Human Cerebellum Gangliosides: A Comprehensive Analysis by Ion Mobility Tandem Mass Spectrometry.

The human cerebellum is an ultraspecialized region of the brain responsible for cognitive functions and movement coordination. The fine mechanisms through which the process of aging impacts such functions are not well understood; therefore, a rigorous exploration of this brain region at the molecular level is deemed necessary. Gangliosides, sialylated glycosphingolipids, highly and specifically expressed in the human central nervous system, represent possible molecular markers of cerebellum development and aging. In this context, for a comprehensive determination of development- and age-specif

On the Arrival Time Distribution of Reacting Systems in Ion Mobility Spectrometry.

Ion mobility spectrometry (IMS) is a widely used gas-phase separation technique, particularly when coupled with mass spectrometry (MS). Modern IMS instruments often apply elevated reduced field strengths for improved ion separation and ion focusing. These alter the collision dynamics and further drive ion reaction processes that can change the analyte's structure. As a result, the measured arrival time distribution (ATD) can change with the applied reduced field strengths. In this work, we systematically study how the ion collision dynamics and the ion reaction dynamics, as a function of the r

Improving the efficiency of ion mobility spectrometry analyses by using multivariate calibration.

The simplicity, sensitivity and expeditiousness of ion mobility spectrometry (IMS) make it especially useful for the determination of active principal ingredients (APIs) present at low concentrations in pharmaceuticals. However, the poor resolution of this technique precludes the identification and/or determination of substances with similar molecular weights, which exhibit also similar drift times and give overlapped peaks as a result. Oral contraceptives are pharmaceutical formulations containing two APIs of similar molecular weights at very low concentrations which therefore give strongly o

Screening of Protein Carbonylation Sites in Human Serum by Ion Mobility Mass Spectrometry.

Excessive oxidative stress, associated with various diseases, can induce protein carbonylation-nonenzymatic modifications involving aldehyde or keto group formation. These modifications are structurally diverse and low in abundance, which complicates their detection and quantitation. Here, we developed a strategy to identify and quantify protein carbonylation in human serum proteins from 39 rheumatoid arthritis patients and 29 healthy donors. Reactive carbonyl groups were derivatized with an aldehyde reactive probe (ARP), digested with trypsin, enriched via avidin affinity chromatography, and

Sweat-based stress screening with gas chromatography-ion mobility spectrometry and electronic nose.

BACKGROUND: Diagnosis of stress generally involves uses of questionnaires which can provide biased results. The more reliable approach relies on observation of individual symptoms by psychiatrists which is time consuming and could not be applicable for massive scale screening tests. This research established alternative approaches with gas chromatography-ion mobility spectrometry (GC-IMS) and electronic nose (e-nose) to perform fast stress screening based on fingerprinting of highly volatile compounds in headspaces of sweat. The investigated samples were obtained from 154 female nurse voluntee

Reliable Detection of Chemical Warfare Agents Using High Kinetic Energy Ion Mobility Spectrometry.

High Kinetic Energy Ion Mobility Spectrometers (HiKE-IMS) ionize and separate ions at reduced pressures of 10-40 mbar and over a wide range of reduced electric field strengths E/N of up to 120 Td. Their reduced operating pressure is distinct from that of conventional drift tube ion mobility spectrometers that operate at ambient pressure for trace compound detection. High E/N can lead to a field-induced fragmentation pattern that provides more specific structural information about the analytes. In addition, operation at high E/N values adds the field dependence of ion mobility as an additional

Gas chromatography-ion mobility spectrometry for the detection of human disease: a review.

Gas chromatography-ion mobility spectrometry (GC-IMS) is an advanced technique used for detecting trace compounds, due to its non-destructive, straightforward, and rapid analytical capabilities. However, the application of GC-IMS in human disease screening is barely reported. This review summarizes the application and related parameters of GC-IMS in human disease diagnosis. GC-IMS detects volatile organic compounds in human breath, feces, urine, bile, etc. It can be applied to diagnose diseases, such as respiratory diseases, cancer, enteropathy, Alzheimer's disease, bacterial infection, and me

Sonic-spray introduction of liquid samples to hand-held Ion mobility spectrometry analyzers.

Sampling hazardous compounds in the form of solids and liquids is a growing need in the fields of homeland security and forensics. Chemical analysis of particles and droplets under field conditions is crucial for various tasks carried out by counter-terrorism and law enforcement units. The use of simple, small and low cost means to achieve this goal is constantly pursued. In this work, an approach for rapid, continuous generation of vapors from liquid samples using sonic spray (SS) as the sample introduction technique, followed by analysis using hand-held ion mobility spectrometry (IMS) vapor

High-Resolution Differential Ion Mobility Separations/Orbitrap Mass Spectrometry without Buffer Gas Limitations.

Strong orthogonality between differential ion mobility spectrometry (FAIMS) and mass spectrometry (MS) makes their hybrid a powerful approach to separate isomers and isobars. Harnessing that power depends on high resolution in both dimensions. The ultimate mass resolution and accuracy are delivered by Fourier Transform MS increasingly realized in Orbitrap MS, whereas FAIMS resolution is generally maximized by buffers rich in He or H2 that elevate ion mobility and lead to prominent non-Blanc effects. However, turbomolecular pumps have lower efficiency for light gas molecules and their flow from

Gas-Phase Dopant-Induced Conformational Changes Monitored with Transversal Modulation Ion Mobility Spectrometry.

The potential of a Transversal Modulation Ion Mobility Spectrometry (TMIMS) instrument for protein analysis applications has been evaluated. The Collision Cross Section (CCS) of cytochrome c measured with the TMIMS is in agreement with values reported in the literature. Additionally, it enables tandem IMS-IMS prefiltration in dry gas and in vapor doped gas. The chemical specificity of the different dopants enables interesting studies on the structure of proteins as CCS changed strongly depending on the specific dopant. Hexane produced an unexpectedly high CCS shift, which can be utilized to ev

Tracking Protein Misfolding and Oligomerization: A Temperature-Controlled Ion Mobility-Mass Spectrometry Approach.

Aberrant protein oligomerization is a hallmark of neurodegenerative disorders, yet the conformational and kinetic underpinnings of early aggregation remain poorly understood due to the inability of structural techniques to capture transient, low-abundance oligomeric intermediates. This necessitates the development of a methodology that can characterize the conformational states related to protein unfolding and thus allow for the investigation of the molecular mechanism responsible for disease progression. Here, we demonstrate how temperature-controlled nanoelectrospray ionization (TC-nESI) com

Rapid and Direct Determination of Methanol in Alcoholic Beverages by Ion Mobility Spectrometry.

This work introduces a new method for direct detection of methanol in alcoholic beverages, using ion mobility spectrometry (IMS), headspace sampling (HS), and Tenax adsorbent preseparation (HS-Tenax-IMS). An ionization method based on the dopant gases was employed to overcome the overlapping of the reactant ions with the methanol peak. Six different ketones dopants were tested, and 2-heptanone was selected as the optimal variant. Tenax-GR column was used for both HS and as a preseparation method of methanol and ethanol prior to IMS detection. The optimum sampling time of 15 min was achieved, a

Noncovalent crown ether-assisted separation of stereoisomeric glycosphingosines using cyclic ion mobility spectrometry.

Glycosphingosines (GlycoSphs), a class of simple glycosphingolipids (GSLs), are abundant in the brain and crucial in neurodegenerative diseases. Specific GlycoSphs, including glucosyl-sphingosine (GlcSph) and galactosyl-sphingosine (GalSph), exhibit distinct biological roles due to subtle structural isomeric differences, including anomeric configurations (α, β). These variations influence solubility, membrane interactions, and pathological outcomes, with GlcSph linked to immune modulation and GalSph to neurodegeneration. Standard mass spectrometry (MS) enables rapid analysis of biochemical mix

Tailored extraction and ion mobility-mass spectrometry enables isotopologue analysis of tetrahydrofolate vitamers.

Climate change directs the focus in biotechnology increasingly on one-carbon metabolism for fixation of CO2 and CO2-derived chemicals (e.g. methanol, formate) to reduce our reliance on both fossil and food-competing carbon sources. The tetrahydrofolate pathway is involved in several one-carbon fixation pathways. To study such pathways, stable isotope-labelled tracer analysis performed with mass spectrometry is state of the art. However, no such method is currently available for tetrahydrofolate vitamers. In the present work, we established a fit-for-purpose extraction method for the methylotro

Opioid cutting agents for use as internal standards in ion mobility spectrometry (IMS).

Opioids have become a serious public health concern over the last decade. These compounds are commonly found mixed, or cut, with safer compounds to make the opioids appear unadulterated while also enhancing the psychoactive effect on the user. Commercial benchtop and handheld IMS devices are capable of detection but published reduced ion mobility (K0) values, used to identify the target analytes with IMS instrumentation, have shown variability. This lack of agreement, even for compounds used for calibration, is often due to the effects of drift tube temperature, drift gas water vapor levels an

Desorption electrospray ionization (DESI) with atmospheric pressure ion mobility spectrometry for drug detection.

Desorption electrospray ionization (DESI) was coupled to an ambient pressure drift tube ion mobility time-of-flight mass spectrometer (IM-TOFMS) for the direct analysis of active ingredients in pharmaceutical samples. The DESI source was also coupled with a standalone IMS demonstrating potential of portable and inexpensive drug-quality testing platforms. The DESI-IMS required no sample pretreatment as ions were generated directly from tablets and cream formulations. The analysis of a range of over-the-counter and prescription tablet formations was demonstrated for amphetamine (methylphenidate)

Charged chiral derivatization for enantioselective imaging of D-,L-2-hydroxyglutaric acid using ion mobility spectrometry/mass spectrometry.

A newly synthesized charged chiral tag-enabled enantioselective imaging of D-,L-2-hydroxyglutaric acid, which are independently associated with the regulation of DNA methylation. The tag-conjugated diastereomers were ionized efficiently through MALDI, separated by ion mobility spectrometry, and further separated from other molecules in mass spectrometry. On-tissue chiral derivatization using the tag facilitated the visualization of different distributions of the two isomers in the mouse testis.

Characterization of polyalphaolefins using halogen anion attachment in atmospheric pressure photoionization coupled with ion mobility spectrometry-mass spectrometry.

Polyalphaolefins (PAOs) are saturated alpha olefin oligomers used as a base stock oil for synthetic lubricants. The synthetic base stocks are manufactured from linear alpha olefins by catalytic oligomerization processes. The aim of this work was the characterization of different PAO grades, synthesized from different linear alpha olefins using two oligomerization processes, acid and metallocene catalyses. Negative ion atmospheric pressure photoionization (APPI) coupled with ion mobility spectrometry-mass spectrometry (IMS-MS) permitted the detection of intact PAO adducts with either chloride,
