Selection and validation of a set of reliable reference genes for quantitative RT-PCR studies in the brain of the Cephalopod Mollusc Octopus vulgaris.

BACKGROUND: Quantitative real-time polymerase chain reaction (RT-qPCR) is valuable for studying the molecular events underlying physiological and behavioral phenomena. Normalization of real-time PCR data is critical for a reliable mRNA quantification. Here we identify reference genes to be utilized in RT-qPCR experiments to normalize and monitor the expression of target genes in the brain of the cephalopod mollusc Octopus vulgaris, an invertebrate. Such an approach is novel for this taxon and of advantage in future experiments given the complexity of the behavioral repertoire of this species w

Recording electrical activity from the brain of behaving octopus.

Octopuses, which are among the most intelligent invertebrates,1,2,3,4 have no skeleton and eight flexible arms whose sensory and motor activities are at once autonomous and coordinated by a complex central nervous system.5,6,7,8 The octopus brain contains a very large number of neurons, organized into numerous distinct lobes, the functions of which have been proposed based largely on the results of lesioning experiments.9,10,11,12,13 In other species, linking brain activity to behavior is done by implanting electrodes and directly correlating electrical activity with observed animal behavior.

The potential impact of Octopus cyanea extracts to improve eggplant resistance against Fusarium-wilt disease: in vivo and in vitro studies.

The novelty of the present research is conducting a new method in the systemic resistance of plant diseases by using distinct marine extracts. The ability of two octopus extracts to reduce the wilt disease caused by Fusarium oxysporum was observed. The applied methods are soaked roots (SR) and foliar shoots (FS). The antioxidant enzyme activities, percent disease index (PDI), and growth parameters were measured. In vitro antifungal potential of the octopus extracts against F. oxysporum was examined. The obtained result shows that SR extracts reduced PDI. Additionally, all the tested treatments

Pseudomonas oryzihabitans cutaneous ulceration from Octopus vulgaris bite: a case report and review of the literature.

BACKGROUND: Octopus vulgaris is a common marine animal that can be found in nearly all tropical and semitropical waters around the world. It is a peaceful sea dweller with a parrotlike beak, and its primary defense is to hide through camouflaging adjustments. Bites from animals of the class Cephalopoda are very rare. We describe a boy who was bitten on his forearm by an Octopus vulgaris. OBSERVATION: A 9 -year-old boy was bitten by an Octopus vulgaris while snorkeling. There was no strong bleeding or systemic symptoms; however, 2 days later, a cherry-sized, black, ulcerous lesion developed, s

Marginolides A-B, polyether macrolide analogues from veined octopus Amphioctopus marginatus: anti-hypertensive leads attenuate angiotensin-converting enzyme.

Angiotensin-I-converting enzyme (ACE) is considered as a major drug target for the treatment of hypertension as it catalyses the production of vasoconstrictor angiotensin II from angiotensin I. ACE inhibitor agents are an effective therapeutic strategy to control high blood pressure. Unprecedented polyether macrolides, marginolide A and B were isolated from the crude extract of marine octopus, Amphioctopus marginatus via bioassay-directed sequential chromatographic fractionation. Marginolide A displayed considerably greater ACE attenuation potential (IC50 0.58 mM) than that exhibited by margin

Mercury in the tissues of five cephalopods species: First data on the nervous system.

Mercury (Hg), one of the elements most toxic to biota, accumulates within organisms throughout their lifespan and biomagnifies along trophic chain. Due to their key role in marine systems, cephalopods constitute a major vector of Hg in predators. Further, they grow rapidly and display complex behaviours, which can be altered by neurotoxic Hg. This study investigated Hg concentrations within 81 cephalopod specimens sampled in the Bay of Biscay, which belonged to five species: Eledone cirrhosa, Sepia officinalis, Loligo vulgaris, Todaropsis eblanae and Illex coindetii. Hg concentrations were mea

The effects of freshwater immersion on muscle qualities and the evaluation of water addition in Amphioctopus fangsiao (Mollusca: Cephalopoda) from the seafood market.

Amphioctopus fangsiao is widely favored by Asian consumers for its high nutritional value and delicious taste, yet fraudulent water addition practices often dissatisfy consumers. This study evaluated its nutritional value, established baseline data for the moisture (80.33 ± 5.28 g/100 g) and protein content (16.47 ± 2.95 g/100 g) of raw samples, and elucidated the effects of freshwater immersion. The muscle of A. fangsiao contained abundant taurine. However, freshwater immersion stress increased the octopus's weight, with 5-h exposure identified as a critical point for weight change. Moreover,

Construct validity of the OCTOPuS stratification algorithm for allocating patients with knee osteoarthritis into subgroups.

BACKGROUND: We recently developed a model of stratified exercise therapy, consisting of (i) a stratification algorithm allocating patients with knee osteoarthritis (OA) into one of the three subgroups ('high muscle strength subgroup' representing a post-traumatic phenotype, 'low muscle strength subgroup' representing an age-induced phenotype, and 'obesity subgroup' representing a metabolic phenotype) and (ii) subgroup-specific exercise therapy. In the present study, we aimed to test the construct validity of this algorithm. METHODS: Data from five studies (four exercise therapy trial cohorts

Multiple optic gland signaling pathways implicated in octopus maternal behaviors and death.

Post-reproductive life in the female octopus is characterized by an extreme pattern of maternal care: the mother cares for her clutch of eggs without feeding until her death. These maternal behaviors are eradicated if the optic glands, the octopus analog of the vertebrate pituitary gland, are removed from brooding females. Despite the optic gland's importance in regulating maternal behavior, the molecular features underlying optic gland function are unknown. Here, we identify major signaling systems of the Octopus bimaculoides optic gland. Through behavioral analyses and transcriptome sequenci

A new species of Microeledone from Galápagos Islands and an amended diagnosis of the Megaleledonidae (Octopoda: Incirrata).

The octopod fauna of the deep tropical Pacific Ocean remains poorly known, as is the octopod family Megaleledonidae Taki. A single female megaleledonid specimen collected at 1773 m depth near the equatorial Galápagos island of Darwin is named Microeledone galapagensis sp. nov. This small, squat, short-armed octopod has few arm suckers and gill lamellae. Its lack of a crop diverticulum, ink sac, and anal flaps suggests that it pertains to Thaumeledone. However, its smooth skin, which dorsally is nearly free of pigment, large rachidian tooth, and large funnel organ ally it with the monotypic Mic

SPATIO-TEMPORAL VARIATION OF PROCHRISTIANELLA SP. (CESTODA: TRYPANORHYNCHA) IN THE HOST OCTOPUS MAYA (CEPHALOPODA: OCTOPODA) AND ITS RELATIONSHIP WITH THE REPRODUCTION.

Although harvest of Octopus maya (Mexican four-eyed octopus) is one of the most important fisheries in the Yucatan Peninsula, little is known about the parasites of these cephalopods and how they affect host physiological processes. We analyzed the spatio-temporal variation of infection of O. maya by the cestode larva Prochristianella sp. (found in the anterior salivary glands) and its relationship with the gonad development stages (GDSs) of the host. From August 2009 to May 2010, 439 specimens of O. maya were caught in 2 localities on the northern coast of the Yucatán Peninsula: Dzilam de Bra

Relations between mercury, methyl-mercury and selenium in tissues of Octopus vulgaris from the Portuguese coast.

Mercury, methyl-mercury (MeHg) and selenium were determined in digestive gland and mantle of Octopus vulgaris, from three areas of the Portuguese coast. To our knowledge these are the first data on MeHg in cephalopods. Concentrations were higher in the digestive gland and percentage of MeHg in mantle. Enhanced Hg and MeHg levels were obtained in digestive gland of specimens from Olhão (3.1-7.4 and 2.0-5.0 microg g(-1), respectively). Differences between areas may be partially related to Hg availability. Relationships between concentrations in mantle and digestive gland pointed to proportional

Fine structure of the gonadal tissue of the horned octopod Eledone cirrhosa (Lamarck, 1798) (Mollusca - Octopoda) during sexual maturity.

The horned octopod Eledone cirrhosa, a medium-sized species found in Arabian Gulf off Saudi Arabia, was collected monthly from the Arabian Gulf off Dammam city during January to December 2022. Samples were dissected and prepared for examination using transmission electron microscopy. During genital maturation, seminiferous tubules are established in the testis, with active spermatogonia dividing. Spermatocytes 1 are observed in the tubule, followed by an increase in spermatogonia and spermatocytes in August. Spermiogenesis begins, with spherical spermatids evolving into elongated spermatids. An Octopus-Inspired Stimulus-Responsive Structural Color Hydrogel for Uterus Cervical Canal Stent.

Soft-bodied aquatic organisms have exhibited remarkable capabilities in navigating and moving within liquid environments serving as a profound inspiration for the development of bionic robots with intricate movements. Traditional rigid components are being replaced by stimulus-responsive soft materials such as hydrogels and shape memory polymers, leading to the creation of dynamically responsive soft robots. In this study, the development of a bionic robot inspired by the shape of an octopus and the adsorptive properties of its tentacles, specifically tailored for targeted stimulation and pH s

The Effect of Prohibitins on Mitochondrial Function during Octopus tankahkeei Spermiogenesis.

Mitochondria are essential for spermiogenesis. Prohibitins (PHBs; prohibitin 1, PHB1 or PHB, and prohibitin 2, PHB2) are evolutionarily conserved and ubiquitously expressed mitochondrial proteins that act as scaffolds in the inner mitochondrial membrane. In this study, we analyzed the molecular structure and dynamic expression characteristics of Ot-PHBs, observed the colocalization of Ot-PHB1 with mitochondria and polyubiquitin, and studied the effect of phb1 knockdown on mitochondrial DNA (mtDNA) content, reactive oxygen species (ROS) levels, and apoptosis-related gene expression in spermatid

Octopus vulgaris ink extracts exhibit antioxidant, antimutagenic, cytoprotective, antiproliferative, and proapoptotic effects in selected human cancer cell lines.

Cancer is a noncommunicable disease of rising worldwide concern. Marine food products such as Octopus vulgaris ink (OI) could be sources of compounds addressing these concerns. This study aimed to evaluate the antimutagenic, cytoprotective, antiproliferative, proapoptotic, and antioxidant capacity of OI extracts on human cancer cell lines (22Rv1, HeLa, A549). The ARPE-19 cell line was used as a reference human cell line to evaluate the ink's cytotoxicity. The water extract exhibited the highest antimutagenic and cytoprotective effect, but the dichloromethane extract (DM) showed the lowest half

Numerical and functional responses to the presence of a competitor--the case of Aggregata sp. (Apicomplexa: Aggregatidae) and Octopicola superba (Copepoda: Octopicolidae).

Evidence of interference competition between the eimeriorin coccidian Aggregata sp. and the octopicolid copepod Octopicola superba at the level of the gills of naturally infected Octopus vulgaris is evaluated. Numerical and functional responses are considered for analysis, and the fundamental and realized spatial niches (FSNs and RSNs) are measured as part of the study. While it was not possible to measure the FSN of Aggregata sp., the analysis of the infection levels of O. superba recorded for non-concomitantly and concomitantly infected hosts suggests that the gills and body skin constitute,

The reaction of Octopus vulgaris hemocyanin with exogenous ligands: proposal of an allosteric model for the binding of cyanide and thiourea to the 11 S subunit.

Octopus vulgaris hemocyanin in 11 S aggregation state binds oxygen following a noncooperative oxygen saturation curve with Hill coefficient n = 1. Under the same conditions the equilibrium and kinetics of the reaction with cyanide and other ligands are indicative of an anticooperative behavior displaying different characteristics for the different ligands. The data are consistent with an induced-fit type allosteric model which assumes for the 11 S subunit of O. vulgaris hemocyanin an annular structure made up by five identical domains each containing one binding site whose reactivity is near-n
