Specificity of lipoprotein lipase binding to endothelial cells.

Lipoprotein lipase (LPL) hydrolyzes circulating lipoprotein triglyceride molecules while it is associated with the luminal surface of capillary endothelial cells. The precise molecular mechanism by which LPL attaches to these cells is unknown. LPL and a number of other molecules, including growth factors and clotting factors, bind to heparin-affinity gels and are eluted using high concentrations of salt. Of these molecules, antithrombin III and basic fibroblast growth factor have been shown to bind to specific cell surface heparan sulfate proteoglycans. Recent data from our laboratory (Sivaram

High muscle lipoprotein lipase activity in thyrotoxic patients.

Serum lipoprotein metabolism was studied in 7 women before and after treatment for thyrotoxicosis. Of the lipoprotein lipids, the triglyceride concentration in the low density lipoproteins (LDL) (P less than 0.01) and the cholesterol concentration in both LDL (P less than 0.01) and the high density lipoproteins (HDL) (P less than 0.05) increased significantly during treatment. These changes were accompanied by increases in apolipoprotein B (P less than 0.01) and A-I (P less than 0.05) concentrations in serum. Muscle lipoprotein lipase activity (LPLA) was increased in the thyrotoxic state by 46

[Changes of lipoprotein lipase and hepatic lipase and their significance in gallstone formation in rabbit model].

This experiment was made to investigate the changes of lipoprotein lipase(LPL) and hepatic lipase (HL) activity and their effects on gallstone formation in rabbit model in which the stones were induced by high cholesterol diet. Activities of plasma LPL and HL were determined; other data including concentration of plasma lipoprotein cholesterol, concentration of bile cholesterol and bile acids were also obtained. The results showed that with the rabbits continuously fed on high cholesterol diet, LPL activity heightened markedly (P < 0.05), and HL activity increased gradually (3 and 4 weeks grou

Lipoprotein lipase in highly vascularized structures of the eye.

Ocular tissues are highly dependent on lipid turnover and metabolism, which requires an uptake mechanism for fatty acids from lipoproteins. We studied the activity and expression of lipoprotein lipase (LPL), which catalyzes the hydrolysis of plasma triglycerides, in different ocular regions. Human and bovine eyes were dissected and various specialized anatomical areas were assayed for LPL activity, mRNA, and immunoreactivity. Variable levels of LPL activity were detected in all structures in human and bovine eyes. LPL activity was much higher in vascularized structures, such as ciliary body, i

Severe hypertriglyceridaemia and pancreatitis in a patient with lipoprotein lipase deficiency based on mutations in lipoprotein lipase (LPL) and apolipoprotein A5 (APOA5) genes.

A 44-year-old woman was admitted with pancreatitis caused by hypertriglyceridaemia (fasting triglycerides 28 mmol/L). She used oral contraceptives and ezetimibe 10 mg. She was overweight (body mass index 29.7 kg/m2). Diabetes mellitus was ruled out, as were nephrotic syndrome, alcohol abuse, hypothyroidism and dysbetalipoproteinaemia. Genetic analysis revealed mutations in two genes involved in triglyceride metabolism (apolipoprotein A5 and lipoprotein lipase [LPL]). The LPL activity was 45% compared with pooled healthy controls. The post-heparin triglyceride reduction was 6%, compared with a

Lipoprotein lipase and hormone-sensitive lipase activity and mRNA in rat adipose tissue during pregnancy.

To investigate the factors controlling maternal depot fat accumulation during early pregnancy and net decrease during late pregnancy, the activity and mRNA expression of adipose tissue lipoprotein lipase (LPL) and hormone-sensitive lipase (HSL) were related to several other lipid metabolic parameters. Virgin control rats, pregnant rats (at days 12, 15, 19, and 21), and lactating rats (at days 5 and 10 postpartum) were studied. In adipose lumbar tissue of late pregnant rats, LPL activity decreased to about one-third that of the virgin control animals, with < 10% of initial LPL mRNA expressed as

Deficiency of the Lipoprotein Lipase Pathway Modifies the Phenotypic Expression of Genetically Determined Lipoprotein(a).

Plasma lipoprotein(a) [Lp(a)] concentrations are primarily genetically determined, but whether disruption of triglyceride-rich lipoprotein metabolism modifies Lp(a) expression is unknown. Familial chylomicronemia syndrome (FCS) provides a human model to investigate this relationship. In the phase 3 Balance trial, 66 participants with genetically confirmed FCS (triglycerides ≥880 mg/dL) underwent Lp(a) measurement, apo(a) isoform phenotyping, and measurement of OxPL-apo(a) and OxPL-apoB. Results were compared with 66 age- and sex-matched participants with moderate hypertriglyceridemia from Esse

Effect of magnesium deficiency on post-heparin lipase activity and tissue lipoprotein lipase in the rat.

Previous studies have provided evidence that Mg deficiency affects lipid metabolism. The present experiments were designed to assess whether the hypertriglyceridemia associated with Mg deficiency was related to alterations in post-heparin lipase activity (PHLA). Mg-deficient and control diets were pair-fed to weanling Wistar rats for eight days and plasma lipoproteins were separated into various density classes by sequential preparative ultracentrifugation. Triglycerides were significantly increased in chylomicrons and in the very low density lipoprotein, low density lipoprotein and high densi

Postprandial adipose tissue lipoprotein lipase activity in primary hypertriglyceridemia.

The fasting activity of adipose tissue lipoprotein lipase has been previously reported to be either normal or reduced in subjects with a primary form of hypertriglyceridemia. The postprandial activity of adipose tissue lipoprotein lipase has not been previously reported in these subjects. In subjects with primary hypertriglyceridemia the fasting lipoprotein lipase activity eluted from pieces of adipose tissue by heparin and the enzyme activity present in extracts of acetone--ether tissue powders were similar to the level of enzyme activity found in normal subjects. There also was no difference

Familial lipoprotein lipase deficiency: a case of compound heterozygosity of a novel duplication (R44Kfs*4) and a common mutation (N291S) in the lipoprotein lipase gene.

Familial lipoprotein lipase (LPL) deficiency (FLLD) is a rare autosomal recessive genetic disorder caused by homozygous or compound heterozygous mutations in the LPL gene. FLLD individuals usually express an impaired or non-functional LPL enzyme with low or absent triglyceride (TG) hydrolysis activity causing severe hypertriglyceridaemia. Here we report a case of FLLD in a 29-year-old man, who initially presented with eruptive cutaneous xanthomata, elevated plasma TG concentration but no other co-morbidities. Subsequent genetic testing of the patient revealed compound heterozygosity of a novel

Lipoprotein lipase gene mutations and the genetic susceptibility of preeclampsia.

In the pathogenesis of preeclampsia, endothelial cell activation or dysfunction is a central theme, and marked dyslipidemia may contribute to endothelial cell dysfunction. The objective of this study was to evaluate the association between preeclampsia and mutations within the lipoprotein lipase (LPL) gene. DNA was extracted from whole blood or cheek swabs of 250 preeclamptic patients, 265 control subjects, and 106 offspring of preeclamptic patients (all white). Control subjects were women who had undergone >/=2 term pregnancies unaffected by preeclampsia. All samples were genotyped for 3 LPL

Postheparin plasma lipoprotein and hepatic lipases in preterm neonates.

Postheparin plasma lipoprotein lipase and hepatic lipase activities of 11 preterm infants aged 1 to 4 days were measured 15 min after a heparin bolus of 100 IU/kg and during an exchange transfusion performed with fresh heparinized blood. Each infant had a birth weight (range, 1210-3490 g) appropriate for gestational age (range, 28-36 (wk). Eight of the infants (group 1) were in good clinical condition while three (group 2) suffered from septic shock. After the heparin bolus and during the exchange transfusion, lipoprotein and hepatic lipase activities in group 1 were higher than in term infant

Lipoprotein Lipase and Its Delivery of Fatty Acids to the Heart.

Ninety percent of plasma fatty acids (FAs) are contained within lipoprotein-triglyceride, and lipoprotein lipase (LPL) is robustly expressed in the heart. Hence, LPL-mediated lipolysis of lipoproteins is suggested to be a key source of FAs for cardiac use. Lipoprotein clearance by LPL occurs at the apical surface of the endothelial cell lining of the coronary lumen. In the heart, the majority of LPL is produced in cardiomyocytes and subsequently is translocated to the apical luminal surface. Here, vascular LPL hydrolyzes lipoprotein-triglyceride to provide the heart with FAs for ATP generation

Lipoprotein Lipase as a Prognostic Marker in Chronic Lymphocytic Leukemia.

The marked clinical heterogeneity of CLL makes early prognosis assessment important. Lipoprotein lipase (LPL) has been shown to confer adverse prognosis in CLL, recent data indicating it might also contribute to CLL cell survival and metabolism. We determined LPL mRNA expression in unselected peripheral blood of 84 CLL patients by RT PCR. Results were correlated with other prognostic markers and outcome. 30/84 (40 %) of cases were LPL positive based on the cutoff established by ROC analysis. In LPL positive patients significantly shorter median survival (136 vs 258 months, p < 0.0001) and time

[Method for the molecular diagnosis of lipoprotein lipase genetic deficiency].

Modifications of plasma lipid profile is one of the major causes of a high cardiovascular risk. They can be the consequences of mutations in the gene encoding lipoprotein lipase (LPL), an enzyme that has an important role in the metabolism of plasma lipoproteins. The aim of the present study was to put into practice a method for detecting the Gly188Glu mutation in the LPL gene. The search was performed on a group of 107 patients with cardiovascular diseases and/or dyslipidemias. DNA investigation consisted, in a first stage, in the enzymatic digestion of exon 5 of the LPL gene, previously ampl
