Inhibitory effects of isatin Mannich bases on carbonic anhydrases, acetylcholinesterase, and butyrylcholinesterase.

The effects of isatin Mannich bases incorporating (1-[piperidin-1-yl (P1)/morpholin-4-yl (P2)/N-methylpiperazin-1-yl (P3)]methyl)-1H-indole-2,3-dione) moieties against human (h) carbonic anhydrase (CA, EC 4.2.1.1) isoenzymes hCA I and hCA II, acetylcholinesterase (AChE), and butyrylcholinesterase (BChE) enzymes were evaluated. P1-P3 demonstrated impressive inhibition profiles against AChE and BChE and also inhibited both CAs at nanomolar level. These inhibitory effects were more powerful in all cases than the reference compounds used for all these enzymes. This study suggests that isatin Manni

Isatin analogs as novel inhibitors of Candida spp. β-carbonic anhydrase enzymes.

Enzyme inhibition data of structurally novel isatin-containing sulfonamides were determined for two carbonic anhydrases (CAs, EC 4.2.1.1) from pathogenic Candida species (CaNce103 from C. albicans and CgNce103 from C. glabrata). The compounds show KI values in the low nanomolar range for the fungal CAs, while they have significantly higher KI values for the human CAs. Homology models were constructed for the CaNce103 and CgNce103 and subsequently the ligands were docked into these models to rationalize their enzyme inhibitory properties.

Cyclic GMP excretion blocked by isatin administration under conditions of fluid overload.

Isatin is a potent inhibitor of atrial natriuretic peptide (ANP) receptors and ANP-induced generation of cGMP in vitro. This study was designed to determine whether it had a similar effect in vivo, using a model of fluid overload known to induce ANP. We confirmed that this model increased urinary output of cGMP 3 hr after volume loading, and showed that this effect was blocked by i.p. injection of isatin (50 mg/kg). Isatin had no effect on urine volume or sodium output. However, isatin did have an effect on plasma protein concentration, both compared with control values, compatible with shifti

The influence of neuroprotector isatin on haloperidolinduced catalepsy and proteomic profile of mice brain.

Isatin (indol-2,3-dione) is an endogenous regulator found in humans and animals. It interacts with numerous target proteins and exhibits a wide range of biological activities, including neuroprotective action in animal models of Parkinson's disease (PD) induced by administration of neurotoxins MPTP (1-methyl-4-phenyl-1,2,3,6- tetrahydropyridine) or rotenone. An antipsychotic drug haloperidol, which impairs neurotransmitter balance in the nigrostriatal pathway, models dopamine deficiency and promotes the development of motor disorders characteristic of PD. In this work, the effect of two doses

Isatin ameliorates ovarian inflammation and apoptosis caused by hormonal imbalances in PCOS mice.

Polycystic ovary syndrome (PCOS) is the most common reproductive endocrine disorder in women of childbearing age. Isatin (ISA) is a bioactive compound naturally found in mammalian body fluids and tissues, known for its potent antioxidant, anti-inflammatory, and anticancer properties. We used letrozole to induce a PCOS mouse model and evaluated the reproductive benefits of ISA. The mice were initially grouped according to the concentration of ISA administered at 25, 50, and 100 mg/kg. Subsequently, research results indicated that the optimal concentration was determined to be 100 mg/kg, while a

Study of antiplasmodial activity, toxicity, pharmacokinetic profiles of n-methyl-isatin (CH3ISACN) derivative.

One of the main factors that have made it difficult to control malaria is the large number of parasites that are resistant to the usual antimalarial drugs. Therefore, the development of new drugs that are more effective and with low toxicity for humans is necessary. In this work, we evaluated the adduct 2-(3-hydroxy-1-methyl-2-oxoindolin-3-yl)acrylonitrile, also called CH3ISACN, as a potential antimalarial through in vitro studies, and evaluated its effects in silico and in vivo toxicology. For this, the compound CH3ISACN was exposed to P. falciparum W2 strain in infected human erythrocytes. T

Spectroscopic and antibacterial studies of new octaazamacrocyclic complexes derived from carbohydrazide and isatin.

A novel series of the macrocyclic complexes of the type: [M(C18H14N10O2)X2], where M = Co(II), Ni(II), Cu(II) and Zn(II); X = Cl(-), NO3(-) and CH3COO(-), has been synthesized by template condensation of carbohydrazide and isatin in methanolic medium. The complexes were characterized by various physico-chemical techniques, such as elemental analyses, molar conductance measurements, magnetic measurements, and electronic, NMR, IR and EPR spectral studies. The low value of molar conductance indicates them to be non-electrolytes. Based on various studies, a distorted octahedral geometry was propos

Evaluation of Novel Isatin-Schiff Base Derivatives: Monoamine Oxidase Inhibition and Antimicrobial Assessment.

Five novel series of isatin-Schiff base derivatives (1a-5e) were synthesized, and their neuroprotective and anti-infective potential was evaluated. Their half-maximal inhibitory concentration (IC50) was determined against both monoamine oxidase A (MAO-A) and MAO-B targets for the treatment of depression and Parkinson's disease, respectively. The anti-infective potential of the synthesized derivatives was evaluated by measuring their IC50 values against urease, an enzyme utilized by specific pathogenic bacteria as a virulence factor. The minimum inhibitory concentration for each compound was as
