Blue-light-activated histidine kinases: two-component sensors in bacteria.

Histidine kinases, used for environmental sensing by bacterial two-component systems, are involved in regulation of bacterial gene expression, chemotaxis, phototaxis, and virulence. Flavin-containing domains function as light-sensory modules in plant and algal phototropins and in fungal blue-light receptors. We have discovered that the prokaryotes Brucella melitensis, Brucella abortus, Erythrobacter litoralis, and Pseudomonas syringae contain light-activated histidine kinases that bind a flavin chromophore and undergo photochemistry indicative of cysteinyl-flavin adduct formation. Infection of

Light in the Darkness: Responses to Light and Diel Activity Rhythm in an Eyeless Cave Flatworm (Dendrocoelum italicum).

Troglobiont species show common traits derived from adaptation to subterranean life. Due to lack of light in cave environments most troglobiont species show eye reduction or even loss, often accompanied by a disruption of diel rhythmicity. Although cave adaptation and eye absence generally imply the loss of capability to perceive light, several cave-adapted species have retained this function, showing some degree of phototaxy or rhythmicity. Flatworms are ubiquitous in natural habitats and the response to light or diel rhythmicity of surface-dwelling species has received considerable scientifi

Light-avoidance-mediating photoreceptors tile the Drosophila larval body wall.

Photoreceptors for visual perception, phototaxis or light avoidance are typically clustered in eyes or related structures such as the Bolwig organ of Drosophila larvae. Unexpectedly, we found that the class IV dendritic arborization neurons of Drosophila melanogaster larvae respond to ultraviolet, violet and blue light, and are major mediators of light avoidance, particularly at high intensities. These class IV dendritic arborization neurons, which are present in every body segment, have dendrites tiling the larval body wall nearly completely without redundancy. Dendritic illumination activate

Seasonal Dimorphism in the Compound Eye Morphology of Scythris sinensis (Felder & Rogenhofer, 1875) (Lepidoptera: Scythrididae).

Scythris sinensis (Felder & Rogenhofer) is a diurnal moth characterized by its distinct spring and autumn forms. In our study, we analyzed the compound eye morphology of both forms utilizing scanning electron microscopy. The eyes are ellipsoidal and symmetrical, consisting of hexagonal ommatidia characterized by convoluted folds but lacking corneal nipples. We found that female moths have slightly larger compound eyes than males. Our counts of ommatidia indicate that males emerging in autumn (589-675) typically possess a higher number than males emerging in spring (492-698). This finding sugge

Molecular characterization and elucidation of the function of Hap38 MAPK in the response of Helicoverpa armigera (Hübner) to UV-A stress.

The cotton bollworm Helicoverpa armigera (Hübner) (Lepidoptera: Noctuidae), an important pest of cotton, is detrimental to cotton production. Light from UV-A ultraviolet lamps is regarded as a form of environmental stress for insects. In order to investigate the response of H. armigera exposed to UV-A, we explored Hap38 MAPK expression and functions. We hope that the findings of this study will lay the foundation for future investigations into the insect's phototaxis mechanism. A p38 MAPK was cloned and named Hap38 MAPK. A phylogenetic tree showed that Hap38 MAPK was highly conserved. The gene

Light-Directed Self-Powered Metal-Organic Framework Based Nanorobots for Deep Tumor Penetration.

Effective intratumoral distribution of anticancer agents with good tumor penetration is of great practical importance for oncotherapy. How to break the limitation of traditional passive drug delivery relying on blood circulatory system into solid tumors remains a challenge. Herein, a light-directed self-powered nanorobot based on zirconium-based porphyrin metal-organic framework (MOF) is reported for smart delivery of chemodrug and photosensitizer for deep tumor penetration. The MOF-based carrier transporting the therapeutics is able to responsively generate hydrogen sulfide as propulsion gas

The protection of invertebrates, fish, and vascular plants against inorganic mercury poisoning by sulfur and selenium derivatives.

Protection of organisms against mercury (Hg) poisoning is most commonly associated with the antagonistic effects of selenium (Se)-compounds against mercury alkyls in higher animals. This study shows that there is no consistent difference among Periodic Group VIA derivatives including S(IV), S(II) organic, Se (IV), Se (II) organic, possibly Se VI, as well and Te (IV) in their ability to protect against mercury poisoning. The organisms used in assays were: Coleus explants (leaf abscission); turnip (germination); pea (growth inhibition and Hg uptake); a planarian (regeneration); the brineshrimp (

Time-resolved detection of transient movement of helix F in spin-labelled pharaonis sensory rhodopsin II.

Sensory rhodopsin II (also called phoborhodopsin) from the archaeal Natronobacterium pharaonis (pSRII) functions as a repellent phototaxis receptor. The excitation of the receptor by light triggers the activation of a transducer molecule (pHtrII) which has close resemblance to the cytoplasmic domain of bacterial chemotaxis receptors. In order to elucidate the first step of the signal transduction chain, the accessibility as well as static and transient mobility of cytoplasmic residues in helices F and G were analysed by electron paramagnetic resonance spectroscopy. The results indicate an outw

Interactions and Cytotoxicity of Human Neurodegeneration- Associated Proteins Tau and α-Synuclein in the Simple Model Dictyostelium discoideum.

The abnormal accumulation of the tau protein into aggregates is a hallmark in neurodegenerative diseases collectively known as tauopathies. In normal conditions, tau binds off and on microtubules aiding in their assembly and stability dependent on the phosphorylation state of the protein. In disease-affected neurons, hyperphosphorylation leads to the accumulation of the tau protein into aggregates, mainly neurofibrillary tangles (NFT) which have been seen to colocalise with other protein aggregates in neurodegeneration. One such protein is α-synuclein, the main constituent of Lewy bodies (LB),

Cystathionine β-synthase Deficiency Impairs Vision in the Fruit Fly, Drosophila melanogaster.

PURPOSE: Deficiency in Cystathionine β-synthase (CBS) leads to an abnormal accumulation of homocysteine and results in classical homocystinuria, a multi-systemic disorder that affects connective tissue, muscles, the central nervous system, and the eyes. However, the genetic players and mechanisms underlying vision alterations in patients with homocystinuria are little understood. MATERIALS AND METHODS: The fruit fly, Drosophila melanogaster, is a useful system to investigate the genetic basis of several human diseases, but no study to date has used Drosophila as model of homocystinuria. Here,

PAS-Domain Protein Orientation at a Polyelectrolyte Surface Revealed by Infrared Nanospectroscopy, Chiral Vibrational Spectroscopy, and Molecular Dynamics Simulations.

PAS domains mediate protein-protein interactions that enable functions such as sensing, signaling, dimerization, and localization. The photoactive yellow protein (PYP) from Halorhodospira halophila is a model PAS-domain protein involved in negative phototaxis, yet its signaling partner remains unidentified. Here, we present a method to resolve protein orientations in PAS-domain signaling by combining nano-FTIR and chiral vibrational sum-frequency generation (VSFG) spectroscopy with molecular dynamics simulations and VSFG spectral calculations. As a demonstration, we used a charged homopolypept

Neurodevelopmental toxicity of black carbon in zebrafish: behavioral and metabolomic insights.

Black carbon (BC), a combustion-derived particulate pollutant, is increasingly detected in aquatic ecosystems. While its respiratory and cardiovascular effects are well documented, its neurotoxicity in aquatic organisms remains poorly understood. In this study, zebrafish (Danio rerio) embryos were exposed to environmentally relevant BC concentrations (0, 0.05, 0.5, and 5 mg/L) for 168 h. BC induced dose-dependent developmental toxicity, manifested as reduced body length and eye size, elevated heart rate, and morphological abnormalities. Behavioral assays revealed deficits in locomotion and pho

Applications of microalga-powered microrobots in targeted drug delivery.

Over the past decade, researchers have proposed a new class of drug delivery systems, bio-hybrid micro-robots, designed with a variety of living cell-driven micro-robots that utilize the unique mobility of natural organisms (bacteria, cells, exosomes, etc.) to transport effective drugs. Microalgae are considered potential drug delivery carriers. Recent studies have shown that microalga-based drug delivery systems exhibit excellent biocompatibility. In addition, microalgae have a large surfactant area, phototaxis, oxygen production, and other characteristics, so they are used as a carrier for t

Self-generated oxygen gradients control collective aggregation of photosynthetic microbes.

For billions of years, photosynthetic microbes have evolved under the variable exposure to sunlight in diverse ecosystems and microhabitats all over our planet. Their abilities to dynamically respond to alterations of the luminous intensity, including phototaxis, surface association and diurnal cell cycles, are pivotal for their survival. If these strategies fail in the absence of light, the microbes can still sustain essential metabolic functionalities and motility by switching their energy production from photosynthesis to oxygen respiration. For suspensions of motile C. reinhardtii cells ab

Attraction of sand flies (Diptera: Psychodidae) to light traps in rural areas of Minas Gerais State, Brazil.

The objective of this study was to detect phlebotomine sand fly vector species by using light traps, and thus determine the risk of cutaneous leishmaniasis (CL) transmission in rural areas of the Viçosa microregion, Minas Gerais State, Brazil. Sand flies were captured with Shannon-like and Falcão light traps hung between 1830 and 2230 h on 22 nights. Four sampling points where CL transmission occurs naturally were chosen. A total of 888 sand flies was collected. The most abundant species was Lutzomyia intermedia. Shannon-like traps caught significantly more L. intermedia, perhaps because of a

Photoactive Tweezers.

The manipulation of micro/nanoscale objects plays a critical role across a wide range of scientific and engineering disciplines. Photoactive colloids, capable of converting low-intensity light into autonomous motion, present a highly programmable platform for micromanipulation. In this work, we introduce a "photoactive tweezers" system based on visible-light-driven, dye-sensitized TiO2 particles. These particles exhibit positive phototaxis and are autonomously confined within structured optical fields via a self-electrophoretic mechanism. Combining experiments and simulations, we reveal that a

Light-driven MOF-based micromotors with self-floating characteristics for water sterilization.

Three-dimensional motion (especially in the Z-axis direction) of metal-organic frameworks (MOFs)-based micromotors (MOFtors) is essential but still in its infancy. Herein, we propose a simple strategy for designing light-driven MOFtors that move in the Z-axis direction and efficiently kill Staphylococcus aureus (S. aureus). The as-prepared polypyrrole nanoparticles (PPy NPs) with excellent photothermal properties are combined with ZIF-8 through a simple in situ encapsulation method, resulting in multi-wavelength photothermally-responsive MOFtors (PPy/ZIF-8). Parasite-induced suppression of aggregation under predation risk in a freshwater amphipod: sociality of infected amphipods.

Recent findings suggest that grouping with conspecifics is part of the behavioural defences developed by amphipod crustaceans to face predation risk by fish. Amphipods commonly serve as intermediate hosts for trophically transmitted parasites. These parasites are known for their ability to alter intermediate host phenotype in a way that promotes predation by definitive hosts, where they reproduce. If aggregation in amphipods dilutes the risk to be preyed on by fish, then it may dilute the probability of transmission for the parasite using fish as definitive hosts. Using experimental infections

The effect of respiration on the phototactic behavior of the purple nonsulfur bacterium Rhodospirillum centenum.

The effect of respiration on the positive phototactic movement of swarming agar colonies of the facultative phototroph Rhodospirillum centenum was studied. When the electron flow was blocked at the bc1 complex level by myxothiazol, the oriented movement of the colonies was totally blocked. Conversely, inhibition of respiration via the cytochrome c oxidase stimulated the phototactic response. No phototaxis was observed in a photosynthesis deficient mutant (YB707) lacking bacteriochlorophylls. Analyses of the respiratory activities as monitored by a oxygen microelectrode in single agar colonies
