[Effects of strophanthin, mefenamic acid and dexamethasone on the inflammatory and reparative processes in the heart in experimental myocarditis].

In isadrin-induced myocarditis, the rat heart shows an increase in the activity of acid phosphatase, glucose-6-phosphate dehydrogenase. The heart weight also rises, whereas ATPase activity and magnesium content fall. Under these conditions strophanthin produces a marked decrease in acid phosphatase activity, stimulates the activity of glucoso-6-phosphate dehydrogenase, and demonstrates a tendency to increasing the activity of ATP-splitting enzymes, to reducing the activity of ATP-synthetase, and to the recovery of the magnesium content. Mefenamic acid and dexamethasone also decrease the activi

Micro- and nanoencapsulation strategies for mefenamic acid delivery: carrier design, release mechanisms, and preclinical performance.

Mefenamic acid, a Biopharmaceutics Classification System (BCS) class II non-steroidal anti-inflammatory drug which is widely used for the treatment of pain and inflammation, but its activity is limited by poor aqueous solubility, rapid metabolism and gastrointestinal toxicity. These constraints increase the first-pass loss, require frequent oral dosing of the drug and reduce bioavailability. We review nanocarrier approaches that enhance dissolution, absorption, and sustained delivery while mitigating the adverse effects. Vesicular systems (niosomes, liposomes, ethosomes) improve encapsulation,

Consensus on the Use of Mefenamic Acid in Pediatric Practice (MAPP): Perspectives From Indian Pediatricians.

Fever and pain are among the most common reasons for pediatric consultations, requiring effective and safe management strategies. Mefenamic acid, a well-established member of the nonsteroidal anti-inflammatory drug (NSAID) class, offers distinct advantages due to its unique pharmacological profile, including preferential cyclooxygenase-2 (COX-2) inhibition, E-type prostanoid (EP) receptor blockade, and nucleotide-binding domain, leucine-rich-containing family, pyrin domain-containing-3 (NLRP3) inflammasome inhibition. These properties enable it to address both inflammatory and non-inflammatory

Testing on continuous production of mefenamic acids-Design of experiment through simulation and process optimisation.

In the pharmaceutical manufacturing industry, continuous production methods have been recognised as providing several benefits compared to traditional batch production. These benefits include increased flexibility, higher product output, enhanced quality assurance through better monitoring techniques, and more consistent distribution of Active Pharmaceutical Ingredients (APIs). Despite these clear advantages, there is a lack of research focused on the simultaneous optimisation of multiple sub-processes in continuous manufacturing. This study explores the optimisation processes of continuous ph

The efficacy of etoricoxib vs mefenamic acid in the treatment of primary dysmenorrhoea: a randomised comparative trial.

Dysmenorrhoea is painful menstruation that occurs in 45-72% of all women. This was a prospective randomised study of the efficacy of etoricoxib (Arcoxia) compared with mefenamic acid (Ponstan) in treating primary dysmenorrhoea. All single, sexually inactive women with primary dysmenorrhoea were randomised into two groups (mefenamic acid and etoricoxib) of pain relief and underwent a cross-over study. The success of treatment as evidenced by pain relief, the side-effects and complications were observed and analysed. Some 80% (20 women) had significantly better pain relief with etoricoxib, compa

Diclofenac sodium and mefenamic acid: potent inducers of the membrane permeability transition in renal cortex mitochondria.

The ability of nonsteroidal anti-inflammatory drugs (NSAIDs) to induce Ca(2+)-mediated/cyclosporin A-sensitive mitochondrial membrane permeability transition (MMPT) was evaluated by monitoring swelling of isolated rat renal cortex mitochondria in the presence of 20 microM CaCl2. Dipyrone and paracetamol did not induce MMPT, while piroxicam and acetylsalicylic acid (and its metabolite salicylate) were poor inducers. In contrast, diclofenac sodium and mefenamic acid were potent triggering agents, inducing MMPT at 2 microM, a concentration below those previously shown to uncouple and/or inhibit o

Single-blind study of diflunisal versus mefenamic acid in the treatment of pain after Colles' fracture.

A single-blind study was carried out in 52 patients with severe pain after Colles' fracture to assess the analgesic efficacy and tolerability of 500 mg diflunisal twice daily compared with that of 500 mg mefenamic acid given 3-times daily over a period of 5 days. The results showed that both treatments were effective in relieving pain, night pain and limitation of movement by pain, and there was no significant difference between the response in the two groups. Both drugs were tolerated and only 3 patients (2 on diflunisal) reported mild drug-related side-effects.

Urinary lipid changes during the development of chemically-induced renal papillary necrosis: a study using mefenamic acid and N-phenylanthranilic acid.

Acute renal papillary necrosis (RPN) in animals is characterized by increased renal lipid accumulation. The excretion of renal lipids into urine has been determined to evaluate their possible use as sensitive early biomarkers for the diagnosis of RPN. This study investigates injury induced by two model nephrotoxins, mefenamic acid (MFA), a non-steroidal anti-inflammatory drug (NSAID), and its analogue N-phenylanthranilic acid (NPAA). Oral NPAA was given repeatedly at doses of 100, 250 and 500 mg kg(-1) daily for 5 days, followed by a 2 day respite over the weekend, and then four further daily

A case of Stevens-Johnson syndrome, cholestatic hepatitis and haemolytic anaemia associated with use of mefenamic acid.

A woman with a history of drug allergy, renal impairment and carcinoma of the breast with pulmonary micrometastases developed haemolytic anaemia and Stevens-Johnson syndrome following the use of mefenamic acid, paracetamol (acetaminophen) and furosemide (frusemide). In addition there was severe cholestatic hepatitis in the absence of clinical evidence of sepsis, biliary obstruction or recurrent metastases. The rash resolved on steroid therapy but the patient eventually died from both renal and liver failure. Acute tubular necrosis with a background of chronic tubulointerstitial nephritis was a

Phenotypic screening of nonsteroidal anti-inflammatory drugs identified mefenamic acid as a drug for the treatment of schistosomiasis.

BACKGROUND: Treatment and control of schistosomiasis, one of the most insidious and serious parasitic diseases, depend almost entirely on a single drug, praziquantel. Since the funding for drug development for poverty-associated diseases is very limited, drug repurposing is a promising strategy. In this study, 73 nonsteroidal anti-inflammatory drugs (NSAIDs) commonly used in medical and veterinary fields were evaluated for their anti-schistosomal properties. METHODS: The efficacy of NSAIDs was first tested against adult Schistosoma mansoni ex vivo using phenotypic screening strategy, effectiv

Exploring the in vitro urease inhibitory potential, molecular docking and dynamics simulations investigation of mefenamic acid derivatives.

AIMS: To biologically evaluate a series of hydrazide derivatives of mefenamic acid as anti-urease agents, elucidating their mechanism of action through integrated computational and experimental approaches. METHOD: The inhibitory mechanism was interpreted through comprehensive computational studies: molecular docking to identify binding modes within the catalytic site of H. pylori urease (PDB: 4UBP), Density Functional Theory (DFT) calculations to evaluate global chemical reactivity and frontier molecular orbitals (FMOs), and Molecular Dynamics (MD) simulations to confirm complex stability. Treatment of Epidermal Pathology in a Pediatric Patient With Keratitis-Ichthyosis-Deafness (KID) Syndrome With Topical Mefenamic Acid.

Keratitis-ichthyosis-deafness (KID) syndrome is a rare genetic condition typically presenting at birth with ichthyosiform erythroderma and bilateral hearing loss and later progressing to diffuse keratodermatous plaques with scaling. The condition is associated with mutations in the GJB2 gene, which lead to aberrant activation of connexin hemichannels in keratinocytes. While no targeted treatment currently exists, a previously published in vivo study demonstrated that flufenamic acid (FFA), a nonspecific connexin inhibitor, reduces epidermal pathology in transgenic mouse models expressing the l

Mefenamic acid in combination with ribavirin shows significant effects in reducing chikungunya virus infection in vitro and in vivo.

Chikungunya virus (CHIKV) infection is a persistent problem worldwide due to efficient adaptation of the viral vectors, Aedes aegypti and Aedes albopictus mosquitoes. Therefore, the absence of effective anti-CHIKV drugs to combat chikungunya outbreaks often leads to a significant impact on public health care. In this study, we investigated the antiviral activity of drugs that are used to alleviate infection symptoms, namely, the non-steroidal anti-inflammatory drugs (NSAIDs), on the premise that active compounds with potential antiviral and anti-inflammatory activities could be directly subjec
