Neuritic plaques and vessels of visual cortex in aging and Alzheimer's dementia.

Changes in microvascular dimensions occurring with normal aging and Alzheimer's dementia were measured in thick sections of postmortem human visual cortex stained for alkaline phosphatase. Capillary density was decreased to the same degree in both normal aged and demented aged subjects. The fields selected for analysis in both groups included all cortical laminae and, where possible, amyloid-cored neuritic plaques. The mean density of such plaques in these selected fields was slightly but not significantly higher in the demented group. In both groups plaques were more plentiful in visual lamin

Neurofibrillary tangles in human upper cervical ganglia. Morphological study with immunohistochemistry and electron microscopy.

Neurofibrillary tangles (NFTs) are one of the main pathological hallmarks of Alzheimer's disease or senile dementia, and are seen in the cerebral cortex and some other nuclei in the central nervous system (CNS). No NFTs have been reported in the human peripheral nervous system, although NFTs were recognized in the dorsal root ganglion of the aged rodents. We report here the presence of NFTs in the upper cervical ganglia (UCGs), but not in the stellate nor in the celiac ganglia, of an elderly patient, who was not demented and had only minimal senile changes in the CNS. Immunohistochemically the

Neurofibrillary tangles in association with congenital hydrocephalus.

Morphologic study of the brain from a retarded patient with progressive congenital hydrocephalus revealed widespread neurofibrillary tangles in the cerebral cortex and the brain stem. Senile plaques were not found. The tangles consisted of paired helical filaments. Scanning electron microscopic energy spectrometric analysis failed to demonstrate abnormal accumulation of aluminum or other elements in the involved neurons. For the first time the association of congenital hydrocephalus and neurofibrillary tangles is reported. This observation is consistent with the hypothesis of interference with

Isolation and partial characterization of neurofibrillary tangles and amyloid plaque core in Alzheimer's disease: immunohistological studies.

Fractions enriched in neurofibrillary tangles (NFT) and amyloid fibrils were isolated from the cerebral cortex of three cases of senile dementia of the Alzheimer type. Distilled water suspensions of these fractions were excluded from all pore size gels and resisted digestion with various proteolytic enzymes. Formic acid/chloroform treatment of each fraction resulted in the appearance of 4,000-6,000, 15,000-17,000 and 24,000 molecular weight proteins, with concomitant diminution in the amount of excluded material at the top of each gel. The 4,000-6,000 dalton band was best seen in fractions con

Senile changes in the human neocortex and hippocampus compared by the use of the electron and light microscopes.

The severity of certain "senile" changes, namely senile plaques, neurofibrillary tangles and granulovacuolar degeneration in the neocortex and hippocampus, was estimated by light microscopy, in brains obtained at autopsy from 16 cases aged 24 to 93 years. An assessment of certain features of plaques and tangles as well as Hirano-bodies (also associated with senile change) was made by electron microscopy. A statistically significant correlation was found between the electron-microscopic and light-microscopic findings. The findings in an additional 4 cases aged 39 to 65 years in which only the c

The claustrum in the dog brain.

The structure of the claustrum was studied in the dog brain using Weigert's, Klüver-Barrera's, Manns' and Nissl's methods. It consists of two main parts arranged one above the other. The dorsal part is situated in the depth of the neocortex and extends from the gyrus orbitalis to the gyrus compositus posterior. The ventral part of the claustrum underlying the olfactory cortex continues from the cuadal fragments of the olfactory peduncle to the entorhinal area, where it fuses with its deep layers. The claustrum can be regarded as a fragment of a bigger cellular formation present in different pa

Manganese-induced neurotoxicity and its role in frontotemporal dementia: Insights from the frontotemporal cortex of BALB/c mice.

BACKGROUND: Manganese (Mn), an essential trace metal, has been identified as a potential contributor to neurotoxicity when misregulated, with implications for cognitive decline and motor dysfunctions. Frontotemporal dementia (FTD) is a devastating neurological condition that causes increasing cognitive, behavioral, and linguistic deficits in the frontal and temporal lobes of the brain. The specific mechanisms behind FTD remain unknown. This study investigates Mn-induced neurotoxicity as it affects frontotemporal cortex of BALB/c mice as a model for FTD. METHODS: BALB/c mice were given varying

Ectopic class II major histocompatibility antigens in Hirschsprung's disease and neuronal intestinal dysplasia.

Although the etiology of Hirschsprung's disease and neuronal intestinal dysplasia remains obscure, both have histological abnormalities involving ganglion cells and neuronal elements. Searching for a common pathway that may inhibit normal maturation of neurogenic precursors, we examined the possible role of an immune mechanism in the maldevelopment of the enteric neural network. Six patients with Hirschsprung's disease were studied by comparing biopsy specimens from diseased colon with ones taken from proximal ganglionic colon in the same patients. These were similarly compared with colonic bi

Comprehensive Overview of Alzheimer's Disease: Etiological Insights and Degradation Strategies.

Alzheimer's disease (AD) is the most prevalent neurodegenerative disorder and affects millions of individuals globally. AD is associated with cognitive decline and memory loss that worsens with aging. A statistical report using U.S. data on AD estimates that approximately 6.9 million individuals suffer from AD, a number projected to surge to 13.8 million by 2060. Thus, there is a critical imperative to pinpoint and address AD and its hallmark tau protein aggregation early to prevent and manage its debilitating effects. Amyloid-β and tau proteins are primarily associated with the formation of p

The ganglion cell response to optic nerve injury in the cat: differential responses revealed by neurofibrillar staining.

The early responses of cat retinal ganglion cells to axotomy have been examined using neurofibrillar and Nissl-stained wholemounts. We were interested to learn whether the enhanced neurofilament expression, seen in a number of neuronal systems, was also present in different neuronal populations of the cat retina and could be used to study the distribution of these cells. We found that beta ganglion cells degenerate very rapidly after axotomy with the nuclei becoming pyknotic within a few days. Few beta cells showed increased neurofibrillar staining of the dendrites. The cell body degenerated p

Apolipoprotein E immunoreactivity in cerebral amyloid deposits and neurofibrillary tangles in Alzheimer's disease and kuru plaque amyloid in Creutzfeldt-Jakob disease.

During the course of our immunohistochemical studies on the change of lipids in Alzheimer's disease brains by using antibody to apolipoprotein E, a protein having a special relevance to nervous tissue, we unexpectedly found that apo E immunoreactivity was associated with amyloid in both senile plaques and cerebral vessels and neurofibrillary tangles. The immunoreactivity was also found in amyloid of kuru plaques in Creutzfeldt-Jakob disease. Pretreatment of the sections with formic acid greatly enhanced immunoreactivity of senile and kuru plaques to antibody to apo E.

Subacute dementia with necrotising encephalitis selectively involving the fornix and splenium. Retrograde development of Alzheimer's neurofibrillar tangles in the subiculum.

An elderly woman developed subacute progressive dementia with predominant impairment in recent memory and CT findings indicating an enhancing lesion near the splenium of the corpus callosum. Autopsy four months after the onset of the symptoms revealed necrotising encephalitis selectively involving the bilateral fornices and the adjacent splenium. Degeneration was marked in the contiguous right fimbria hippocampi. Particularly numerous Alzheimer's neurofibrillar tangles (NFTs) were present in the right subiculum, while they were scattered in Sommer's sectors and parahippocampal gyri and were pr

Phenotypic heterogeneity in familial Alzheimer's disease: a study of 24 kindreds.

We report the clinical and neuropathological characteristics occurring in 180 demented individuals from 24 kindreds with familial Alzheimer's disease (FAD). Each family had at least two affected generations, and at least one autopsy or brain biopsy was compatible with the diagnosis of AD. Forty-nine neuropathological specimens or reports were reviewed. Mean age of onset for the total group was 54.7 years +/- 11.5, with a large range of 30 to 84 years. Mean age at death was 63.5 years +/- 12.2, with a range of 46 to 85. Mean duration of disease was 8.8 years +/- 4.4, with a range of 1 to 23 yea

Relationship between numbers of cortical argentophilic and congophilic senile plaques in the brains of elderly people with and without senile dementia of Alzheimer type.

Four neocortical regions (frontal, parietal, temporal, occipital) of 44 cases were impregnated by a silver stain to show neuritic plaques and stained with Congo red to show amyloid plaques. In 11 cases with large numbers of argentophilic plaques these plaques decreased significantly with age while the reverse appeared to be true for the congophilic plaques. In the remaining 34 cases which had few argentophilic plaques the numbers of congophilic plaques increased significantly with age in the parietal and occipital regions. These results suggest that the neuritic component of senile plaques pre

Serum antibodies to neurofilament antigens in patients with neurological and other diseases and in healthy controls.

A total of 529 sera from patients with a wide variety of neurological and non-neurological diseases, and 101 sera from healthy control subjects, were examined by indirect immunofluorescence for the presence of autoantibodies to neurofilament antigens. Antibodies were found in approximately 50% of sera from patients with spongiform encephalopathies, 15-30% of sera from patients with other neurological and non-neurological diseases, and 7% of sera from healthy controls. The antigenic stimulus to these autoantibodies in very diverse disease processes is unknown, but as presently assayed, they are

Atypical senile dementia with widespread Lewy type inclusion in the cerebral cortex.

An atypical case of senile dementia is reported which shows senile plaques, neurofibrillary tangles, granulovacuolar degeneration, and Hirano bodies and numerous Lewy bodies in the pigmented neurons of the brain stem, and Lewy-body-like inclusions in the cerebral cortex. The inclusions in the cerebral cortex had a strong resemblance to the Lewy bodies in the substantia nigra, though a few histochemical and ultrastructural differences were observed. The pathological findings were unexpected since the parkinsonian syndrome was not recognized.

Heme oxygenase-1 is associated with the neurofibrillary pathology of Alzheimer's disease.

Heme oxygenase-1 is an important enzyme that degrades heme, a pro-oxidant, leading to the formation of antioxidant molecules. In this study we demonstrate by immunocytochemistry close association of heme oxygenase-1 with Alzheimer neurofibrillary pathology and with the neurofibrillary tangles found in progressive supranuclear palsy and subacute sclerosing panencephalitis. In Alzheimer's disease, using two different rabbit antisera against heme oxygenase-1 protein, we localized, using immunocytochemical methods, heme oxygenase-1 to neurofibrillary tangles, senile plaque neurites, granulovacuola

Age-dependent deterioration of neuronal membranes and the pathogenesis of Alzheimer's disease: a hypothesis.

Senile dementia of the Alzheimer's type (SDAT) is considered either a specific pathological condition unrelated to normal aging or an accumulation of brain aging alterations leading to clinical dysfunction. In the present paper a hypothesis is formulated to reconcile these two issues to a common denominator, i.e. the increased age-dependent deterioration of the neuronal membrane. If this holds true, normal aging and SDAT may be considered as two different aspects of the same problem and specific interventions to slow down the former can delay the onset of the latter to older ages.

A case of combined Pick's disease and Alzheimer's disease.

The diagnosis of combined Pick's and Alzheimer's disease is rare, and over the years different authors have used different criteria to arrive at such a diagnosis. A case is reported of presenile dementia in which the histological changes of Pick's disease and Alzheimer's disease were mingled. The brain showed no focal atrophy, but the Pick changes were most numerous in the hippocampus and in the temporal lobe. An antibody against the 155 kilodalton component of neurofilaments demonstrated not only neurofibrillary tangles and components of senile plaques, but also Pick's inclusions.
