Postmortem neuropathological features secondary to boron neutron capture therapy for glioblastoma multiforme.

This postmortem study of 12 patients with glioblastoma multiforme (GBM) treated with boron neutron capture therapy (BNCT) employing an epithermal neutron beam and p-boronophenylalanine describes the neuropathological findings in patients receiving a relatively high radiation dose to the tumor, but a relatively low radiation dose to the normal brain. In addition to a standardized neuropathology panel of sections, we used individual treatment dosimetry maps to select sections along the projected maximum radiation beam pathway. We found that the normal neuroparenchyma exposed to the highest radia

Boron encapsulated in a liposome can be used for combinational neutron capture therapy.

Boron neutron capture therapy (BNCT) is an attractive approach to treat invasive malignant tumours due to binary heavy-particle irradiation, but its clinical applications have been hindered by boron delivery agents with low in vivo stability, poor biocompatibility, and limited application of combinational modalities. Here, we report boronsome, a carboranyl-phosphatidylcholine based liposome for combinational BNCT and chemotherapy. Theoretical simulations and experimental approaches illustrate high stability of boronsome. Then positron emission tomography (PET) imaging with Cu-64 labelled boron

A realistic appraisal of boron neutron capture therapy as a cancer treatment modality.

Boron neutron capture therapy (BNCT) is a binary therapeutic modality based on the nuclear capture and fission reactions that occur when the stable isotope boron-10 is irradiated with neutrons to produce high-energy alpha particles and recoiling lithium-7 nuclei. In this Commentary we will focus on a number of papers that were presented at a Symposium entitled "Current Clinical Status of Boron Neutron Capture Therapy and Paths to the Future", which was held in September 2017 at the China National Convention Center in Beijing. Results were presented by clinicians from Japan, Finland, the United

10Boron-doped carbon nanoparticles as delivery platforms for boron neutron capture therapy and photothermal therapy.

The current clinical application of the boron drug boronophenylalanine (BPA) in BNCT faces various issues owing to its low boron loading (approximately 5%), which limits its therapeutic efficacy. Therefore, the development of boron drugs with higher boron contents is essential. Enhancing the boron content in boron drug materials is the focus of this study. Two-dimensional (2D) boron nitride-doped nano graphene (BNNG) with a high boron content of 24.97% ± 1.14% w/w was synthesized via the chemical vapor deposition (CVD) method. The size of BNNG was controlled through gradient density centrifuga

Compassionate boron neutron capture therapy for locally recurrent nasopharyngeal cancer: A retrospective study.

BACKGROUND: Boron neutron capture therapy (BNCT) is a targeted form of particle radiotherapy (RT) that better spares normal tissues than does conventional photon RT. We describe our experience with compassionate-use BNCT for locally recurrent nasopharyngeal cancer (rNPC) after prior RT. METHODS: Data from patients with rNPC who received BNCT outside of clinical trials at the Tsing-Hua Open-Pool Reactor between 2020 and 2024 were retrospectively analyzed. RESULTS: Ten patients (eight men and two women) with a median age of 54 years and recurrent stage T2-T4N0-N1 disease were included. The med

Size-Tunable Boron Nanoreactors for Boron Neutron Capture Synergistic Chemodynamic Therapy of Tumor.

Boron neutron capture therapy (BNCT) stands out as a noninvasive potential modality for invasive malignant tumors, with boron drugs playing a crucial role in its efficacy. Nevertheless, the development of boron drugs with biodegradability, as well as high permeability and retention effects, continues to present significant challenges. Here, we fabricate a size-tunable boron nanoreactor (TBNR) via assembling boron nitride quantum dots (BNQDs) and Fe3+ for tumor BNCT and chemodynamic (CDT) synergistic treatment. The obtained TBNR with an appropriate size exhibits superior tumor accumulation and

Salvage boron neutron capture therapy for pediatric patients with recurrent diffuse midline glioma.

PURPOSE: Pediatric diffuse malignant glioma located in the brainstem was officially named "diffuse midline glioma" (DMG) by the World Health Organization in 2016. For this disease, radical surgery is not beneficial, and the only major treatment strategy is radiotherapy. However, the dose limitations to brainstem tissue mean that treatment by radiotherapy can only control and not eradicate the tumors, and there is no effective treatment for recurrence, resulting in short overall survival of 6-12 months. This paper reports our experience with boron neutron capture therapy (BNCT), a new treatment

Carborane bearing pullulan nanogel-boron oxide nanoparticle hybrid for boron neutron capture therapy.

Boron neutron capture therapy shows is a promising approach to cancer therapy, but the delivery of effective boron agents is challenging. To address the requirements for efficient boron delivery, we used a hybrid nanoparticle comprising a carborane = bearing pullulan nanogel and hydrophobized boron oxide nanoparticle (HBNGs) enabling the preparation of highly concentrated boron agents for efficient delivery. The HBNGs showed better anti-cancer effects on Colon26 cells than a clinically boron agent, L-BPA/fructose complex, by enhancing the accumulation and retention amount of the boron agent wi

A dose calculation algorithm for boron neutron capture therapy using convolution/superposition method.

The convolution/superposition (C/S) method originally designed for photon dose calculation was first applied for developing a treatment planning system for boron neutron capture therapy. The original concept of TEGMA (total energy generated per unit mass) was proposed to represent distinctive dose components from neutron reactions with the elements in the patient's tissue. First, neutron fluence distributions in a homogeneous brain phantom irradiated with an energy-groupwise pencil beam of 2.5 × 2.5 mm2 were calculated using the MCNP6.2 code. Then, a library of energy-groupwise TEGMA and KERMA

A case of radiation-induced osteosarcoma treated effectively by boron neutron capture therapy.

We treated a 54-year-old Japanese female with a recurrent radiation-induced osteosarcoma arising from left occipital skull, by reactor-based boron neutron capture therapy (BNCT). Her tumor grew rapidly with subcutaneous and epidural extension. She eventually could not walk because of cerebellar ataxia. The tumor was inoperable and radioresistant. BNCT showed a marked initial therapeutic effect: the subcutaneous/epidural tumor reduced without radiation damage of the scalp except hair loss and the patient could walk again only 3 weeks after BNCT. BNCT seems to be a safe and very effective modali

Impact of arc therapy in boron neutron capture therapy (BNCT) on dose uniformity and motion robustness.

Monte Carlo simulations were conducted to evaluate the dose distribution characteristics and motion robustness of fixed-angle versus arc irradiation techniques in boron neutron capture therapy (BNCT), using a head phantom. For fixed-angle irradiation, an anterior-posterior (AP) beam was oriented towards the facial plane. In contrast, the arc technique involved a continuous 180° rotation, from the left to the right, around the central axis traversing the face. The target was located in the nasal cavity region. Results demonstrated the dosimetric superiority of the arc technique, which increased

Elemental Boron Nanoparticles for Boron-Neutron Capture Therapy of BT-474 Human Breast Cancer.

Breast cancer remains one of the most common malignancies worldwide. Highly invasive HER2-positive breast cancer carries a high risk of metastasis and poses a significant therapeutic challenge. Boron neutron capture therapy (BNCT) may be an option for patients with this severe diagnosis. Here we evaluated the effectiveness of BNCT with elemental boron nanoparticles obtained by laser fragmentation and coated Silane-PEG-COOH and conjugated with the Affibody ZHER2:342 guide protein (BPs) against the HER2-positive breast cancer. The MTT assay after BNCT with BPs revealed an almost twofold reductio

Carborane-Containing Matrix Metalloprotease (MMP) Ligands as Candidates for Boron Neutron-Capture Therapy (BNCT).

Based on the previously reported potent and selective sulfone hydroxamate inhibitors SC-76276, SC-78080 (SD-2590), and SC-77964, potent MMP inhibitors have been designed and synthesized to append a boron-rich carborane cluster by employing click chemistry to target tumor cells that are known to upregulate gelatinases. Docking against MMP-2 suggests binding involving the hydroxamate zinc-binding group, key H-bonds by the sulfone moiety with the peptide backbone residues Leu82 and Leu83, and a hydrophobic interaction with the deep P1' pocket. The more potent of the two triazole regioisomers exhi

Covalent Organic Polymer as a Carborane Carrier for Imaging-Facilitated Boron Neutron Capture Therapy.

Boron neutron capture therapy (BNCT) is an atomic targeted radiotherapy that shows fantastic suppression impact on locally intrusive threatening tumors. One key factor for effective BNCT is to aggregate an adequate concentration (>20 ppm) of 10B in the cytoplasm of the tumor. Carborane-loaded polymer nanoparticles are promising because of their outstanding biocompatibility and plasma steadiness. In this study, a new class of carborane-loaded nanoscale covalent organic polymers (BCOPs) was prepared by a Schiff base condensation reaction, and their solubility was greatly improved in common solve
