consistent with their ability on their own to immortalize primary keratinocytes. The fact that HPV6 can also transform mammalian cells, at a low frequency, in cooperation with

Papillomaviruses are a large and diverse group of viruses. It includes approximately 200 fully described types that have been detected in humans. Human papilloma viruses (HPV) are etiologic agents during various, benign and malignant lesions of mucous membrane and skin epithelium. Very importantly, persistent HPV infection of certain types is a leading cause of carcinoma of uterine cervix, penis, vulva; vagina, anal canal and fauces (including tongue base and tonsils). HPV infection prophylaxis is the best means to control HPV-conditioned diseases, and vaccination, as had been demonstrated, --

In this work, we investigate the potential of highly sulfated synthetic glycomimetics to act as inhibitors of viral binding/infection. Our results indicate that both long-chain glycopolymers and short-chain glycooligomers are capable of preventing viral infection. Notably, glycopolymers efficiently inhibit Human Papillomavirus (HPV16) infection in vitro and maintain their antiviral activity in vivo, while the glycooligomers exert their inhibitory function post attachment of viruses to cells. Moreover, when we tested the potential for broader activity against several other human pathogenic viru

To overcome the limitations of conventional vaccines, new platforms for vaccine design have emerged such as those based on viral vectors and virus-like particles (VLPs). Viral vector vaccines are highly efficient and the onset of protection is quick. Many recombinant vaccine candidates for humans are based on viruses belonging to different families such as Adenoviridae, Retroviridae, Paramyxoviridae, Rhabdoviridae, and Parvoviridae. Also, the first viral vector vaccine licensed for human vaccination was the Japanese encephalitis virus vaccine. Since then, several viral vectors have been approv

Esophageal adenocarcinoma (EAC) has had the fastest increasing incidence of any solid tumor in the United States in the last 30 years. Long standing gastroesophageal reflux disease is a well-established risk factor with strong associations with obesity, alcohol and tobacco. However, there are likely additional contributing factors. Viruses such as human papillomavirus, ebstein-barr virus and herpes simplex virus have been implicated in the pathogenesis of esophageal cancer. This review will discuss the known literature linking viruses to esophageal adenocarcinoma and consider future relationsh

Biallelic mutations in the ITK gene cause a T-cell primary immunodeficiency with Epstein-Barr virus (EBV)-lymphoproliferative disorders. We describe a novel association of a homozygous ITK mutation with β-human papillomavirus (HPV)-positive epidermodysplasia verruciformis. Thus, loss of function in ITK can result in broad dysregulation of T-cell responses to oncogenic viruses, including β-HPV and EBV.

Oncogenic viruses, with their significant capacity to drive malignant transformation, are currently known as a major factor involved in cardiovascular diseases (CVDs) by intricate immunometabolic and inflammatory processes. In this context, viral pathogens such as Hepatitis B and C viruses (HBV/HCV), Human Cytomegalovirus (HCMV), Human Papillomavirus (HPV), Epstein-Barr Virus (EBV), and Human T-Lymphotropic Virus (HTLV) have been detected in vascular tissues and are hypothesised to contribute to endothelial dysfunction, atherogenesis, and myocarditis. These viruses can modulate host cell signa
