Plants exhibit striking developmental plasticity, driven by stem cell populations within meristematic tissues such as the shoot apical meristem (SAM). While the genetic networks governing stem cell homeostasis are well-characterized, recent studies reveal that the SAM also functions as an active environmental sensor to modulate growth plasticity. This Tansley review synthesizes advances in how the SAM perceives and integrates key environmental signals - including light, temperature, oxygen, and humidity - to direct adaptive growth. We detail the molecular mechanisms, such as photoreceptor-medi

Ultraviolet-B (UV-B, 280-315 nm) radiation acts as a key environmental signal that regulates plant development and acclimation responses. The UV-B photoreceptor UV RESISTANCE LOCUS 8 (UVR8) plays a central role in perceiving and transducing these signals. In the absence of UV-B, UVR8 exists predominantly as a homodimer in the cytoplasm. Upon UV-B absorption, UVR8 dissociates into active monomers that translocate to the nucleus, where they initiate downstream signaling via interactions with core regulatory components. In this review, we systematically elaborate on the UVR8-mediated UV-B signal

Extreme temperatures are among the most important stressors limiting plant growth and development. Results indicate that light substantially influences the acclimation processes to both low and high temperatures, and it may affect the level of stress injury. The interaction between light and temperature in the regulation of stress acclimation mechanisms is complex, and both light intensity and spectral composition play an important role. Higher light intensities may lead to overexcitation of the photosynthetic electron transport chain; while different wavelengths may act through different phot

The functionality of the UV-B photoreceptor UV RESISTANCE LOCUS8 (UVR8) has been widely demonstrated in angiosperms under controlled conditions, but studies in nature or using bryophytes are scarce. To fill this gap, we compared the UV responses of the model liverwort Marchantia polymorpha and a mutant lacking a functional UVR8, to provide for the first time an evolutionary perspective of UVR8 performance in both controlled and field conditions. Under exposure to UV in the laboratory, UVR8 in wild type plants was essential to induce the expression of genes (CHS1, FNSI, HY5, SIG5, and ELIP2) an

The photosynthetic capacity of mature leaves increases after several days' exposure to constant or intermittent episodes of high light (HL) and is manifested primarily as changes in chloroplast physiology. How this chloroplast-level acclimation to HL is initiated and controlled is unknown. From expanded Arabidopsis leaves, we determined HL-dependent changes in transcript abundance of 3844 genes in a 0-6 h time-series transcriptomics experiment. It was hypothesized that among such genes were those that contribute to the initiation of HL acclimation. By focusing on differentially expressed trans

In plants, the regulation of transgene transcription is typically achieved using chemical agents. A safe alternative to chemically induced systems may be optogenetic systems. The BphP1-QPAS1 system has distinct advantages over other optogenetic systems, as it is activated by near-infrared (NIR, 780 nm) light, which is beyond the spectrum of plant photoreceptors. This system is based on the use of a split transcription factor (TF), consisting of the DNA-binding and dimerization domains of the yeast TF Gal4, fused to the QPAS1 component, along with the transactivation domain VP16 fused to BphP1.

BACKGROUND: Virus-induced gene silencing (VIGS) is a rapid and powerful method for gene functional analysis in plants that pose challenges in stable transformation. Numerous VIGS systems based on Agrobacterium infiltration has been widely developed for tender tissues of various plant species, yet none is available for recalcitrant perennial woody plants with firmly lignified capsules, such as tea oil camellia. Therefore, there is an urgent need for an efficient, robust, and cost-effective VIGS system for recalcitrant tissues.
RESULTS: Herein, we initiated the Tobacco rattle virus (TRV)-elicit

The etiolation process, which occurs after germination, is terminated once light is perceived and then de-etiolation commences. During the de-etiolation period, monochromatic lights (blue, red and far-red) induce differences in gene expression profiles and plant behavior through their respective photoreceptors. ELONGATED HYPOCOTYL 5 (HY5), a bZIP-type transcription factor (TF), regulates gene expression in the de-etiolation process, and other bZIP TFs are also involved in this regulation. However, transcriptomic changes that occur in etiolated seedlings upon monochromatic light irradiation and

The function of the phytochrome B photoreceptor is enhanced by its homologous counterpart, phytochrome D, at low temperatures, thereby increasing phenotypic plasticity in response to fluctuating environmental conditions. Light is a dynamically changing environmental factor that influences nearly every aspect of plant growth and development. Photoreceptors detect light, with phytochromes (PHY) specifically monitoring red and far-red wavelengths. Arabidopsis thaliana has five phytochromes, named phyA through phyE. phyA and phyB are extensively studied key mediators of most phytochrome responses;

Maintaining cellular photosensitivity while preventing damage from excessive light energy is a crucial challenge for plants, and negative feedback inhibition of photoreceptors plays a vital role in achieving this balance. Here, we uncover a regulatory mechanism involved in the UVR8-mediated UV-B signalling pathway. Through a combination of genetic, biochemical and cross-species analyses, we identified Arabidopsis BIC1 and BIC2, along with their orthologs in eggplant (Solanum melongena L.), as functionally conserved negative regulators in UV-B signalling. Through physical association with UVR8,

Histidine kinases, used for environmental sensing by bacterial two-component systems, are involved in regulation of bacterial gene expression, chemotaxis, phototaxis, and virulence. Flavin-containing domains function as light-sensory modules in plant and algal phototropins and in fungal blue-light receptors. We have discovered that the prokaryotes Brucella melitensis, Brucella abortus, Erythrobacter litoralis, and Pseudomonas syringae contain light-activated histidine kinases that bind a flavin chromophore and undergo photochemistry indicative of cysteinyl-flavin adduct formation. Infection of

Three different families of blue-light receptors have been characterized from higher plants: three cryptochromes, two phototropins, and the three members of the ZTL/ADO family. Phototropins and the ZTL/ADO proteins have chromophore modules, designated LOV domains, that bind flavin mononucleotide and undergo formation of a C(4a) flavin-cysteinyl adduct. All contain the highly conserved amino acid motif GXNCRFLQ. Over 90 prokaryote proteins also contain LOV domains with this motif upstream from one of several different functional groups. All of these that have been investigated to date act as ph

Phototropin (PHOT) is a photoreceptor involved in a variety of blue-light-elicited physiological processes including phototropism, chloroplast movement and stomatal opening in plants. The work presented here tests whether PHOT is involved in expression of light-regulated genes in Chlamydomonas reinhardtii. When C. reinhardtii was transferred from the dark to very low-fluence rate white light, there was a substantial increase in the level of transcripts encoding glutamate-1-semialdehyde aminotransferase (GSAT), phytoene desaturase (PDS) and light-harvesting polypeptides (e.g. LHCBM6). Increased

OsiCRY2 is involved in light-regulated plant development and plays a role in regulating photomorphogenesis, plant height, flowering and most strikingly partial photomorphogenesis in dark. Cryptochrome 2 (CRY2), the blue/UV-A light photoreceptor in plants, has been reported to regulate photoperiod-dependent flowering and seedling photomorphogenesis (under low-intensity light). Among monocots, CRY2 has been reported from japonica rice, wheat, sorghum and barley. The two sub-species of rice, indica and japonica, exhibit a high degree of genetic variation and morphological and physiological differ

All organisms need to sense their surroundings and respond. This is especially true for plants, which are rooted to the ground and must contend with daily fluctuations of both above- and belowground environments. Of these, temperature is perhaps the most variable, with shifts throughout the day and microenvironments that affect one part of the plant differently from another (e.g., a leaf in the canopy vs. one in the shade). Heat stress is of particular concern and negatively affects basic cellular processes including protein folding and function, membrane fluidity, and cytoskeletal organizatio

Experiments have shown that many phenylpropanoid genes are highly expressed in light-grown Arabidopsis roots. Studies employing reporter gene constructs have indicated that the expression of these genes is localized not only to the lignifying root vasculature, but also to non-lignifying tissues, such as the root cortex, suggesting that the proteins encoded by these genes may be involved in aspects of phenylpropanoid metabolism other than lignification. Consistent with this hypothesis, roots of etiolated and soil-grown plants contain almost no soluble phenylpropanoids, but exposure to light lea

Chlorophyll is an indispensable photoreceptor in plant photosynthesis. Its anabolic imbalance is detrimental to individual growth and development. As an essential epigenetic modification, DNA methylation can induce phenotypic variations, such as leaf color transformation, by regulating gene expression. Albino line XN1376B is a natural mutation of winter wheat cultivar XN1376; however, the regulatory mechanism of its albinism is still unclear. In this study, we found that low temperatures induced albinism in XN1376B. The number of chloroplasts decreased as the phenomenon of bleaching intensifie

Peronospora belbahrii is a biotrophic oomycete attacking sweet basil. It propagates asexually by producing spores on dichotomously branched sporophores emerging from leaf stomata. Sporulation occurs when infected plants are incubated for at least 7.5h in the dark in moisture-saturated atmosphere at 10-27°C. Exposure to light suppresses spore formation but allows sporophores to emerge from stomata. Incandescent or CW fluorescent light of 3.5 or 6 µmoles.m(2).s(-1) respectively, caused 100% inhibition of spore formation on lower leaf surface even when only the upper leaf surface was exposed to l

Light triggers diurnal dynamics in stomatal apertures. To balance carbon dioxide uptake for photosynthesis against water loss through transpiration, plants employ mechanisms to prevent excessive stomatal opening. However, the dynamic regulatory mechanism governing stomatal apertures in diurnal stomatal movements remains poorly understood. Here, we report that ROP (RHO OF PLANTS) GUANINE NUCLEOTIDE EXCHANGE FACTOR 2 (RopGEF2), a negative regulator of light-induced stomatal opening, exhibits dynamic phosphorylation in Arabidopsis (Arabidopsis thaliana) guard cells along with changes in stomatal
