Laccase Immobilization on Copper-Magnetic Nanoparticles for Efficient Bisphenol Degradation.

Laccase activity is influenced by copper (Cu) as an inducer. In this study, laccase was immobilized on Cu and Cu-magnetic (Cu/Fe2O4) nanoparticles (NPs) to improve enzyme stability and potential applications. The Cu/Fe2O4 NPs functionally activated by 3-aminopropyltriethoxysilane and glutaraldehyde exhibited an immobilization yield and relative activity (RA) of 93.1 and 140%, respectively. Under optimized conditions, Cu/Fe2O4 NPs showed high loading of laccase up to 285 mg/g of support and maximum RA of 140% at a pH 5.0 after 24 h of incubation (4°C). Immobilized laccase, as Cu/Fe2O4-laccase,

Laccase catalyzed synthesis of iodinated phenolic compounds with antifungal activity.

Iodine is a well known antimicrobial compound. Laccase, an oxidoreductase which couples the one electron oxidation of diverse phenolic and non-phenolic substrates to the reduction of oxygen to water, is capable of oxidizing unreactive iodide to reactive iodine. We have shown previously that laccase-iodide treatment of spruce wood results in a wash-out resistant antimicrobial surface. In this study, we investigated whether phenolic compounds such as vanillin, which resembles sub-structures of softwood lignin, can be directly iodinated by reacting with laccase and iodide, resulting in compounds

Novel penicillins synthesized by biotransformation using laccase from Trametes spec.

Eight novel penicillins were synthesized by heteromolecular reaction of ampicillin or amoxicillin with 2,5-dihydroxybenzoic acid derivatives using a laccase from Trametes spec. All products inhibited the growth of several gram positive bacterial strains in the agar diffusion assay, among them methicillin-resistant Staphylococcus aureus and vancomycin-resistant Enterococci. The products protected mice against an infection with Staphylococcus aureus lethal to the untreated animals. Cytotoxicity and acute toxicity of the new compounds were neglectable. The results show the usefulness of laccase f

Redox chemistry in laccase-catalyzed oxidation of N-hydroxy compounds.

1-Hydroxybenzotriazole, violuric acid, and N-hydroxyacetanilide are three N-OH compounds capable of mediating a range of laccase-catalyzed biotransformations, such as paper pulp delignification and degradation of polycyclic hydrocarbons. The mechanism of their enzymatic oxidation was studied with seven fungal laccases. The oxidation had a bell-shaped pH-activity profile with an optimal pH ranging from 4 to 7. The oxidation rate was found to be dependent on the redox potential difference between the N-OH substrate and laccase. A laccase with a higher redox potential or an N-OH compound with a l

Laccase protects Cryptococcus neoformans from antifungal activity of alveolar macrophages.

While laccase of Cryptococcus neoformans is implicated in the virulence of the organism, our recent studies showing absence of melanin in the infected mouse brain has led us to a search for alternative roles for laccase in cryptococcosis. We investigated the role of laccase in protection of C. neoformans against murine alveolar macrophage (AM)-mediated antifungal activity by using a pair of congenic laccase-positive (2E-TUC) and laccase-deficient (2E-TU) strains. The laccase-positive cells with laccase derepression were more resistant to the antifungal activity of AM than a laccase-deficient s

Fabrication of nanobiocatalyst using encapsulated laccase onto chitosan-nanobiochar composite.

Laccase is one of the widely used enzymes for biotechnological processes. Immobilization of enzymes is a universally accepted approach to increase their reusability and stability. In this study, laccase enzyme from Trametes versicolor was encapsulated for the first time in a chitosan-nanobiochar matrix. The chitosan-tripolyphosphate gel formation technique was employed to produce homogeneous biocatalyst nanoparticles, with 35% effective binding efficiency and 3.5 Units/g apparent activity under the best configuration. The reusability of the encapsulated laccase was demonstrated towards the oxi

[Laccase as a useful biotechnological tool in the synthesis of biologically active compounds].

The synthesis of organic compounds using oxidoreductive enzymes as biocatalysts is increasingly being considered as an environmentally friendly alternative to classical chemical synthesis. An example of such an enzyme, which exhibits low substrate specificity and operates under mild conditions of pH, pressure and temperature, is laccase, a versatile phenolic oxidase that uses oxygen as a natural reaction co-substrate. It can oxidise both phenolic derivatives and aromatic amines, which in homo- or heteromolecular reactions are coupled to form new organic compounds with unique properties and app

CH3COOAg with Laccase-like Activity for Differentiation and Detection of Aminoglycoside Antibiotics.

Aminoglycoside antibiotics (AGs) are widely used in medicine and animal husbandry, but they pose significant risks due to residual toxicity and antibiotic resistance. In this study, a novel chemical sensor based on the laccase-like activity of CH3COOAg was developed for the selective detection of AGs. CH3COOAg exhibited varying degrees of laccase-like activity in different buffers (MES, HEPES, and NaAc) and H2O, and five AGs showed distinct intensities of the inhibitory effect on the laccase-like activity of CH3COOA in different buffers and H2O. Therefore, a four-channel colorimetric sensor ar

The role of cyanide in the removal of type 2 copper from laccase.

We have improved a method for the removal of the type 2 copper from tree laccase under anaerobic reducing conditions and developed a mechanistic model. We identify two key steps in the reaction: (i) dissociation of copper(I) catalyzed by trace levels of cyanide in a weakly acidic medium and (ii) sequestration of the released metal by an appropriate chelator such as 2,9-dimethyl-1,10-phenanthroline. We maintain a steady-state concentration of cyanide in a pH 5.5 acetate buffer under a constantly exchanging nitrogen atmosphere by introducing a cyanometalate ion as a cofactor or by continuously i

Depolymerisation of Kraft Lignin by Tailor-Made Alkaliphilic Fungal Laccases.

Lignins released in the black liquors of kraft pulp mills are an underutilised source of aromatics. Due to their phenol oxidase activity, laccases from ligninolytic fungi are suitable biocatalysts to depolymerise kraft lignins, which are characterised by their elevated phenolic content. However, the alkaline conditions necessary to solubilise kraft lignins make it difficult to use fungal laccases whose activity is inherently acidic. We recently developed through enzyme-directed evolution high-redox potential laccases active and stable at pH 10. Here, the ability of these tailor-made alkaliphil

Modification of Deoxynivalenol by a Fungal Laccase Paired with Redox Mediator TEMPO.

Mycotoxins such as deoxynivalenol introduce a health risk to the food supply and are costly to manage or avoid. Technologies for reducing or eliminating the toxicity of deoxynivalenol could be useful in a variety of processes, such as in preserving the value as animal feed of byproducts of ethanol production. We characterized transformation products of deoxynivalenol that were formed by the combination of a fungal laccase paired with the chemical mediator 2,2,6,6-tetramethylpiperidine-N-oxyl (TEMPO), using chromatography, mass spectrometry, and nuclear magnetic resonance spectroscopy. Role of the ESCRT Pathway in Laccase Trafficking and Virulence of Cryptococcus neoformans.

The endosomal sorting complex required for transport (ESCRT) plays a crucial role in the transportation and degradation of proteins. We determined that Vps27, a key protein of the ESCRT-0 complex, is required for the transport of the virulence factor laccase to the cell wall in Cryptococcus neoformans Laccase activity was perturbed, as was melanin production, in vps27Δ strains. In the absence of VPS27, there was an accumulation of multivesicular bodies with vacuolar fragmentation and mistargeting of the vacuolar carboxypeptidase CPY/Prc1, resulting in an extracellular localization. Laccases in Food Industry: Bioprocessing, Potential Industrial and Biotechnological Applications.

Laccase is a multi-copper oxidase that catalyzes the oxidation of one electron of a wide range of phenolic compounds. The enzyme is considered eco-friendly because it requires molecular oxygen as co-substrate for the catalysis and it yields water as the sole by-product. Laccase is commonly produced by fungi but also by some bacteria, insects and plants. Due it is capable of using a wide variety of phenolic and non-phenolic substrates, laccase has potential applications in the food, pharmaceutical and environmental industries; in addition, it has been used since many years in the bleaching of p

Laccase-mediator system in the decolorization of different types of recalcitrant dyes.

Phloroglucinol, thymol, and violuric acid (VIO) were selected as laccase mediators after screening 14 different compounds with indigo carmine (indigoid dye) as a substrate. With the presence of these three mediators, a nearly complete decolorization (90-100%) was attained in 1 h. Thus, these three compounds were used as mediators for the decolorization of other four dyes. The results indicated that VIO was effective mediator in decolorization of Remazol brilliant blue R (RBBR, anthraquinoid dye) and Coomassie brilliant blue G-250 (CBB, triphenylmethane dyes), and Acid red (diazo dye). Locked ultrasound-induced active conformation of laccase in covalent organic frameworks.

Improving enzyme catalytic activity is a key challenge in biocatalysis. To this end, the present study explored a simple and versatile strategy to enhance the catalytic activity of laccase (LAC). LAC was pretreated with ultrasound (US) for 20 min, and the activity of the pretreated LAC (designated as LACUS) was increased by 2.6-fold compared with that of pristine LAC. This increase in activity was attributed to the partial depolymerization of the spatial structure of LAC during ultrasound pretreatment, leading to a shift from the α-helix structure to an irregularly coiled structure. This struc

Catalytic activation of Bacillus laccase after temperature treatment: Structural & biochemical characterization.

Laccases belong to a family of multicopper oxidases that have strong oxidation ability towards phenolic compounds. Here, more detailed investigations were carried out on a Bacillus laccase with remarkable behavior of activation after thermal treatment. The kcat of the enzyme was increased 2.5 fold after 50 min incubation at 70 °C. Copper content determination revealed a molar copper to protein ratio of 3.2 in the both sample. The present paper concerns the differences which are induced in enzyme structure after thermal treatment using common biochemical methods Intrinsic fluorescence of the en

Laccase Immobilization on Tannic Acid-Coated Nylon Membrane for Dye Decolorization.

Laccase catalyzes the oxidation of a wide array of substrates, yet its industrial applications are hindered by its low thermal and chemical stability, poor reusability, and high production costs. Enzyme immobilization is a cost-effective strategy for overcoming these limitations, enhancing stability and operational efficiency. In this study, laccase from Trametes versicolor (LTV) was immobilized onto a nylon membrane coated with 3-aminopropyltriethoxysilane (APTES) and tannic acid (TA). A mathematical model was employed to optimize enzyme immobilization parameters (e.g., pH, LTV concentration,
