Field-grown maize is frequently exposed to fluctuating light (FL) combined with heat and drought during summer, yet the underlying photosynthetic regulatory mechanisms remain poorly understood. We examined chlorophyll fluorescence, the P700 redox state, and electrochromic shift signals in maize leaves subjected to FL under individual and compound stresses of moderate heat and drought. Following short-term FL treatment, photosystem I (PSI) photoinhibition was not exacerbated by any stress treatment, whereas photosystem II (PSII) photoinhibition was significantly aggravated by heat and compound

Ocean acidification (OA) exerts diverse effects on marine macroalgae, with calcified species being particularly vulnerable. Due to calcified skeletons can contribute to physical screening against solar ultraviolet radiation (UVR), OA-driven calcification loss may increase exposure of the photosynthetic apparatus to UVR. Here, we cultured Corallina officinalis under ambient CO₂ (∼420 μatm) or elevated CO₂ (∼1000 μatm), with or without UVR, under natural solar radiation. Our results confirmed that OA reduced calcification and, under UVR, enhanced donor-side impairment of photosystem II (PSII), a

Acclimation to changing light intensities poses major challenges to plant metabolism and has been shown to involve regulatory adjustments in chloroplast gene expression. However, this regulation has not been examined at a plastid genome-wide level and for many genes, it is unknown whether their expression responds to altered light intensities. Here, we applied comparative ribosome profiling and transcriptomic experiments to analyze changes in chloroplast transcript accumulation and translation in leaves of tobacco (Nicotiana tabacum) seedlings after transfer from moderate light to physiologica

Light is the primary energy source for photosynthetic organisms, but in excess, it can generate reactive oxygen species and lead to cell damage. Plants evolved multiple mechanisms to modulate light use efficiency depending on illumination intensity to thrive in a highly dynamic natural environment. One of the main mechanisms for protection from intense illumination is the dissipation of excess excitation energy as heat, a process called nonphotochemical quenching. In plants, nonphotochemical quenching induction depends on the generation of a pH gradient across thylakoid membranes and on the pr

The investigation of spatial heterogeneity within the thylakoid membrane (TM) proteins has gained increasing attention in photosynthetic research. The recent advances in live-cell imaging have allowed the identification of heterogeneous organisation of photosystems in small cyanobacterial cells. These sub-micrometre TM regions, termed microdomains in cyanobacteria, exhibit functional similarities with granal (Photosystem II dominant) and stromal (Photosystem I dominant) regions observed in TM of higher plants. This study delves into microdomain heterogeneity using super-resolution Airyscan-bas

We report on the changes the photosynthetic apparatus of Chlamydomonas reinhardtii undergoes upon acclimation to different light intensity. When grown in high light, cells had a faster growth rate and higher biomass production compared with low and control light conditions. However, cells acclimated to low light intensity are indeed able to produce more biomass per photon available as compared with high light-acclimated cells, which dissipate as heat a large part of light absorbed, thus reducing their photosynthetic efficiency. This dissipative state is strictly dependent on the accumulation o

In the next 10-20 years, several observatories will aim to detect the signatures of oxygenic photosynthesis on exoplanets, though targets must be carefully selected. Most known potentially habitable exo-planets orbit cool M-dwarf stars, which have limited emission in the photosynthetically active region of the spectrum (PAR, 400 < λ < 700 nm) used by Earth's oxygenic photoautotrophs. Still, recent experiments have shown that model cyanobacteria, algae, and non-vascular plants grow comfortably under simulated M-dwarf light, though vascular plants struggle. Here, we hypothesize that this is par

Photosynthesis constantly adapts to fluctuating light and temperature. Under high light, non-photochemical quenching (NPQ) protects the photosynthetic machinery by dissipating excess excitation energy as heat. With rising global temperatures and more frequent heat events, understanding how photosynthesis responds to combined light and temperature stress is critical. In vascular plants, NPQ is regulated by the protonation of Photosystem II subunit S (PsbS) and the enzymatic formation of zeaxanthin, but it remains unclear how temperature influences the complete NPQ response and the roles of thes

Magnesium (Mg), as an essential and central mineral element for chlorophyll biosynthesis, plays a crucial role in plant photosynthesis. Magnesium deficiency inevitably affects the photosynthetic ability of leaves, thereby impairing the yield and quality of crops. However, few studies have revealed the intrinsic mechanisms by which Mg deficiency hinders growth and photosynthesis, particularly by analyzing the processes of light capture, dissipation, absorption, and utilization in tomato. The experiment studied the effects of Mg deficiency on internal structure of leaves, light absorption, elect

Photosensitivity and photosynthetic characteristics have been analyzed in wild type (KC) and its psbAII mutant (I6) of Synechocystis having three point amino acid substitutions, i.e., N322I, I326F and F328S, which are localized in the C-terminal extension of D1 protein of the photosystem II reaction center. Wild type and mutant cells show almost an identical growth pattern under normal/low light (30 mumol m-2s-1, 30 degrees C) liquid culture (BG-11) condition. However, upon shifting the cultures to high light (500 mumol m-2s-1, 30 degrees C), these two types of cells exhibit entirely different

Photosystem II (PSII) is highly sensitive to light-induced damage. Photoinhibition, the light-dependent inactivation of PSII, is associated with an increase of excitation-energy quenching. Recovery from photoinhibition involves migration of PSII complexes from the appressed to the non-appressed region of the thylakoid membrane, where D1 (the PSII core protein most sensitive to photodamage) is degraded and repair occurs. However, it remains unclear whether damaged PSII core complexes accumulate in the stroma lamellae when repair is blocked. Here, we combined confocal Fluorescence Lifetime Imagi

Indian camphorweed (Pluchea indica (L.) Less.) is used as herbal tea due to the presence of volatile aromatic oils and several phytochemical compounds. The aim of this study was to assess the impact of copper (Cu) contamination on the physiology and morphology of P. indica, and the health risks associated with its consumption as tea. The cuttings of P. indica were subjected to 0 mM (control), 5 mM (low Cu), and 20 mM (excess Cu) of CuSO4 treatments for 1, 2, and 4 weeks. Thereafter, Cu contamination as well as physiological and morphological parameters were assessed. Cu accumulation was higher

To investigate effects of UV radiation (UVR, 280-400 nm) on coccolithophorids under nutrient-limited conditions, we grew Gephyrocapsa oceanica to determine its resilience to consecutive daily short-term exposures to +UVR (irradiances >295 nm) under a range of nitrate availabilities (100, 24, 12, 6 and 3 μM). +UVR alone significantly hampered the growth of G. oceanica, with the synergistic negative effects of +UVR and N-limitation being about 58% and 22% greater than under UVR or N-limitation alone, respectively. Most 3 μM nitrate cultures died, but those exposed to UVR succumbed sooner. This w

Marine phytoplankton, and in particular diatoms, are responsible for almost half of all primary production on Earth. Diatom species thrive from polar to tropical waters and across light environments that are highly complex to relatively benign, and so have evolved highly divergent strategies for regulating light capture and utilization. It is increasingly well established that diatoms have achieved such successful ecosystem dominance by regulating excitation energy available for generating photosynthetic energy via highly flexible light harvesting strategies. However, how different light harve

We carried out experiments using long-term (5-7 days) exposure of marine phytoplankton species to solar radiation, in order to assess the joint effects of ultraviolet radiation (UVR) and temperature on the photochemical responses and photoprotective mechanisms. In the experiments, carried out at Atlantic coast of Patagonia (43°18.7'S; 65°2.5'W) in spring-summer 2011, we used three species as model organisms: the dinoflagellate Prorocentrum micans, the chlorophyte Dunaliella salina and the haptophyte Isochrysis galbana. They were exposed under: (1) two radiation quality treatments (by using dif

The DNA-binding protein WHIRLY1, sharing structural similarities with ferritin, plays a role in the formation of iron cofactor proteins within chloroplasts. Previous studies indicated that barley plants with a knockdown of HvWHIRLY1 containing a minimal amount of the protein are compromised in chloroplast development and photosynthesis, and get chlorotic leaves when grown at high irradiance. Thereby, the leaves display signs of iron deficiency. Metal determination revealed, however, that leaves of WHIRLY1-deficient plants had a regular iron content. Nevertheless, WHIRLY1-deficiency affected th

Microplastics (MPs) and cadmium (Cd) has attracted increasing attention due to their combined toxicity to terrestrial vegetation. Photosynthesis which utilizes light energy to synthesize organic substances is crucial for crop production. However, the plant photosynthetic response to the joint toxicity of MPs and Cd is still unknown. Here, we studied the effects of polyethylene (PE) MPs on the photosynthetic performance of two maize cultivars Xianyu 335 (XY) and Zhengdan 958 (ZD) grown in a Cd contaminated soil. Results showed that the leaf Cd concentration in XY and ZD reached 26.1 and 31.9 μg

We have investigated the nature of the photocurrent generated by Photosystem II (PSII), the water oxidizing enzyme, isolated from Thermosynechococcus elongatus, when immobilized on nanostructured titanium dioxide on an indium tin oxide electrode (TiO2/ITO). We investigated the properties of the photocurrent from PSII when immobilized as a monolayer versus multilayers, in the presence and absence of an inhibitor that binds to the site of the exchangeable quinone (QB) and in the presence and absence of exogenous mobile electron carriers (mediators). The findings indicate that electron transfer o

The response of the heat-sensitive dgd1-2 and dgd1-3 Arabidopsis mutants depleted in the galactolipid DGDG to photoinhibition of chloroplasts photosystem II was studied to verify if there is a relationship between heat stress vulnerability due to depletion in DGDG and the susceptibility to photoinhibitory damage. Non-photochemical quenching (NPQ) is known to dissipate excessive absorbed light energy as heat to protect plants against photodamage. The main component of NPQ is dependent of the transthylakoid pH gradient and is modulated by zeaxanthin (Zx) synthesis. These processes together with

Responses of electron transport to three levels of irradiation (20, 200, and 1200 μmol photons m-2 s-1 PAR; exposures called LL, ML and HL, respectively) were investigated in eelgrass (Zostera marina L.) utilizing the chlorophyll a fluorescence technique. Exposure to ML and HL reduced the maximum quantum yield of photosystem II (PSII) (Fv/Fm) and the maximum slope decrease of MR/MRO (VPSI), indicating the occurrence of photoinhibition of both PSII and photosystem I (PSI). A comparatively slow recovery rate of Fv/Fm due to longer half-life recovery time of PSII and 40% lower descending amplitud
