Microsporidia, a Highly Adaptive Organism and Its Host Expansion to Humans.

Emerging infectious disease has become the center of attention since the outbreak of COVID-19. For the coronavirus, bats are suspected to be the origin of the pandemic. Consequently, the spotlight has fallen on zoonotic diseases, and the focus now expands to organisms other than viruses. Microsporidia is a single-cell organism that can infect a wide range of hosts such as insects, mammals, and humans. Its pathogenicity differs among species, and host immunological status plays an important role in infectivity and disease severity. Disseminated disease from microsporidiosis can be fatal, especi

[Investigation of Microsporidia prevalence by different staining methods in cases of diarrhea].

Microsporidia, depending on their different species, generally lead to self-limited, sporadic and mild infections such as diarrhea, corneal ulcer and myositis. They are considered as opportunistic pathogens in HIV-positive patients however in recent years Microsporidia have been detected also in immunocompetent individuals as a cause of diarrhea. Diagnosis of Microsporidia depends on the detection of spores or different developmental stages of protozoon in stool, urine, sinus aspirates, nasal discharge, bronchoalveolar lavage or tissue biopsies. Diagnosis of Microsporidia infections is usually

[Natural transmission of microsporidia (Encephalitozoon cuniculi) by way of the chicken egg].

100 chicken embryos were examined for naturally occurring infections with Encephalitozoon cuniculi (E.c.). Two embryos were found to be dead, the others were killed at an age of 18 days. Samples from the oesophagus, intestine, liver, kidneys, heart and brain were collected and examined by immunohistochemical methods. E.c. was found in about 40% of the embryos. This microsporidian was observed in all organs, particularly, however, in the oesophagus, intestine, brain and heart. The number of E.c. was significantly larger in the two embryos that died than in the others. The investigations demonst

Systemic dissemination by a newly recognized intestinal microsporidia species in AIDS.

OBJECTIVE: Primarily to determine whether an intestinal microsporidian recently identified in AIDS patients disseminates from the bowel to infect other organs. DESIGN: Disseminated microsporidiosis has been reported in immunocompromised humans, but never due to Enterocytozoon bieneusi, the most common species in AIDS patients and one that evidently infects only enterocytes. In animals, dissemination follows ingestion of Encephalitozoon cuniculi spores, apparently via macrophages, and pathology occurs in, for example, kidneys and brain. A second, un-named Encephalitozoon-like intestinal micros

Molecular Diagnosis of Microsporidia among Immunocompromised Patients in Kuala Lumpur, Malaysia.

Microsporidia are obligate intracellular parasitic fungi causing chronic diarrhea, particularly among immunocompromised patients. The current method used for diagnosis is based on conventional microscopy, which does not differentiate parasites at the species level. The present study was carried out to identify microsporidian species in immunocompromised patients. From March 2016 to March 2017, a total of 289 archived stool samples were examined microscopically for microsporidian spores using Gram-chromotrope Kinyoun (GCK) stain. Positive stool samples by microscopy were subjected to polymerase

An Investigation of Microsporidia Species in Humans Living in Düzce and Bolu Provinces of Türkiye.

OBJECTIVE: The literature about microsporidia clearly shows that the prevalence of microsporidia varies at different rates depending on the sampling status and region in the studies. However, the majority of studies have been conducted in immunocompromised individuals. It is vital to determine the prevalence of pathogens in society in order to protect immunocompromised individuals from opportunistic pathogens. Consequently, the present study investigated stool samples from Düzce and Bolu provinces for microsporidia, irrespective of disease group. METHODS: A study of 400 stool samples from Düz

Monitoring the Capacity of Microsporidia MB Transgenerational Spread in Anopheles arabiensis Populations.

Microsporidia MB is a natural symbiont of Anopheles arabiensis Patton, 1905 that blocks Plasmodium transmission without compromising mosquito fitness. Developing this symbiont for malaria control requires a thorough understanding of its transmission dynamics across generations under environmentally relevant conditions. This study aimed to evaluate the transgenerational spread of Microsporidia MB in semi-field settings. From field-collected infected females (G0), several independent replicates of Microsporidia MB-infected An. arabiensis were established and maintained under semi-field condition

Plastic parasites: extreme dimorphism creates a taxonomic conundrum in the phylum Microsporidia.

In this paper, we combine field observations of highly statistically significant co-occurrence with histopathological, ultrastructural and molecular phylogenetic analyses, to provide evidence for extreme morphological plasticity in a microsporidium parasite infecting the musculature of marine crabs. The parasite appears to alternate between lineages that culminate in production of either bizarre needle-like spores in the peripheral sarcoplasm of heart and skeletal muscle fibres (reminiscent of Nadelspora canceri infecting Cancer magister) or alternatively, Ameson-like spores with pronounced su

Public health importance of Brachiola algerae (Microsporidia)--an emerging pathogen of humans.

Brachiola algerae, a parasite of Anopheles mosquitoes, has also been isolated from a human cornea, a cutaneous nodule and deep muscle tissue. All three human isolates of B. algerae are morphologically, serologically, and genetically similar to the mosquito-derived isolates including the original isolate of Vavra and Undeen. All of these isolates grew well in mammalian cell cultures at 37 degrees C and produced spores. Transmission electron microscopy revealed that all developmental stages including meronts, sporoblasts and spores were diplokaryotic and developed in direct contact with the host

The dissemination potential of Microsporidia MB in Anopheles arabiensis mosquitoes is modulated by temperature.

Microsporidia MB, a vertically transmitted endosymbiont of Anopheles mosquitoes, shows strong potential as a malaria control agent due to its ability to inhibit Plasmodium development within the mosquito host. To support its deployment in malaria transmission reduction strategies, it is critical to understand how environmental factors, particularly temperature, influence its infection dynamics. In this study, we investigated the impact of four temperature regimes (22 °C, 27 °C, 32 °C, and 37 °C) on Microsporidia MB prevalence and infection intensity by rearing mosquito larvae under controlled

[Importance of antiproteases in the treatment of microsporidia and/or cryptosporidia infections in HIV-seropositive patients].

Diarrhea due to infection with Microsporidium (M) or Cryptosporidium (C) raises significant therapeutic challenges in HIV-infected patients. The usefulness of protease inhibitor therapy was evaluated in 20 HIV-positive patients with positive tests for M and/or C. There were 17 men and three women with a mean age of 42.5 years (range, 26-64 years). Two patients had category B disease and 18 category C disease according to the 1993 CDC classification scheme (CD4 count before therapy, 72/mm3; mean viral burden, 4.6 log). Seventeen patients had chronic diarrhea (due to M in 12 cases and to C in fi

Use of the fluorochrome calcofluor white in the screening of stool specimens for spores of microsporidia.

Microsporidia are obligate intracellular protozoal pathogens associated with chronic diarrhea in individuals infected with HIV. Direct detection methods for microsporidial spores in stool include chromotrope-based, fluorochrome, and immunofluorescent stains. The authors compared the ability to detect microsporidial spores in 168 stool specimens using two stains: a chromotrope-based modified trichrome stain and a fluorochrome stain, calcofluor white (Cellufluor, Polysciences, Warrington, PA). In addition to being faster and easier to perform, the calcofluor white stain was found to be more sens

Microsporidia in duodenal biopsies from 72 HIV-infected patients with abdominal complaints.

In order to evaluate the capacity of routine histological examination to detect microsporidiosis, a retrospective study was performed on 72 duodenal biopsies from 72 HIV-infected patients with upper abdominal symptoms of unknown cause. Two light microscopic cytological staining techniques, modified trichrome stain and the fluorochrome Calcofluor, were used. Two cases of microsporidiosis were detected among the 20 patients with prolonged diarrhoea of unknown origin in whom no etiological agent had been demonstrated by stool examination, mycobacterial and cytomegalovirus culture of biopsies, and

Host range of Microsporidia MB in aquatic macrofauna inhabiting Anopheles breeding sites.

Microsporidia MB, a recently discovered endosymbiont Anopheles arabiensis mosquitoes, shows great potential as a malaria-blocking tool due to its Plasmodium-blocking phenotype. The symbiont utilizes vertical (mother-to-offspring) and horizontal (sexual) transmission routes to propagate across anopheline populations. Microsporidia MB occurs throughout the Anopheles life cycle, with no evidence to date of the existence of a secondary host. In this study, we conducted a comprehensive molecular screening of Microsporidia MB in aquatic macrofauna associated with Anopheles larval habitats to determi
