Flow cytometry is the primary tool for phenotyping leukemias and related conditions. With the ever increasing numbers of antibodies commercially available, the ability to study and understand leukemias, myelodysplastic syndromes, and the myelodysplastic/myeloproliferative diseases is improving. Although the data generated by flow cytometry are not comprehensive enough to completely subtype leukemias or myelodysplastic syndromes into their myriad divisions, this wealth of information does provide phenotype, reproducible enumeration of blasts, certain prognostic information, and it can reveal th

We present here a patient with acute myeloid leukemia (M2) who developed fatal infective endocarditis. On admission, the patient (67-year-old male) had mitral stenosis and atrial fibrillation. Complete remission was achieved after induction chemotherapy. During the course of consolidation therapy, he developed sepsis caused by coagulase-negative staphylococcus, which was successfully treated with antibiotics. Thereafter, blood culture yielded multidrug-resistant staphylococcus epidermidis. An echocardiogram revealed mitral valve regurgitation with vegetation. He was diagnosed as having infecti

Two cases of acute myeloid leukemia (AML) presenting with bulky adenopathy are reported. Both patients were febrile at admission and showed massive and diffuse lymph node involvement, hepatomegaly, and splenomegaly. Erythematopapular leukemic skin lesions were present in one case at the onset and developed in the other at the time of relapse. Anemia, thrombocytopenia, and moderate leukocytosis were present in both. The presence of immature cells in peripheral blood and bone marrow allowed a rapid diagnosis of AML, FAB M1, in one patient. In the other case, owing to the paucity of immature cell

Clonal chromosome translocations, deletions, and inversions have been repeatedly observed for decades in approximately two thirds of all cases of acute myeloid leukemia (AML). With the dramatic advances in molecular biology that have occurred during the past two decades, these structural cytogenetic abnormalities have now provided invaluable clues as to the location of genes known or suspected of inducing leukemia. In most instances, leukemogenesis in AML results from gene fusion, when segments from two different genes are fused together to give rise to a chimeric structure consisting of the 5

The traditional classification and model of acute myeloid leukemia (AML), in common usage for much of the twentieth century, correlates poorly with treatment outcome, biologic studies, and genetic markers in AML, fails to accommodate large subgroups such as typical AML in the elderly or AML following myelodysplastic syndrome (MDS), and (except for acute promyelocytic leukemia) is not used in clinical decisions. Available data suggest an alternative classification and model that initially divides AML into two groups not recognized by traditional classification: MDS-related (MDR)-AML and true de

Leukemia cutis (LC) refers to cutaneous infiltration by leukemic cells, most often by acute myeloid leukemia (AML) in adults and by lymphoblastic leukemia in children. Although uncommon, LC is a clinically significant manifestation of extramedullary disease and may precede, coincide with, or follow bone marrow involvement. Recognition of AML-associated LC is essential, as it can be diagnostically challenging due to histopathologic overlap with inflammatory dermatoses and other cutaneous hematolymphoid neoplasms. Recent cohort-based studies have systematically characterized AML involving the sk

Relapsed or refractory acute myeloid leukemia (AML) continues to have a poor prognosis, highlighting the urgent need for novel therapeutic approaches. Immunotherapy, with its distinct mechanisms of action from chemotherapy or molecularly targeted therapies, offers a promising option for treatment-resistant cases. To date, numerous chimeric antigen receptor (CAR) T cell products targeting surface antigens such as CD33, CD123, and CLL-1 have been developed and evaluated in clinical trials. T cell receptor (TCR)-T therapies targeting intracellular antigens such as WT1 are also in development, wit

Acute myeloid leukemia (AML) is an aggressive hematologic malignancy defined by the clonal expansion of undifferentiated myeloid blasts with a block in differentiation and aberrant self-renewal. While recurrent genomic mutations are well-documented in AML, epigenetic dysregulation has emerged as an equally pivotal driver of leukemogenesis, a notion corroborated by the frequent recurrence of mutations in epigenetic regulators. Leukemic cells exhibit pervasive epigenetic alterations-including abnormal DNA methylation patterns, dysregulated histone modification, disrupted chromatin architecture a

NUP98::DDX10 is a rare fusion gene associated with acute myeloid leukemia (AML), for which the prognosis and indication for allogeneic hematopoietic stem cell transplantation are unknown. A 48-year-old woman was diagnosed with AML harboring NUP98::DDX10. The results of quantitative RT-PCR of the fusion mRNA as a minimal residual disease (MRD) marker guided the treatment. In August 2019, the patient achieved hematological remission following standard remission induction therapy with idarubicin and cytarabine. After four cycles of consolidation therapies, MRD was detected, and she underwent allo

Panhypopituitarism is an extremely rare disorder in acute myeloid leukemia (AML). To our knowledge, only six cases have been reported. Of note, central diabetes insipidus is known to be associated with AML in patients with certain cytogenic abnormalities, such as monosomy chromosome 7 and inv(3)(q21;q26). We describe a 30-year-old incarcerated woman with newly diagnosed AML who developed panhypopituitarism. Her cytogenetic studies revealed a normal karyotype, were negative for inv(3)(q21;q26), but were positive for FMS-like tyrosine kinase 3 internal tandem duplication.

Acute myeloid leukemia (AML) is a neoplasm of immature myeloid cells and is associated with a wide variety of clinical presentations, morphological features, immunophenotypes, and genetic findings. Recent advances in identification of cytogenetic abnormalities and mutations have provided novel insights into the pathogenesis of AML. Based on the above-mentioned parameters, the World Health Organization (WHO) classified AML into 25 subtypes, including 2 provisional entities, which differ in prognosis and treatment. In addition, certain mutations are associated with germline predisposition and in

CD33 is variably expressed on leukemia blasts in almost all patients with acute myeloid leukemia (AML) and possibly leukemia stem cells in some. Efforts to target CD33 therapeutically have focused on gemtuzumab ozogamicin (GO; Mylotarg), an antibody-drug conjugate delivering a DNA-damaging calicheamicin derivative. GO is most effective in acute promyelocytic leukemia but induces remissions in other AML types and received accelerated approval in the US in 2000. However, because a large follow-up study showed no survival improvement and increased early deaths the drug manufacturer voluntarily wi

Acute myeloid leukemia (AML) is a clonal malignancy characterized by ineffective hematopoiesis. Most AML patients present different cytogenetic and molecular defects associated with certain biologic and clinical features of the disease. Approximately 50–60 % of de novo AML and 80–95 % of secondary AML patients demonstrate chromosomal aberrations. Structural chromosomal aberrations are the most common cytogenetic abnormalities in about of 40 % of de novo AML patients. A relatively large group of intermediate risk patients with cytogenetically normal (CN) AML demonstrates a variety of outcomes.

PURPOSE: Leukemic hypopyon uveitis in acute myeloid leukemia (AML) is a very rare condition. We report this case of an unusual finding in a 2½-year-old boy in second remission after chemotherapy for relapsed AML.
METHODS: A young patient with an AML FAB M5 in second remission developed pain, photophobia, and conjunctival injections in the right eye. Recent bone marrow aspiration showed no blast increase and recent peripheral blood sampling presented no evidence of relapse.
RESULTS: The patient showed a viscous hypopyon and a myotic pupil in the right eye that was refractory to corticosteroid

In this report we focus on the importance of an accurate diagnosis of gastrointestinal complications during chemotherapy for acute myeloid leukemia. The leukemic infiltrtion of the digestive system may cause mucosal ulcers which can lead to bleeding or perforation. The immune system deficiency in this cohort of patients may result in necrotic enterocolitis (leukemic typhlitis), perianal inflammation, abscesses, and peritonitis. We describe a 37-year old male who presented in June 2004 with 2-month history of fever, weakness and sore throat, treated with antibiotic therapy. Physical examination
